Translocator protein has received attention for its involvement in the pathogenesis of depression. This study assessed the effects of the new translocator protein ligand, YL-IPA08, on alleviating inflammation-induced depression-like behavior in mice and investigated its mechanism of action. Mice were intracerebroventricularly injected with 1, 10, 100 or 1000 ng lipopolysaccharide. The tail-suspension test and the forced swimming test confirmed that 100 ng lipopolysaccharide induced depression-like behavior. A mouse model was then established by intraventricular injection of 100 ng lipopolysaccharide. On days 16-24 after model establishment, mice were intragastrically administered 3 mg/kg YL-IPA08 daily. Immunohistochemistry was used to determine BrdU and NeuN expression in the hippocampus. YL-IPA08 effectively reversed the depression-like behavior of lipopolysaccharide-treated mice, restored body mass, increased the number of BrdU-positive cells, and the number and proportion of BrdU and NeuN double-positive cells. These findings indicate that YL-IPA08 can attenuate lipopolysaccharide-induced depression-like behavior in mice by promoting the formation of hippocampal neurons.

Data-driven innovation has shown great power in solving problems in multifactor correlation,convergence and optimization,synergistic and antagonistic effects,pattern and boundary identification,critical behavior and phase transition,which are ubiquitous in polymer science.Either for the in-depth understanding of physical problems or in the discovery of new polymer materials,integrating data and machine learning into conventional experimental,theoritical,modeling and simulation approaches becomes blooming.Here we present a perspective based on our research interests,highlight some key issues and provide a prospection in this emerging direction.We focus on a number of typical advances in the description and identification of polymer conformation and structures,and the interpretation and prediction of structureproperty correlations,that have applied data and machine learning in polymer science.

This study investigated the transformations of heavy metals(HMs) and polycyclic aromatic hydrocarbons(PAHs) in rhizosphere soils and adaptive responses of rhizobacterial community under the real field conditions during four growth stages(e.g., greening, tillering, heading, and maturity) of early rice(Zhongjiazao 17) and late rice(Zhongyou 9918) in Jiangshe village(JSV) and Yangji village(YJV). Results showed that rhizosphere soils of YJV were mildly polluted by Cd and PAHs compared to that of JSV. The relative abundance of bioavailable Cd(bio-Cd) and bioavailable As(bio-As) in rhizosphere soil increased before the heading stage but decreased at the subsequent growth stage, but the content of ΣPAHs in rhizosphere soil decreased gradually during whole growth period. The dominant rhizobacteria genera at YJV(e.g., Bacillus, Massilia, Sphingomonas, and Geobacter) increased at an abundance level from the tillering to heading stage. Rhizobacteria interacted with the above co-pollutant more intensely at the tillering and heading stage, where genes involved in HM-resistance and PAH-degradation appeared to have a significant enhancement. The contents of bioCd and bio-As in rhizosphere soil of early rice were higher than that of late rice at each growth stage, especially at the heading stage. Bio-Cd, ΣPAHs, and organic matter were key factors influencing the community structure of rhizobacteria. Results of this study provide valuable insights about the interactions between HM-PAH co-pollutant and rhizobacterial community under real field conditions and thus develop in-situ rhizosphere remediation techniques for contaminated paddy fields.

