Preparation of human decellularized peripheral nerve allograft using amphoteric detergent and nuclease

Animal studies have shown that amphoteric detergent and nuclease(DNase I and ribonuclease A) is the most reliable decellularization method of the peripheral nerve. However, the optimal combination of chemical reagents for decellularization of human nerve allograft needs further investigation. To find the optimal protocol to remove the immunogenic cellular components of the nerve tissue and preserve the basal lamina and extracellular matrix and whether the optimal protocol can be applied to larger-diameter human peripheral nerves, in this study, we decellularized the median and sural nerves from the cadavers with two different methods: nonionic and anionic detergents(Triton X-100 and sodium deoxycholate) and amphoteric detergent and nuclease(3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate(CHAPS), deoxyribonuclease I, and ribonuclease A). All cellular components were successfully removed from the median and sural nerves by amphoteric detergent and nuclease. Not all cellular components were removed from the median nerve by nonionic and anionic detergent. Both median and sural nerves treated with amphoteric detergent and nuclease maintained a completely intact extracellular matrix. Treatment with nonionic and anionic detergent decreased collagen content in both median and sural nerves, while the amphoteric detergent and nuclease treatment did not reduce collagen content. In addition, a contact cytotoxicity assay revealed that the nerves decellularized by amphoteric detergent and nuclease was biocompatible. Strength failure testing demonstrated that the biomechanical properties of nerves decellularized with amphoteric detergent and nuclease were comparable to those of fresh controls. Decellularization with amphoteric detergent and nuclease better remove cellular components and better preserve extracellular matrix than decellularization with nonionic and anionic detergents, even in large-diameter human peripheral nerves. The influence of fabrication on the tensile and fatigue behavior of copper films manufactured by 3 kinds of fabrication methods was investigated. The tensile and high cycle fatigue tests were performed using the test machine developed by authors. Young＇s moduli （72, 71 and 69 GPa, respectively） are lower than the literature values （108-145 GPa）, while the yield strengths were as high as 358, 350 and 346 MPa, respectively and the ultimate strengths as 462, 456 and 446 MPa, respectively. There is not much difference in the tensile properties of the 3 kinds of films. There is little difference in the fatigue properties of the 3 kinds of films but one of them has shorter fatigue life than others in high cycle region and longer fatigue life than others in low cycle region.

Functionalization of aniline molecules on zinc oxide(ZnO)nanoparticles is reported using a simple impregnation *** ZnO samples were systematically characterized based on morphology,surface and optical properties,and photocatalytic performance towards methyl orange(MO).Aniline functionalization increased the surface charge of the modified *** to pristine ZnO,the aniline-functionalized ZnO yielded faster photodegradation of MO,degrading 98.29%of MO in 60 min and superoxide radicals were the major species during the MO photodegradation *** results indicate that the improvement of photocatalytic degradation could be attributed to opposite charge-induced surface ***,protonated amine as a positively charged molecule was expected to increase the surface adsorption of MO(as an anionic dye)on ZnO nanoparticles surfaces,thereby increasing their photocatalytic degradation performance.

Introducing Ni in Ru oxide is a promising approach to enhance the catalytic activity for the oxygen evolution reaction(OER).However,the role of Ni(which has a poor intrinsic activity)is not fully ***,a Ru NiO_(x)electrode fabricated via a modified dip coating method exhibited excellent OER performance in acidic media,and neutral media for CO_(2)reduction *** combined in-situ/operando X-ray absorption near-edge structure and on-line inductively coupled plasma mass spectrometry studies to unveil the role of the Ni introduced in the Ru *** propose that the Ni not only transforms the electronic structure of the Ru oxide,but also produces a large number of oxygen vacancies by distorting the oxygen lattice structure at low overpotentials,increasing the participation of lattice oxygen for *** work demonstrates the real behavior of bimetallic oxide materials under applied potentials and provides new insights into the development of efficient electrocatalysts.

