Glycogen metabolism-mediated intercellular communication in the tumor microenvironment influences liver cancer prognosis

Glycogen metabolism plays a key role in the development of hepatoellular carcinoma(HCC),but the function of glycogen metabolism genes in the tumor microenvironment(TME)is still to be elucidated.Single cell RNA-seq data were obtained from ten HCC tumor samples totaling 64,545 cells and 65 glycogen metabolism genes were analyzed bya nonnegative matrix factorization(NMF).The prognosis and immune response of new glycogen TME cell dusters were predicted by using HCC and immunotherapy cohorts from public databases.HOC single cell analysis was divided into fibroblasts,NT T cells,macrophages,endothelial clls,and B cells,which were separately divided into new cell clusters by glycogen metabolism gene annotation.Pseudo temporal trajectory analysis demonstrated the temporal differentiation trajectory of different glycogen subtype cell dusters.Cellular communication analysis revealed extensive interactions between endothelial cells with glycogen metabolizing TME cell.related subtypes and diferent glycogen subtype cell clusters.SCENIC analysis of transcription factors upstream of TME cell clusters with different glycogen metabolism.In addition,TME cell dusters of glycogen metabolism were found to be enriched in expression in CAF subtypes,CD8 depleted,M1,and M2 types.Bulk seq analysis showed the prognostic signifcance of glycogen metabolism.mediated TME cell dusters in HCC,while a significant immune response was found in the immunotherapy cohort in patients treated with immune checkpoint blockade(ICB),especially for CAFs,T cells,and macrophages In summary,our study reveals for the first time that glycogen metabolism mediates intercellular communication in the hepatocellular carcinoma microenvironment while elucidating the anti-tumor mechanisms and immune prognostic responses of different subtypes of cell dusters.

Particle size dependence of the microsegregation and microstructure in the atomized Ni-based superalloy powders:Theoretical and experimental study

The microstructure and microsegregation of atomized powder,which depend on their sizes,are of great importance to the mechanical properties of the consolidated bulk materials.Therefore,it is necessary to reveal the relationship between particle size and powder attributes.The effects of particle size on the so-lidification characterization of the atomized Ni-based superalloy powders were studied via finite element simulation.Based on the simulations,a model was developed to predict the microsegregation and mi-crostructure of atomized powders with different sizes and study the influence of thermal history on the powder attributes during the atomization processes.The radiation heat transfer and temperature gradi-ent within the rapid solidification alloy powders were taken into account in this model.For validating the accuracy of the model,the predictions of the present model were compared with the microsegregation and microstructure of the specific size powder close to the screen mesh size.The results showed that mi-crostructure depended primarily on the temperature gradient within the powder,while the solidification rate had a more significant effect on the microsegregation.The model predicted microstructure features in agreement with the experiment,and for microsegregation,the deviations of prediction for most ele-ments were less than 10%.This work provides a new model to precisely predict the microsegregation and microstructure of the atomized alloy powders and sets a foundation to control the powder features for various engineering applications.

ATSSC:An Attack Tolerant System in Serverless Computing

Serverless computing is a promising paradigm in cloud computing that greatly simplifies cloud programming.With serverless computing,developers only provide function code to serverless platform,and these functions are invoked by its driven events.Nonetheless,security threats in serverless computing such as vulnerability-based security threats have become the pain point hindering its wide adoption.The ideas in proactive defense such as redundancy,diversity and dynamic provide promising approaches to protect against cyberattacks.However,these security technologies are mostly applied to serverless platform based on“stacked”mode,as they are designed independent with serverless computing.The lack of security consideration in the initial design makes it especially challenging to achieve the all life cycle protection for serverless application with limited cost.In this paper,we present ATSSC,a proactive defense enabled attack tolerant serverless platform.ATSSC integrates the characteristic of redundancy,diversity and dynamic into serverless seamless to achieve high-level security and efficiency.Specifically,ATSSC constructs multiple diverse function replicas to process the driven events and performs cross-validation to verify the results.In order to create diverse function replicas,both software diversity and environment diversity are adopted.Furthermore,a dynamic function refresh strategy is proposed to keep the clean state of serverless functions.We implement ATSSC based on Kubernetes and Knative.Analysis and experimental results demonstrate that ATSSC can effectively protect serverless computing against cyberattacks with acceptable costs.

