Relationship between Lingual Papilla Surface Roughness and Oral Condition in Elderly Individuals

Objectives: Recently, an increasing number of elderly patients have complained of tongue pain and fissured tongue, while atrophy of the lingual papillae with low nutrition has also become a commonly encountered condition. The viewpoint that lingual papilla atrophy reflects systemic and pathological conditions, such as diabetes and in the circulatory system abnormalities, is supported by many clinicians. The present study was conducted to clarify the relationship of degree of atrophy of tongue papillae with oral symptoms, with an aim to evaluate the usefulness of clinical diagnostic criteria for tongue papillae atrophy. Subjects and Methods: The subjects were asked to protrude the tongue forward. The tongue was held at rest and impressions were taken of the tongue dorsum 15 mm posterior to the apex of the tongue. Surface impressions were measured using a Surfcoder SE300 and the obtained values were used to define surface roughness (Ra). Multivariate analyses of the relationships between the Ra measurements and the living environment, subjective symptoms of oral health, and survey results of measurements were performed to examine factors associated with Ra. The tongue was photographed with a digital camera, then RGB color value of four random points 15 mm from the tongue apex was determined. Using those findings, redness was calculated. Results and Conclusions: One hundred and six subjects with a mean age of 79.5 ± 9.2 years were analyzed. In the smooth group, there was more redness in the tongue dorsum as compared to the rough group. Total and Sagittal Ra values of subjects with dysphagia were lower than those without dysphagia, suggesting that the degree of oral mucosal atrophy is related to subjective symptoms of dysphagi

The early Cambrian pentamerous microfossil Olivooides/Punctatus in South China, which is characterized by a diagnostic stellate tubular apex, has been well-known for its almost complete development sequence that can be confidently traced from embryos and hatched juveniles, to conical adults. However, its zoological affinity remains highly controversial. Here we describe the internal microanatomic structures of the soft-body inside the peridermal theca of Olivooides multisulcatus Qian, 1977, including interradial pairs of tentacles, adradial and perradial frenula, perradial oral marginal lappets and twins of perradial gastric saccule-like humps as well as a circular velarium with striated coronal muscles. Particularly, one specimen shows bifurcated velarial canals along the bell aperture. Both the components of the soft-body and the external theca （or cyst） are arranged in perfect pentaradial symmetry. These characteristics are more compatible with those of living cubomedusans and co-occurring Cambrian athecate embryonic cubozoans. Concerning the presence of peridermal theca, Olivooides most likely represents an extinct thecate stem-group cubomedusae but devoid of both perradial eyes and specialized pedalia. The well-grown soft body inside the peridermal tube displays a set of mixed features of both polyp and medusa.

AIM: To investigate the anti-neoplastic effects of MK615, an extract from the Japanese apricot (Prunus mume), against colon cancer cells. METHODS: Three colon cancer cell lines, SW480, COLO, and WiDr, were cultured with MK615. Growth inhibition was evaluated by cell proliferation assay and killing activity was determined by lactate dehydrogenase assay. Induction of apoptosis was evaluated by annexin Ⅴ flow cytometry. Morphological changes were studied by light and electron microscopy, and immunofluorescence staining with Atg8. RESULTS: MK615 inhibited growth and lysed SW480, COLO and WiDr cells in a dose-dependent manner. Annexin Ⅴ flow cytometry showed that MK615 induced apoptosis after 6 h incubation, at which point the occurrence of apoptotic cells was 68.0%, 65.7% and 64.7% for SW480, COLO, and WiDr cells, respectively. Light and electron microscopy, and immunofluorescence staining with Atg8 revealed that MK615 induced massive cytoplasmic vacuoles (autophagosomes) in all three cell lines. CONCLUSION: MK615 has an anti-neoplastic effect against colon cancer cells. The effect may be exerted by induction of apoptosis and autophagy.

