ABSTRACT： CD98 has been described to play a crucial role in tumor progression and survival. However, the role of CD98 in biliary tract cancer remains unclear. We found that 36.7% of all patients with biliary tract cancer had a high CD98 expression. Statistical analysis using Spearman＇s rank correlation showed that CD98 was significantly correlated with L-type amino acid transporter 1 （LAT1, r=0.562, P〈0.001）, Ki-67 （r=0.230, P=0.006） and CD34 （r=0.290, P=0.005）. Multivariate analysis confirmed that a high CD98 expression was an independent prognostic factor for predicting poor outcome. CD98 is closely associated with tumor growth, biological aggressiveness, and survival of patients. With these data we proposed that CD98 expression is necessary for the development and pathogenesis of biliary tract cancer.

Background: Fibroepithelial polyp in the vulva is a rare disease mainly seen in female adolescents and young adults, and several reports were published in the past. Some cases showed large polyps that might be misinterpreted as malignant tumors such as aggressive angiomyxoma. Aim: We report a case of bilateral fibroepithelial polyps in the labia minora in an adolescent age girl. Case Presentation: A 16 year-old-girl was referred to our department for the treatment of an enlarged skin-tag appearance lesion at the right sided labia minora. The lesion was resected and the microscopic finding was a fibroepithelial polyp. One year after the first operation the patient was again referred to our department for the treatment of a skin-tag lesion at the left sided labia minora. It was again resected and the pathological diagnosis was a fibroepithelial polyp. Conclusion: Fibroepithelial polyp of the labia minora is not common and it is often seen in adolescent girls and in young adults. Bilateral lesions and skin-tag appearance are especially rare. The lesion should be excised for obtaining the correct diagnosis.

Radio-photoluminescence （RPL） is a phenomenon seen in luminescent materials in which the appearance of new photolu- minescence （PL） emission is induced by an incident ionizing radiation such as X-rays; and the signal is stable even after the irradia- tion and during the PL measurement. Since the induced PL intensity is proportional to the irradiation dose, the RPL can be used in ra- diation measurements. The distinct advantage of RPL over the conventional thermally- or photo-stimulated luminescence （abbrevi- ated as TSL or PSL） dosimeters is the stability of response signal. With an RPL detector, it allows us to readout the signal multiple time without signal fading. In this work, we discovered that CsBr：Sm showed an RPL phenomenon by X-ray irradiation, and we characterized this new material as an RPL detector. While the sample showed PL emissions mainly in the visible range, after an X-ray irradiation additional emissions could be observed in the red to near-infrared range around 650-850 nm and 900-1000 nm and longer. The RPL response was fairly stable overall, but very interestingly the 650-850 nm signal slightly increased while the 900-1000 nm decreased during PL readout. The dynamic range was confirmed over 1-104 mGy with linear response.

The silkworm,Bombyx mori,played an important role in the old Silk Road that connected ancient Asia and Europe.However,to date,there have been few studies of the origins and domestication of this species using molecular methods.In this study,DNA sequences of mitochondrial and nuclear loci were used to infer the phylogeny and evolutionary history of the domesticated silkworm and its relatives.All of the phylogenetic analyses indicated a close relationship between the domesticated silkworm and the Chinese wild silkworm.Domestication was estimated to have occurred about 4100 years ago(ya),and the radiation of the different geographic strains of B.mori about 2000 ya.The Chinese wild silkworm and the Japanese wild silkworm split about 23600 ya.These estimates are in good agreement with the fossil evidence and historical records.In addition,we show that the domesticated silkworm experienced a population expansion around 1000 ya.The divergence times and the population dynamics of silkworms presented in this study will be useful for studies of lepidopteran phylogenetics,in the genetic analysis of domestic animals,and for understanding the spread of human civilizations.

Development of a competitive enzyme-linked immunosorbent assay for therapeutic drug monitoring of afatinib

Afatinib is an oral tyrosine kinase inhibitor(TKI) approved for treating advanced non-small cell lung cancer. It is necessary to develop a simple quantification method for TKIs in order to facilitate therapeutic drug monitoring(TDM) in clinical settings. This study sought to develop a simple and sensitive competitive enzyme-linked immunosorbent assay(ELISA) to quantify afatinib in plasma for routine pharmacokinetic applications. An anti-afatinib antibody was obtained using(S)-N-4-(3-chloro-4-fluorophenyl)-7-(tetrahydrofuran-3-yloxy)-quinazoline-4,6-diamine(CTQD), which has the same substructure as afatinib, as a hapten. Enzyme labeling of afatinib with horseradish peroxidase was similarly performed using CTQD. A simple competitive ELISA for afatinib was developed based on the principle of direct competition between afatinib and the enzyme marker for the anti-afatinib antibody, which had been immobilized on the plastic surface of a microtiter plate. Plasma afatinib concentrations below the limit of quantification of 30 pg/mL were reproducibly measurable. Also, the values of plasma afatinib levels measured from 20 patients were comparable with those measured by high-performance liquid chromatography, and there was a strong correlation between the values determined by both methods(Y=0.976 X – 0.207, r=0.975). As indicated by its specificity and sensitivity, this newly developed ELISA for afatinib is an important tool for TDM and studies of the pharmacokinetics of afatinib.