Preparation of CIP@TiO_(2) composite with broadband electromagnetic wave absorption properties

Scholars aim for the improved impedance matching (Z) of materials while maintaining their excellent wave absorption properties. Based on the hydrolysis characteristics of isopropyl titanate, a simple preparation process for the coating of carbonyl iron powder(CIP) with TiO_(2) was designed. Given the TiO2coating, the Z of the CIP@TiO_(2) composite was adjusted well by decreasing the dielectric constant. Moreover, the interfacial polarization of CIP@TiO_(2) was enhanced. Ultimately, the electromagnetic-wave (EMW) absorption property of the CIP@TiO_(2)composite was improved substantially, the minimum reflection loss reached-46.07 dB, and the effective absorption bandwidth can reach 8 GHz at the composite thickness of 1.5 mm. Moreover, compared with CIP, the oxidation resistance of CIP@TiO_(2)showed remarkable improvement. The results revealed that the oxidation starting temperature of CIP@TiO_(2) as about 400℃,whereas the uncoated CIP had an oxidation starting temperature of approximately 250℃. Moreover, the largest oxidation rate temperature of CIP@TiO_(2) increased to around 550℃. This work opens up a novel strategy for the production of high-performance EMW absorbers via structural design.

Molecular simulation study on the evolution process of hydrate residual structures into hydrate

The clathrate hydrate memory effect is a fascinating phenomenon with potential applications in carbon capture,utilization and storage(CCUS),gas separation,and gas storage as it can accelerate the secondary formation of clathrate hydrate.However,the underlying mechanism of this effect remains unclear.To gain a better understanding of the mechanism,we conducted molecular dynamic simulations to simulate the initial formation and reformation processes of methane hydrate.In this work,we showed the evolution process of hydrate residual structures into hydrate cages.The simulation results indicate that the residual structures are closely related to the existence of hydrate memory effect,and the higher the contribution of hydrate dissociated water to the hydrate nucleation process,the faster the hydrate nucleation.After hydrate dissociation,the locally ordered structures still exist after hydrate dissociation and can promote the formation of cluster structures,thus accelerating hydrate nucleation.Additionally,the nucleation process of hydrate and the formation process of clusters are inseparable.The size of clusters composed of cup-cage structures is critical for hydrate nucleation.The residence time at high temperature after hydrate decomposition will affect the strength of the hydrate memory effect.Our simulation results provide microscopic insights into the occurrence of the hydrate memory effect and shed light on the hydrate reformation process at the molecular scale.

Development of MoS_(2)-stainless steel catalyst by 3D printing for efficient destruction of organics via peroxymonosulfate activation

Herein,a novel MoS_(2)-stainless steel composite material was first synthetized via a 3D printing method(3DP MoS_(2)-SS)for peroxymonosulfate(PMS)activation and organics degradation.Compared with MoS_(2)-SS powder/PMS system(0.37 g/(m^(2)/min)),4.3-fold higher k_(FLO)/S_(BET)value was obtained in 3DP MoS_(2)-SS/PMS system(1.60 g/(m^(2)/min),resulting from the superior utilization of active sites.We observed that 3DP MoS_(2)-SS significantly outperformed the 3DP SS due to the enhanced electron transfer rate and increased active sites.Moreover,Mo^(4+)facilitated the Fe^(2+)/Fe^(3+)cycle,resulting in the rapid degradation of florfenicol(FLO).Quenching experiments and electron paramagnetic resonance spectra indicated that·OH,SO_(4)·^(-),O_(2)·^(-)and^(1)O_(2)were involved in the degradation of FLO.The effect of influencing factors on the degradation of FLO were evaluated,and the optimized degradation efficiency of 98.69%was achieved at 1 mM PMS and pH of 3.0.Six degradation products were detected by UPLC/MS analyses and several possible degradation pathways were proposed to be the cleavage of C-N bonds,dechlorination,hydrolysis,defluorination and hydroxylation.In addition,3DP MoS_(2)-SS/PMS system also demonstrated superior degradation performance for 2-chlorophenol,acetaminophen,ibuprofen and carbamazepine.This study provided deep insights into the MoS_(2)-SS catalyst prepared by 3DP technology for PMS activation and FLO-polluted water treatment.

