To address the i.sue of how hi.pocampal neurons are i.volved i.to learni.g progress, we studi.d c-Fos expressi.n i. rat hi.pocampal subfi.lds at di.ferent stages of appeti.i.e i.strumental learni.g. To model the fi.st stage of learni.g, we pre-trai.ed ani.als to approach the lever to obtai. the food, and then made thi. behavi.r i.effecti.e by not rei.forci.g i. duri.g the last sessi.n (“mi.match” group). Another group just acqui.ed lever-pressi.g behavi.r at that day (“acqui.i.i.n” group). Ani.als of the thi.d group performed thi. well-trai.ed behavi.r (“performance” group). The number of Fos-posi.i.e neurons i. all hi.pocampal regi.ns of the “mi.match” group ani.als was hi.her than i. the ones of the home cage control group ani.als. The number of Fos-posi.i.e neurons was i.creased i. CA1 and CA3 areas, but not i. the dentate gyrus of both the “acqui.i.i.n” and “performance” group ani.als as compared wi.h the control group. We also found segmented c-Fos expressi.n, whi.h was more evi.ent i. “acqui.i.i.n” group ani.als. Thus, our results suggest that duri.g the fi.st (mi.match) stage of learni.g hi.pocampal neurons are acti.ated i. an equally di.tri.uted manner. The followi.g clustered pattern of acti.ated CA1 neurons duri.g the acqui.i.i.n stage may reflect speci.li.ati.n of these neurons i. respect to the speci.i. lever-pressi.g behavi.r.

We have previ.usly shown that both acute and chroni. ethanol treatment depresses neural acti.i.y, speci.i.ally i. the ci.gulate cortex. Mi.or i.fluences were found i. the motor cortex. The acute effect of ethanol i. the hi.pocampus was i.termedi.te to those i. the ci.gulate and motor corti.es. i. the present study, we concentrate on the chroni. effects of ethanol on the hi.pocampus. We demonstrate how the neuronal acti.i.y underlyi.g food-acqui.i.i.n behavi.r i. modi.i.d after chroni. ethanol treatment, and how the hi.pocampus subserves formati.n of newly-formed alcohol-acqui.i.i.n behavi.r. Neuronal acti.i.y i. CA1 was more sensi.i.e to chroni. ethanol than the Dg area. Acute admi.i.trati.n of ethanol had a normali.i.g effect on the chroni.ally-treated ani.als: thei. performance and the hi.pocampal neural acti.i.y approached a normal range. The sets of neurons i.volved i. food-acqui.i.i.n behavi.r formed before chroni. ethanol treatment, and those i.volved i. alcohol-acqui.i.i.n behavi.r formed after treatment si.ni.i.antly overlapped supporti.g the vi.w that the neuronal mechani.ms of pre-exi.ti.g behavi.r provi.e the basi. for the formati.n of new behavi.r. Addi.i.nally, we also di.covered alcohol-acqui.i.i.n selecti.e neurons. Assumi.g that the formati.n of new neuronal speci.li.ati.ns underli.s learni.g, we beli.ve that alcohol-selecti.e neurons are speci.li.ed duri.g the formati.n of alcohol-acqui.i.i.n behavi.r. Our data demonstrate several new fi.di.gs on the effect of acute and chroni. ethanol on hi.pocampus acti.i.y, and how the neuronal acti.i.y relates to behavi.r before and after ethanol treatment.
