Pseudomonas aeruginosa is a ubiquitous and metabolically versatile microorganism naturally found in soil and *** is also an opportunistic pathogen in plants,insects,animals,and *** response to increasing cell density,*** uses two acylhomoserine lactone(AHL)quorum-sensing(QS)signals(i.e.,N-3-oxo-dodecanoyl homoserine lactone[3-oxo-C12-HSL]and Nbutanoyl-homoserine lactone[C4-HsL]),which regulate the expression of hundreds of ***,how the biosynthesis of these two QS signals is coordinated remains *** studied the regulation of these two QS signals in the rhizosphere strain ***1201 sequentially produced 3-oxo-C12-HSL and C4-HSL at the early and late growth stages,*** highest 3-oxo-C12-HSL-dependent elastase activity was observed at the early stage,while the highest C4-HSL-dependent rhamnolipid production was observed at the late *** atypical regulator RsaL played a pivotal role in coordinating 3-oxo-C12-HSL and C4-HSL biosynthesis and QS-associated *** repressed las/transcription by binding the-10 and-35 boxes of the lasl *** contrast,RsaL activated rhll transcription by binding the region encoding the 5'-untranslated region of the rhll ***,RsaL repressed its own expression by binding a nucleotide motif located in the-35 box of the rsaL ***,RsaL acts as a molecular switch that coordinates the sequential biosynthesis of AHL QS signals and differential virulence in ***,C4-HSL activation by RsaL was independent of the Las and Pseudomonas quinolone signal(PQS)QS signaling ***,we propose a new model of the QS regulatory network in PA1201,in which RsaL represents a superior player acting at the top of the hierarchy.

The Triticum-Aegilops complex groups demonstrated high cross-affinity with each other to overcome the barriers of distant hybridization(Loureiro et al.,2023).Distant hybridization involves two distinct yet closely related events:hybridization and genome *** studies have indicated that bursts of transposable elements(TEs)can occur as a consequence or concomitant to hybridization or genome duplication(Parisod et al.,2010).This raises an important scientific question regarding how the TEs-rich centromere region copes with genomic shock(McClintock,1984).The Triticum-Aegilops species complexes,particularly in the F1,So,and subsequent early generations resulting from successive selfcrossing,offer an opportunity to investigate whether the centromere environment undergoes reconstruction and the associated mechanisms that maintain genomic stability.

Sex control techniques are in great demand for aquaculture because of the huge benefits of mono-sex *** a previous study on yellow catfish(Tachysurus fulvidraco),intersex individuals were produced by administrating 17α-methyltestosterone(MT)or aromatase inhibitor letrozole(LZ).The ovarian and testicular portions of intersexual gonads developed well and were completely separated,arousing our interest in producing self-fertilizable fish and clone-like fish *** this study,yellow catfish fry were treated with a combination of MT and LZ at a low dose(5 mg kg^(-1)MT,300 mg kg^(-1)LZ)at a low water temperature(27.0℃,mixed sex-producing temperature,LL group)and a high temperature(33.5◦C,male-producing temperature,HL group),and with a high dose(10 mg kg^(-1) MT,600 mg kg^(-1) LZ)at a low water temperature(LH group),during 12-65 days post-hatching,the critical period of sex *** XX and XY intersex were produced with different percentages at the same time through co-treatment with MT and LZ,with or without the help of male-producing *** highest percentage of XY intersex(53%in XY,36%in XX and XY)and XX intersex(47%in XX,25%in XX and XY)was produced in LH and HL groups,*** of gonadal morphology,histology,and sperm dynamic analysis indicate that some XX and XY intersex individuals were capable of self-fertilization through artificial *** reproductive performance of these intersex individuals was then investigated by injecting spawning agents,dissecting gonads,and artificially fertilizing eggs with their own *** XX,XY,and YY genotypic individuals were produced by the self-fertilization of XY intersex *** present study successfully produced high percentages of XX and XY intersex and conducted self-fertilization by mating the eggs and sperm of a single intersex ***-fertilization in fish will show great potential in aquaculture genetics and breeding,for example,generating high

Machine learning-based prediction of pregnancy outcomes in couples with non-obstructive azoospermia using micro-TESE for ICSI: a retrospective cohort study

Objective:To develop a clinically applicable tool for predicting clinical pregnancy,providing individualized patient counseling,and helping couples with non-obstructive azoospermia(NOA)decide whether to use fresh or cryopreserved spermatozoa for oocyte insemination before microdissection testicular sperm extraction(mTESE).Methods:A total of 240 couples with NOA who underwent mTESE-ICSI were divided into two groups based on the type of spermatozoa used for intracytoplasmic sperm injection(ICSI):the fresh and cryopreserved *** evaluating several machine learning algorithms,logistic regression was *** LASSO regression and 10-fold cross-validation,the factors associated with clinical pregnancy were ***:The area under the curves(AUCs)for the fresh and cryopreserved groups in the Logistic Regression-based prediction model were 0.977 and 0.759,*** with various modeling algorithms,Logistic Regression outperformed machine learning in both groups,with an AUC of 0.945 for the fresh group and 0.788 for the cryopreserved ***:The model accurately predicted clinical pregnancies in NOA couples.

