Silencing of signal transducer and activator of transcription 3 expression by RNA interference suppresses growth of human hepatocellular carcinoma in tumor-bearing nude mice

AIM： To explore the effect of silencing of signal transducer and activator of transcription 3 （STAT3） expression by RNA interference （RNAi） on growth of human hepatocellular carcinoma （HCC） in tumorbearing nude mice in vivo.METHODS： To construct the recombinant plasmid of pSilencer 3.0-H1-STAT3-siRNA-GFP （pSHI-siRNA- STAT3） and establish the tumor-bearing nude mouse model of the HCC cell line SMMC7721, we used intratumoral injection together with electroblotting to transfect the recombinant plasmid pSHI-siRNA- STAT3 into the transplanted tumor. The weight of the nude mice and tumor volumes were recorded. STAT3 gene transcription was detected by semi-quantitative reverse transcription polymerase chain reaction （RT- PCR）. Level of protein expression and location of STAT3 were determined by Western blotting and immunohistochemical staining. STAT3-related genes such as survivin, c-myc, VEGF, p53 and caspase3 mRNA and protein expression were detected in tumor tissues at the same time. The terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling （TUNEL） assay was used to detect apoptosis of tumor cells.RESULTS： The weight of the treated nude mice increased, and the tumor volume decreased markedly compared with those of the mock-treated and negative control groups （P 〈 0.01）. The results of RT-PCR and Western blotting showed that mRNA and protein levels of STAT3 declined markedly in the treated group. The change in STAT3-related gene expression in tumor tissues at the mRNA and protein level also varied, the expression of survivin, VEGF and c-myc were obviously reduced, and expression of p53 and caspase3 increased （P 〈 0.01）. Most of the tumor tissue ceils in the treated group developed apoptosis that was detected by TUNEL assay.CONCLUSION： Silencing of STAT3 expression by RNAi significantly inhibits expression of STAT3 mRNA and protein, and suppresses growth of human HCC in tumor-bearing nude mice. Heat Transfer and Deformation Behavior of Shell Solidification in Wide and Thick Slab Continuous Casting Mold

With the considerations of the behaviors of shell deformation, mold flux film and air gap dynamic distribution in shell/mold gap, a two dimensional slice-travel transient thermo-mechanical coupled model of simulation shell solidification in wide and thick slab continuous casting mold was developed by using the commercial program ANSYS. The evolutions of strand-mold system thermal behaviors, including the air gap formation and the mold flux film dynamical distribution in shell/mold gap and shell temperature field, and the evolutions of shell deformation and stress distribution of peritectic steel solidified in a 2120 mm wide and 266 mm thick slab continuous casting mold were analyzed. The results show that the air gap formation and the thick mold flux film distribution mainly concentrate in the regions 0–21 mm and 0–7 mm, 0–120 mm and 0–100 mm off the shell wide and narrow faces corners, and thus the hot spots are given rise to form in the regions 15–55 mm and 15–50 mm off the shell wide and narrow face corners. The shell server deformation occurs in the off-corners in the middle and lower parts of the mold. The stress evolution in shell surface is tensile stress, while that in shell solidification front is compressive stress.

Numerical Simulation and Experimental Study of F-EMS for Continuously Cast Billet of High Carbon Steel

A final electromagnetic stirring model was developed for billet continuous casting of high carbon steel using the commercial software ANSYS and CFX, and the numerical model was validated by the magnetic flux density measured under a Teslameter CT-3. The magnetic flux density and fluid flow in the liquid pool at the location of final electromagnetic stirring(F-EMS) were calculated by the present numerical model. Meanwhile, the plant trials were carried out to determine the optimum current intensity and frequency of F-EMS for the continuously cast billet of high carbon steel. The numerical results show that, through increasing the current intensity by 100 A, the corresponding increases of magnetic induction intensity, tangential electromagnetic force and flow velocity at the solid/liquid interface in the strand are 0.025 T, 1933 N/m3 and 6.9 cm/s, respectively. Moreover, the industrial trial results showed that for the continuously cast billet of 60 steel, the optimum current intensity and frequency of F-EMS, which is 8.2 m from the meniscus, are respectively 380 A and 6 Hz. With the optimum F-EMS parameters, the significant improvement of center segregation of billet is achieved, and the center carbon segregation index in billet reaches 1.04.

