Elevated DLL3 in stomach cancer by tumor-associated macrophages enhances cancer-cell proliferation and cytokine secretion of macrophages

Background The notch signal pathway is important in the development of both tumor-associated macrophages(TAMs)and stomach cancer,but how Notch signaling affects TAMs in stomach cancer is barely understood.Methods The expressions of Notch1,Notch2,Notch3,Notch4,hes family bHLH transcription factor 1(Hes1),and delta-like canonical Notch ligand 3(DLL3)were detected by Western blot and the expressions of interleukin(IL)-10,IL-12,and IL1-b were detected using enzyme-linked immunosorbent assay after the co-culture of macrophages and stomach-cancer cells.The proliferation and migration of cancer cells were detected using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay and scratch assay,respectively,and the cell cycle was detected using Annexin V/propidium iodide assay.The protein interactions with DLL3 were detected using co-immunoprecipitation and mass spectrometry.Results The co-culture of macrophages and stomach-cancer cells MKN45 and BGC823 could enhance cell proliferation accompanied by the activation of Notch1/Notch2 signaling and upregulation of DLL3.Notch signaling gamma-secretase inhibitor(DAPT)blocked this process.The overexpression of DLL3 in stomach-cancer cells could promote the proliferation of cancer cells,enhance the activation of Notch1/Notch2 signaling,induce the expression of IL-33,lead to the degradation of galectin-3–binding protein(LG3BP)and heat shock cognate 71 kDa protein(HSPA8),and result in elevated IL-1b,IL-12,and IL-10 secretion by macrophages.Higher expression of DLL3 or IL-33 could lead to a lower survival rate based on University of California,Santa Cruz Xena Functional Genomics Explorer and The Cancer Genome Atlas data set.Conclusions This is evidence that DLL3 regulates macrophages in stomach cancer,suggesting that DLL3 may be a novel and potential target for stomach-cancer therapy.

MicroRNAs(miRNAs),a class of non-coding single-strand RNA molecules encoded by endogenous genes,are about 21–24 nucleotides long and are involved in the post-transcriptional regulation of gene expression in plants and animals.Generally,the types and quantities of miRNAs in the different tissues of an organism are diverse,and these divergences may be related to their specific functions.Here we have identified 296 known miRNAs and 46 novel miRNAs in the antennae of the parasitoid wasp Microplitis mediator by high-throughput sequencing.Thirty-three miRNAs were predicted to target olfactory-associated genes,including odorant binding proteins(OBPs),chemosensory proteins,odorant receptors(ORs),ionotropic receptors(IRs)and gustatory receptors.Among these,17 miRNAs were significantly highly expressed in the antennae,four miRNAs were highly expressed both in the antennae and head or wings,while the remaining 12 miRNAs were mainly expressed in the head,thorax,abdomen,legs and wings.Notably,miR-9a-5p and miR-2525-3p were highly expressed in male antennae,whereas miR-1000-5p and novel-miR-13 were enriched in female antennae.The 17 miRNAs highly expressed in antennae are likely to be associated with olfaction,and were predicted to target one OBP(targeted by miR-3751-3p),one IR(targeted by miR-7-5p)and 14 ORs(targeted by 15 miRNAs including miR-6-3p,miR-9a-5p,miR-9b-5p,miR-29-5p,miR-71-5p,miR-275-3p,miR-1000-5p,miR-1000-3p,miR-2525-3p,miR-6012-3p,miR-9719-3p,novel-miR-10,novel-miR-13,novel-miR-14 and novel-miR-28).These candidate olfactory-associated miRNAs are all likely to be involved in chemoreception through the regulation of chemosensory gene expression in the antennae of M.mediator.

