Background:Ziwan and Taoren(ZT)is a classic medicine pair in the formula of Mai Dong Di Shao Decoction,has been used to treat primary Sjogren’s syndrome(pSS)for more than 20 *** its action mechanism is still *** study is aimed to reveal the potential mechanism of ZT treated pSS and discover its active compounds of ZT and therapeutic target for ***:Firstly,the potential pathways of ZT for pSS treatment were predicted through network pharmacology and GO and KEGG enrichment ***,the inter-structural relationships between active compounds of ZT and target proteins were visualized using molecular docking ***,efficacy and mechanism were conducted through in vivo experiments,such as water intake,spleen index,hematoxylin-eosin staining pathological changes,ELISA,Western Blot analysis,and immunofluorescence ***:Nine active compounds were extracted from network pharmacology,including quercitrin,luteolin,kaempferol,β-sitosterol,isorhamnetin,galangin,hederagenin,diosmetin and gibberellin *** disease targets were identified:RELA,TP53,AKT1,interleukin(IL)6,MAPK1,ESR1,IL10;with RELA being the most core *** and GO enrichment analysis indicated that ZT may act through the TLR/NF-κB/RELA inflammatory mechanism *** results of molecular docking showed that ZT’s active compounds bind well to the RELA(p65)*** vivo results demonstrated that a high dose of ZT significantly improved water intake and reduced lymphocytes infiltration in submandibular gland pathology in NOD *** expression content of AQP5 and vasoactive intestinal peptide in the submaxillary gland was significantly increased,while levels of inflammatory factors such as tumor necrosis factor-α,IL-6,and IL-1βalong with protein expressions including toll-like receptor4,p-p65 and p-IKKα/βin NF-κB pathway were ***:The ZT treatment exhibits a promising efficacy in mitigating dryness symptoms of pSS,potentially attr

Global Protein Expression Analysis of Molecular Markers of DS-1-47,a Component of Implantation-promoting Traditional Chinese Medicine

This study investigated the molecular markers of DS-1-47,a component of an implantation-promoting traditional Chinese medicine consisting of Astragalus mongholicus,Atractylodes macrocephala,Scutellaria baicalensis and Dipsacales,in an attempt to clarify the molecular mechanism and action targets of *** ovarian stimulation（COS） method was used to establish the implantation dysfunction models of *** were divided into normal pregnant group,COS model group and DS-1-47 *** capture microdissection-double dimensional electrophoresis-mass spectrum（LCM-DE-MS） was used to analyze the uterine protein molecules that were possibly involved in the promotion of ***-three proteins in DS-1-47 group were significantly changed as compared to those in COS model group,with 7 proteins down-regulated and 16 proteins *** for some constituent proteins,the down-regulated proteins included collagen α-1（Ⅵ） chain,keratin 7,keratin 14,myosin regulatory light chain 12 B,myosin light polypeptide 9,heat shock protein β-7,and C-U-editing enzyme APOBEC-2;the up-regulated proteins included apolipoprotein A-I,calcium regulated protein-3,proliferating cell nuclear antigen,L-xylulose reductase,and calcium binding *** 23 proteins that were regulated by DS-1-47 represented a broad diversity of molecule *** down-regulated proteins were associated with stress and immune response,and those up-regulated proteins were related to *** was suggested that these proteins were important in regulating the uterine environment for the blastocyst *** identification of DS-1-47 markers,proteomic analysis coupled with functional assays is demonstrated to be a promising approach to better understand the molecular mechanism of traditional Chinese medicine.