Efficient detection of pathogen virus in sand dabs,Paralichthys olivaceus using loop-mediated isothermal amplification(LAMP)

Viral hemorrhagic septicemia virus（VHSV） and marine birnavirus（MABV） are the causative pathogens for some of the most explosive epidemics of emerging viral diseases in many Asian countries, leading to huge economic losses in aquaculture. Rapid molecular detection for surveillance or diagnosis has been a critical component in reducing the prevalence of pathogen infection. The loop-mediated isothermal amplification（LAMP） of DNA is currently one of the most commonly used molecular diagnostic tools, as it is simple, quick, and easy to amplify target DNA under isothermal conditions. In the present study, a novel and highly specific LAMP assay for the sensitive and rapid detection of VHSV and MABV infection in fish was developed. Using a set of synthesized primers matching a specific region of the genome, the efficiency and specificity of the LAMP assay were optimized in terms of the reaction temperature and DNA polymerase concentration, as they are the main determinants of the sensitivity and specificity of the LAMP assay. In particular, we demonstrated that our assay could be applied to efficient detection of VHSV and MABV infection in the wild fish, Paralichthys olivaceus. Our results demonstrate the simplicity and convenience of this method for the detection of viral infection in aquatic organisms.

Comparative analysis of radiofrequency ablation and resection for resectable colorectal liver metastases

Identification of Aristolactam Derivatives That Act as Inhibitors of Human Immunodeficiency Virus Type 1 Infection and Replication by Targeting Tat-Mediated Viral Transcription

Despite the success of antiretroviral therapy(ART),efforts to develop new classes of antiviral agents have been hampered by the emergence of drug ***-indole-bearing aristolactams are compounds that have been isolated from various plants species and which show several clinically relevant effects,including anti-inflammatory,antiplatelet,and antimycobacterial ***,the effect of these compounds on human immunodeficiency virus type 1(HIV-1)infection has not yet been *** this study,we discovered an aristolactam derivative bearing dibenzo[cd,f]indol-4(5 H)-one that had a potent anti-HIV-1 effect.A structure-activity relationship(SAR)study using nine synthetic derivatives of aristolactam identified the differing effects of residue substitutions on the inhibition of HIV-1 infection and cell *** the compounds tested,1,2,8,9-tetramethoxy-5-(2-(piperidin-1-yl)ethyl)-dibenzo[cd,f]indol-4(5 H)-one(Compound 2)exhibited the most potent activity by inhibiting HIV-1 infection with a half-maximal inhibitory concentration(IC50)of 1.03 lmol/L and a half-maximal cytotoxic concentration(CC50)of 16.91 lmol/L(selectivity index,16.45).The inhibitory effect of the compounds on HIV-1 infection was linked to inhibition of the viral replication ***-of-action studies showed that the aristolactam derivatives did not affect reverse transcription or integration;instead,they specifically inhibited Tat-mediated viral *** together,these findings show that several aristolactam derivatives impaired HIV-1 infection by inhibiting the activity of Tat-mediated viral transcription,and suggest that these derivatives could be antiviral drug candidates.

Major media outlets have run many articles on the COVID-19 *** the public suffers cognitive and emotional effects related to COVID-19 from such reports,we analyzed and reviewed the topics of news *** searched newspaper articles with the term‘COVID-19’term in four Korean daily newspapers from January 20,2020,when thefirst patient in Korea was found,to June 15,*** modeling analysis was conducted through text mining using *** themes were found:“Changes in people’s everyday life,”“Socio-economic shock,”“Trends in infection,”“Role of the government and business,”and“Increased psychological anxiety,”which all showed sharp increases in articles from mid-February to early March and then *** the increased psychological anxiety people suffered from the COVID-19 pandemic,this topic showed the fewest articles.“Changes in people’s everyday life”showed the most,focusing attention on stimulating lifestyle articles of general *** the COVID-19 pandemic can lead to mental health problems due to severe changes and isolation in everyday life,a comprehensive response to the news focusing on the impact on the mental health of the population around the world should be made.