Secular trends and urban–rural disparities in the median age at menarche among Chinese han girls from 1985 to 2019

Background Menarche is a substantial milestone of female puberty.Timing of age at menarche is considered the key to understanding the potential linkages with women’s health outcomes later in life.This study aimed to explore the secular trends and urban‒rural disparities in the median age at menarche among Chinese Han girls from 1985 to 2019.Methods Data were extracted from the 1985,1995,2005,2014,and 2019 Chinese National Surveys on Students’Constitution and Health,which were nationally representative cross-sectional studies,and a total of 173,535 Han girls aged 9–18 years were examined.Girls were asked whether menarche had occurred.The median age at menarche was estimated by probit analysis.Z tests were used to compare the differences between survey years and between urban and rural areas.Results The median age at menarche among Chinese Han girls decreased from 13.37 years in 1985 to 12.00 years in 2019,and the overall decrease was more significant in rural areas(1.77 years)than in urban areas(0.99 years).The average five-year change in the decrease in the median age at menarche showed an accelerating and then slowing pace;and it was observed similarly in both urban and rural areas.The urban‒rural disparities shrank from 0.64 years in 1985 to 0.44 years in 1995,then to 0.27 years in 2005,0.24 years in 2014,and finally to−0.14 years in 2019.Conclusions The median age at menarche among Chinese Han girls continued to decline from 1985 to 2019 but at a slowing pace in the last five years.Urban‒rural disparities gradually narrowed.Sexual and reproductive health education and interventions to prevent the decline in the age of menarche are needed,especially in rural areas.

Sensory neuron membrane proteins(SNMPs),homologs of the human fatty acid transport protein CD36 family,are observed to play a significant role in chemoreception,especially in detecting sex pheromone in Drosophila and some lepidopteran species.In the current study,two full‐length SNMP transcripts,MmedSNMP1 and MmedSNMP2,were identified in the parasitoid Microplitis mediator(Hymenoptera:Braconidae).Quantitative real‐time polymerase chain reaction analysis showed that the expression of MmedSNMP1 was significantly higher in antennae than in other tissues of both sexes.In addition,the MmedSNMP1 transcript was increased dramatically in newly emerged adults and there were no significant differences between adults with or without mating and parasitic experiences.However,compared with MmedSNMP1,the expression of MmedSNMP2 was widely found in various tissues,significantly increased at half‐pigmented pupae stage and remained at a relatively constant level during the following developmental stages.It was found that MmedSNMP1 contained eight exons and seven introns,which was highly conserved compared with other insect species.In situ hybridization assay demonstrated that MmedSNMP1 transcript was distributed widely in antennal flagella.Among selected chemosensory genes(odorant binding protein,odorant receptor,and ionotropic receptor genes),MmedSNMP1 only partially overlapped with MmedORco in olfactory sensory neurons of antennae.Subsequent immunolocalization results further indicated that MmedSNMP1 was mainly expressed in sensilla placodea of antennae and possibly involved in perceiving plant volatiles and sex pheromones.These findings lay a foundation for further investigating the roles of SNMPs in the chemosensation of parasitoids.

Niemann-Pick type C2 （NPC2） is a type of small soluble protein involved in lipid metabolism and triglyceride accumulation in vertebrates and arthropods. Recent stud- ies have determined that NPC2 also participates in chemical communication of arthropods. In this work, two novel NPC2 proteins （MmedNPC2a and MmedNPC2b） in Microplitis mediator were identified. Real-time quantitative PCR （qPCR） analysis revealed that Mmed- NPC2a was expressed higher in the antennae than in other tissues of adult wasps compared with MmedNPC2b. Subsequent immunolocalization results demonstrated that NPC2a was located in the lymph cavities of sensilla placodea. To further explore the binding charac- terization of recombinant MmedNPC2a to 54 candidate odor molecules, a fluorescence binding assay was performed. It was found MmedNPC2a could not bind with selected fatty acids, such as linoleic acid, palmitic acid, stearic acid and octadecenoic acid. How- ever, seven cotton volatiles, 4-ethylbenzaldehyde, 3,4-dimethylbenzaldehyde, fl-ionone, linalool, m-xylene, benzaldehyde and trans-2-hexen-l-al showed certain binding abilities with MmedNPC2a. Moreover, the predicted 3D model of MmedNPC2a was composed of seven r-sheets and three pairs of disulfide bridges. In this model, the key binding residues for oleic acid in CjapNPC2 of Camponotus japonicus, Lue68, Lys69, Lys70, Phe97, Thr103 and Phe127, are replaced with Phe85, Ser86, His87, Leu113, Tyr119 and Ile143 in MmedNPC2a, respectively. We proposed that MmedNPC2a in M. mediator may play roles in perception of plant volatiles.

