Identification of blaIMI-mediated carbapenem-resistant Enterobacter from a duck farm in China

Carbapenem-and colistin-resistant Enterobacter has been a clinical and therapy problem in recent years.Here,we report the carbapenem-and colistin-resistant Enterobacter harboring bla_(IMI) isolated from intestinal samples and the environment of a duck farm in China.Four bla_(IMI)-positive Enterobacter isolates were resistant to carbapenem and colistin.Three bla_(IMI) subtypes were detected in different molecular categories of Enterobacter.The detection of the various IMI producers highlights the diversity of carbapenemases in a duck farm.Whole-genome sequencing demonstrated the bla_(IMI) genes were present in chromosomes or plasmids in these strains.The conjugation experiment demonstrated the ability of bla_(IMI)-carrying plasmid to transmit horizontally.The molecular evolution characteristics were examined through comparative genetic analysis.The study demonstrated the presence of chromosomal and plasmid bla_(IMI) and the bla_(IMI)-carrying plasmid exhibits a horizontal transmission between Enterobacter and Escherichia coli C600.The similar genetic content was discovered between two bla_(IMI-16)-positive Enterobacter asburiae.In addition,a bla_(IMI-16)-carrying plasmid is an IncFII(Yp)plasmid,and a substantial amount of mobile genetic elements were identified around bla_(IMI-16).The IS-like elements and IncFII(Yp)plasmid are significant in the propagation of bla_(IMI).Our study provides evidence for the transmission of diverse bla_(IMI) genes in China and supplies additional reference data for bla_(IMI)-positive antimicrobialresistant Enterobacter.Routine surveys of bla_(IMI)-positive Enterobacter from animal-raising environments must be given more focus.

With good mobility and flexibility,mobile manipulators have shown broad applications in construction scenarios.Base position(BP)planning,which refers to the robot autonomously determining its working station in the environment,is an important technique for mobile manipulators when performing the construction assembly task,especially in a large-scale construction environment.However,the BP planning process is tedious and time-consuming for a human worker to carry out.Thus,to improve the efficiency of construction assembly tasks,a novel BP planning method is proposed in this paper,which can lead to appropriate BPs and minimize the number of BPs at the same time.Firstly,the feasible BP regions are generated based on the grid division and the variable workspace of the mobile manipulator.Then,the positioning uncertainties of the mobile manipulator are considered in calculating the preferred BP areas using clustering.Lastly,a set coverage optimization model is established to obtain the minimum number of BPs using an optimization algorithm according to the greedy principle.The simulated experiment based on a 9-degree of free(DoF)mobile manipulator has been performed.The results illustrated that the time for BP planning was significantly reduced and the number of BPs was reduced by 63.41%compared to existing manual planning,which demonstrated the effectiveness of the proposed method.

DEAR EDITOR,Extracellular vesicles(EVs)are important for the transport of biologically active materials and for intercellular communication.As an exposed mucosa,amphibian skin participates in many essential physiological processes.To date,however,little is known about EVs in amphibian skin.Here,we successfully isolated EVs from the skin secretions of Bombina maxima,and characterized the EVs using nanoparticle tracking,western blotting,and electron microscopy.

Behavior of apatite in granitic melts derived from partial melting of muscovite in metasedimentary sources

Fluid-absent and fluid-fluxed melting of muscovite in metasedimentary sources are two types of crustal anatexis to produce the Himalaya Cenozoic leucogranites.Apatite grains separated from melts derived from the two types of parting melting have different geochemical compositions.The leucogranites derived from fluid-fluxed melting have relict apatite grains and magmatic crystallized apatite grains,by contrast,there are only crystallized apatite grains in the leucogranites derived from fluid-absent melting.Moreover,apatite grains crystallized from fluid-fluxed melting of muscovite contain higher Sr,but lower Th and LREE than those from fluid-absent melting of muscovite,which could be controlled by the distribution of partitioning coefficient(D_(Ap/Melt))between apatite and leucogranite.D_(Ap/Melt) in granites derived from fluidabsent melting is higher than those from fluid-fluxed melting.So,not only SiO_(2) and A/CNK,but also types of crustal anatexis are sensitive to trace element partition coefficients for apatite.In addition,due to being not susceptible to alteration,apatite has a high potential to yield information about petrogenetic processes that are invisible at the whole-rock scale and thus is a useful tool as a petrogenetic indicator.

