Whole-genome genotyping methods are important for breeding.However,it has been challenging to develop a robust method for simultaneous foreground and background genotyping that can easily be adapted to different genes and species.In our study,we accidently discovered that in adapter ligation-mediated PCR,the amplification by primertemplate mismatched annealing(PTMA)along the genome could generate thousands of stable PCR products.Based on this observation,we consequently developed a novel method for simultaneous foreground and background integrated genotyping by sequencing(FBI-seq)using one specific primer,in which foreground genotyping is performed by primer-template perfect annealing(PTPA),while background genotyping employs PTMA.Unlike DNA arrays,multiple PCR,or genome target enrichments,FBI-seq requires little preliminary work for primer design and synthesis,and it is easily adaptable to different foreground genes and species.FBI-seq therefore provides a prolific,robust,and accurate method for simultaneous foreground and background genotyping to facilitate breeding in the postgenomics era.

In 2020 and 2021,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),a novel coronavirus,caused a global pandemic.Vaccines are expected to reduce the pressure of prevention and control,and have become the most effective strategy to solve the pandemic crisis.SARS-CoV-2 infects the host by binding to the cellular receptor angiotensin converting enzyme 2(ACE2)via the receptor-binding domain(RBD)of the surface spike(S)glycoprotein.In this study,a candidate vaccine based on a RBD recombinant subunit was prepared by means of a novel glycoengineered yeast Pichia pastoris expression system with characteristics of glycosylation modification similar to those of mammalian cells.The candidate vaccine effectively stimulated mice to produce high-titer anti-RBD specific antibody.Furthermore,the specific antibody titer and virus-neutralizing antibody(NAb)titer induced by the vaccine were increased significantly by the combination of the double adjuvants Al(OH)_(3) and CpG.Our results showed that the virus-NAb lasted for more than six months in mice.To summarize,we have obtained a SARS-CoV-2 vaccine based on the RBD of the S glycoprotein expressed in glycoengineered Pichia pastoris,which stimulates neutralizing and protective antibody responses.A technical route for fucose-free complex-type N-glycosylation modified recombinant subunit vaccine preparation has been established.

Fractional derivative multivariable grey model for nonstationary sequence and its application

Most of the existing multivariable grey models are based on the 1-order derivative and 1-order accumulation, which makes the parameters unable to be adjusted according to the data characteristics of the actual problems. The results about fractional derivative multivariable grey models are very few at present. In this paper, a multivariable Caputo fractional derivative grey model with convolution integral CFGMC(q, N) is proposed. First, the Caputo fractional difference is used to discretize the model, and the least square method is used to solve the parameters. The orders of accumulations and differential equations are determined by using particle swarm optimization(PSO). Then, the analytical solution of the model is obtained by using the Laplace transform, and the convergence and divergence of series in analytical solutions are also discussed. Finally, the CFGMC(q, N) model is used to predict the municipal solid waste(MSW). Compared with other competition models, the model has the best prediction effect. This study enriches the model form of the multivariable grey model, expands the scope of application, and provides a new idea for the development of fractional derivative grey model.

Recently,stretchable and wearable health monitoring equipment has greatly improved human’s daily life,which sets higher demands for portable power source in stretchability,sustainability,and biocompatibility.In this work,we proposed a stretchable triboelectric nanogenerator(TENG)based on stretchable poly(3,4-ethylenedioxythiophene):poly(styrenesulfonate)(PEDOT:PSS)/porous carbon hybrid for oxyhemoglobin saturation(SpO2)monitoring.To combine advantages of carbon material for its high conductivity and organic electrode for its high stretchability,we spin-coated a solution of PEDOT:PSS/porous carbon onto a plasma-treated pre-stretched Ecoflex film to fabricate a stretchable electrode with rough surface.Due to its roughness and high potential difference with the dielectric material,the stretchable-electrode-based TENG exhibited better performance compared to the pristine TENG based on carbon or PEDOT:PSS material.The output voltage and current reached up to 51.5 V and 13.2μA as the carbon concentration increased.More importantly,the performance further increased under large strain(100%)which is suitable for wearable systems.Finally,the device demonstrated its application potential for powering a flexible blood oxygen monitor.This simple and cost-effective method can enhance the stretchability and stability of organic/inorganic electrode-based TENG,which paves the development of high-performance stretchable TENG.

