L-Arginine/nitric oxide regulates skeletal muscle development via muscle fibre-specific nitric oxide/mTOR pathway in chickens

L-Arginine (L-Arg), the precursor of nitric oxide (NO), plays an important role in muscle function. Fasttwitchglycolytic fibres are more susceptible to age-related atrophy than slow-twitch oxidative fibres.The effect of L-Arg/NO on protein metabolism of fast- and slow-twitch muscle fibres was evaluated inchickens. In Exp. 1, 48 chicks at 1 day old were divided into 4 groups of 12 birds and subjected to 4treatments: basal diet without supplementation or supplemented with 1% L-Arg, and water supplementedwith or without L-nitro-arginine methyl ester (L-NAME, 18.5 mM). In Exp. 2, 48 chicks weredivided into 4 groups of 12 birds fed with the basal diet and subjected to the following treatments: tapwater (control), tap water supplemented with L-NAME (18.5 mM), or molsidomine (MS, 0.1 mM), or18.5 mM L-NAME t 0.1 mM MS (NAMS). The regulatory effect of L-Arg/NO was further investigatedin vitro with myoblasts obtained from chicken embryo pectoralis major (PM) and biceps femoris (BF).In vivo, dietary L-Arg supplementation increased breast (t14.94%, P < 0.05) and thigh muscle mass(t23.40%, P < 0.05);whereas, MS treatment had no detectable influence. However, L-NAME treatmentblocked the beneficial influence of L-Arg on muscle development. L-Arg decreased (P < 0.05) proteinsynthesis rate, phosphorylated mTOR and ribosomal protein S6 kinase beta-1 (p70S6K) levels in breastmuscle, which was recovered by L-NAME treatment. In vitro, L-Arg or sodium nitroprusside (SNP)reduced protein synthesis rate, suppressed phosphorylated mTOR/p70S6K and decreased atrogin-1 andmuscle RING finger 1 (MuRF1) in myoblasts from PM muscle (P < 0.05). L-NAME abolished the inhibitoryeffect of L-Arg on protein synthesis and the mTOR/p70S6K pathway. However, myoblasts from BF muscleshowed the weak influence. Moreover, blocking the mTOR/p70S6K pathway with rapamycin suppressedprotein synthesis of the 2 types of myoblasts;whereas, the protein expression of atrogin-1 and MuRF1levels were restricted only in myoblasts from PM muscle. As a key epigenetic modification,5-formylcytosine(5fC)controls many important cell functions,such as gene expression and cell differentiation.However,5fC exists in mammalian tissues and cells with low content,and its structure is highly similar to 5-methylcytosine(5mC),5-hydroxymethylcytosine(5hmC)and 5-carboxycytosine(5caC), which requires the detection method for detecti ng 5fC with high sensitivity and specificity.In this experiment,WS_(2)/Bi/Bi_(2)O_(2)CO_(3) was used as the photoactive material,5fC antibody was used as the target molecule recog nition reage nt,and rolling amplification(RCA)was employed as the signal amplification strategy to con struct a no vel photoelectrochemical biose nsor for detecti ng 5-formylcytosi ne deox ynu cleotide(SfdCTP).

Cardiovascular diseases(CVDs)are one of the most urgent threats to humans worldwide,which are responsible for almost one-third of global mortality.Over the last decade,research on flexible electronics for monitoring and treatment of CVDs has attracted tremendous attention.In contrast to conventional medical instruments in hospitals that are usually bulky,hard to move,monofunctional,and time-consuming,flexible electronics are capable of continuous,noninvasive,real-time,and portable monitoring.Notable progress has been made in this emerging field,and thus a number of significant achievements and concomitant research prospects deserve attention for practical implementation.Here,we comprehensively review the latest progress of flexible electronics for CVDs,focusing on new functions provided by flexible electronics.First,the characteristics of CVDs and flexible electronics and the foundation of their combination are briefly reviewed.Then,four representative applications of flexible electronics for CVDs are elaborated:blood pressure(BP)monitoring,electrocardiogram(ECG)monitoring,echocardiogram monitoring,and direct epicardium monitoring.Their operational principles,progress,merits and demerits,and future efforts are discussed.Finally,the remaining challenges and opportunities for flexible electronics for cardiovascular healthcare are outlined.

