FLT3 and NPM1 mutations in Chinese patients with acute myeloid leukemia and normal cytogenetics

Mutations of fins-like tyrosine kinase 3 （FLT3） and nucleophosmin （NPM1） exon 12 genes are the most common abnormalities in adult acute myeloid leukemia （AML） with normal cytogenetics. To assess the prognostic impact of the two gene mutations in Chinese AML patients, we used multiplex polymerase chain reaction （PCR） and capillary electrophoresis to screen 76 AML patients with normal cytogenetics for mutations in FLT3 internal tandem duplication （FLT3/ITD） and exon 12 of the NPM1 gene. FLT3/ITD mutation was detected in 15 （19.7%） of 76 subjects, and NPM1 mutation in 20 （26.3%） subjects. Seven （9.2%） cases were positive for both FLT3/ITD and NPM1 mutations Significantly more FLT3/ITD aberration was detected in subjects with French-American-British （FAB） M1 （42.8%）. NPM1 mutation was frequently detected in subjects with M5 （47.1%） and infrequently in subjects with M2 （11.1%）. FLT3 and NPM1 mutations were significantly associated with a higher white blood cell count in peripheral blood and a lower CD34 antigen expression, but not age, sex, or platelet count. Statistical analysis revealed that the FLT3/ITD- positive group had a lower complete remission （CR） rate （53.3% vs. 83.6%）. Survival analysis showed that the FLT3/ITD-positive/NPM1 mutation-negative group had worse overall survival （OS） and relapse-free survival （RFS）. The FLT3/ITD-positive/NPM1 mutation-positive group showed a trend towards favorable survival compared with the FLT3/ITD-positive/NPM1 mutation-negative group （P=0.069）. Our results indicate that the FLT3/ITD mutation might be a prognostic factor for an unfavorable outcome in Chinese AML subjects with normal cytogenetics, while NPM1 mutation may be a favorable prognostic factor for OS and RFS in the presence of FLT3/ITD.

In this study,we investigate the detectability of the secondary spin in an extreme mass ratio inspiral(EMRI) system within a modified gravity model coupled with a scalar *** central black hole,which reduces to a Kerr one,is circularly spiralled by a scalar-charged spinning secondary body on the equatorial *** analysis reveals that the presence of the scalar field amplifies the secondary spin effect,allowing for a lower limit of the detectability and an improved resolution of the secondary spin when the scalar charge is sufficiently *** findings suggest that secondary spin detection is more feasible when the primary mass is not large,and TianQin is the optimal choice for detection.

BACKGROUND Gastric cancer(GC)is the most common malignant tumor and ranks third for cancer-related deaths among the *** disease poses a serious public health problem in China,ranking fifth for incidence and third for *** of the invasive depth of the tumor is vital to treatment *** To evaluate the diagnostic performance of double contrast-enhanced ultrasonography(DCEUS)for preoperative T staging in patients with GC by comparing with multi-detector computed tomography(MDCT).METHODS This single prospective study enrolled patients with GC confirmed by preoperative gastroscopy from July 2021 to March *** underwent DCEUS,including ultrasonography(US)and intravenous contrast-enhanced ultrasonography(CEUS),and MDCT examinations for the assessment of preoperative T *** of GC were identified on DCEUS and criteria developed to evaluate T staging according to the 8th edition of AJCC cancer staging *** diagnostic performance of DCEUS was evaluated by comparing it with that of MDCT and surgical-pathological findings were considered as the gold *** A total of 229 patients with GC(80 T1,33 T2,59 T3 and 57 T4)were *** accuracies were 86.9%for DCEUS and 61.1%for MDCT(P<0.001).DCEUS was superior to MDCT for T1(92.5%vs 70.0%,P<0.001),T2(72.7%vs 51.5%,P=0.041),T3(86.4%vs 45.8%,P<0.001)and T4(87.7%vs 70.2%,P=0.022)staging of *** DCEUS improved the diagnostic accuracy of preoperative T staging in patients with GC compared with MDCT,and constitutes a promising imaging modality for preoperative evaluation of GC to aid individualized treatment decision-making.

Objective:Qu Du Qiang Fei 1 Hao Fang(QDQF1)is a novel Chinese herbal medicine formula used to treat coronavirus disease 2019(COVID-19).However,the pharmacological mechanisms of action of QDQF1 remain *** objective of this study was to identify the effective ingredients and biological targets of QDQF1 for COVID-19 *** and Methods:The effective ingredients and mechanisms of action of QDQF1 were analyzed by using network pharmacology methods,which included an analysis of the effective ingredients and corresponding targets,COVID-19-related target acquisition,compound-target network analyses,protein-protein interaction network analysis,Kyoto Encyclopedia of Genes and Genomes(KEGG)and Gene Ontology(GO)enrichment analyses,and molecular docking ***:In total,288 effective QDQF1 ingredients were *** identified 51 core targets from the 148 targets through an overlap between putative QDQF1 targets and COVID-19-related *** key components,including formononetin,kaempferol,luteolin,naringenin,quercetin,and wogonin were identified through component-target network *** functional enrichment analysis of the core targets revealed 1296 items,while KEGG pathway enrichment analysis identified 148 signaling *** central targets(CCL2,CXCL8,IL1B,IL6,MAPK1,MAPK3,MAPK8,STAT3,and TNF)related to the COVID-19 pathway were identified in the KEGG pathway enrichment ***,molecular docking analysis suggested that the docking scores of the six key components to the nine central targets were better than those to ***:QDQF1 may regulate multiple immune-and inflammation-related targets to inhibit the progression of severe acute respiratory syndrome coronavirus 2,and thus,may be suitable for the treatment of COVID-19.