Macrophages in tumors(tumor-associated macrophages,TAMs),a major population within most tumors,play key homeostatic functions by stimulating angiogenesis,enhancing tumor cell growth,and suppressing antitumor immunity.Resetting TAMs by simple,efficacious and safe approach(s)is highly desirable to enhance antitumor immunity and attenuate tumor cell malignancy.Previously,we used tumor cell-derived microparticles to package chemotherapeutic drugs(drug-MPs),which resulted in a significant treatment outcome in human malignant pleural effusions via neutrophil recruitments,implicating that drug-MPs might reset TAMs,considering the inhibitory effects of M2 macrophages on neutrophil recruitment and activation.Here,we show that drug-MPs can function as an antitumor immunomodulator by resetting TAMs with M1 phenotype and IFN-βrelease.Mechanistically,drug molecules in tumor MPs activate macrophage lysosomal P450 monooxygenases,resulting in superoxide anion formation,which further amplifies lysosomal ROS production and pH value by activating lysosomal NOX2.Consequently,lysosomal Ca^(2+)signaling is activated,thus polarizing macrophages towards M1.Meanwhile,the drug molecules are delivered from lysosomes into the nucleus where they activate DNA sensor hnRNPA2B1 for IFN-βproduction.This lysosomal-nuclear machinery fully arouses the antitumor activity of macrophages by targeting both lysosomal pH and the nuclear innate immunity.These findings highlight that drug-MPs can act as a new immunotherapeutic approach by revitalizing antitumor activity of macrophages.This mechanistic elucidation can be translated to treat malignant ascites by drug-MPs combined with PD-1 blockade.

目的探讨贝伐珠单抗(bevacizumab,BEV)单用及贝伐珠单抗联合替莫唑胺(temozolomide,TMZ)治疗复发性高级别脑胶质瘤(high-grade glioma,HGG)的近期疗效、安全性及影响复发性高级别脑胶质瘤患者总生存期(overall survival,OS)的预后因素。方法按照脑胶质瘤治疗效果评估(response assessment in neuro oncology,RANO)标准,以客观有效率(objective response rate,ORR)、疾病控制率(disease control rate,DCR)为指标,回顾性分析云南省肿瘤医院2020年8月至2023年7月31例复发性HGG患者的临床病历资料,分为对照组(BEV)和治疗组(BEV+TMZ),评价2组近期疗效。采用Cox单因素和多因素分析方法分析组别(对照组与治疗组)、性别、年龄、疾病分级、入院评分、组织病理型分类等对31例复发性HGG患者OS的影响;通过χ^(2)检验比较2组不良反应(adverse reactions,ADR),评价安全性。结果治疗组ORR=63.16%,对照组ORR=16.67%,差异有统计学意义(χ^(2)=6.419,P=0.011);治疗组DCR=89.47%,对照组DCR=75.00%,DCR比较差异无统计学意义(χ^(2)=0.320,P=0.571)。对照组中位生存期(mOS)为7个月(95%CI:3.605~10.395),治疗组中位生存期(mOS)为14个月(95%CI:3.853~24.147),2组mOS差异无统计学意义(χ^(2)=0.829,P=0.363)。31例复发性HGG患者Cox单因素分析入院评分和组织病理型分类差异有统计学意义(P0.05);血小板及白细胞降低率比较,差异有统计学意义(P<0.05),2组出现的ADR对症处理后均得到了缓解。结论治疗组治疗复发性HGG近期疗效优于对照组,2组治疗复发性HGG后的ADR可耐受,安全性良好。组织病理型分类可能是影响31例复发性HGG患者OS的预后因素。

目的基于CiteSpace探析中医“肾藏精”理论的研究历程、热点演变及前沿方向。方法检索中国知网、万方数据知识服务平台及维普网自建库至2023年6月研究主题符合“肾藏精”的文献,运用CiteSpace软件对作者、机构及关键词进行可视化分析。结果共纳入文献646篇,年发文量呈整体上升趋势。核心作者46名,发文最多者为郑洪新,初步形成以郑洪新、陈云志、王小琴等为代表的核心团队,主要在辽宁中医药大学、北京中医药大学、上海中医药大学附属龙华医院及天津中医药大学等机构间形成合作网络。纳入关键词1393个,研究热点可聚为理论探讨、临床诊疗及现代生物学机制3个集群,维生素D及骨髓间充质干细胞为研究前沿。结论“肾藏精”领域不断发展,逐步与现代医学接轨。未来需加强跨地域、多学科合作,结合现代前沿技术,不断深化“肾藏精”理论的物质基础及科学内涵研究。
