Aberrant expression of nuclear matrix proteins during HMBA-induced differentiation of gastric cancer cells

AIM: To investigate the aberrant expression of nuclear matrix proteins in human gastric cancer cells before and after hexamethylene bisacetamide (HMBA) ***: Proteomics analysis of differential nuclear matrix proteins was performed by two dimensional electrophoresis polyacrylamide gel electrophoresis and matrix-assisted laser desorption/ionization time-of-flight mass *** expression levels of three nuclear matrix proteins were further confirmed by Western blotting and their locations in nuclear matrix filament were observed by quantum dots-based ***: Proteomics analysis showed that 43 protein spots were significantly changed due to HMBA *** proteins were identified in the HMBAinduced differentiation of gastric tumor *** proteins spots were down-regulated while seven were *** these proteins,prohibitin,nucleophosmin and hnRNP A2/B1 were significantly decreased in HMBA-treated human gastric cancer cells,and their locations in nuclear matrix were altered by *** results proved the alteration of specific nuclear matrix proteins during the differentiation of human gastric cancer *** the aberrant expressions of nuclear matrix proteins were of significance in revealing the regulatory mechanism of tumor cell proliferation and ***: The aberrant expressions and intracellular redistributions of nuclear matrix proteins before and after HMBA treatment indicated that nuclear matrix proteins play a pivotal role in the differentiation of gastric cancer cells.

Rice is one of the most important cereal crops in the world, and a substantial increase in grain yield is necessary for food security. However, high yields of semidwarf modern rice varieties are heavily dependent on the application of mineral nitrogenous fertilizer (Tilman et al., 2002;Sun et al., 2014). Nitrogen (N)-insensitive sponses associated with reduced N-use efficiency (NUE) is a major characteristic of the green revolution varieties (GRVs), in which the growth-inhibiting protein SLENDER RICE1 (SLR1) accumulates (Li et al., 2018). Unfortunately, increasing the level of N fertilizer use to reach the full yield potential of GRVs is subject to diminishing returns, quite apart from its deleterious effect on the environments (Rahn et al., 2009;Liu et al., 2015). Therefore, there is an urgent need to develop new rice GRVs that increase NUE while maintaining their high yields. Recently, several genes (e.g., DEP1, ***, OsNRT2.3b, ARE1 and GRF4) responsible for improved NUE have been identified in rice (Sun et al.. 2014;Hu et al., 2015;Fan et al., 2016;wang et al., 2018;Li et al., 2018). More importantly, boosting the activity of the transcription factor GRF4 overcomes the ability of SLR1 to prevent the GRF4-GIF1 interaction, which in turn promotes the coordinated expression of the genes involved in N assimilation and carbon fixation and consequently enhances the NUE of rice GRVs, thereby improving our ability to grow crops sustainably (Li et al., 2018). However, current understanding of the genetic basis for improving NUE remains at the level of identification of a number of quantitative trait loci (QTLs), without any understanding of the nature of the gene products.

Ankylosing spondylitis(AS)is chronic inflammatory arthritis with a progressive fusion of axial ***-inflammatory treatments such as anti-TNF-αantibody therapy suppress inflammation but do not effectively halt the progression of spine fusion in AS *** we report that the autoimmune inflammation of AS generates a microenvironment that promotes chondrogenesis in spine ligaments as the process of spine *** differentiation was observed in the ligaments of patients with earlystage AS,and cartilage formation was followed by ***,a large number of giant osteoclasts were found in the inflammatory environment of ligaments and on bony surfaces of calcified *** activity by these giant osteoclasts generated marrow with high levels of active TGF-β,which induced new bone formation in the ***,no Osterix+osteoprogenitors were found in osteoclast resorption areas,indicating uncoupled bone resorption and *** at the late and maturation stages,the uncoupled osteoclast resorption in bony interspinous ligament activates TGF-βto induce the progression of ossification in AS *** resorption of calcified cartilage-initiated ossification in the progression of AS is a similar pathologic process of acquired heterotopic ossification(HO).Our finding of cartilage formation in the ligaments of AS patients revealed that the pathogenesis of spinal fusion is a process of HO and explained why anti-inflammatory treatments do not slow ankylosing once there is new bone formation in spinal soft ***,inhibition of HO formation,such as osteoclast activity,cartilage formation,or TGF-βactivity could be a potential therapy for AS.

