Relationship of serum folic acid and vitamin B12 contents with maternal endothelial injury and placental ischemia hypoxia in patients with preeclampsia

Objective:To investigate the relationship of serum folic acid and vitamin B12 contents with maternal endothelial injury and placental ischemia hypoxia in patients with preeclampsia. Methods:A total of 76 puerperae with preeclampsia who gave birth in this hospital between March 2016 and October 2017 were selected as the preeclampsia group, and 100 healthy puerperae who gave birth in this hospital during the same period were selected as the normal control group. The differences in serum contents of folic acid, vitamin B12, endothelial injury indexes and oxidative stress indexes were compared between the two groups 24 h before delivery. Pearson test was used to assess the intrinsic relationship of serum folic acid and vitamin B12 contents with the disease severity in puerperae with preeclampsia.Results:Serum folic acid and vitamin B12 contents of preeclampsia group were lower than those of control group;endothelial injury markers sEng, sFlt-1, ET-1 and TM contents were higher than those of control group whereas ADM content was lower than that of control group;oxidative stress indexes AOPPs and LHP contents were higher than those of normal control group whereas T-AOC content was lower than that of control group. The Pearson test showed that serum folic acid and vitamin B12 contents in puerperae with preeclampsia were directly correlated with vascular endothelial injury and placental ischemia hypoxia.Conclusion: There is folic acid and vitamin B12 deficiency in patients with preeclampsia, and it can aggravate the vascular endothelial injury and placental ischemia hypoxia.

Changes of miR-26b expression in preeclampsia placenta and its downstream target genes

Objective:To study the changes of miR-26b expression in the preeclampsia placenta and its downstream target genes.Methods: Preeclampsia placenta tissue and healthy placenta tissue from puerperae who gave birth in People's Hospital of Songzi between August 2014 and February 2017 were collected, miRNA was extracted to determine miR-26b expression, and protein was extracted to detect he expression of cell invasion molecules MEST, LAMA4, uPA, HtrA4 and CTSB as well as angiogenesis molecules VEGF, ANG1, ANG2 and PLGF. Results: miR-26b expression in preeclampsia placenta tissue was significantly higher than that in healthy placenta tissue whereas MEST, LAMA4, uPA, HtrA4, CTSB, VEGF, ANG1, ANG2 and PLGF protein expression were significantly lower than those in healthy placenta tissue;MEST, LAMA4, uPA, HtrA4, CTSB, VEGF, ANG1, ANG2 and PLGF protein expression in preeclampsia placenta tissue with high miR-26b expression were significantly lower than those in preeclampsia placenta tissues with low miR-26 expression.Conclusion: miR-26b is highly expressed in the preeclampsia placenta tissue and the target genes are cell invasion molecules and angiogenesis molecules.

Kinesin 26B modulates M2 polarization of macrophage by activating cancer-associated fibroblasts to aggravate gastric cancer occurrence and metastasis

BACKGROUND The regulatory effects of KIF26B on gastric cancer(GC)have been confirmed,but the specific mechanism still needs further exploration.Pan-cancer analysis shows that the KIF26B expression is highly related to immune infiltration of cancerassociated fibroblasts(CAFs),and CAFs promote macrophage M2 polarization and affect cancers’progression.AIM To investigate the regulatory functions of KIF26B on immune and metastasis of GC.METHODS We analyzed genes’mRNA levels by quantitative real-time polymerase chain reaction.Expression levels of target proteins were detected by immunohistochemistry,ELISA,and Western blotting.We injected AGS cells into nude mice for the establishment of a xenograft tumor model and observed the occurrence and metastasis of GC.The degree of inflammatory infiltration in pulmonary nodes was observed through hematoxylin-eosin staining.Transwell and wound healing assays were performed for the evaluation of cell invasion and migration ability.Tube formation assay was used for detecting angiogenesis.M2-polarized macrophages were estimated by immunofluorescence and flow cytometry.RESULTS KIF26B was significantly overexpressed in cells and tissues of GC,and the higher expression of KIF26B was related to GC metastasis and prognosis.According to in vivo experiments,KIF26B promoted tumor formation and metastasis of GC.KIF26B expression was positively associated with CAFs’degree of infiltration.Moreover,CAFs could regulate M2-type polarization of macrophages,affecting GC cells’migration,angiogenesis,invasion,and epithelial-mesenchymal transition process.CONCLUSION KIF26B regulated M2 polarization of macrophage through activating CAFs,regulating the occurrence and metastasis of GC.