The role of wildlife (wild birds)in the global transmission of antimicrobial resistance genes

Antimicrobial resistance is an urgent global health challenge in human and veterinary medicine. Wild animals are not directly exposed to clinically relevant antibiotics; however, antibacterial resistance in wild animals has been increasingly reported worldwide in parallel to the situation in human and veterinary medicine. This underlies the complexity of bacterial resistance in wild animals and the possible interspecies transmission between humans, domestic animals, the environment, and wildlife. This review summarizes the current data on expanded- spectrum 13-1actamase （ESBL）, AmpC IB-lactamase, carbapenemase, and colistin resistance genes in Enterobacteriaceae isolates of wildlife origin. The aim of this review is to better understand the important role of wild animals as reservoirs and vectors in the global dissemination of crucial clinical antibacterial resistance. In this regard, continued surveillance is urgently needed worldwide.

Insights into the effects of pulsed antimicrobials on the chicken resistome and microbiota from fecal metagenomes

Antimicrobial resistance has become a global problem that poses great threats to human health. Antimicrobials are widely used in broiler chicken production and consequently affect their gut microbiota and resistome. To better understand how continuous antimicrobial use in farm animals alters their microbial ecology, we used a metagenomic approach to investigate the effects of pulsed antimicrobial administration on the bacterial community, antibiotic resistance genes(ARGs) and ARG bacterial hosts in the feces of broiler chickens. Chickens received three 5-day courses of individual or combined antimicrobials, including amoxicillin, chlortetracycline and florfenicol. The florfenicol administration significantly increased the abundance of mcr-1 gene accompanied by floR gene, while amoxicillin significantly increased the abundance of genes encoding the AcrAB-tolC multidrug efflux pump(marA, soxS, sdiA, rob, evgS and phoP).These three antimicrobials all led to an increase in Proteobacteria. The increase in ARG host, Escherichia, was mainly attributed to the β-lactam, chloramphenicol and tetracycline resistance genes harbored by Escherichia under the pulsed antimicrobial treatments. These results indicated that pulsed antimicrobial administration with amoxicillin,chlortetracycline, florfenicol or their combinations significantly increased the abundance of Proteobacteria and enhanced the abundance of particular ARGs. The ARG types were occupied by the multidrug resistance genes and had significant correlations with the total ARGs in the antimicrobial-treated groups. The results of this study provide comprehensive insight into pulsed antimicrobial-mediated alteration of chicken fecal microbiota and resistome.

Monitoring the prevalence of antimicrobial resistance genes(ARGs)is vital for addressing the global crisis of antibiotic-resistant bacterial infections.Despite its importance,the characterization of ARGs and microbiome structures,as well as the identification of indicators for routine ARG monitoring in pig farms,are still lacking,particularly concerning variations in antimicrobial exposure in different countries or regions.Here,metagenomics and random forest machine learning were used to elucidate the ARG profiles,microbiome structures,and ARG contamination indicators in pig manure under different antimicrobial pressures between China and Europe.Results showed that Chinese pigs exposed to high-level antimicrobials exhibited higher total and plasmid-mediated ARG abundances compared to those in European pigs(P<0.05).ANT(6)-Ib,APH(3')-IIIa,and tet(40)were identified as shared core ARGs between the two pig populations.Furthermore,the core ARGs identified in pig populations were correlated with those found in human populations within the same geographical regions.Lactobacillus and Prevotella were identified as the dominant genera in the core microbiomes of Chinese and European pigs,respectively.Forty ARG markers and 43 biomarkers were able to differentiate between the Chinese and European pig manure samples with accuracies of 100%and 98.7%,respectively.Indicators for assessing ARG contamination in Chinese and European pigs also achieved high accuracy(r=0.72-0.88).Escherichia flexneri in both Chinese and European pig populations carried between 21 and 37 ARGs.The results of this study emphasize the importance of global collaboration in reducing antimicrobial resistance risk and provide validated indicators for evaluating the risk of ARG contamination in pig farms.