Flexible ureteroscopic treatment of kidney stones: How do the new laser systems change our concepts?

Objective: Flexible ureteroscopy (fURS) has become a widely accepted and effective technique for treating kidney stones. With the development of new laser systems, the fURS approach has evolved significantly. This literature review aims to examine the current state of knowledge on fURS treatment of kidney stones, with a particular focus on the impact of the latest laser technologies on clinical outcomes and patient ***: We conducted a search of the PubMed/PMC, Web of Science Core Collection, Scopus, Embase (Ovid), and Cochrane Databases for all randomized controlled trial articles on laser lithotripsy in September 2023 without time ***: We found a total of 22 relevant pieces of literature. Holmium laser has been used for intracavitary laser lithotripsy for nearly 30 years and has become the golden standard for the treatment of urinary stones. However, the existing holmium laser cannot completely powder the stone, and the retropulsion of the stone after the laser emission and the thermal damage to the tissue have caused many problems for clinicians. The introduction of thulium fiber laser and Moses technology brings highly efficient dusting lithotripsy effect through laser innovation, limiting pulse energy and broadening pulse ***: While the holmium:yttrium-aluminum-garnet laser remains the primary choice for endoscopic laser lithotripsy, recent technological advancements hint at a potential new gold standard. Parameter range, retropulsion effect, laser fiber adaptability, and overall system performance demand comprehensive attention. The ablation efficacy of high-pulse-frequency devices relies on precise targeting, which may pose practical challenges.

Knockdown of the nucleoporin Nup50 protects cells against ionizing radiation through enhancing DNA-PKcs-mediated DNA damage repair

Objective:To investigate the effect and mechanism of Nup50 on radiation-induced DNA damage repair to radiation and explore the potential role of Nup50 as radioprotective ***:The Nup50 gene was knocked down in HUVEC cells using *** formation,CCK-8,and flow cytometry were performed to determine the viability,proliferation and apoptosis of HUVEC cells treated withγ-rays,*** extent of DNA damage was evaluated by using comet assay and immu-nofluorescence staining againstγ-*** addition,we explored the role of Nup50 in DNA damage response(DDR)pathways through western blotting ***,nuclear and chromatin fractionation were performed to determine the potential molecular mechanism underlying the radiation protection function of Nup50 ***:Nup50 knockdown increased the cellular resistance to ionizing *** CCK-8 data showed that cell viability was significantly increased in the Nup50 knockdown group after radiation(t=4.23,P<0.01).The Nup50 knockdown group also showed more survived colonies(t=10.06,P<0.001),less apoptosis rate(t=3.78,P<0.05)and less unrepaired DNA ***,Nup50 knockdown increased radiation-activated phosphorylation levels of DNA-PKcs in HUVEC ***,the nuclear and chromatin fractionation data showed that inhibiting Nup50 increased the recruitment of DNA-PKcs to chromatin after DNA ***:Our findings revealed that Nup50 knockdown promoted radioresistance in normal HUVEC cells by regulating DNA-PKcs pathway,suggesting Nup50 as a potential target for radiation protection.

Alkaline water electrolysis is an environmentally friendly and promising approach to produce ***,high cost,low efficiency,and poor stability are roadblocks to commercialization of *** work aims to design and develop a highly efficient,durable,and cost-effective electrocatalyst toward water splitting through modifying metal–organic *** electrocatalytic performance and stability surpass those of noble metal benchmarks at high current density(1–10 A·cm^(−2)).Theoretical calculations and in situ Raman spectra reveal the electronic structure of the synthesized catalyst and the mechanism of the catalytic reaction process,which rationalizes that the high catalytic activity and stability at high current are attributed to the unique electronic structure of cobalt regulated by copper and the protection provided by carbon nanotubes formed in situ,*** addition,this paper proposes that the desorption ability of the catalyst toward the products(H_(2)and O_(2)),rather than the adsorption ability toward the reactants(H^(+)or OH^(−)),is more important to the sustainable and stable electrochemical water splitting progress at high current density,which is a kinetic rather than thermodynamic dominating *** findings provide alternative insights to design and employ high performance catalysts to fuel hydrogen production as a clean energy source to tackle the global energy crisis.