D-dopachrome tautomerase(DDT),a member of the macrophage migration inhibitory factor(MIF)protein superfamily,is a newly described cytokine with chemokine-like characteristics.However,research on fish DDT remains limited.In this study,we identified a DDT homolog(LjDDT)from the Japanese sea bass,Lateolabrax japonicus.Sequence analysis showed that LjDDT had typical sequence features of known DDT and MIF homologs and was most closely related to DDT of rock bream(Oplegnathus fasciatus).LjDDT transcripts were detected in all tested tissues of healthy Japanese sea bass,with the highest expression found in the liver.Upon infection with Vibrio harveyi,LjDDT transcripts were significantly down-regulated in the three tested tissues,including the liver,spleen,and head kidney.Recombinant LjDDT(rLjDDT)and the corresponding antibody(anti-rLjDDT)were subsequently prepared.The administration of 100μg/g anti-rLjDDT had a statistically significant protective effect on the survival of V.harveyi-infected fish.Moreover,rLjDDT was able to induce the migration of monocytes/macrophages(MO/MФ)and lymphocytes both in vitro and in vivo,but without significant influence on the migration of neutrophils.rLjDDT exhibited chemotactic activity for lipopolysaccharide(LPS)-stimulated M1-type MO/MΦin vitro,but not for cAMP-stimulated M2-type MO/MΦ.Furthermore,the knockdown of LjCD74,but not LjCXCR4,significantly down-regulated the rLjDDT-enhanced migration of MO/MΦand relieved the rLjMIF-inhibited migration of MO/MΦ.These results indicate that LjCD74 may be the major chemotactic receptor of LjDDT and LjMIF in Japanese sea bass MO/MΦ.Combined rLjDDT+rLjMIF treatment had no significant effect on the migration of MsiRNA,LjCD74si-,or LjCXCR4sitreated MO/MΦcompared to the control group,suggesting that the roles of LjDDT and LjMIF may be antagonistic.In conclusion,our study demonstrates for the first time that DDT may play a role in the immune responses of fish against bacterial infection through chemotactic recruitment of MO/MΦvia mediation of CD74 as an

In this paper we survey some results on the symmetric semi-perfect obstruction theory on a Deligne-Mumford stack X constructed by Chang-li,and Behrend’s theorem equating the weighted Euler characteristic of X and the virtual count of X by symmetric semi-perfect obstruction theories.As an application,we prove that Joyce’s d-critical scheme admits a symmetric semi-perfect obstruction theory,which can be applied to the virtual Euler characteristics by Jiang-Thomas.

Objective To elucidate the renoprotective effect of resveratrol(RSV)on sphingosine kinase 1(SphK1)signaling pathway and expression of its downstream molecules including activator protein 1(AP-1)and transformation growth factor-β1(TGF-β1)in lipopolysaccharide(LPS)-induced glomerular mesangial cells(GMCs).Methods The rat GMCs line(HBZY-1)were cultured and randomly divided into 5 groups,including control,LPS(100 ng/mL),and 5,10,20µmol/L RSV-treated groups.In addition,SphK1 inhibitor(SK-II)was used as positive control.GMCs were pretreated with RSV for 2 h and treated with LPS for another 24 h.GMCs proliferation was determined by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT)assay.The proteins expression of SphK1,p-c-Jun and TGF-β1 in GMCs were detected by Western blot,and DNA-binding activity of AP-1 was performed by electrophoretic mobility shift assay(EMSA).The binding activity between RSV and SphK1 protein was detected by AutoDock Vina and visualized by Discovery Studio 2016.Results LPS could obviously stimulate GMCs proliferation,elevate SphK1,p-c-Jun and TGF-β1 expression levels and increase the DNA-binding activity of AP-1(P0.05).Moreover,RSV exhibited good binding affinity towards SphK1,with docking scores of−8.1 kcal/moL and formed hydrogen bonds with ASP-178 and LEU-268 in SphK1.Conclusion RSV inhibited LPS-induced GMCs proliferation and TGF-β1 expression,which may be independent of its hypoglycemic effect on preventing the development of mesangial cell fibrosis and closely related to the direct inhibition of SphK1 pathway.

