Mammalian pancreatic β-cells play a pivotal role in development and glucose homeostasis through the production and secretion of insulin. Functional failure or decrease in β-cell number leads to type 2 diabetes （T2D）. Despite the physiological importance of β-cells, the viability of β-cells is often challenged mainly due to its poor ability to adapt to their changing microenvironment. One of the factors that negatively affect β-cell viability is high concentration of free fatty acids （FFAs） such as palmitate. In this work, we demonstrated that Yes-associated protein （Yap1） is activated when β-cells are treated with palmitate. Our loss- and gain-of-function analyses using rodent insulinoma cell lines revealed that Yap1 suppresses palmitate-induced apoptosis in β-cells without regulating their proliferation. We also found that upon palmitate treatment, re-arrangement of F-actin mediates Yap1 activation. Palmitate treatment increases expression of one of the Yap1 target genes, connective tissue growth factor （CTGF）. Our gain-offunction analysis with CTGF suggests CTGF may be the downstream factor of Yap1 in the protective mechanism against FFA-induced apoptosis.

Objective:To determine the risk factors for hypertension in pregnant women,which is a significant cause of maternal and newborn morbidity and mortality in ***:This was a cross-sectional study used secondary data from the 2018 Indonesian Basic Health *** women aged 15-54 years in the second and third trimesters were *** history of hypertension in pregnant women was the dependent *** independent variables consisted of the demographic characteristics of pregnant women including pregnancy age,gestational age,education,occupation,place of residence,and region of *** study used multivariate logistic regression to determine the risk factors for hypertension in pregnant women in ***:.Among 6479 respondents included in this study,11(7.0%)had a history of diabetes mellitus(DM)and hypertension compared to pregnant women who did not have a history of DM(0.1%).The dominant risk factors for hypertension in pregnant women in Indonesia include maternal age above 35 years(OR 3.67,95%CI 2.54-5.32),third-trimester pregnancy(OR 2.40,95%CI 1.72-3.35),primigravida(OR 1.78,95%CI 1.19-2.68),excessive consumption of salty foods(OR 1.44,95%CI 1.03-2.03),and diabetes mellitus(OR 10.20,95%CI 3.04-32.23).Conclusions:Great efforts must be made to increase public awareness about the dangers of hypertension in pregnant women through early diagnosis and treatment,education on sodium intake,and appropriate care for pregnant women with DM.

<i>Momordica charantia</i>Ameliorates Atopic Dermatitis by Inhibiting the Expression of Inducible Nitric Oxidase Synthase in NC/Nga Mice

Introduction: Momordica charantia (MC) has been reported to possess various beneficial effects. Improvement in natural aging of the skin has been observed with the use of MC. However, few studies have detailed the effects of MC on atopic dermatitis (AD). Therefore, in this study, we investigated the effects of MC on the skin symptoms of AD. Methods: Specific pathogen-free and conventional NC/Nga mice were orally administered a 50 mg/kg/day dose of MC every day for 2 weeks. Results: The expression levels of lipopolysaccharide (LPS), inducible nitric oxidase synthase (iNOS), and prostaglandin E2 (PGE2) remarkably increased in AD, but were suppressed by MC administration. As a result, the degradation of filaggrin by PGE2 was suppressed. Furthermore, in AD, iNOS induced macrophage type 1 and increased NO levels. In contrast, due to suppression of iNOS with MC administration, macrophages shifted to type 2 and an increase in L-ornithine was observed, which subsequently promoted filaggrin synthesis. Conclusions: These findings indicate that the AD-like skin symptoms were decelerated by MC via the regulation of the LPS/iNOS/PGE2/filaggrin and LPS/iNOS/Arginase 1/L-ornithine/ filaggrin signaling pathways.

Role of the ACTH/MC2R System in the Hair Cycle in Mice

Adrenocorticotropin hormone (ACTH), which is secreted in response to psychological stress, plays an important role in the hair cycle. This study examined the mechanism by which ACTH affects the hair cycle using mice deficient in melanocortin receptor-2(MC2R-/-), which is a main receptor for ACTH. We observed the hair cycle using female MC2R-/- mice at 15 weeks old and five days old to determine whether there were any age-dependent differences. The 15-week-old MC2R-/- mice showed the anagen phase for all mice. On the other hand, all of the MC2R+/+ mice showed the telogen phase at the same age. Moreover, in the five-day-old mice, the hair growth of the MC2R-/- mice occurred earlier than in the MC2R+/+ mice. Both the 15-week-old and five-day-old MC2R-/- mice had higher levels of ACTH and alpha-melanocyte stimulating hormone in the blood than did the MC2R+/+ mice. In addition, in the 15-week-old MC2R-/- mice, the hair cycle shifted to the telogen phase following the administration of a cyclic guanosine monophosphate (cGMP) inhibitor and MC1R/MC5R inhibitor. In the five-day-old MC2R-/- mice, the hair growth was slowed by the administration of corticosterone. These results suggest that the ACTH/MC2R system has an important role in the hair cycle.