Pharmacokinetics of Cyadox and Its Major Metabolites in Swine After Intravenous and Oral Administration

Pharmacokinetics of cyadox （CYX） and its major metabolites in healthy swine was investigated in this paper. 1,4- Bisdesoxycyadox （BDCYX）, cyadox-1-monoxide （CYX-1-O） and quinoxaline-2-carboxylic acid （QCA）, three main metabolites of cyadox, were synthesized by College of Science, China Agricultural University. Cyadox （CYX） was administered to 8 healthy cross-bread swine intravenously （i.v.） and orally （p.o.） at a dosage of 1 mg kg^-1 body weight and 40 mg kg-1 body weight respectively in a randomized crossover design test with 2-wk washout period. A sensitive high-performance liquid chromatography-tandem mass spectrometry （LC-ESI-MS/MS） method was developed for the determination of cyadox and its major metabolites in plasma. CYX and its major metabolites BDCYX, and CYX-1-O can be detected after intravenous administration of cyadox while CYX and its metabolites BDCYX, CYX-1-O and QCA can be detected after oral administration of CYX. Plasma concentration vs. time profiles of CYX and its major metabolites were analyzed by non-compartmental pharmacokinetic method. Following i.v. administration, the areas under the plasma concentration-time curve （AUC0-∞） were （0.38±0.03） lag mL-I h （CYX）, （0.018±0.002） lag mL-1 h （BDCYX） and （0.17±0.02） lag mL^-1 h （CYX- 1-O）, respectively. The terminal elimination half-lives （t1/2kz） were determined to be （0.93±0.07） h （CYX）, （1.45±0.04） h （BDCYX）, and （0.92±0.04） h （CYX-1-O）, respectively. Steady-state distribution volume （Vss） of （2.14±0.11） L kg^-1 and total body clearance （CL） of （2.84±0.19） L h^-1 kg^-1 were determined for CYX after i.v. dosing. The bioavailability （F） of CYX was 2.85% for oral administration. After single i.v. administration, peak plasma concentrations （Cmax） of （1.08±0.06） lag mL^-1 （CYX）, （0.0068~ 0.0004） lag mL-1 （BDCYX） and （0.25~0.03） gg mL-~ （CYX-I-O） were observed at Tmax of 0.033 h （CYX）, 1 h （BDCYX） and 0.033 h （CYX-1-O）, respectively. DEAR EDITOR,Pore-forming proteins(PFPs)are widely distributed among all kingdoms of life and can oligomerize to form pores/channels in membrane systems.Extracellular vesicles(EVs)circulate in all biological fluids and can trigger biological responses at a distance,thus emerging as an additional means of intercellular communication through the release of cellular cargo,such as lipids,nucleic acids,metabolites,and proteins.To date,however,the mechanism by which EV contents are released into extracellular space remains unclear.In our previous study on toads(Bombina maxima),we identified a PFP and trefoil factor complexβγ-CAT(βγ-crystallin fused aerolysin-like protein(α-subunit)and trefoil factor(β-subunit)complex,hence namedβγ-CAT),which assembled under strict regulation in response to environmental cues.Here,we observed that the PFPβγ-CAT colocalized with EVs in the skin of B.maxima in vivo.Using small EVs(sEVs)isolated from B.maxima dermal fibroblast cells and murine fibroblast cells,we found thatβγ-CAT could specifically target and oligomerize on purified EVs,rather than disrupt membrane integrity,and promote the release of different metabolites.Our analysis revealed that a secretory PFP drove metabolite release from EVs through channel formation,providing novel clues for the delivery of EV content into extracellular space.