Evaluation of Antibodies Induced by the Injection of Single Capsid Protein or Purified Virus Particle of Coxsackievirus B3 in Mice

Four capsid proteins (VP1, VP2, VP3, and VP4) of coxsackievirus B3 (CVB3) were expressed as recombinant proteins in an Escherichia coli expression system and used as antigens for subunit vaccines against CVB3 in ICR mice. Antigens were expressed as thioredoxin-histidine (TrxHis)-tagged protein and purified before immunization. Although all VPs other than VP4 induced anti-CVB3 specific antibodies in mice (detected by ELISA and western blotting), they did not neutralize the infectious CVB3 in a virus neutralization assay. Meanwhile, 2 virus strains were purified from CVB3 virus stock on the basis of their plaque size on HeLa cells. ICR mice were infected with the 2 purified virus strains (S-strain and L-strain) and unpurified virus stock (wild type) to analyze the difference in antibody responses against infections of purified and unpurified virus strains. The reactivity of antisera against each virus strain was tested by ELISA, and the results showed that the inoculation of purified virus strain induced a strong antibody response against the inoculated strain. As a result, the antibody response against wild-type and other virus strains was suppressed. These results suggest using unpurified virus stock as an antigen is advantageous for inducing a broad antibody response in inoculated animals.

To determine the type of surface roughness pattern that is suitable for adaptive suppression of the drag of an obstacle, we observed flow structures introduced by such obstacles. Several roughness patterns were tested: geometric patterns, fractal patterns, reptile-skin patterns, and patterns of circular cylinders arranged in a lattice and in a zigzag manner. A suitable pattern for adaptive control of flow is one that generates longitudinal vortices with nonconstant distances. The preferred instability mode of a laminar boundary layer is expected to be selected automatically from fluctuations involving many frequencies and caused by fractal patterns. Snake- and reptile-skin patterns may have a similar ability as fractal patterns because they consist of multiscale patterns. The longitudinal vortices generated from peculiar positions and concave corners in patterns were observed. The distance between these vortices is not constant because the onset of vortices is at concave corners in fractal patterns. These vortices have differing strengths and easily cause nonlinear interactions, so they can disturb a laminar boundary layer with several higher-harmonic frequencies. The velocity profiles of the laminar boundary-layer flow over the fractal patterns were measured by using hydrogen bubbles. The results show the down-wash flow between the longitudinal vortices, which means that the vortices may effectively suppress the boundary layer separation in an adverse pressure gradient.

Underweight and Obesity in Low Birth Weight Children in Early Infancy in Japan

Introduction: Although there are several reports on the prevalence of underweight or obesity in low birth weight (LBW, th percentile and ≥90th percentile of the body mass index (BMI), respectively, based on reference values for Japanese children. Results: LBW children had accelerated growth from birth to age 3.5 years, but at Age 5, both sexes had significantly lesser height and weight z-scores, with notably lesser BMI percentile scores for girls, compared with NBW children. The prevalence of underweight was significantly higher in the LBW children throughout the study period. There were no significant differences in the prevalence of obesity between the two groups, except for at birth. Multiple regression analysis revealed that small-for-gestational age was associated with underweight at Age 5 years. Conclusions: This study indicates that, up to Age 5, underweight is a more serious problem than obesity in LBW children. Our results suggest that careful observation of growth from early infancy is necessary to prevent the occurrence of underweight in early infancy.