Molecular cloning,characterization and promoter analysis of LbgCWIN1 and its expression profiles in response to exogenous sucrose during in vitro bulblet initiation in lily

Lily(Lilium spp.) is an important ornamental flower, which is mainly propagated by bulbs. Cell wall invertases(CWINs), which catalyze the irreversibly conversion of sucrose into glucose and fructose in the extracellular space, are key enzymes participating in sucrose allocation in higher plants. Previous studies have shown that CWINs play an essential role in bulblet initiation process in bulbous crops, but the underlying molecular mechanism remains unclear. Here, a CWIN gene of Lilium brownii var. giganteum(Lbg) was identified and amplified from genomic DNA. Quantitative RT-PCR assays revealed that the expression level of LbgCWIN1 was highly upregulated exactly when the endogenous starch degraded in non-sucrose medium during in vitro bulblet initiation in Lbg. Phylogenetic relationship, motif, and domain analysis of LbgCWIN1 protein and CWINs in other plant species showed that all sequences of these CWIN proteins were highly conserved. The promoter sequence of LbgCWIN1 possessed a number of alpha-amylase-, phytohormone-, light-and stress-responsive cis-elements. Meanwhile, β-glucuronidase(GUS) assay showed that the 459 bp upstream fragment from the translational start site displayed maximal promoter activity. These results revealed that LbgCWIN1 might function in the process of in vitro bulblet initiation and be in the response to degradation of endogenous starch.

Insufficient TRPM5 Mediates Lipotoxicity-induced Pancreaticβ-cell Dysfunction

Objective:While the reduction of transient receptor potential channel subfamily M member 5(TRPM5)has been reported in islet cells from type 2 diabetic(T2D)mouse models,its role in lipotoxicity-induced pancreaticβ-cell dysfunction remains *** study aims to study its ***:Pancreas slices were prepared from mice subjected to a high-fat-diet(HFD)at different time points,and TRPM5 expression in the pancreaticβcells was examined using immunofluorescence ***-stimulated insulin secretion(GSIS)defects caused by lipotoxicity were mimicked by saturated fatty acid palmitate(Palm).Primary mouse islets and mouse insulinoma MIN6 cells were treated with Palm,and the TRPM5 expression was detected using qRT-PCR and Western ***-induced GSIS defects were measured following siRNA-based Trpm5 *** detrimental effects of Palm on primary mouse islets were also assessed after overexpressing Trpm5 via an adenovirus-derived Trpm5(Ad-Trpm5).Results:HFD feeding decreased the mRNA levels and protein expression of TRPM5 in mouse pancreatic *** reduced TRPM5 protein expression in a time-and dose-dependent manner in MIN6 *** also inhibited TRPM5 expression in primary mouse *** of Trpm5 inhibited insulin secretion upon high glucose stimulation but had little effect on insulin *** of Trpm5 reversed Palm-induced GSIS defects and the production of functional maturation molecules unique toβ***:Our findings suggest that lipotoxicity inhibits TRPM5 expression in pancreaticβcells both in vivo and in vitro and,in turn,drivesβ-cell dysfunction.

Effect of the vortical structures on the hydrodynamic performance of a pitching hydrofoil

The objective is to study the vortical structural behaviors of a transient pitching hydrofoil and their effects on the hydrodynamic performance. The pitching motion of the hydrofoil is set to pitch up with an almost constant rate from 5° to 15° and then back to 5°, with the Reynolds number 4.4×10^(5) and the frequency 2 Hz. The results show that the main coherent structures around the pitching hydrofoil include small-scale laminar separation bubble (LSB), large-scale second vortex (SV) and trailing edge vortex (TEV) which are all vortical. The relationship between the vortical structure and the lift is investigated with the finite-domain impulse theory. It indicates that the major part of the lift is contributed by the LSB, whereas the shedding and the formation of the SV and TEV cause the fluctuation of the lift. The proper orthogonal decomposition (POD) method is applied to capture the most energetic modes, revealing that the LSB mode occupies a large amount of energy in the flow field. The dynamic mode decomposition (DMD) method accurately extracts the dominant frequency and modal characteristics, with the first mode corresponding to the mean flow, the second mode corresponding to the LSB structure and the third and fourth modes corresponding to the vortex shedding.

Differences in insomnia-related self-reported outcomes among elderly hospitalized patients