Breakage of the C-N bond is a structure sensitive process,and the catalyst size significantly affects its activity.On the active metal nanoparticle scale,the role of catalyst size in C-N bond cleavage has not been clearly elucidated.So,Ru catalysts with variable nanoparticle sizes were obtained by modulating the reduction temperature,and the catalytic activity was evaluated using 1,2,3,4-tetrahydroquinoline and o-propylaniline with different C-N bond hybridization patterns as reactants.Results showed a 13 times higher reaction rate for sp3-hybridized C-N bond cleavage than sp2-hybridized C-N bond cleavage,while the reaction rate tended to increase first and then decrease as the catalyst nanoparticle size increased.Different concentrations of terrace,step,and corner sites were found in different sizes of Ru nanoparticles.The relationship between catalytic site variation and C-N bond cleavage activity was further investigated by calculating the turnover frequency values for each site.This analysis indicates that the variation of different sites on the catalyst is the intrinsic factor of the size dependence of C-N bond cleavage activity,and the step atoms are the active sites for the C-N bond cleavage.When Ru nanoparticles are smaller than 1.9 nm,they have a strong adsorption effect on the reactants,which will affect the catalytic performance of the Ru catalyst.Furthermore,these findings were also confirmed on other metallic Pd/Pt catalysts.The role of step sites in C-N bond cleavage was proposed using the density function theory calculations.The reactants have stronger adsorption energies on the step atoms,and step atoms have d-band center nearer to the Fermi level.In this case,the interaction with the reactant is stronger,which is beneficial for activating the C-N bond of the reactant.

In this study, we focused on the effect of the underwater explosion parameters of multi-point array explosion. The shock wave and bubble parameters of aggregate charge, two charges, and four charges were measured through an underwater explosion test, and their influence on the explosion power field of charge quantity and array distance was analyzed. Results show that the multi-shock wave collision of array explosion can be approximated to a linear superposition, and the interaction of delayed shock wave can be deemed as the increase of the shock wave baseline. Shock wave focusing and delayed superposition increase the shock wave peak pressure. Compared with the aggregate charge, the greater the number of array explosion points is, the higher the impulse and the gain of the bubble peak pressure are. At the same array distance, the smaller the charge quantity is, the higher the bubble impulse will be. At the same charge quantity, the smaller the array distance is, the higher the bubble impulse will be. The bubble period decreases gradually with the increase of the charge quantity, but the test orientation has little effect on the bubble period.

Coniopteris simplex is a common component in many Jurassic floras.However,due to morphological variations in sterile fronds and incomplete preservation of reproductive organs,its classification has been controversial for a long time.Here,we present new material collected from the Middle Jurassic Yan’an Formation in the Ordos Basin,Inner Mongolia.The new material reveals important morphological characters,including hemi-dimorphic fronds,two forms of sterile pinnae,short-stalked sori and sporangia on fertile fronds,annulus of sporangium composed of about 30 cells,in situ rounded triangular spores with protruded suture,and the lips of trilete laesura encircled by one or two rows of small ostioles.Based on these observations,we emended the diagnosis of this common species.The spatio-temporal distribution of Coniopteris simplex shows that the iconic species was restricted to the paleolatitudes ranging from 19.5°N to 48.9°N in the Northern Hemisphere during the Early—Late Jurassic,mainly distributed in the tropical to paratropical zones,and the paleolongitude ranging from 9.6°E to 129.3°E,which may be limited by the opening of the Viking Corridor and the splitting of the Central Atlantic Ocean.

We provide a construction of the moduli space of stable coherent sheaves in the world of non-archimedean geometry,where we use the notion of Berkovich non-archimedean analytic spaces.The motivation for our construction is Tony Yue Yu’s non-archimedean enumerative geometry in Gromov-Witten theory.The construction of the moduli space of stable sheaves using Berkovich analytic spaces will give rise to the non-archimedean version of Donaldson-Thomas invariants.In this paper we give the moduli construction over a non-archimedean field K.We use the machinery of formal schemes,that is,we define and construct the formal moduli stack of(semi)-stable coherent sheaves over a discrete valuation ring R,and taking generic fiber we get the non-archimedean analytic moduli space of semistable coherent sheaves over the fractional non-archimedean field K.We generalize Joyce’s dcritical scheme structure in[37]or Kiem-li’s virtual critical manifolds in[38]to the world of formal schemes,and Berkovich non-archimedean analytic spaces.As an application,we provide a proof for the motivic localization formula for a d-critical non-archimedean K-analytic space using global motive of vanishing cycles and motivic integration on oriented formal d-critical schemes.This generalizes Maulik’s motivic localization formula for the motivic Donaldson-Thomas invariants.