Facile fabrication of graphene/g-C_(3)N_(4) for electromagnetic wave absorption

With the development of the miniaturization of electronic equipment and lightweight weapon equipment,there are new requirements for electromagnetic wave absorption material(EMWAM).EMWAM has outstanding electromagnetic wave absorption properties and lightweight characteristics become an important direction of research.In this study,graphene/g-C_(3)N_(4)(GGCN)EMWAM was first synthesized in situ by simple heat treatment,in which the g-C_(3)N_(4) had a porous structure and dispersed on the surface of graphene.The impedance matching of the GGCN was well adjusted by decreasing the dielectric constant and attenuation constant due to the g-C_(3)N_(4) semiconductor property and the graphite-like structure.The EMW loss mechanism of GGCN was also analyzed by simulating GGCN’s electric field mode distribution and resistance loss power density.The analysis result shows that the distribution of g-C_(3)N_(4) among GGCN sheets can produce more polarization effects and relaxation effects by increasing the lamellar spacing.Furthermore,the polarization loss of GGCN could be increased successfully by porous g-C_(3)N_(4).Ultimately,the EMW absorption property of GGCN is optimized significantly,and GGCN exhibits excellent EMW absorption performance.When the thickness is 2 mm,the effective absorption bandwidth(EAB)can reach 4.6 GHz,and when the thickness is 4.5 mm,the minimum reflection loss(RLmin)at 4.56 GHz can reach-34.69 dB.Moreover,the practical application of EMWAM was studied by radar cross-section(RCS)simulation,showing that GGCN has a good application prospect.

Real-time Abnormal Detection of GWAC Light Curve based on Wavelet Transform Combined with GRU-Attention

Nowadays, astronomy has entered the era of Time-Domain Astronomy, and the study of the time-varying light curves of various types of objects is of great significance in revealing the physical properties and evolutionary history of celestial bodies. The Ground-based Wide Angle Cameras telescope, on which this paper is based, has observed more than 10 million light curves, and the detection of anomalies in the light curves can be used to rapidly detect transient rare phenomena such as microgravity lensing events from the massive data. However, the traditional statistically based anomaly detection methods cannot realize the fast processing of massive data. In this paper, we propose a Discrete Wavelet(DW)-Gate Recurrent Unit-Attention(GRU-Attention) light curve warning model. Wavelet transform has good effect on data noise reduction processing and feature extraction, which can provide richer and more stable input features for a neural network, and the neural network can provide more flexible and powerful output model for wavelet transform. Comparison experiments show an average improvement of 61% compared to the previous pure long-short-term memory unit(LSTM) model, and an average improvement of 53.5% compared to the previous GRU model. The efficiency and accuracy of anomaly detection in previous paper work are not good enough, the method proposed in this paper possesses higher efficiency and accuracy,which incorporates the Attention mechanism to find out the key parts of the light curve that determine the anomalies. These parts are assigned higher weights, and in the actual anomaly detection, the star is detected with83.35% anomalies on average, and the DW-GRU-Attention model is compared with the DW-LSTM model, and the detection result f1 is improved by 5.75% on average, while having less training time, thus providing valuable information and guidance for astronomical observation and research.

Discriminative feature encoding for intrinsic image decomposition

Intrinsic image decomposition is an important and long-standing computer vision problem.Given an input image,recovering the physical scene properties is ill-posed.Several physically motivated priors have been used to restrict the solution space of the optimization problem for intrinsic image decomposition.This work takes advantage of deep learning,and shows that it can solve this challenging computer vision problem with high efficiency.The focus lies in the feature encoding phase to extract discriminative features for different intrinsic layers from an input image.To achieve this goal,we explore the distinctive characteristics of different intrinsic components in the high-dimensional feature embedding space.We define feature distribution divergence to efficiently separate the feature vectors of different intrinsic components.The feature distributions are also constrained to fit the real ones through a feature distribution consistency.In addition,a data refinement approach is provided to remove data inconsistency from the Sintel dataset,making it more suitable for intrinsic image decomposition.Our method is also extended to intrinsic video decomposition based on pixel-wise correspondences between adjacent frames.Experimental results indicate that our proposed network structure can outperform the existing state-of-the-art.
