HLA-mismatched allogeneic adoptive immune therapy in severely immunosuppressed AIDS patients

Severely immunosuppressed AIDS patients with recurrent opportunistic infections(Ols)represent an unmet medical need even in the era of antiretroviral therapy(ART).Here we report the development of a human leukocyte antigen(HLA)-mismatched allogeneic adaptive immune therapy(AAIT)for severely immunosuppressed AIDS *** severely immunosuppressed AIDS patients with severe Ols were enrolled in this single-arm *** donors received subcutaneous recombinant granulocyte-colony-stimulating factor twice daily for 4-5 days to stimulate *** blood mononuclear cells were collected from these donors via leukapheresis and transfused into the coupled ***,immunological,and virological parameters were monitored during a 12-month follow-up *** found AAIT combined with ART was safe and well-tolerated at the examined doses and transfusion regimen in all 12 *** in clinical symptoms were evident throughout the study *** patients exhibited a steady increase of peripheral CD4^(+)T cells from a median 10.5 to 207.5 cells/μ*** increase in peripheral CD8^(+)T-cell count from a median 416.5 to 1206.5 cells/μl was found in the first 90 days since initiation of *** addition,their inflammatory cytokine levels and HIV RNA viral load decreased.A short-term microchimerism with donor cells was *** were no adverse events associated with graft-versus-host disease throughout the study ***,AAIT treatment was safe,and might help severely immunosuppressed AIDS patients to achieve a better immune restoration.A further clinical trial with control is necessary to confirm the efficacy of AAIT medication.

FK506(Tacrolimus)is a systemic immunosuppressant approved by the *** and Drug ***506 has been shown to promote peripheral nerve regeneration,however,its precise mechanism of action and its pathways remain *** this study,we established a rat model of sciatic nerve injury and found that FK506 improved the morphology of the injured sciatic nerve,increased the numbers of motor and sensory neurons,reduced inflammatory responses,markedly improved the conduction function of the injured nerve,and promoted motor function *** findings suggest that FK506 promotes peripheral nerve structure recovery and functional regeneration by reducing the intensity of inflammation after neuronal injury and increasing the number of surviving neurons.

Selective modification of peptides has always been a challenging issue. The selective alkylation of peptides can alter the biological activity and physical properties of molecules, and it has gained significant research interest. We herein disclose an intriguing gold-catalyzed alkylation of peptides using bench-stable unactivated alkyl bromides under photoredox catalysis. A wide range of structurally diverse primary, secondary and tertiary alkyl bromides serve as effective coupling partners to precisely connect with the α-C(sp3)–H in glycine residue of peptides. This protocol demonstrates an exceptionally broad scope for alkyl halides and excellent tolerance for a wide range of useful functional groups. A series of relay transformations to construct cyclic peptides, to proceed click reaction and to realize peptide fluorescence labeling further enhance its synthetic practicality. In addition, mechanistic studies and density functional theory(DFT) calculations reveal an inner-sphere and subsequent outersphere single electron transfer(SET) mechanism.

Effect of SO_2 on the performance of Ag-Pd/Al_2O_3 for the selective catalytic reduction of NOx with C_2H_5OH

The influence of SO2 on the performance of Ag-Pd/Al2O3 for the selective catalytic reduction （SCR） of NOx with C2H5OH was investigated experimentally. The activity test results suggest that Ag-Pd/Al2O3 shows a small activity loss in the presence of SO2 when using C2H5OH as a reductant. In situ DRIFTS spectra show that the activity loss originates from the formation of surface sulphate species on the Ag-Pd/Al2O3. The surface sulphate species formation inhibits the formation of nitrate, whereas hardly changes the partial oxidation of C2H5OH. Compared with the NOx reduction by C3H6, an obvious suppression of the surface sulphate species formation was observed by DRIFTS experiment when using C2H5OH as a reductant. This phenomenon reveals the better catalytic performance and strong SO2 tolerance of Ag-Pd/Al2O3-C2H50H system.

