Olfactory ensheathing cells promote nerve regeneration and functional recovery after facial nerve defects

Olfactory ensheathing cells from the olfactory bulb and olfactory mucosa have been tbund to increase axonal sprouting and pathfinding and promote the recovery of vibrissae motor performance in facial nerve transection injured rats. However, it is not yet clear whether olfactory ensheathing cells promote the reparation of facial nerve defects in rats. In this study, a collagen sponge and silicone tube neural conduit was implanted into the 6-mm defect of the buccal branch of the facial nerve in adult rats. Olfactory ensheathing cells isolated from the olfactory bulb of newborn Sprague-Dawley rats were injected into the neural conduits connecting the ends of tile broken nerves, the morphology and function of the regenerated nerves were compared between the rats implanted with olfactory ensheathing cells with the rats injected with saline. Facial paralysis was assessed. Nerve electrography was used to measure facial nerve-induced action potentials. Visual inspection, anatomical microscopy and hematoxylin-eosin staining were used to assess the histomorphology around the trans planted neural conduit and the morphology of the regenerated nerve. Using fluorogold retrograde tracing, toluidine blue staining and lead uranyl acetate staining, we also measured the number of neurons in the anterior exterior lateral f：acial nerve motor nucleus, the number of myelinated nerve fibers, and nerve fiber diameter and myelin sheath thickness, respectively. After surgery, olfactory ensheathing cells de- creased facial paralysis and the latency of the facial nerve-induced action potentials. There were no differences in the general morphology of the regenerating nerves between the rats implanted with olfactory ensheathing cells and the rats injected with saline. In this work a novel strategy has been developed to prepare well-dispersed amine-functionalized SiO2 nanodot-coated layered double hydroxide nano- composite （NH2-SiO2@LDH） via electrostatic interactions and condensation of （3-aminopropyl）triethoxysilane （APTES）. This nanocomposite system is well dispersed in culture media and phosphate buffered saline, and exhibits low cytotoxicity and good biocompatibility. The fluorescence microscopy images and flow cytometry data indicate that such an NH2-SiO2@LDH nanocomposite is able to efficiently deliver small interfering RNA （siRNA） into the U2OS cell line to inhibit cell proliferation. Thus, NH2-SiOR@LDH nanocomposite has a great potential as a nanocarrier for efficient gene delivery.

This paper proposes using a Nonlinear Energy Sink(NES) to suppress the nonlinear aeroelastic response of laminated composite panels in supersonic airflows. Relevant aeroelastic equations are established using Hamilton’s principle and a finite element approach, drawing upon Von Karman’s large deflection theory and first order piston theory. The idea of the NES suppression region is proposed and the effects of NES parameters on the NES suppression region are studied in detail. The results show that the nonlinear aeroelastic responses of the panel can be completely suppressed by the Transient Resonance Capture(TRC);the appropriate NES parameter values can increase the critical dynamic pressure for flutter and suppress the nonlinear aeroelastic response effectively. Increasing the mass ratio of the NES can improve the NES suppression region;the nonlinear stiffness coefficient and damping of the NES within a specific range can suppress the nonlinear aeroelastic response. The most effective installation position for a NES is in a specific region behind the center-line of the panel in the direction of the airflow.

A new type of simple emissive organic cage consisting of diphenyl sulfone units and nitrogen atoms was developed via one-pot C–N coupling condensation or two-step reactions along with a series of sulfoxide cages obtained, which exhibited excellent chemical modifiability and structural stability. By precisely controlling the participation of donor and acceptor units, a highly soluble organic cage with delayed fluorescence was obtained for the first time. More interestingly, due to the restriction of intramolecular flipping by locking the building blocks within a rigid framework, this emissive organic cage further showed narrowband ultra-deep blue electroluminescence emission at 413 nm with a full-width at half-maximum of 35 nm. This finding not only expands the family of electroluminescent organic cages, but also opens up a new platform for delayed fluorescence materials with high solubility and color purity.

Background: Interleukin-37 b(IL-37 b), a vital negative regulator of the innate immune system, has been reported to be a tumor inhibitor in different type of cancers. However, little is known about the relationship between IL-37 b and hepatocellular carcinoma(HCC). The present study aimed to investigate the potential roles of IL-37 b in HCC progression. Methods: Subjects( n = 237) were recruited, and serum IL-37 b was measured using ELISA. The tumorsuppressive capacity and underlying mechanisms of IL-37 b in HCC were investigated in vitro and in vivo. Results: Compared to healthy controls, serum IL-37 b levels were elevated in chronic hepatitis B(CHB) patients but decreased significantly in HBV-HCC patients, especially for those with portal venous tumor thrombus. Low level serum IL-37 b in HBV-HCC patients correlated with high HCC stage and poor overall survival and disease-free survival. In vitro and in vivo, recombinant human IL-37 b inhibited proliferation and metastasis in HCC cells. Furthermore, IL-37 b inhibited epithelial mesenchymal transition in HCC cells in vitro by downregulating IL-6, pSTAT3(Y705), N-cadherin, and vimentin expression and by upregulating E-cadherin expression. These effects were partially reversed by transfection of adenovirus encoding human IL-6. Conclusions: IL-37 b inhibits HCC growth, metastasis and epithelial mesenchymal transition by regulating IL-6/STAT3 signaling. Serum IL-37 b may be a biomarker for HBV-HCC and its staging.
