Ginkgo biloba extract alleviates fatty liver hemorrhagic syndrome in laying hens via reshaping gut microbiota

Background Ginkgo biloba extract(GBE)is evidenced to be effective in the prevention and alleviation of metabolic disorders,including obesity,diabetes and fatty liver ***,the role of GBE in alleviating fatty liver hemorrhagic syndrome(FLHS)in laying hens and the underlying mechanisms remain to be ***,we investigated the effects of GBE on relieving FLHS with an emphasis on the modulatory role of GBE in chicken gut *** The results showed that GBE treatment ameliorated biochemical blood indicators in high-fat diet(HFD)-induced FLHS laying hen model by decreasing the levels of TG,TC,ALT and *** lipid accumulation and pathological score of liver were also relieved after GBE ***,GBE treatment enhanced the antioxidant activity of liver and serum by increasing GSH,SOD,T-AOC,GSH-PX and reducing MDA,and downregulated the expression of genes related to lipid synthesis(FAS,LXRα,GPAT1,PPARγand Ch REBP1)and inflammatory cytokines(TNF-α,IL-6,TLR4 and NF-κB)in the *** profiling analysis revealed that GBE treatment reshaped the HFD-perturbed gut microbiota,particularly elevated the abundance of Megasphaera in the ***,targeted metabolomic analysis of SCFAs revealed that GBE treatment significantly promoted the production of total SCFAs,acetate and propionate,which were positively correlated with the GBE-enriched gut ***,we confirmed that the GBE-altered gut microbiota was sufficient to alleviate FLHS by fecal microbiota transplantation(FMT).Conclusions We provided evidence that GBE alleviated FLHS in HFD-induced laying hens through reshaping the composition of gut *** findings shed light on mechanism underlying the anti-FLHS efficacy of GBE and lay foundations for future use of GBE as additive to prevent and control FLHS in laying hen industry.

The wet multi-disc clutches are extensively used in various transmission systems,withone of the most prevalent failure modes being the buckling deformation of friction *** Hilbert-Huang transform method(IHHT)is proposed to address the limitations of tradi-tional time-domain vibration analyses,such as low accuracy and mode *** paper first clas-sifies the buckling degree of the friction ***,wavelet packet transform(WPT)isapplied to the vibration signals of different buckling plates to partition them into distinct fre-quency ***,the instantaneous features are extracted by empirical mode decomposition(EMD)and Hilbert transform(HT)to discarding extraneous intrinsic mode function(IMF)*** analyses of Hilbert spectral entropy and time-domain features confirm theenhanced precision of IHHT under specific classifiers,which is better than traditional methods.

Probiotics have been demonstrated to be effective and safe for hyperuricemia (HUA) and gout treatment. Hence， identifying microbes with high uric acid- or purine-metabolizing ability is highly important. Due to the high load of uric acid in chicken feces， we hypothesized that the chicken gut would be an excellent source for new uric acid or purine-metabolizing microbes. In this study， we examined chicken gut contents to isolate an Enterococcus faecalis strain (designated CML390) that displayed high uric acid-degrading ability. In vitro experiments indicated that E. faecalis CML390 could effectively metabolize uric acid under both aerobic and anaerobic conditions， and the richer the nutrients， the higher the rate of uric acid degradation. By integrating genomics， transcriptomics， and metabolomics analyses， we revealed two pathways involved in aerobic and anaerobic uric acid degradation， respectively， in this isolate. E. faecalis CML390 relies on oxygen to metabolize uric acid into substances such as allantoin under aerobic conditions， while it also harbors a conserved uric acid-inducible gene cluster for anaerobic uric acid metabolism. An in vivo experiment showed that gavage with live E. faecalis CML390 in a mouse model of HUA (established using a combination of uric acid and potassium oxonate) significantly reduced serum uric acid levels and alleviated HUA. Our findings contribute to the exploration of the pathways and mechanisms of uric acid metabolism in intestinal microorganisms and provide a candidate probiotic for the treatment of HUA and gout.

在全球变暖的背景下,区域干旱化程度加强,严重限制了干旱和半干旱地区树木的生长。安太堡矿区位于农牧交错带,生态环境脆弱,对气候变化敏感。通过树木年代学研究方法,建立了安太堡煤矿复垦区人工刺槐林(Robinia pseudoacacia)的树轮宽度标准年表,树轮气候响应分析发现水分是限制研究区刺槐生长的主要因素,上一年9月至当年8月的水文年降水以及主要生长季(6~8月)的降水对树木的生长有重要的促进作用,但是当年9月降水有限制作用;生长季前冬季(12月、1月)的较高温度也是限制刺槐径向生长的重要因素。

为了降低板材料库的生产和加工成本,对料库托盘进行结构简化和轻量化改进。从型材选型入手,对结构方管的承重性能进行参数敏感度分析,汇总不同方管的质量与性能。通过拓扑优化计算,得到托盘承载的材料最佳分配方案,设计最优的托盘结构,并对该结构进行参数优化,运用稀疏网格模型拟合试验参数建立响应面模型,通过MOGA算法进行优化求解,得到结构参数的最优解。通过搭配不同方管和最优解方案,实现了不同目标的优化效果。

Crystallographic facet-dependent stress responses by polyhedral lead sulfide nanocrystals and the potential ＂safe-by-design＂ approach

The particular physicochemical properties of nanomaterials are able to elicit unique biological responses. The property activity relationship is usually established for in-depth understanding of toxicity mechanisms and designing safer nanomaterials. In this study, the toxic role of specific crystallographic facets of a series of polyhedral lead sulfide （PbS） nanocrystals, including truncated octahedrons, cuboctahedrons, truncated cubes, and cubes, was investigated in human bronchial epithelial cells （BEAS-2B） and murine alveolar macrophages （RAW 264.7） cells./100} facets were found capable of triggering facet-dependent cellular oxidative stress and heavy metal stress responses, such as glutathione depletion, lipid peroxidation, reactive oxygen species （ROS） production, heme oxygenase-1 （HO-1） and metallothionein （MT） expression, and mitochondrial dysfunction, while {111} facets remained inert under biological conditions. The {100}-facet-dependent toxicity was ascribed to {100}-facet-dependent lead dissolution, while the low lead dissolution of {111} facets was due to the strong protection afforded by poly（vinyl pyrrolidone） during synthesis. Based on this facet-toxicity relationship, a ＂safe-by-design＂ strategy was designed to prevent lead dissolution from {100} facets through the formation of atomically thin lead-chloride adlayers, resulting in safer polyhedral PbS nanocrystals.
