Ginkgo biloba extract alleviates fatty liver hemorrhagic syndrome in laying hens via reshaping gut microbiota

Background Ginkgo biloba extract(GBE)is evidenced to be effective in the prevention and alleviation of metabolic disorders,including obesity,diabetes and fatty liver disease.However,the role of GBE in alleviating fatty liver hemorrhagic syndrome(FLHS)in laying hens and the underlying mechanisms remain to be elucidated.Here,we investigated the effects of GBE on relieving FLHS with an emphasis on the modulatory role of GBE in chicken gut microbiota.Results The results showed that GBE treatment ameliorated biochemical blood indicators in high-fat diet(HFD)-induced FLHS laying hen model by decreasing the levels of TG,TC,ALT and ALP.The lipid accumulation and pathological score of liver were also relieved after GBE treatment.Moreover,GBE treatment enhanced the antioxidant activity of liver and serum by increasing GSH,SOD,T-AOC,GSH-PX and reducing MDA,and downregulated the expression of genes related to lipid synthesis(FAS,LXRα,GPAT1,PPARγand Ch REBP1)and inflammatory cytokines(TNF-α,IL-6,TLR4 and NF-κB)in the liver.Microbial profiling analysis revealed that GBE treatment reshaped the HFD-perturbed gut microbiota,particularly elevated the abundance of Megasphaera in the cecum.Meanwhile,targeted metabolomic analysis of SCFAs revealed that GBE treatment significantly promoted the production of total SCFAs,acetate and propionate,which were positively correlated with the GBE-enriched gut microbiota.Finally,we confirmed that the GBE-altered gut microbiota was sufficient to alleviate FLHS by fecal microbiota transplantation(FMT).Conclusions We provided evidence that GBE alleviated FLHS in HFD-induced laying hens through reshaping the composition of gut microbiota.Our findings shed light on mechanism underlying the anti-FLHS efficacy of GBE and lay foundations for future use of GBE as additive to prevent and control FLHS in laying hen industry.

In this study,we investigated how empathic neural responses unfold over time in different empathy networks when viewing same-race and other-race individuals in dynamic painful conditions.We recorded magnetoencephalography signals from Chinese adults when viewing video clips showing a dynamic painful(or non-painful)stimulation to Asian and White models’faces to trigger painful(or neutral)expressions.We found that perceived dynamic pain in Asian models modulated neural activities in the visual cortex at 100 ms–200 ms,in the orbitofrontal and subgenual anterior cingulate cortices at 150 ms–200 ms,in the anterior cingulate cortex around 250 ms–350 ms,and in the temporoparietal junction and middle temporal gyrus around 600 ms after video onset.Perceived dynamic pain in White models modulated activities in the visual,anterior cingulate,and primary sensory cortices after 500 ms.Our findings unraveled earlier dynamic activities in multiple neural circuits in response to same-race(vs other-race)individuals in dynamic painful situations.

Portable power is an effective solution to realize self-powered sensors for wearable devices,promoting future sustainable development.Membrane-based triboelectric nanogenerators(M-TENGs)have emerged as a promising technology for harvesting biomechanical energy from human motion owing to their advantages,such as simple structure,lightweight design,and efficient energy conversion.However,the poor durability,low adaptability,and un-washability of two-dimensional membrane materials have largely hindered their application in wearable electronics.In this study,we propose a sheath-core polyvinylidene fluoride(PVDF)/graphene(G)-carbon fiber(CF)yarn fabricated via conjugate electrospinning,comprising a commercial CF core and an electrospun graphene-doped PVDF sheath,which improves the fatigue resistance of electrospun nanofiber films under prolonged friction and keeps a high degree of freedom.The resulting electronic textile,woven with the large-scale electrospun PVDF/G-CF yarn,demonstrates a remarkable power density of 25.5 mW·m^(-2).The tight distribution of PVDF/G nanofibers on the textile surface ensures excellent softness,washability,and durability.Furthermore,the electrospun PVDF/G-CF textile exhibits significant potential in pressure sensing,self-powered operation,and motion detection,making it highly suitable for wearable electronics applications.