Relationship and significance between anti-b2-glycoproteinⅠantibodies and platelet activation state in patients with ulcerative colitis

AIM： To study the relationship between anti-β2- glycoprotein Ⅰ （aβ2GPⅠ） antibodies and platelet activation state in patients with ulcerative colitis （UC） and its significance. METHODS： Peripheral blood samples were collected from 56 UC patients （34 males and 22 females, aged 43.5 years, range 21-66 years）, including 36 at active stage and 20 at remission stage, and 25 sex-and age-matched controls. The level of aβ2GP Ⅰ was measured by ELISA. The platelet activation markers, platelet activation complex- Ⅰ （PAC- Ⅰ ） and P-selectin （CD62P） were detected by flow cytometry. RESULTS： The A value for IgG aβ2GP Ⅰ in the active UC group was 0.61 ± 0.13, significantly higher than that in the remittent UC and control groups （0.50 ± 0.13 and 0.22 ± 0.14, P 〈 0.01）. There was a significant difference between the two groups （P 〈 0.01）. The A value for IgM aβ2GP Ⅰ in the active and remittent UC groups was 0.43 ± 0.13 and 0.38 ± 0.12, significantly higher than that in the control group （0.20 ± 0.12, P 〈 0.01）. However, there was no significant difference between the two groups （P 〉 0.05）. The PAC- Ⅰ positive rate for the active and remittent UC groups was 30.6% ± 7.6% and 19.6% ± 7.8% respectively, significantly higher than that for the control group （6.3% ± 1.7%,P 〈 0.01）. There was a significant difference between the two groups （P 〈 0.01）. The CD62P positive rate for the active and remittent UC groups was 45.0% ± 8.8% and 31.9% ± 7.8% respectively, significantly higher than that for the control group （9.2% ± 2.7%, P 〈 0.01）. There was a significant difference between the two groups （P 〈 0.01）. In the active UC group, the more severe the state of illness was, the higher the A value for IgG aβ2GP Ⅰ was, and the positive rate for PAC-Ⅰ and CD62P was positively correlated with the state of illness （Faβ2GP Ⅰ = 3.679, P 〈 0.05; FPAC-Ⅰ （%） = 5.346, P 〈 0.01; and FCD62P （%） = 5. 418, P 〈 0.01）. Recently, observational hints for supermassive black holes have been accumulating, prompting the question: Can primordial black holes（PBHs） be supermassive, particularly with masses M?10～9M⊙? A supercritical bubble,containing an inflating baby universe, that nucleated during inflation can evolve into a PBH in our observable universe. We find that when the inflaton slowly transitions past a neighboring vacuum, the nucleation rate of supercritical bubbles inevitably peaks, leading to a mass distribution of multiverse PBHs with a peak mass up to M?1011M⊙. Thus, our mechanism naturally provides a primordial origin for supermassive black holes.

We perform a unified description of the experimental data of the π^(+)π^(-)and J/ψπ^(±)invariant mass spectra for e^(+)e^(-)→J/ψπ^(+)π^(-)and the D^(0)D^(*-)mass spectrum for e^(+)e^(-)→D^(0)D^(*-)π^(+)at e^(+)e^(-)center-of-mass energies 4.23 and 4.26 *** analysis takes into account open-charm meson loops that contain triangle singularities,the J/ψπ-DD^(*)coupled-channel interaction respecting unitarity,and the strong ππ-KK final state interaction using dispersion *** analysis leads to a precise determination of the Z_(c)(3900)pole with the pole mass and width(3880.7±1.7_(stat)±22.4_(syst))MeV and(35.9±1.4_(stat)±15.3_(syst))MeV,respectively,and hints at that the DD^(*)molecular and non-molecular components are of similar importance in the Z_(c)(3900)formation.