The optimal dietary arginine level of laying hens fed with low-protein diets

Background:Arginine(Arg)is an essential amino acid(EAA)in poultry,an important substrate for protein synthesis and a precursor of several molecules.Supplementation of EAAs with low protein(LP)diet increases the utilization efficiency of dietary crude protein(CP).However,if the EAA requirement is changed in hens fed a LP diet remains to be elucidated.The aim of the present study was to evaluate the optimal level of dietary Arg in the LP diet of hens.A total of 1350 Hy-Line Brown laying hens were randomly allocated to six dietary treatments:a basal diet(16%CP,positive control),or an isoenergetic LP diet(14%CP,0.80%Arg)supplemented 0,0.05%,0.10%,0.15%,and 0.20%L-Arg,corresponding to 0.80%,0.85%,0.90%,0.95%and 1.00%dietary Arg,respectively.Results:The feed efficiency was decreased(P<0.05)by 0.80%and 1.00%Arg-LP diets,compared to control.Within LP diets,dietary Arg level had significant quadratic effects(P<0.05)on laying rate,egg mass,and feed efficiency.Compared to control,the plasma CAT activity or T-AOC content were decreased by 0.80%(P<0.001).However,the hens offered 0.85%and 0.90%Arg-LP diets had higher CAT activity(P<0.001)than 0.80%Arg-LP diet.In contrast,1.00%Arg-LP group had the highest MDA and the lowest T-AOC content in plasma,liver,duodenal and jejunal mucosa(P<0.05).Compared to control,the villus height was decreased by 0.80%,0.95%and 1.00%Arg-LP diets,while the villus height to crypt depth(V/C)ratio was reduced by 0.95%and 1.00%Arg-LP diets in duodenum.Conclusion:The result demonstrates that LP diet(14%CP)deficient in Arg(0.80%Arg)result in augmented oxidative damage and impaired development of intestinal mucosa.According to the quadratic broken-line regression model,the optimal dietary arginine levels for Hy-Line Brown laying hens fed with low protein diet(14%CP)aged 33 to 40 weeks are 0.85%,0.86%,and 0.86%to obtained the maximum laying rate,egg mass,and feed efficiency,respectively.

A Printable and Conductive Yield-Stress Fluid as an Ultrastretchable Transparent Conductor

In contrast to ionically conductive liquids and gels,a new type of yield-stress fluid featuring reversible transitions between solid and liquid states is introduced in this study as a printable,ultrastretchable,and transparent conductor.The fluid is formulated by dispersing silica nanoparticles into the concentrated aqueous electrolyte.The as-printed features show solid-state appearances to allow facile encapsulation with elastomers.The transition into liquid-like behavior upon tensile deformations is the enabler for ultrahigh stretchability up to the fracture strain of the elastomer.Successful integrations of yield-stress fluid electrodes in highly stretchable strain sensors and light-emitting devices illustrate the practical suitability.The yield-stress fluid represents an attractive building block for stretchable electronic devices and systems in terms of giant deformability,high ionic conductivity,excellent optical transmittance,and compatibility with various elastomers.

为了减轻在光伏板表面缺陷检测中由图像噪声和目标过小造成的影响,提出了改进Dn-YOLOv7(de-noising you only look once version 7)算法。该算法结合去噪卷积神经网络(de-noising convolutional neural network,DnCNN),提出了一个降噪模块(de-noise block,DnBlock),该模块使用了噪声容限更大的损失函数,并利用空间坐标卷积(coordinates convolution,CoordConv)对噪声通道进行卷积整合,增强了网络的降噪能力。同时,使用归一化高斯瓦瑟斯坦距离(normalized Gaussian Wasserstein distance,NWD)替代传统交并比(intersection over union,IoU)损失函数,提高了模型对小目标的检测能力。结果表明,改进后的模型具有降噪能力并提高了检测精确率,在无噪声水平下平均精确率均值达到了96.6%,在较强的高斯噪声和脉冲噪声下平均精确率均值分别达到91.4%和85.4%,检测速度达到78.0帧/s。该算法在航拍图像的光伏板缺陷检测中有一定实用价值。