Continuous Vagus Nerve Monitoring during Carotid Endarterectomy

Backgrounds: Injury to the vagus nerve or one of its branches during carotid endarterectomy can result in vocal fold paralysis but the exact mechanism of injury responsible for vocal fold paralysis after carotid endarterectomy is unclear. Aims: This study was performed to identify potential predictors of vagus nerve injury and obtain feedback by application of intraoperative continuous vagus nerve monitoring. Materials and Methods: Seventy-four patients undergoing carotid endarterectomy were enrolled. A new vagus nerve electrode was designed for less invasive continuous vagus nerve stimulation and monitoring of the vocal fold electromyogram without disturbing the surgical procedure. The device was rectangular (13 mm × 9 mm), with two small round electrodes set on a flexible silicon plate and tube. The electrode was fully implantable during carotid endarterectomy and was positioned at the most distal site of the vagus nerve by suturing to the connective tissue without nerve dissection. All patients underwent laryngoscopy to assess postoperative vocal fold and pharyngeal wall palsy at one week after carotid endarterectomy. Results: Sudden loss of the vocal fold electromyogram was noted in two patients (during plaque removal and during arterial wall suture in one each). In these two patients, incomplete vocal fold and pharyngeal palsy was confirmed by laryngoscopy. The cause of vagus nerve injury may have been traction at the time of distal internal carotid artery manipulation. The vocal fold electromyogram remained normal during the operation in the other 72 patients. However laryngoscopy revealed postoperative vocal fold and pharyngeal palsy in six patients. These findings suggested that delayed vagus nerve injury can occur after carotid endarterectomy. Conclusion: The continuous vagus nerve monitoring may be worthwhile for elucidating the mechanism of vagus nerve injury related to carotid endarterectomy.

Upregulated genes in toll-like receptor (TLR) signaling pathway in periodontitis-affected gingival tissues

Toll-like receptor (TLR) signaling is thought to be one of the most important pathways initiating periodontitis onset. We have previously reported that the TLR signaling pathway is upregulated in periodontitis-affected gingival tissues by microarray pathway frequency analysis. The aim of the present study was to quantitatively analyze specific upregulated genes in the TLR signaling pathway, as compared to healthy controls. Healthy and periodontitis-affected gingival tissues were taken from distinct sites of 3 patients with severe chronic periodontitis. Total RNAs from 6 gingival tissue samples were used for microarray. Samples were taken from 14 chronic periodontitis patients and 14 healthy individuals for quantitative reverse transcription real-time polymerase chain reaction (qRT-PCR) analysis. Data-mining analyses, such as pathway analyses, were performed and significant biological pathways in periodontitis were identified. In addition, qRT-PCR analysis was performed for 5 genes—cluster of differentiation 14 (CD14), lymphocyte antigen 96 (MD-2), interleukin-1 beta (IL-1β), interleukin 8 (IL-8), and chemokine ligand 9 (CXCL-9), which are associated with TLR signaling, in order to confirm the results of pathway analysis. qRT-PCR verified that the transcripts for 5 genes in the TLR signaling pathway were significantly upregulated (MD-2 p = 0.0082, CD14 p = 0.0322, IL-1β p = 0.0126, IL-8 p = 0.0438, CXCL-9 p = 0.0325), which was consistent with pathway analyses. We confirmed upregulated MD-2 gene expression levels and associated TLR pathway gene expression, including CD14, IL-1β, IL-8 and CXCL-9, in periodontitis-affected gingival tissues, as compared with healthy controls.

An Efficient Liver-Segmentation System Based on a Level-Set Method and Consequent Processes

This paper presents an efficient liver-segmentation system developed by combining three ideas under the operations of a level-set method and consequent processes. First, an effective initial process creates mask and seed regions. The mask regions assist in prevention of leakage regions due to an overlap of gray-intensities between liver and another soft-tissue around ribs and verte-brae. The seed regions are allocated inside the liver to measure statistical values of its gray-intensities. Second, we introduce liver-corrective images to represent statistical regions of the liver and preserve edge information. These images help a geodesic active contour (GAC) to move without obstruction from high level of image noises. Lastly, the computation time in a level-set based on reaction-diffusion evolution and the GAC method is reduced by using a concept of multi-resolution. We applied the proposed system to 40 sets of 3D CT-liver data, which were acquired from four patients (10 different sets per patient) by a 4D-CT imaging system. The segmentation results showed 86.38% ± 4.26% (DSC: 91.38% ± 2.99%) of similarities to outlines of manual delineation provided by a radiologist. Meanwhile, the results of liver segmentation only using edge images presented 79.17% ± 5.15% or statistical regions showed 74.04% ± 9.77% of similarities.
