AIM: To clarify the role of high in normal-1 (HIN-1) gene promoter methylation during gastric cancer development. METHODS: Gastric cancer cell lines and tissue specimens were analyzed for expression of HIN-1 mRNA and protein using the semi-quantitative reverse transcription polymerase chain reaction and immunohistochemistry. The methylation of the HIN-1 gene promoter was detected in gastric carcinoma cells and tissues using methylation-specific polymerase chain reaction. The 3-(4,5-dimethylthiazol-2yl)-5-(3-carboxymethoxyphenyl)-2-(4-sulfophenyl)-2H-tetrazolium cell viability assay and flow cytometry were used to assess the changes in behaviors of gastric cancer cells with or without 5-aza-2’-deoxycytidine treatment. RESULTS: HIN-1 was not expressed in 4 of 5 gastric cancer cell lines. The demethylation reagent 5-aza-2’-deoxycytidine was able to induce or upregulate HIN-1 expression in gastric cancer cell lines, which is associated with reduction of tumor cell viability. Furthermore, methylation of the HIN-1 gene promoter was shown in 57.8% (26/45) of the primary gastric cancer and 42.1% (17/38) of adjacent tissue samples, but was not shown in normal gastric mucosa (0/10). From the clinicopathological data of the patients, methylation of the HIN-1 gene promoter was found to be associated with tumor differentiation (P = 0.000). CONCLUSION: High methylation of HIN-1 gene promoter results in silence of HIN-1 expression in gastric cancer. 5-aza-2’-deoxycytidine reverses HIN-1 methylation and reduces viability of gastric cancer cells.

Spinal cord injury（SCI） from trauma or disease severely impairs sensory and motor function. Neurorehabilitation after SCI is a complex medical process that focuses on improving neurologic function and repairing damaged connections in the central nervous system. An increasing number of preclinical studies suggest that melatonin may be useful for the treatment of SCI. Melatonin is an indolamine that is primarily secreted by the pineal gland and known to be regulated by photoperiodicity. However, it is also a versatile hormone with antioxidative, antiapoptotic, neuroprotective, and anti-inflammatory properties. Here, we review the neuroprotective properties of melatonin and the potential mechanisms by which it might be beneficial in the treatment of SCI. We also describe therapies that combine melatonin with exercise, oxytetracycline, and dexamethasone to attenuate the secondary injury after SCI and limit potential side effects. Finally, we discuss how injury at different spinal levels may differentially affect the secretion of melatonin.

The pyndazine hexafluorophosphate[C4H5N2]~＋[PF6]^-（1） undergoes a reversible phase transition around140 K,which was confirmed by the DSC measurement.Variable-temperature crystal structures determined at 293 K and 93 K show that the compound crystallizes in the same space group P21/c,indicating that 1 undergoes an iso-structural phase *** the temperature decreases,dielectric measurement of the title compound shows no significant change around the phase transition *** hydrogen bonds are found between molecules at 293 K and 93 l〈 with similar packing *** most distinct difference between the low temperature and room temperature structures is the order-disorder transition of the hexafluorophosphate anion,which is probably the driving force of the phase transition.

Objectives To investigate the effect of nerve growth factor (NGF) on the action potential and potassium currents of non-infarcted myocardium in the myocardial infarcted rabbit *** Rabbitswith occlusion of the left anterior descending coronary arterywere prepared and allowed to recover for eight weeks (healed myocardial infarction, Hmi). During ligation surgery of the left coronary artery, a polyethylene tube was placed near the left stellate ganglion in the subcutis of the neck for the purpose of administering NGF 400 U/d for eight weeks (Hmi + NGF group). Cardiomyocytes were isolated from regions of the non-infarcted left ventricular wall and the action potentials and ion currents in these cellswere recorded using whole-cell patch clamps. Results Comparedwith Hmi and control cardiomyocytes, significant prolongation of APD50 or APD90 (Action potential duration (APD) measured at 50% and 90% of repolarization) in Hmi + NGF cardiomyocytes was found. The results showed that the 4-aminopyridine sensitive transient outward potassium current (Ito), the rapidly activated omponent of delayed rectifier potassium current (IKr), the slowly activated component of delayed rectifier potassium current (IKs), and the L-type calcium current (ICaL) were significantly altered in NGF + Hmi cardiomyocytes compared with Hmi and control cells. Conclusions Our results suggest thatNGF treatment significantly prolongsAPDinHmi cardiomyocytes and that a decrease in outward potassium currents and an increase of inward Ca2+ current are likely the underlying mechanism of action.

Objective:To investigate the hemostatic effect of modified Sijunzi Granules(MSG)in primary immune thrombocytopenia(ITP)zebrafish model and explore the potential ***:AB strain wild type zebrafish were treated with simvastatin(6μmol/L)for 24 h to establish the hemorrhage model(model control group).The zebrafish were treated with MSG at different doses(55.6,167,and 500μg/mL),*** hemostatic effect was assessed by examining the intestinal bleeding and hemostatic rate.5-hydroxytryptamine(5-HT)content was determined using enzyme-linked immunosorbent assay(ELISA)*** expressions of5-HT2aR,5-HT2bR,and SERT genes were detected by quantitative real-time polymerase chain reaction(PCR).The protein expressions of protein kinase B(Akt),p-Akt,extracellular regulated protein kinases(Erk),and p-Erk were examined using Western blot ***:The intestinal bleeding rate was 37%,40%,and 80%in the55.6,167,and 500μg/mL dose of MSG,respectively,in which 55.6 and 167μg/mL MSG dose groups were associated with significantly decreased intestinal bleeding rate when compared with the model control group(70%,Pdetected after treatment with MSG(Pdence and severity of intestinal bleeding in zebrafish by activating MAPK/Erk and PI3K/Akt signal pathways through regulating the levels of 5-HT and its receptors,which may provide evidence for the treatment of ITP.

The conventional distillation is hard to accomplish the separation of acetonitrile/ethyl acetate/n-hexane mixture. Herein, a heterogeneous azeotropic distillation(HAD) without adding entrainer is proposed to separate ternary mixture. The proposed scheme is optimized via the simulated annealing algorithm and minimum total annual cost(TAC) is used as objective functions. To minimize energy consumption,heat pump is added on the basis of optimal heterogeneous azeotropic distillation and heat integration technology is used to further improve the energy recovery. The TAC, gas emission, energy consumption and exergy destruction are used to discuss the economy and environmental protection of *** all the processes, the heat pump with higher preheating temperature(HPT) assisted HAD process by combining with heat integration(HAD-HPT-HI) has best performances on economic, environment,energy and exergy. Compared with conventional HAD process, the HAD-HPT-HI achieves the reductions of 52.17%, 68.86%, 65.87% and 65.46% on TAC, total energy consumption, gas emissions and exergy destruction, respectively.

The growth of Sn whiskers from Ti_(2)SnC/Sn-X(X=Sn,Bi,Pb,Ga)samples was studied in this work to understand the effect of alloying elements on whiskering *** mobility of source Sn atoms relating with the formation and migration of defects was found to be the critical factor dominating the growth rate,with the gathering of which to grow whiskers on the surface acts as an effective means to lower the *** a result,the source Sn atoms would migrate to whisker root spontaneously,making the growth of whiskers the nature of materials,and external factors like compressive stress or oxidation are no more necessary but facilitate the whiskering process by promoting the mobility of source Sn atoms.