Mitochondria are critical cellular energy resources and are central to the life of the neuron.Mitophagy selectively clears damaged or dysfunctional mitochondria through autophagic machinery to maintain mitochondrial quality control and homeostasis.Mature neurons are postmitotic and consume substantial energy,thus require highly efficient mitophagy pathways to turn over damaged or dysfunctional mitochondria.Recent evidence indicates that mitophagy is pivotal to the pathogenesis of neurological diseases.However,more work is needed to study mitophagy pathway components as potential therapeutic targets.In this review,we briefly discuss the characteristics of nonselective autophagy and selective autophagy,including ERphagy,aggrephagy,and mitophagy.We then introduce the mechanisms of Parkin-dependent and Parkin-independent mitophagy pathways under physiological conditions.Next,we summarize the diverse repertoire of mitochondrial membrane receptors and phospholipids that mediate mitophagy.Importantly,we review the critical role of mitophagy in the pathogenesis of neurodegenerative diseases including Alzheimer’s disease,Parkinson’s disease,and amyotrophic lateral sclerosis.Last,we discuss recent studies considering mitophagy as a potential therapeutic target for treating neurodegenerative diseases.Together,our review may provide novel views to better understand the roles of mitophagy in neurodegenerative disease pathogenesis.

Physical mechanism of the rapid increase in intense and long-lived extreme heatwaves in the Northern Hemisphere since 1980

Since 1980,both the intensity and duration of summer heatwaves in the middle and high latitudes of the Northern Hemisphere have significantly increased,leading this region to become a critical area for a significant increase in the frequency of intense and long-lived extreme heatwaves.We found that stronger and more persistent high-pressure systems and lower soil moisture before the events were the main drivers of intense and long-lived extreme heatwaves in western Europe and the middle and high latitudes of North America.However,in eastern Europe and Siberia,lower cloud cover before events is also a main driver of this type of extreme heatwave,in addition to the above drivers.These factors are coupled with each other and can change heatwave intensity and duration by influencing surface radiation processes during events.Using the self-organizing map classification method,we found that 6 weather patterns with increased frequency,intensity,and duration were the main dynamic reasons leading to the increase in intense and long-lived extreme heatwaves after 1980.In addition,the decrease in summer average soil moisture in most areas of the mid-high latitudes of the Northern Hemisphere and the decrease in average cloud cover in eastern Europe and Siberia are found to be the main thermodynamic reasons leading to the increase in these extreme heatwaves.

Accurate localization of cranial nerves and responsible blood vessels is important for diagnosing trigeminal neuralgia(TN)and hemifacial spasm(HFS).Manual delineation of the nerves and vessels on medical images is time-consuming and labor-intensive.Due to the development of convolutional neural networks(CNNs),the performance of medical image segmentation has been improved.In this work,we investigate the plans for automated segmentation of cranial nerves and responsible vessels for TN and HFS,which has not been comprehensively studied before.Different inputs are given to the CNN to find the best training configuration of segmenting trigeminal nerves,facial nerves,responsible vessels and brainstem,including the image modality and the number of segmentation targets.According to multiple experiments with seven training plans,we suggest training with the combination of three-dimensional fast imaging employing steady-state acquisition(3D-FIESTA)and three-dimensional time-of-flight magnetic resonance angiography(3DTOF-MRA),and separate segmentation of cranial nerves and vessels.

Afforestation effectively improved soil microbial communities and significantly increased soil nitro-gen mineralization rate(Rm).Soil microorganisms drive Rm by regulating soil N-cycling genes.Soil nitrification genes had a major effect on soil Rm than denitrification genes after afforestation.Assessing the function of forest ecosystems requires an understanding of the mechanism of soil nitrogen mineralization.However,it remains unclear how soil N-cycling genes drive soil nitrogen mineralization during afforestation.In this study,we collected soil samples from a chrono-sequence of 14,20,30,and 45 years of Robinia pseudoacacia L.(RP14,RP20,RP30,and RP45)with a sloped farmland(FL)as a control.Through metagenomic sequencing analysis,we found significant changes in the diversity and composition of soil microbial communities involved in N-cycling along the afforestation time series,with afforestation effectively increasing the diversity(both alpha and beta diversity)of soil microbial communities.We conducted indoor culture experiments and analyzed correlations,which revealed a significant increase in both soil nitrification rate(Rn)and soil nitrogen mineralization rate(Rm)with increasing stand age.Furthermore,we found a strong correlation between soil Rm and soil microbial diversity(both alpha and beta diversity)and with the abundance of soil N-cycling genes.Partial least squares path modeling(PLS-PM)analysis showed that nitrification genes(narH,narY,nxrB,narG,narZ,nxrA,hao,pmoC-amoC)and denitrification genes(norB,nosZ,nirK)had a greater direct effect on soil Rm compared to their effect on soil microbial communities.Our results reveal the relationships between soil nitrogen mineralization rate and soil microbial communities and between the mineralization rate and functional genes involved in N-cycling,in the context of Robinia pseudoacacia L.restoration on the Loess Plateau.This study enriches the understanding of the effects of microorganisms on soil nitrogen mineralization rate during afforestation and provides a ne