Robotic anterior resection of rectal cancer： technique and early outcome

Background The Da Vinci system is a newly developed device for colorectal surgery. With advanced stereoscopic vision, lack of tremor, and the ability to rotate the instruments surgeons find that robotic systems are ideal laparoscopic tools. Since conventional laparoscopic total mesorectal excision is a challenging procedure, we have sought to assess the utility of the Da Vinci robotic system in anterior resections for rectal cancer. Methods Between November 2010 and December 2011, a total of 22 patients affected by rectal cancer were operated on with robotic technique, using the Da Vinci robot. Data regarding the outcome and pathology reports were prospectively collected in a dedicated database. Results There were no conversions to open surgery and no postoperative mortality of any patient. Mean operative time was （220±46） minutes （range, 152-286 minutes）. The median number of lymph nodes harvested was （14.6±6.5）（range, 8-32）, and the circumferential margin was negative in all cases. The distal margin was （2.6±1.2） cm （range, 1.0-5.5 cm）. The mean length of hospital stay was （7.8±2.6） days （range, 7.0-13.0 days）. Macroscopic grading of the specimen was complete in 19 cases and nearly complete in three patients. Conclusions Robotic anterior resection for rectal surgery is safe and feasible in experienced hands. Outcome and pathology findings are comparable with those observed in open and laparoscopy procedures. This technique may facilitate minimally invasive radical rectal surgery.

Association between the Neutrophil-to-lymphocyte Ratio and New-onset Subclinical Macrovascular and Microvascular Diseases in the Chinese Population

Objective The association between neutrophil-to-lymphocyte ratio(NLR)with subclinical macrovascular and microvascular diseases has been less investigated.We sought to examine the association between NLR and new-onset subclinical macrovascular and microvascular abnormalities in the Chinese population.Methods From a community cohort,we included 6,430 adults aged≥40 years without subclinical macrovascular and microvascular diseases at baseline.We measured subclinical macrovascular and microvascular abnormalities separately using the ankle-brachial index(ABI),brachial-ankle pulse wave velocity(baPWV),and albuminuria.Results During a mean follow-up of 4.3 years,110 participants developed incident abnormal ABI,746 participants developed incident elevated baPWV,and 503 participants developed incident albuminuria.Poisson regression analysis indicated that NLR was significantly associated with an increased risk of newonset abnormal ABI,elevated baPWV,and albuminuria.Compared to overweight/obese participants,we found a much stronger association between NLR and subclinical vascular abnormalities in participants with normal weight.Furthermore,we found an interaction between the NLR and body mass index(BMI)on the risk of new-onset abnormal ABI(P for interaction:0.01).Conclusion NLR was associated with subclinical macrovascular and microvascular diseases in the Chinese population.Furthermore,in participants with normal weight,the association between NLR and subclinical vascular abnormalities was much stronger.

目的通过对直肠癌淋巴结跳跃转移的临床病理特点进行分析探讨直肠癌根治术高位血管结扎的意义。方法回顾性分析2009年10月至2010年10月行根治性直肠癌切除的207例患者资料，观察肠系膜下动脉周围及其根部淋巴结发生跳跃转移的情况，分析临床病理因素与跳跃转移间的相关性。结果207例直肠癌患者手术切除标本中共检出淋巴结2305枚，有转移的患者168例。跳跃转移与肿瘤的分化程度（X^2=113．65，P=0．037）及肿瘤浸润深度（X^2=108．22，P=0．042）相关，与性别、年龄、部位、大小、术前血清癌胚抗原水平、大体类型和组织类型因素均无明显相关性。结论术前对直肠癌肿瘤分化程度及浸润深度进行评估，有助于提示是否存在淋巴结跳跃转移；对于有跳跃转移风险的患者应行血管高位结扎淋巴结清扫术。
