Intragingival injection of Porphyromonas gingivalis-derived lipopolysaccharide induces a transient increase in gingival tumour necrosis factor-a, but not interleukin-6,in anaesthetised rats

This study used in vivo microdialysis to examine the effects of intragingival application of lipopolysaccharide（LPS） derived from Porphyromonas gingivalis（Pg-LPS） on gingival tumour necrosis factor（TNF）-a and interleukin（IL）-6 levels in rats. A microdialysis probe with an injection needle attached to the surface of the dialysis membrane was implanted into the gingiva of the upper incisor. For comparison, the effects of LPS derived from Escherichia coli（Ec-LPS） on IL-6 and TNF-a levels were also analysed. Pg-LPS（1 mg/1 m L） or Ec-LPS（1 or 6 mg/1 m L） was applied by microsyringe, with gingival dialysates collected every hour. Enzyme-linked immunosorbent assay（ELISA） revealed that gingival dialysates contained approximately 389 pg？m L21 of IL-6 basally; basal TNF-a levels were lower than the detection limit of the ELISA. Pg-LPS failed to alter IL-6 levels but markedly increased TNF-a levels, which remained elevated for 2 h after treatment. Neither IL-6 nor TNF-a were affected by Ec-LPS. Reverse transcriptase-polymerase chain reaction（RT-PCR） analysis revealed that the gingiva expresses Toll-like receptor（TLR） 2 and TLR4 m RNA. Immunohistochemical examination showed that TLR2 and TLR4 are expressed by gingival epithelial cells. The present study provides in vivo evidence that locally applied Pg-LPS, but not Ec-LPS, into the gingiva transiently increases gingival TNF-a without affecting IL-6. The present results suggest that TLR2 but not TLR4 expressed on gingival epithelial cells may mediate the Pg-LPS-induced increase in gingival TNF-a in rats.

Machine perfusion of the liver: Which is the best technique to mitigate ischaemia-reperfusion injury?

Longstanding research describes the mechanisms whereby the restoration of blood flow and reoxygenation(reperfusion) aggravates the ischaemic injury caused by a period of anoxia to a donor liver. This phenomenon, called ischaemia-reperfusion injury(IRI), leads to parenchymal cell death,microcirculatory failure, and inflammatory immune response. Clinically, IRI is the main factor responsible for the occurrence of posttransplant graft dysfunction and ischaemic-type biliary lesions. While extended criteria donor livers are more vulnerable to IRI, their utilisation is required to address the shortfall in donor organs. Thus, the mitigation of IRI should drive the setting of a new benchmark for marginal organ preservation. Herein, strategies incorporating different modalities of machine perfusion of the liver to alleviate IRI are discussed in conjunction with advantages and disadvantages of individual *** leading to reperfusion of the liver during machine perfusion(in situ normothermic regional perfusion and ex situ normothermic machine perfusion)may mitigate IRI by shortening the ischaemic period of the organs. This benefit potentially escalates from the minimum level, obtained following just partial alleviation of the ischaemic period, to the maximum level, which can be potentially achieved with ischaemia-free organ transplantation. Techniques that do not lead to reperfusion of the liver during machine perfusion(hypothermic,subnormothermic, and controlled-oxygenated rewarming) optimise mitochondrial oxidative function and replenish cellular energy stores, thereby lowering reactive oxygen species production as well as the activation ofdownstream inflammatory pathways during reperfusion. Further mechanistic insights into IRI may guide the development of donor-specific protocols of machine perfusion on the basis of the limitations of individual categories of extended criteria donor organs.

Computational modeling methods have been increasingly employed to quantify aortic hemodynamic parameters that are challenging to in vivo measurements but important for the diagnosis/treatment of aortic *** the presence of turbulence-like behaviors of blood flow in normal or diseased aorta has long been confirmed,the majority of existing computational model studies adopted the laminar flow assumption(LFA)in the treatment of sub-grid flow *** far,it remains unclear whether LFA would significantly compromise the reliability of hemodynamic *** the present study,we addressed the issue in the context of a specific aortopathy,namely aortic dilation,which is usually accompanied by disturbed flow *** patient-specific aortas with treated/untreated dilation of the ascending segment were investigated,and their geometrical models were reconstructed from computed tomography angiographic images,with the boundary conditions being prescribed based on flow velocity information measured in vivo with the phase contrast magnetic resonance imaging *** the modeling of blood flow,apart from the traditional LFA-based method in which sub-grid flow dynamics is ignored,the large eddy simulation(LES)method capable of incorporating the dissipative energy loss induced by turbulent eddies at the sub-grid level,was adopted and taken as a reference for examining the performance of the LFA-based *** results showed that the simulated large-scale flow patterns with the two methods had high similarity,both agreeing well with in vivo measurements,although locally large between-method discrepancies in computed hemodynamic quantities existed in regions with high intensity of flow ***,a switch from the LES to the LFAbased modeling method led to mild(<6%)changes in computed space-averaged wall shear stress metrics(i.e.,SA-TAWSS,SA-OSI)in the ascending aortic segment where intensive vortex evolution accompanied by high stat

Nonlinear optics is a well-established field of research that traditionally relies on the interaction of light with macroscopic nonlinear media over distances significantly greater than the wavelength of light. However, the recently emerged field of optical metasurfaces provides a novel platform for studying nonlinear phenomena in planar geometries. Nonlinear optical metasurfaces introduce new functionalities to the field of nonlinear optics extending them beyond perturbative regimes of harmonic generation and parametric frequency conversion,being driven by mode-matching, resonances, and relaxed phase-matching conditions. Here we review the very recent advances in the rapidly developing field of nonlinear metasurface photonics, emphasizing multi-frequency and cascading effects, asymmetric and chiral frequency conversion, nonperturbative nonlinear regimes, and nonlinear quantum photonics, empowered by the physics of Mie resonances and optical bound states in the continuum.

The paper is devoted to an approach for image inpainting developed on the basis of neurogeometry of vision and sub-Riemannian *** is realized by completing damaged isophotes(level lines of brightness)by optimal curves for the left-invariant sub-Riemannian problem on the group of rototranslations(motions)of a plane SE(2).The approach is considered as anthropomorphic inpainting since these curves satisfy the variational principle discovered by neurogeometry of vision.A parallel algorithm and software to restore monochrome binary or halftone images represented as series of isophotes were *** approach and the algorithm for computation of completing arcs are presented in detail.

An Experimental Prototype of an Innovative Fluid-driven Electromagnetic Stirring Technique

A new electromagnetic stirring technique that is driven by hydrodynamic forces was presented. This technique offers the following advantages. First,the stirrer can be immersed in the liquid metal,thereby significantly increasing the penetration depth of the electromagnetic forces and significantly improving the stirring efficiency; thus,this technique is particularly suitable for large-scale liquid metal. Second,under certain conditions,this technique can overcome difficulties that are encountered with traditional stirrers,such as accessing regions that are difficult to reach in working spaces with complex or narrow shapes. This stirrer also has a simpler structure than a traditional stirrer; thus,the design can be easily modified,and no external power supply is required. An experimental prototype was also presented for controlling the fluid flow rate,thereby controlling the electromagnetic force and velocity field of the driven liquid metal. The velocity distribution in a liquid Ga In Sn alloy under fluid-driven electromagnetic stirring was quantitatively measured using ultrasonic Doppler velocimetry（ UDV）. The primary results show that a remarkable velocity field has been achieved and that fluid-driven electromagnetic stirring is an effective means of stirring liquid metal. Finally,the potential applications of this technique in industry,along with key challenges,were discussed.