Spermatogenic cell heterogeneity is determined by the complex process of spermatogenesis differentiation.However,effectively revealing the regulatory mechanisms underlying mammalian spermatogenic cell development and differentiation via traditional methods is difficult.Advances in technology have led to the emergence of many single-cell transcriptome sequencing protocols,which have partially addressed these challenges.In this review,we detail the principles of 10x Genomics technology and summarize the methods for downstream analysis of single-cell transcriptome sequencing data.Furthermore,we explore the role of single-cell transcriptome sequencing in revealing the heterogeneity of testicular ecological niche cells,delineating the establishment and disruption of testicular immune homeostasis during human spermatogenesis,investigating abnormal spermatogenesis in humans,and,ultimately,elucidating the molecular evolution of mammalian spermatogenesis.

GNSS gyroscopes:determination of angular velocity and acceleration with very high-rate GNSS

Although global navigation satellite systems(GNSS)have been routinely applied to determine attitudes,there exists no literature on determining angular velocity and/or angular acceleration from GNSS.Motivated by the invention of computerized accelerometers of the correspondence author and following the success of accurately recovering translational velocity and acceleration waveforms from very high-rate GNSS precise positioning by Xu and his collaborators in 2021,we propose the concept of GNSS gyroscopes and reconstruct angular velocity and acceleration from very high-rate GNSS attitudes by applying regularization under the criterion of minimum mean squared errors.The major results from the experiments can be summarized in the following:(i)angular velocity and acceleration waveforms computed by applying the difference methods to high-rate GNSS attitudes are too noisy and can be physically not meaningful and numerically incorrect.The same can be said about inertial measurement unit(IMU)attitudes,if IMU gyros are not of very high accuracy;(ii)regularization is successfully applied to reconstruct the high-rate angular velocity and acceleration waveforms from 50 Hz GNSS attitudes and significantly outperforms the difference methods,validating the proposed concept of GNSS gyroscopes.By comparing the angular velocity and acceleration results by using the difference methods and regularization,we find that the peak values of angular velocity and acceleration by regularization are much smaller by a maximum factor of 1.57 in the angular velocity to a maximum factor of 8662.53 times in the angular acceleration in the case of high-rate GNSS,and by a maximum factor of 1.26 in the angular velocity to a maximum factor of 2819.85 times in the angular acceleration in the case of IMU,respectively;and(iii)the IMU attitudes apparently lead to better regularized angular velocity and acceleration waveforms than the high-rate GNSS attitudes,which can well be explained by the fact that the former is of better accuracy than the latter.As

Highly Porous Yet Transparent Mechanically Flexible Aerogels Realizing Solar-Thermal Regulatory Cooling

The demand for highly porous yet transparent aerogels with mechanical flexibility and solar-thermal dual-regulation for energy-saving windows is significant but challenging.Herein,a delaminated aerogel film(DAF)is fabricated through filtration-induced delaminated gelation and ambient drying.The delaminated gelation process involves the assembly of fluorinated cellulose nanofiber(FCNF)at the solid-liquid interface between the filter and the filtrate during filtration,resulting in the formation of lamellar FCNF hydrogels with strong intra-plane and weak interlayer hydrogen bonding.By exchanging the solvents from water to hexane,the hydrogen bonding in the FCNF hydrogel is further enhanced,enabling the formation of the DAF with intra-layer mesopores upon ambient drying.The resulting aerogel film is lightweight and ultra-flexible,which pos-sesses desirable properties of high visible-light transmittance(91.0%),low thermal conductivity(33 mW m^(-1) K^(-1)),and high atmospheric-window emissivity(90.1%).Furthermore,the DAF exhibits reduced surface energy and exceptional hydrophobicity due to the presence of fluorine-containing groups,enhancing its durability and UV resistance.Consequently,the DAF has demonstrated its potential as solar-thermal regulatory cooling window materials capable of simultaneously providing indoor lighting,thermal insulation,and daytime radiative cooling under direct sunlight.Significantly,the enclosed space protected by the DAF exhibits a temperature reduction of 2.6℃ compared to that shielded by conventional architectural glass.

Astragaloside IV Alleviates Acute Liver Failure Induced by D-GalN/LPS by Upregulating Autophagy and Reducing Inflammation

Acute liver failure(ALF)is a life-threatening condition that manifests in an extremely serious manner and progresses rapidly.The following study investigated the protective effect of astragaloside IV(AS-IV),a traditional Chinese drug,on ALF,and its underlying mechanisms,focusing on autophagy and inflammation regulation.Mice were randomly divided into a saline group,a D-galactosamine and lipopolysac-charide(D-GalN/LPS)group and an AS-IV group.Biochemical analysis,immunohistochemistry,cytometric bead array,high-throughput quantitative PCR,flow cytometry and Western analysis were used to assess inflammation and liver damage 5 hours after D-GalN/LPS ex-posure.Astragaloside IV treatment reduced mortality by alleviating D-GalN/LPS–induced hepatic damage and decreasing inflammation(decreasing Ly6c+monocyte levels,reducing inflammatory cytokines and increasing anti-inflammatory factors)as well as upregulating au-tophagy.Furthermore,PCR array was used to detect expression of autophagy-related genes,which demonstrated a Log2 fold change in gene expression between the AS-IV and D-GalN/LPS groups ranging from 1.19 to-3.53,with Tnfsf10 showing the largest alteration be-tween the two groups.These data suggest that AS-IV may alleviate ALF by upregulating autophagy and reducing inflammation,and it may therefore be an interesting drug for alleviating ALF.