Study on the clogging mechanism of punching screen in sand control by the punching structure parameters

As an independent sand control unit or a common protective shell of a high-quality screen,the punching screen is the outermost sand retaining unit of the sand control pipe which is used in geothermal well or oil and gas ***,most screens only consider the influence of the internal sand retaining medium parameters in the sand control performance design while ignoring the influence of the plugging of the punching screen on the overall sand retaining performance of the *** explore the clogging mechanism of the punching screen,this paper established the clogging mechanism calculation model of a single punching screen sand control unit by using the computational fluid mechanics-discrete element method(CFD-DEM)combined *** to the combined motion of particles and fluids,the influence of the internal flow state on particle motion and accumulation was *** results showed that(1)the clogging process of the punching sand control unit is divided into three stages:initial clogging,aggravation of clogging and stability of *** the initial stage of blockage,coarse particles form a loose bridge structure,and blockage often occurs preferentially at the streamline gathering place below chamfering inside the sand control *** the stage of blockage intensification,the particle mass develops into a relatively complete sand bridge,which develops from both ends of the opening to the center of the *** the stable plugging stage,the sand deposits show a“fan shape”and form a“V-shaped”gully inside the punching slot element.(2)Under a certain reservoir particle-size distribution,The slit length and opening height have a large influence on the permeability and blockage rate,while the slit width size has little influence on the permeability and blockage *** microscopic clogging mechanism and its law of the punching screen prevention unit are proposed in this study,which has some field guidance significance for the design of punching screen

近视性孔源性视网膜脱离288例临床疗效分析

目的:了解影响近视性孔源性视网膜脱离(RRD)手术治疗预后的因素,为临床治疗提供指导。方法:回顾性分析2007-04/2014-04我科收治并行手术的近视性RRD患者288眼(排除外伤、无晶状体眼、先天性疾患、牵拉性及渗出性视网膜脱离),统计手术解剖复位情况,分析裂孔大小、视网膜脱离时间长短对视力恢复和手术成功率的影响。结果:本文统计结果显示:一次性手术解剖复位245眼,复位率为85.1%,经二次、三次手术后总复位率为95.1%。近视程度及老龄因素对视网膜复位有影响:即近视程度高、年龄大的近视性RRD手术复位率低(P<0.05)。通过手术治疗,术后视力明显好于术前视力(P<0.01);近视程度高的RRD,术后视力恢复差(P<0.01),术后视力有提高的仅有152眼(52.9%),术后视力在0.05以上的有106眼。结论:手术是治疗近视性RRD的有效手段。年龄大的视网膜复位率低;近视程度高的视网膜脱离手术复位率低、术后视力恢复差;近视性孔源性视网膜脱离应重视视功能的恢复。

Application of neoadjuvant chemoradiotherapy and neoadjuvant chemotherapy in curative surgery for esophageal cancer:A metaanalysis

BACKGROUND The effectiveness of neoadjuvant therapy in esophageal cancer(EC)treatment is still a subject of *** To compare the clinical efficacy and toxic side effects between neoadjuvant chemoradiotherapy(nCRT)and neoadjuvant chemotherapy(nCT)for locally advanced EC(LAEC).METHODS A comprehensive search was conducted using multiple databases,including PubMed,EMBASE,MEDLINE,Science Direct,The Cochrane Library,China National Knowledge Infrastructure,Wanfang Database,Chinese Science and Technology Journal Database,and Chinese Biomedical Literature Database *** up to December 2022 comparing nCRT and nCT in patients with EC were *** The analysis revealed significant differences between nCRT and nCT in terms of disease-free *** results indicated that nCRT provided better outcomes in terms of the 3-year overall survival rate(OSR)[odds ratio(OR)=0.95],complete response rate(OR=3.15),and R0 clearance rate(CR)(OR=2.25).However,nCT demonstrated a better 5-year OSR(OR=1.02)than ***,when compared to nCRT,nCT showed reduced risks of cardiac complications(OR=1.15)and pulmonary complications(OR=1.30).CONCLUSION Overall,both nCRT and nCT were effective in terms of survival outcomes for ***,nCT exhibited better performance in terms of postoperative complications.