Regulation of <i>α</i>-Melanocyte-Stimulating Hormone by Testosterone Is Associated with the Onset of Atopic Dermatitis Symptoms during Exercise

Objective: Atopic dermatitis (AD) symptoms are altered with exercise. However, no study has investigated the association between testosterone and AD. The aim of this study was to investigate the role of testosterone by which the strength and weakness of exercise affects the skin symptoms of AD. Methods: Specific pathogen-free (SPF) and conventional NC/Nga mice were used. NC/Nga mice spontaneously developed dermal symptoms similar to AD patients. Two exercises, mild (20 m/min, 60 min) and rigorous (25 m/min, 90 min), were carried out using a treadmill four times every alternate day. Furthermore, we administered testosterone (0, 5, 50, 500, and 5000 pg/mice) to non-exercised conventional NC/Nga mice. On the final day of this experiment, we analyzed the plasma levels of immunoglobulin E (IgE), interleukin (IL)-6, IL-4, IL-13, testosterone and α-melanocyte-stimulating hormone (α-MSH) by ELISA kit. Results: Symptoms manifested by NC/Nga mice were strongly exacerbated upon severe exercise but were ameliorated during mild exercise. Between mild and severe exercised conventional mice, the plasma level of IgE was not changed. On administering an equivalent amount of testosterone, depending on the exercise, AD-like symptoms in non-exercising NC/Nga mice were ameliorated with mild exercise and exacerbated through rigorous exercise. Plasma IL-6, IL-4, and IL-13 levels remained unchanged between +50 pg (mild) and +500 pg (severe) testosterone administration. Plasma α-MSH levels were elevated with +500 pg testosterone but decreased with +50 pg testosterone administration. Conclusion: The present results suggest that exercise largely mimics AD symptoms depending on the α-MSH and testosterone levels.

Nodal segments from secondary branches of saplings of Phyllostachys bambusoides were inoculated in MS medium to assess the in vitro morphogenic response of leaf sheath through the induction to callogenesis by Picloram（4-amino-3,5,6-trichloropicolinic acid） at different concentrations of carbohydrate under the same conditions with presence or absence of *** our experiment,secondary explants were kept in MS medium containing 8.0 mg·L-1 of Picloram for the callus *** were transferred in MS medium supplemented with sucrose,fructose and glucose（control,2%,4% and 6%）.Results show that Picloram induced the callogenesis in leaf *** secondary embryogenesis was formed in yellow-globular *** sucrose as carbohydrate source in the absence of light was more efficient to induce *** was more efficiency in the presence of *** induction and further embryogenesis evidenced the competence and determination of leaf sheath cells.

It has been reported that atopic dermatitis (AD) and ulcerative colitis are related. However, the mechanism underlying this association has not been clarified. We therefore explored how AD induces ulcerative colitis. We developed an AD mouse model (NC/Nga mice) with ulcerative colitis by administering dextran sodium sulfate (DSS) for five days. DSS-induced ulcerative colitis was deteriorated in our conventional AD mouse model compared with specific-pathogen-free (SPF) mice. The plasma levels of thymic stromal lymphopoietin (TSLP) and tumor necrosis factor-α increased the most in DSS-treated conventional mice. Furthermore, the expression of dendritic cells (DC), retinoid-related orphan receptor (ROR)γt (marker of T helper 17 cells [Th17]), interleukin (IL)-17, GATA binding protein 3 (GATA3) (marker of Th2), and IL-4 increased the most in the colon of the DSS-treated conventional mice compared with DSS-treated SPF mice. In addition, TSLP inhibitor (TSLP neutralizing antibody) did not exacerbate ulcerative colitis in DSS-treated conventional mice. These results indicate that TSLP/DC/Th2 and Th17 play major roles in the exacerbation of ulcerative colitis by AD.