Microcavity photon dynamics in curved space is an emerging interesting area at the crossing point of nanophotonics,chaotic science,and non-Euclidean *** report the sharp difference between the regular and chaotic motions of cavity photons subjected to the varying space *** the island modes of regular motion rise in the phase diagram in the curved space,the chaotic modes show special mechanisms to adapt to the space curvature,including the fast diffusion of ray dynamics,and the localization and hybridization of the Husimi wavepackets among different periodic *** observations are unique effects enabled by the combination of the chaotic trajectory,the wave nature of light,and the non-Euclidean orbital motion,and therefore make the system a versatile optical simulator for chaotic science under quantum mechanics in curved space-time.

Relationship between genetic polymorphism of cytochrome P450IIE1 and fatty liver

AIM: To study the correlation between genetic polymorphism of cytochrome P450IIE1 (CYPIIE1) and fatty ***: Peripheral blood mononuclear cells were collected in 56 patients with fatty liver, 26 patients without fatty liver and 20 normal controls. Then PCR-RFLP was used to analyze genetic polymorphism of CYPIIE1 in monocytes on the region of Pst I and Rsa ***: The frequency of homozygotic C1 gene in patients with alcoholic fatty liver (28.6 %), obese fatty liver (38.5 %), or diabetic fatty liver (33.3 %) was significantly lower than that of the corresponding patients without fatty liver (100 %, 100 % and 80 % respectively), while the frequency of C2 genes, including C1/C2 and C2/C2, was significantly higher (71.4 %/0 %, 61.5 %/0 %, and 66.7 %/20 %) (P0.05).CONCLUSION: The genetic polymorphism of CYPIIE1 on the position of Pst I and Rsa I is related to the susceptibility of fatty liver. Besides, C2 gene may play a key role in the pathogenesis of fatty liver.

Inhibitory effect of all-trans retinoic acid on human hepatocellular carcinoma cell proliferation

AIM: To study the inhibitory effect of all-trans retinoic acid on human hepatocellular carcinoma cell line SMMC-7721and to explore the mechanism of its ***: SMMC-7721 cells were divided into two groups, one treated with all-trans retinoic acid (ATRA) for 5 days and the other as a control group. Light microscope and electron microscope were used to observe the morphological changes. Telomerase activity was analyzed with silver-stained telomere repeated assay protocal (TRAP). Expression of Caspase-3 was demonstrated with western ***: ATRA-treated cells showed differentiation features including small and pyknotic nuclei, densely stained chromatin and fewer microvilli. Besides, ATRA could inhibit the activity of telomerase, promote the expression of Caspase-3 and its ***: Telomerase activity and Caspase-3expression are changed in human hepatocellular carcinoma cell line SMMC-7721 treated with all-trans retinioc *** inhibition of telomerase activity and the activation of Caspase-3 may be the key steps through which ATRA inhibits the proliferation of SMMC-7721 cell line.

Along with the development of 5G network and Internet of Things technologies,there has been an explosion in personalized healthcare *** the 5G and Artificial Intelligence(Al)is introduced into diabetes management architecture,it can increase the efficiency of existing systems and complications of diabetes can be handled more effectively by taking advantage of *** this article,we propose a 5G-based Artificial Intelligence Diabetes Management architecture(AIDM),which can help physicians and patients to manage both acute complications and chronic *** AIDM contains five layers:the sensing layer,the transmission layer,the storage layer,the computing layer,and the application *** build a test bed for the transmission and application ***,we apply a delay-aware RA optimization based on a double-queue model to improve access efficiency in smart hospital wards in the transmission *** application layer,we build a prediction model using a deep forest *** on real-world data show that our AIDM can enhance the efficiency of diabetes management and improve the screening rate of diabetes as well.