The effects of alloying elements on zincate treatment and adhesion of electroless Ni-P coating onto various aluminum alloy substrates were examined.Surface morphology of zinc deposits in the 1st zincate treatment and its adhesion were changed depending on the alloying element.The zinc deposits in the 2nd zincate treatment became thinly uniform,and the adhesion between aluminum alloy substrate and Ni-P coating was improved irrespective of the alloying element.XPS analysis revealed the existence of zinc on the surface of each aluminum alloy substrate after the pickling in 5% nitric acid.This zinc on the surface should be an important factor influencing the morphology of zinc deposit at the 2nd zincate treatment and its adhesion.

AIM:To reveal the characteristics of CD133+ cells in the liver.METHODS:This study examined the histological characteristics of CD133+ cells in non-neoplastic and neoplastic liver tissues by immunostaining,and also analyzed the biological characteristics of CD133+ cells derived from human hepatocellular carcinoma(HCC) or cholangiocarcinoma cell lines.RESULTS:Immunostaining revealed constant expression of CD133 in non-neoplastic and neoplastic biliary epithelium,and these cells had the immunophenotype CD133+/CK19+/HepPar-1-.A small number of CD133+/CK19-/HepPar-1+ cells were also identified in HCC and combined hepatocellular and cholangiocarcinoma.In addition,small ductal structures,resembling the canal of Hering,partly surrounded by hepatocytes were positive for CD133.CD133 expression was observed in three HCC(HuH7,PLC5 and HepG2) and two cholangiocarcinoma cell lines(HuCCT1 and CCKS1).Fluorescence-activated cell sorting(FACS) revealed that CD133+ and CD133-cells derived from HuH7 and HuCCT1 cells similarly produced CD133+ and CD133-cells during subculture.To examine the relationship between CD133+ cells and the side population(SP) phenotype,FACS was performed using Hoechst 33342 and a monoclonal antibody against CD133.The ratios of CD133+/CD133-cells were almost identical in the SP and non-SP in HuH7.In addition,four different cellular populations(SP/CD133+,SP/CD133-,non-SP/CD133+,and non-SP/CD133-) could similarly produce CD133+ and CD133-cells during subculture.CONCLUSION:This study revealed that CD133 could be a biliary and progenitor cell marker in vivo.However,CD133 alone is not sufficient to detect tumor-initiating cells in cell lines.

Achieving greener cloud computing is non-negligible for the open-source cloud platform.In this paper,we propose a novel virtual machine allocation scheme with a sleep-delay and establish a corresponding mathematical model.Taking into account the number of tasks and the state of the physical machine,we construct a two-dimensional Markov chain and derive the average latency of tasks and the energy-saving degree of the system in the steady state.Moreover,we provide numerical experiments to show the effectiveness of the proposed scheme.Furthermore,we study the Nash equilibrium behavior and the socially optimal behavior of tasks and carry out an improved adaptive genetic algorithm to obtain the socially optimal arrival rate of tasks.Finally,we present a pricing policy for tasks to maximize the social profit when managing the network resource within the cloud environment.

Performance of 18-fluoro-2-deoxyglucose positron emission tomography for esophageal cancer screening

AIM To evaluate the performance of 18-fluoro-2-deoxyglucose positron emission tomography(FDG-PET) for esophageal cancer(EC) screening.METHODS We retrospectively analyzed the data of consecutive asymptomatic individuals who underwent FDG-PET and esophagogastroduodenoscopy(EGD) simultaneously for cancer screening at our institution from February 2004 to March 2013. In total, 14790 FDG-PET and EGD procedures performed for 8468 individuals were included in this study, and the performance of FDGPET for EC screening was assessed by comparing the results of FDG-PET and EGD, considering the latter as the reference.RESULTS Thirty-two EC lesions were detected in 28 individuals(31 squamous cell carcinomas and 1 adenocarcinoma). The median tumor size was 12.5 mm, and the depths of the lesions were as follows: Tis(n = 12), T1a(n = 15), and T1b(n = 5). Among the 14790 FDG-PET procedures, 51 examinations(0.3%) showed positive findings in the esophagus; only 1 was a true-positive finding. The screen sensitivity, specificity, positive predictive value, and negative predictive value of FDGPET for ECs were 3.6%(95%CI: 0.1-18.3), 99.7%(95%CI: 99.6-99.7), 2.0%(95%CI: 0.0-10.4), and 99.8%(95%CI: 99.7-99.9), respectively. Of the 50 FDG-PET false-positive cases, 31 were observed in the lower esophagus, and gastroesophageal reflux disease was observed in 17 of these 31 cases.CONCLUSION This study is the first to clarify the FDG-PET performance for EC screening. Based on the low screen sensitivity, FDG-PET is considered to be difficult to use as a screening modality for ECs.