Surface wettability plays a significant role in reducing solid–liquid frictional resistance,especially the superhydrophilic/hydrophilic interface because of its excellent thermodynamic stability.In this work,poly(acrylic acid)-poly(acrylamide)(PAA–PAM)hydrogel coatings with different thicknesses were prepared in situ by polydopamine(PDA)-UV assisted surface catalytically initiated radical polymerization.Fluid drag reduction performance of hydrogel surface was measured using a rotational rheometer by the plate–plate mode.The experimental results showed that the average drag reduction of hydrogel surface could reach up to about 56%in Couette flow,which was mainly due to the interfacial polymerization phenomenon that enhanced the ability of hydration layer to delay the momentum dissipation between fluid layers and the diffusion behavior of surface.The proposed drag reduction mechanism of hydrogel surface was expected to shed new light on hydrogel–liquid interface interaction and provide a new way for the development of steady-state drag reduction methods.

Exercise-with-melatonin therapy improves sleep disorder and motor dysfunction in a rat model of ischemic stroke

Exercise-with-melatonin therapy has complementary and synergistic effects on spinal cord injury and Alzheimer’s disease, but its effect on stroke is still poorly understood. In this study, we established a rat model of ischemic stroke by occluding the middle cerebral artery for 60 minutes. We treated the rats with exercise and melatonin therapy for 7 consecutive days. Results showed that exercise-with-melatonin therapy significantly prolonged sleep duration in the model rats, increased delta power values, and regularized delta power rhythm. Additionally, exercise-with-melatonin therapy improved coordination, endurance, and grip strength, as well as learning and memory abilities. At the same time, it led to higher hippocampal CA1 neuron activity and postsynaptic density thickness and lower expression of glutamate receptor 2 than did exercise or melatonin therapy alone. These findings suggest that exercise-withmelatonin therapy can alleviate sleep disorder and motor dysfunction by increasing glutamate receptor 2 protein expression and regulating hippocampal CA1 synaptic plasticity.

The short-term photovoltaic power generation forecasting is of great significance for the power system and energy management system(EMS). In this paper, the short-term forecasting model of PV generation power based on the RBF neural network is proposed, which forecast the power of PV generation system for the next 24 hours. Factors of position, environment, and inner performance of the system are fully considered. A novel prediction strategy combined with mechanism model is used, and modulations of parameters are executed according to online training of neural network. Experimental results prove that the proposed model reduces the deviation between the predict power and the actual power significantly, and can achieve fast and accurate prediction even the amount of number is very small.

Strong metal-support interaction between AuPd nanoparticles and oxygen-rich defect ZrO_(2) for enhanced catalytic 5-hydroxymethylfurfural oxidation

The unique properties of metal oxide surfaces,crystal surfaces and defects play vital roles in biomass upgrading reactions.In this work,hierarchical porous bowl-shaped ZrO_(2)(HB-ZrO_(2))with mixed crystal phase was designed and employed as the support for loading AuPd bimetal with different proportions to synthesize AuPd/HB-ZrO_(2) catalysts.The effects of surface chemistry,oxygen defects,bimetal interaction and metal-support interaction of AuPd/HB-ZrO_(2) on catalytic performance for the selective oxidation of 5-hydroxymethylfurfural(HMF)to 2,5-furandicarboxylic acid(FDCA)were systematically investigated.The Au 2 Pd1/HB-ZrO_(2) catalyst afforded a satisfactory FDCA yield of 99.9%from HMF oxidation using O_(2) as the oxidant in water,accompanied with an excellent FDCA productivity at 97.6 mmol g^(−1) h^(−1).This work offers fresh insights into rationally designing efficient catalysts with oxygen-rich defects for the catalytic upgrading of biomass platform chemicals.