Experimental Study on the Performance of ORC System Based on Ultra-Low Temperature Heat Sources

This paper discussed the experimental results of the performance of an organic Rankine cycle(ORC)system with an ultra-low temperature heat source.The low boiling point working medium R134a was adopted in the system.The simulated heat source temperature(SHST)in this work was set from 39.51°C to 48.60°C by the simulated heat source module.The influence of load percentage of simulated heat source(LPSHS)between 50%and 70%,the rotary valve opening(RVO)between 20%and 100%,the resistive load between 36Ωand 180Ωor the no-load of the generator,as well as the autumn and winter ambient temperature on the system performance were studied.The results showed that the stability of the system was promoted when the generator had a resistive load.The power generation(PG)and generator speed(GS)of the system in autumn were better than in winter,but the expander pressure ratio(EPR)was lower than in winter.Keep RVO unchanged,the SHST,the mass flow rate(MFR)of the working medium,GS,and the PG of the system increased with the increasing of LPSHS for different generator resistance load values.When the RVO was 60%,LPSHS was 70%,the SHST was 44.15°C and the resistive load was 72Ω,the highest PG reached 15.11 W.Finally,a simulation formula was obtained for LPSHS,resistance load,and PG,and its correlation coefficient was between 0.9818 and 0.9901.The formula can accurately predict the PG.The experimental results showed that the standard deviation between the experimental and simulated values was below 0.0792,and the relative error was within±5%.

In recent years,remarkable breakthroughs have been reported on antibody-drug conjugates(ADCs),with 15 ADCs successfully entering the market over the past decade.This substantial development has positioned ADCs as one of the fastest-growing domains in the realm of anticancer drugs,demonstrating their efficacy in treating awide array of malignancies.Nonetheless,there is still an unmet clin-ical need forwider application,better efficacy,and fewer side effects of ADCs.An ADC generally comprises an antibody,a linker and a payload,and the combina-tion has profound effects on drug structure,pharmacokinetic profile and efficacy.Hence,optimization of the key components provides an opportunity to develop ADCs with higher potency and fewer side effects.In this review,we comprehen-sively reviewed the current development and the prospects of ADC,provided an analysis of marketed ADCs and the ongoing pipelines globally as well as in China,highlighted several ADC platforms and technologies specific to different pharmaceutical enterprises and biotech companies,and also discussed the new related technologies,possibility of next-generation ADCs and the directions of clinical research.

Calcium-based polymers for suppression of soil acidification by improving acid-buffering capacity and inhibiting nitrification

Soil acidification is a major threat to agricultural sustainability in tropical and subtropical regions.Biodegradable and environmentally friendly materials,such as calcium lignosulfonate(CaLS),calcium poly(aspartic acid)(PASP-Ca),and calcium polyγ-glutamic acid(γ-PGA-Ca),are known to effectively ameliorate soil acidity.However,their effectiveness in inhibiting soil acidification has not been studied.This study aimed to evaluate the effect of CaLS,PASP-Ca,andγ-PGA-Ca on the resistance of soil toward acidification as directly and indirectly(i.e.,via nitrification)caused by the application of HNO_(3)and urea,respectively.For comparison,Ca(OH)_(2)and lignin were used as the inorganic and organic controls,respectively.Among the materials,γ-PGA-Ca drove the substantial improvements in the pH buffering capacity(pHBC)of the soil and exhibited the greatest potential in inhibiting HNO_(3)-induced soil acidification via protonation of carboxyl,complexing with Al~(3+),and cation exchange processes.Under acidification induced by urea,CaLS was the optimal one in inhibiting acidification and increasing exchangeable acidity during incubation.Furthermore,the sharp reduction in the population sizes of ammonia-oxidizing bacteria(AOB)and ammonia-oxidizing archaea(AOA)confirmed the inhibition of nitrification via CaLS application.Therefore,compared to improving soil pHBC,CaLS may play a more important role in suppressing indirect acidification.Overall,γ-PGA-Ca was superior to PASP-Ca and CaLS in enhancing the soil pHBC and the its resistance to acidification induced by HNO_(3) addition,whereas CaLS was the best at suppressing urea-driven soil acidification by inhibiting nitrification.In conclusion,these results provide a reference for inhibiting soil re-acidification in intensive agricultural systems.