Anammox is widely considered as the most cost-effective and sustainable process for nitrogen removal.However,how to achieve the enrichment of anammox biomass remains a challenge for its large-scale application,especially in mainstream wastewater treatment.In this study,the feasibility of enrichment of anammox biomass was explored through the realization of partial denitrification and the addition of bio-carriers.By using ordinary activated sludge,a sequencing batch reactor(SBR)followed by an up-fow anaerobic sludge bed(UASB)was operated at 25±2℃ for 214 days.The long-term operation was divided into five phases,in which SBR and UASB were started-up in Phases I and II,respectively.By eliminating oxygen and adjusting the infow ratios in Phases III-V,advanced nitrogen removal was achieved with the effuent total nitrogen being 4.7 mg/L and the nitrogen removal efficiency being 90.5%in Phase V.Both in-situ and ex-situ activity tests demonstrated the occurrence of partial denitrification and anammox.Moreover,16S rRNA high-throughput sequencing analysis revealed that Candidatus Brocadia was enriched from below the detection limit to in biofilms(0.4%in SBR,2.2%in UASB)and the foc sludge(0.2%in SBR,1.3%in UASB),while Thauera was mainly detected in the foc sludge(8.1%in SBR,8.8%in UASB),which might play a key role in partial denitrification.Overall,this study provides a novel strategy to enrich anammox biomass driven by rapid achievement of partial denitrification through the addition of bio-carriers,which will improve large-scale application of anammox processes in mainstream wastewater treatment.

Clinical efficacy and safety of kyphoplasty for the treatment of osteoporotic vertebral compression fractures at different surgical timings based on the theory of“dynamic-static integration”

Objective:To investigate the clinical efficacy and safety of percutaneous kyphoplasty at different surgical timings in the treatment of osteoporotic vertebral compression fracture(OVCF)based on the theory of“dynamic-static integration”.Methods:Patients with OVCF who underwent percutaneous kyphoplasty in our hospital were selected and divided into Groups A,B,and C for those undergoing surgery within 7,7—21,and>21 days of fracture occurrence.The variations in the amount of bone cement injected,pre-and post-operative pain levels,functional activity,deformity correction of the injured vertebrae,bone cement leakage,and vertebral body height loss were compared among the three groups.Results:Regarding pain relief and functional activity,the postoperative Visual Analog Scale and Oswestry Disability Index scores of the three groups significantly improved.Furthermore,the deformities of the injured vertebrae in the three groups were significantly corrected,with Groups A and B exhibiting superior correction compared to Group C.Moreover,the bone cement leakage rates in groups A and C were higher than that in Group B.At the 3-month follow-up,the loss of vertebral height in Group C was significantly higher than those in groups A and B.Conclusion:Kyphoplasty is effective for OVCF treatment.Early surgery can effectively restore the vertebral height of the injured vertebra,reduce kyphosis,and reduce height loss of the injured vertebra after surgery;nevertheless,treatment within 1—3 weeks of the fracture can reduce the occurrence of bone cement leakage,making the surgery safer.Therefore,surgical treatment within 1—3 weeks of fracture is safer and can achieve satisfactory therapeutic effects.From the perspective of traditional Chinese medicine,PKP surgery can transform the fracture end from a micromotion state to a fixed state,which fully embodies the theory of“dynamic-static integration”.