Background We explored the differences in baseline characteristics, pathogens, complications, outcomes, and risk factorsbetween children with hospital-acquired septic shock (HASS) and community-acquired septic shock (CASS) in the pediatricintensive care unit (PICU).Methods This retrospective study enrolled children with septic shock at the PICU of Beijing Children’s Hospital from January1, 2016, to December 31, 2019. The patients were followed up until 28 days after shock or death and were divided intothe HASS and CASS group. Logistic regression analysis was used to identify risk factors for *** A total of 298 children were enrolled. Among them, 65.9% (n = 91) of HASS patients had hematologic/oncologicdiseases, mainly with Gram-negative bacterial bloodstream infections (47.3%). Additionally, 67.7% (n = 207) of CASSpatients had no obvious underlying disease, and most experienced Gram-positive bacterial infections (30.9%) of the respiratoryor central nervous system. The 28-day mortality was 62.6% and 32.7% in the HASS and CASS groups, respectively(P < 0.001). Platelet [odds ratio (OR) = 0.996, 95% confidence interval (CI) = 0.992–1.000, P = 0.028], positive pathogendetection (OR = 3.557, 95% CI = 1.307–9.684, P = 0.013), and multiple organ dysfunction syndrome (OR = 10.953, 95%CI = 1.974–60.775, P = 0.006) were risk factors for 28-day mortality in HASS patients. Lactate (OR = 1.104, 95% CI = 1.022–1.192, P = 0.012) and mechanical ventilation (OR = 8.114, 95% CI = 1.806–36.465, P = 0.006) were risk factors for 28-daymortality in patients with *** The underlying diseases, pathogens, complications, prognosis, and mortality rates varied widely between theHASS and CASS groups. On Coupled KdV Equations with Self-consistent Sources

The coupled Korteweg-de Vries （CKdV） equation with self-consistent sources （CKdVESCS） and its Lax representation are derived. We present a generalized binary Darboux transformation （GBDT） with an arbitrary time- dependent function for the CKdVESCS as well as the formula for the N-times repeated GBDT. This GBDT provides non-auto-Biicklund transformation between two CKdVESCSs with different degrees of sources and enables us to construct more generM solutions with N arbitrary t-dependent functions. We obtain positon, negaton, complexiton, and negaton- positon solutions of the CKdVESCS.

Prediction model for mechanical properties of hot-rolled strips by deep learning

The prediction of the mechanical properties of hot-rolled strips is a very complex,highly dimensional and nonlinear problem,and the published models might lack reliability,practicability and ***,a new model was proposed for predicting the mechanical properties of hot-rolled strips by deep ***,the one-dimensional numerical data were transformed into two-dimensional data for expressing the complex interaction between the influencing ***,a new convolutional network was proposed to establish the prediction model of tensile strength of hot-rolled strips,and an improved inception module was introduced into this network to abstract features from different *** comparative experiments were carried out to find the optimal network structure and its ***,the prediction experiments were carried out on different models to evaluate the performance of the new convolutional network,which includes the stepwise regression,ridge regression,support vector machine,random forest,shallow neural network,Bayesian neural network,deep feed-forward network and improved LeNet-5 convolutional neural *** results show that the proposed convolutional network has better prediction accuracy of the mechanical properties of hot-rolled strips compared with other models.

Independent N-Electron Method and High Accuracy Calculations of the Ground State Energy of the Nonrelativistic Light Atoms (3 Ne 10)

A new method based on the independent electron-pair approximation is developed to calculate the correlation energy and the basis size truncation error. The formalisms for the nondegenerate systems are described in this paper. Because all important terms are very efficiently selected and the calculations can be separated into several parts, this new method gives very accurate results and does not require expensive computing facilities. As a test, the ground state energies (GSE) of the nonrelativistic atoms for (3 ≤ Ne ≤ 10) are calculated using this new method. The calculated results are found to be in close agreement with the most recent available version of the exact GSE which is derived from the experimental data after subtracting the contributions from the relativistic effect, QED radiative corrections, finite nucleon mass corrections, and the finite radius of the nuclear size. Those accurate results have not been reproduced for Ne ≥ 5 by any direct theoretical calculations in literatures. Since this method is not restrained to nonrelativistic or Coulomb systems, it should become powerful to study other quantum interacting systems.

Nanofiltration Membranes via Layer-by-layer Assembly and Cross-linking of Polyethyleneimine/Sodium Lignosulfonate for Heavy Metal Removal

Layer-by-layer(LbL)assembly technology is a facile method for constructing thin film composite ***,a novel nanofiltration(NF)membrane was prepared by LbL assembly of polyethyleneimine(PEI)and sodium lignosulfonate(LS)followed by *** surface composition,morphology,and property of PEI/LS bilayer were detailedly investigated by FTIR/ATR,XPS,SEM,AFM,and water contact angle *** PEI/LS bilayer full of amino and hydroxyl groups presents increased roughness and improved ***,the NF performance of PEI/LS LbL assembly membranes can be modulated by bilayer number,polyelectrolyte concentration,and salt *** water flux reduced while the salt rejection greatly improved as increasing the bilayer numbers,PEI concentration,or NaCl *** than 95%MgSO4 and MgCl2,as well as 80%NaCl can be rejected by a NF membrane prepared by 6 PEI/LS bilayers,1 wt%PEI,0.5 wt%LS,and 1 mol/L ***,this NF membrane can be used to remove more than 95%heavy metal ions(Cd2+,Zn2+,Mn2+,Cr2+,Cu2+,and Ni2+).This work proposed a promising NF membrane by using PEI/LS as low cost polyelectrolytes and facile LbL assembly method,which should receive much attention in water purification.