Establishment of a Highly Efficient Regeneration System for the Mature Embryo Culture of Wheat

Establishment of a highly efficient regeneration system for the mature embryo of wheat will provide a convenient tool for wheat tissue culture and transformation, thereby facilitating the transformation of foreign genes into wheat. By using the mature embryos derived from 20 different wheat lines including Shi 4185, Yumai 66, Lunxuan 987, CB037, Yangmai 6, Xinchun 9, Bobwhite, Han 6172, Zheng 9023, Jimai 20, Ningchun 4, and Jing 411, the effects of some factors including inoculation methods, initiating culture media, organic additives, antioxidants, and auxins on the regeneration from the explants were evaluated. The results indicated that the scraping embryo culture was better than the whole embryo culture, the Aa medium was better than the SD2 medium and dicamba was better than 2,4-D in increasing the regeneration frequency. An Adi medium was established in this study by adding silver nitrate, cysteine, ascorbic acid, dicamba, glutamine into the Aa medium at the concentration of 4,40, 100, 2, and 5 mg L^-1, respectively. By using the Adi medium and the scraping technique, the regeneration frequencies of the mature embryos of CB037, Lunxuan 987, Hart 6172, Yangmai 6, Bobwhite, Zheng 9023, Shi 4 185, and Jimai 20 became 85.6, 60,1, 46.0, 42.1,42.0, 34.0, 33.0, and 32.0%, respectively, which were about 5-8 times higher than that obtained from the conventional culture mediums and techniques. This novel regeneration system could be helpful in wheat transformation.

Hepatic fibrosis is a common feature of almost all chronic liver diseases. Formation of new vessels （angiogenesis） is a process strictly related to the progressive fibrogenesis which leads to cirrhosis and liver cancer. This review mainly concerns the relationship between angiogenesis and hepatic fibrosis, by considering the mechanism of angiogenesis, cells in angiogenesis, anti-angiogenic and Chinese medicine therapies.

Potential hepatic and renal toxicity induced by the biflavonoids from Ginkgo biloba

Evidence continues to grow on potential health risks associated with Ginkgo biloba and its *** biflavonoid is a subclass of the flavonoid family in Ginkgo biloba with a plenty of pharmacological properties,the potential toxicological effects of biflavonoids remains largely ***,the aim of this study was to investigate the in vitro and in vivo toxicological effects of the biflavonoids from Ginkgo biloba(i.e.,amentoflavone,sciadopitysin,ginkgetin,isoginkgetin,and bilobetin).In the in vitro cytotoxicity test,the five biflavonoids all reduced cell viability in a dose-dependent manner in human renal tubular epithelial cells(HK-2)and human normal hepatocytes(L-02),indicating they might have potential liver and kidney *** the in vivo experiments,after intragastrical administration of these biflavonoids at 20 mg·kg^–1·d^–1 for 7 days,serum biochemical analysis and histopathological examinations were *** activity of alkaline phosphatase was significantly increased after all the bifl avonoid administrations and widespread hydropic degeneration of hepatocytes was observed in ginkgetin or b ilobetin-treated ***,the five biflavonoids all induced acute kidney injury in treated mice and the main pathological lesions were confirmed to the tubule,glomeruli,and interstitium *** the in vitro and in vivo results suggested that these biflavonoids may be more toxic to the kidney than the liver,we further detected the mechanism of biflavonoids-induced *** increased TUNEL-positive cells were detected in kidney tissues of biflavonoids-treated mice,accompanied by elevated expression of proapoptotic protein BAX and unchanged levels of antiapoptotic protein BCL-2,indicating apoptosis was involved in biflavonoids-induced *** together,our results suggested that the five biflavonoids from Ginkgo biloba may have potential hepatic and renal toxicity and more attentions should be paid to ensure Ginkgo biloba p

Suppose that there are two populations x and y with missing data on both of them, where x has a distribution function F（·） which is unknown and y has a distribution function Gθ（·） with a probability density function gθ（·） with known form depending on some unknown parameter θ. Fractional imputation is used to fill in missing data. The asymptotic distributions of the semi-empirical likelihood ration statistic are obtained under some mild conditions. Then, empirical likelihood confidence intervals on the differences of x and y are constructed.

Ferroptosis is an iron-dependent form of cell death characterized by an accumulation of lipid peroxides.A growing body of recent evidence supports the notion that ferroptosis plays an important role in mediating a wide variety of cellular processes in ***,ferroptosis can play a significant role in mediating various functions in immune cells and ***,we discuss our current understanding regarding the regulatory role of ferroptosis in immune cells,including T cells,B cells,granulocytes,monocytes,and *** addition,we discuss the general effect of immune cell ferroptosis on human pathophysiology and immunotherapies,thereby suggesting new strategies for targeting ferroptosis in order to modulate the immune system and unravel the mechanisms that underlie ferroptosis in the immune response。

Theoretical derivation of the crystallographic parameters of polytypes of long-period stacking ordered structures with the period of 13 and 14 in hexagonal close-packed system

Based on crystallographic theory, there are 63 kinds of polytypes of 13H long-period stacking order （LPSO） structure, 126 kinds of polytypes of 14H LPSO structure, 120 kinds of polytypes of 39R LPSO structure, and 223 kinds of polytypes of 42R LPSO structure in a hexagonal close-packed （HCP） system, and their stacking sequences and space groups have been derived in detail. The result provides a theoretical explanation for the various polytypes of the LPSO structure.
