A matched-pair comparison between bilateral intrafascial and interfascial nerve-sparing techniques in extraperitoneal laparoscopic radical prostatectomy

The aim of this study was to validate the advantages of the intrafascial nerve-sparing technique compared with the interfascial nerve-sparing technique in extraperitoneal laparoscopic radical prostatectomy. From March 2010 to August 2011, 65 patients with localized prostate cancer （PCa） underwent bilateral intrafascial nerve-sparing extraperitoneal laparoscopic radical prostatectomy. These patients were matched in a 1.2 ratio to 130 patients with localized PCa who had undergone bilateral interfascial nerve-sparing extraperitoneal laparoscopic radical prostatectomy between January 2008 and August 2011. Operative data and oncological and functional results of both groups were compared. There was no difference in operative data, pathological stages and overall rates of positive surgical margins between the groups. There were 9 and 13 patients lost to follow-up in the intrafascial group and interfascial group, respectively. The intrafascial technique provided earlier recovery of continence at both 3 and 6 months than the interfascial technique. Equal results in terms of continence were found in both groups at 12 months. Better rates of potency at 6 months and 12 months were found in younger patients （age 65 years） and overall patients who had undergone the intrafascial nerve-sparing extraperitoneal laparoscopic radical prostatectomy. Biochemical progression-free survival rates I year postoperatively were similar in both groups. Using strict indications, compared with the interfascial nerve-sparing technique, the intrafascial technique provided similar operative outcomes and short-term oncological results, quicker recovery of continence and better potency. The intrafascial nerve-sparing technique is recommended as a preferred approach for young PCa patients who are clinical stages cT1 to cT2a and have normal preoperative potency.

Sodium current kinetics of transitional myocytes in Koch triangle of rabbit hearts

Background Few studies have explored the inward sodium current （INa） kinetics of transitional cardiomyocytes. This study aimed to explore the kinetics of transitional cardiomyocytes types α and β. Methods The whole-cell patch clamp technique was used to study the rapid INa of isolated transitional cardiomyocytes in the Koch triangle of rabbit hearts. Results Maximal amplitude and density of INa in type a and type β was （-1627±288） pA （α）, （-35.17±6.56） pA/pF （β） and （-3845±467） pA （α）, （-65.64±10.23） pA/pF （β）（P 〈0.05）. Steady state activation curves of INa, fitted to a Boltzmann distribution for both types, were sigmoid in shape. Half activation voltage and slope factors did not significantly differ between types at （-43.46±0.85） mV （α）, （-41.39±0.47） mV （β） or （9.04±0.66） mV （α）, （11.08±0.89） mV （β）. Steady state inactivation curves of INa, fitted to a Boltzmann distribution in both types were inverse ＂S＂ shape. Half inactivation voltage and slope factors were （-109.9±0.62） mV （α）, （-107.5±0.49） mV （β） and （11.78±0.36） mV （α）, （11.57±0.27） mV （β）, （P〉0.05）, but time constants of inactivation were significantly different at （1.10±0.19） mV （α） and （2.37±0.33） ms （β）, （P 〈0.05）. Time constants of recovery from inactivation of INa for both types were （122.16±27.43） mV （α） and （103.84±2.8.97） ms （β）（P 〈0.05）. Conclusions Transitional cardiomyocytes in rabbit hearts show a heterogeneous, voltage gated and time dependent fast inward sodium current. Types α and β show the features of INa similar to those in slow- and fast-response myocytes, with probably better automaticity and conductivity, respectively.

A large ground-based optical/infrared telescope is being planned for a world-class astronomical site in *** cloud-free night percentage is the primary meteorological consideration for evaluating candidate *** data from GMS and NOAA satellites and the MODIS instrument were utilized in this research,covering the period from 1996 to *** data analysis benefits from overlapping results from different independent teams as well as a uniform analysis of selected sites using GMS+NOAA *** significant ground-based monitoring is needed to validate these findings,we identify three different geographical regions with a high percentage of cloud-free conditions(~83%on average),which is slightly lower than at Mauna Kea and Cerro Armazones(~85%on average)and were chosen for the large international projects TMT and ELT *** study finds evidence that cloud distributions and the seasonal changes affected by the prevailing westerly winds and summer monsoons reduce the cloud cover in areas influenced by the *** is consistent with the expectations from climate change models and is suggestive that most of the identified sites will have reduced cloud cover in the future.

In vertebrates,the type 1 parathyroid hormone receptor(PTH1R)is a critical regulator of skeletal development and homeostasis;however,how it is modulated is incompletely *** we report that deleting Kindlin-2 in osteoblastic cells using the mouse 10-kb Dmp1-Cre largely neutralizes the intermittent PTH-stimulated increasing of bone volume fraction and bone mineral density by impairing both osteoblast and osteoclast formation in murine adult ***-cell profiling reveals that Kindlin-2 loss increases the proportion of osteoblasts,but not mesenchymal stem cells,chondrocytes and fibroblasts,in non-hematopoietic bone marrow cells,with concomitant depletion of osteoblasts on the bone surfaces,especially those stimulated by ***,haploinsufficiency of Kindlin-2 and Pth1r genes,but not that of either gene,in mice significantly decreases basal and,to a larger extent,PTH-stimulated bone mass,supporting the notion that both factors function in the same genetic ***,Kindlin-2 interacts with the C-terminal cytoplasmic domain of PTH1R via aa 474–475 and Gsα.Kindlin-2 loss suppresses PTH induction of cAMP production and CREB phosphorylation in cultured osteoblasts and in ***,PTH promotes Kindlin-2 expression in vitro and in vivo,thus creating a positive feedback regulatory ***,estrogen deficiency induced by ovariectomy drastically decreases expression of Kindlin-2 protein in osteocytes embedded in the bone matrix and Kindlin-2 loss essentially abolishes the PTH anabolic activity in bone in ovariectomized ***,we demonstrate that Kindlin-2 functions as an intrinsic component of the PTH1R signaling pathway in osteoblastic cells to regulate bone mass accrual and homeostasis.

A comprehensive understanding of excited-state dynamics of semiconductor quantum dots or nanomaterials at the atomic or molecular level is of scientific *** inorganic(or non-covalently protected)seimiconductor molecular nanoclusters with atomically precise structure are contributive to establish accurate correlation of excited-state dynamics with their composition/structure,however,the related studies are almost blank because of unresolved solvent dispersion ***,we designedly created the largest discrete chalcogenide seimiconductor molecular nanoclusters(denoted P2-CuMSnS,M=In or/and Ga)with great dispersibility,and revealed an interesting intracluster“core–shell”charge transfer relaxation dynamics.A systematic red shift in absorption spectra with the gradual substitution of In by Ga was experimentally and computationally investigated,and femtosecond transient absorption measurements further manifested there were three ultrafast processes in excited-state dynamics of P2 nanoclusters with the corresponding amplitudes directed by composition *** results hold the great promise of the solution-processible applications of semiconductor-NC-based quantum dots and facilitate the development of atomically precise nano-chemistry.
