Semireplication-competent vesicular stomatitis virus as a novel platform for oncolytic virotherapy

Among oncolytic viruses, the vesicular stomatitis virus(VSV) is especially potent and a highly promising agent for the treatment of cancer. But, even though effective against multiple tumor entities in preclinical animal models, replicationcompetent VSV exhibits inherent neurovirulence, which has so far hindered clinical development. To overcome this limitation,replication-defective VSV vectors for cancer gene therapy have been tested and proven to be safe. However, gene delivery was inefficient and only minor antitumor efficacy was observed. Here, we present semireplicationcompetent vector systems for VSV(sr VSV), composed of two trans-complementing, propagation-deficient VSV vectors. The de novo generated deletion mutants of the two VSV polymerase proteins P(phosphoprotein) and L(large catalytic subunit), VSVΔP and VSVΔL respectively, were used mutually or in combination with VSVΔG vectors. These sr VSV systems copropagated in vitro and in vivo without recombinatory reversion to replication-competent virus. The sr VSV systems were highly lytic for human glioblastoma cell lines,spheroids, and subcutaneous xenografts. Especially the combination of VSVΔG/VSVΔL vectors was as potent as wild-type VSV(VSV-WT) in vitro and induced long-term tumor regression in vivo without any associated adverse effects. In contrast, 90% of VSV-WT-treated animals succumbed to neurological disease shortly after tumor clearance. Most importantly, even when injected into the brain, VSVΔG/VSVΔL did not show any neurotoxicity. In conclusion, sr VSVis a promising platform for virotherapeutic approaches and also for VSV-based vector vaccines, combining improved safety with an increased coding capacity for therapeutic transgenes, potentially allowing for multipronged approaches.

We introduce a novel method to create mid-infrared(MIR)thermal emitters using fully epitaxial,metal-free structures.Through the strategic use of epsilon-near-zero(ENZ)thin films in InAs layers,we achieve a narrow-band,wide-angle,and p-polarized thermal emission spectra.This approach,employing molecular beam epitaxy,circumvents the complexities associated with current layered structures and yields temperature-resistant emission wavelengths.Our findings contribute a promising route towards simpler,more efficient MIR optoelectronic devices.

Histone lysine methyltransferases(HKMTs)deposit methyl groups onto lysine residues on histones and play important roles in regulating chromatin structure and gene expression.The structures and functions of HKMTs have been extensively investigated in recent decades,significantly advancing our understanding of the dynamic regulation of histone methylation.Here,we review the recent progress in structural studies of representative HKMTs in complex with nucleosomes(H3K4,H3K27,H3K36,H3K79,and H4K20 methyltransferases),with emphasis on the molecular mechanisms of nucleosome recognition and trans-histone crosstalk by these HKMTs.These structural studies inform HKMTs'roles in tumorigenesis and provide the foundations for developing new therapeutic approachestargetingHKMTs incancers.

Linearized model for optimization of coupled electricity and natural gas systems

In this paper,a combined optimization of a coupled electricity and gas system is presented.For the electricity network a unit commitment problem with optimization of energy and reserves under a power pool,considering all system operational and unit technical constraints is solved.The gas network subproblem is a medium-scale mixed-integer nonconvex and nonlinear programming problem.The coupling constraints between the two networks are nonlinear as well.The resulting mixed-integer nonlinear program is linearized with the extended incremental method and an outer approximation technique.The resulting model is evaluated using the Greek power and gas system comprising fourteen gas-fired units under four different approximation accuracy levels.The results indicate the efficiency of the proposed mixed-integer linear program model and the interplay between computational requirements and accuracy.

Estimation of Throughfall Erosivity in a Highly Diverse Forest Ecosystem Using Sand-Filled Splash Cups

Sand-fflled splash cups were used to study the erosive power of rainfall and throughfall in the humid subtropics of Southeast China. The splash cup measurements yielded precise and reproducible results under both open field conditions and forest vegetation. The splash cups were exposed to specific forest stands of different ages and to selected species （Schima superba, Castanopsis eyrei, Daphniphyllum oldhamii, Lithocarpus glaber） in the Gutianshan （古田山） National Nature Reserve （GNNR）. The results of the measurements under forest vegetation show that the erosive power of throughfall drops to be 2.59 times higher compared to the open field. This accentuates the importance of shrub, herb and litter layers in forest ecosystems to protect the soil against erosion. Coalescing drops from leaves and branches （drips） are responsible for this notable gain in erosive power. Moreover, differences in sandloss between the investigated tree species （deciduous, evergreen） revealed that the erosion potential and the spatial heterogeneity of throughfall are species-specific. This highlights the importance of selecting specific species for afforestation projects considering the prevention of soil erosion.

Homology Priority Task Scheduling in μC/OS-Ⅱ Real-Time Kernel

μC/OS-Ⅱ is an open source real-time kernel adopting priority preemptive schedule strategy. Aiming at the problem of μC/OS-Ⅱ failing to support homology priority tasks scheduling, an approach for solution is proposed. The basic idea is adding round-robin scheduling strategy in its original scheduler in order to schedule homology priority tasks through time slice roundrobin. Implementation approach is given in detail. Firstly, the Task Control Block （TCB） is extended. And then, a new priority index table is created, in which each index pointer points to a set of homology priority tasks. Eventually, on the basis of reconstructing μC/OS-Ⅱ real-time kernel, task scheduling module is rewritten. Otherwise, schedulability of homology task supported by modified kernel had been analyzed, and deadline formula of created homology tasks is given. By theoretical analysis and experiment verification, the modified kernel can support homology priority tasks scheduling, meanwhile, it also remains preemptive property of original μC/OS-Ⅱ.

Simulation of hydrogen distribution and effect of Engineering Safety Features (ESFs) on its mitigation in a WWER-1000 containment

In this study, thermal–hydraulic parameters inside the containment of aWWER-1000/v446 nuclear power plant are simulated in a double-ended cold leg accident for short and long times (by using CONTAIN 2.0 and MELCOR 1.8.6 codes), and the effect of the spray system as an engineering safety feature on parameters mitigation is analyzed with the former code. Along with the development of the accident from design basis accident to beyond design basis accident, the Zircaloy–steam reaction becomes the source of in-vessel hydrogen generation. Hydrogen distribution inside the containment is simulated for a long time (using CONTAIN and MELCOR), and the effect of recombiners on its mitigation is analyzed (using MELCOR). Thermal–hydraulic parameters and hydrogen distribution profiles are presented as the outcome of the investigation. By activating the spray system, the peak points of pressure and temperature occur in the short time and remain belowthe maximumdesign values along the accident time. It is also shown that recombiners have a reliable effect on reducing the hydrogen concentration below flame propagation limit in the accident localization area. The parameters predicted by CONTAIN and MELCOR are in good agreement with the final safety analysis report. The noted discrepancies are discussed and explained.
