Objective This study aimed to compare the current Essen rabies post-exposure immunization schedule(0-3-7-14-28)in China and the simple 4-dose schedule(0-3-7-14)newly recommended by the World Health Organization in terms of their safety,efficacy,and *** Mice were vaccinated according to different immunization schedules,and blood was collected for detection of rabies virus neutralizing antibodies(RVNAs)on days 14,21,28,35,and 120after the first ***,different groups of mice were injected with lethal doses of the CVS-11 virus on day 0,subjected to different rabies immunization schedules,and assessed for morbidity and death *** a clinical trial,185 rabies-exposed individuals were selected for post-exposure vaccination according to the Essen schedule,and blood was collected for RVNAs detection on days 28and 42 after the first *** A statistically significant difference in RVNAs between mice in the Essen and 0-3-7-14 schedule groups was observed on the 35th day(P0.05)in RVNAs levels at any time *** post-exposure immune protective test showed that the survival rate of mice in the control group was 20%,whereas that in the immunization groups was 40%.In the clinical trial,the RVNAs positive conversion rates on days 28(14 days after 4 doses)and 42(14 days after 5 doses)were both 100%,and no significant difference in RVNAs levels was observed(P>0.05).Conclusion The simple 4-dose schedule can produce sufficient RVNAs levels,with no significant effect of a delayed fourth vaccine dose(14–28 d)on the immunization potential.

Objective Ferritin,initially acting as an iron-storage protein,was found to be associated with metabolic *** study was designed to investigate the association between serum ferritin and metabolic-associated fatty liver disease(MAFLD)using data from the National Health and Nutrition Examination Survey(NHANES)of the United State of *** A cross-sectional study was conducted,enrolling a total of 2145 participants from the NHANES in the 2017–2018 *** steatosis and liver fibrosis were assessed by ultrasound images and several non-invasive *** regression analysis was conducted to determine the associations between serum ferritin concentration and MAFLD and liver *** The analysis revealed that participants with higher serum ferritin levels(Q3 and Q4 groups)had a higher prevalence of MAFLD than those with the lowest serum ferritin levels[Q3 vs.Q1:OR=2.17(1.33,3.53),P<0.05 in fatty liver index(FLI);Q4 vs.Q1:OR=3.13(1.91,5.13),P<0.05 in FLI].Additionally,participants with the highest serum ferritin levels(Q4 group)displayed a higher prevalence of liver fibrosis[Q4 vs.Q1:OR=2.59(1.19,5.62),P<0.05 in liver stiffness measurement;OR=5.06(1.12,22.94),P<0.05 in fibrosis-4 index],with significantly increased risk observed in participants with concomitant diabetes[OR=7.45(1.55,35.72),P=0.012].Conclusion Our study revealed that elevated serum ferritin levels are associated with a higher prevalence of MAFLD and advanced liver fibrosis in *** serum ferritin levels combined with diabetes are important risk factors for liver fibrosis.

Cryopreservation of Queen Honeybee (Apis mellifera carnica) Born Worker Eggs by Vitrification

Many species of insect egg can be targeted individually or （and） collectively for cryopreservation by vitrification. However, there has been no report on cryopreservation of honeybee eggs by vitrification. In an attempt to define a preliminary procedure of cryopreservation of honeybee eggs by vitrification, queen honeybee born worker eggs （worker eggs） were stored through vitrification in liquid nitrogen up to 1 h, and then post-vitrification survival of the vitrified worker eggs in vitro and their hatching rates during maturation in vivo were observed using microscopic and close visual inspections. The procedure of cryopreservation by vitrification included dechorionation with sodium hypochlorite and permeabilization with isopropyl alcohol; equilibration by addition of loading solution （i.e., 25% vitrification storage solution） and dehydration by gradual replacement of loading solution with vitrification storage solution; cooling in liquid nitrogen vapor right before droplet vitrification in liquid nitrogen; and recovery in liquid nitrogen vapor right after storage in liquid nitrogen, thawing at temperature of thawing medium （5% sucrose in TC 100-insect medium） and rehydration by gradual replacement of vitrification storage solution with rehydration solution （5% fetal bovine serum in TC 100-insect medium）. It was found that among the worker eggs experiencing cyropreservation by vitrification, 1.25% of them were successfully passed through the four life stages, viz., egg, larva, pupa, and adult. In summary, it can be inferred that although a majority of worker eggs were dead after cyropreservation by vitrification, a few of them were developed into larvae, pupae, and finally emerged as adults.

Background Our previous study had cloned two glioma cell lines SWOZ1 and SWOZ2 isolated from parental glioma cell line SWO38. The 1,3-bis（2-chloroethyl）-l-nitrosourea （BCNU） resistance of SWOZ1 was higher than that of SWOZ2. Since Oe-methylguanine-DNA methyltransferase （MGMT） was thought to be closely related to BCNU resistance in glioma this study aimed to explore the function of MGMT in glioma resistant to BCNU. Methods A BCNU resistant glioma cell line SWOZ2-BCNU was established. The expression of MGMT was detected in SWOZ1, SWOZ2 and SWOZ2-BCNU. Small interferencing RNA targeting MGMT was used to silence the expression of MGMT in resistant cell lines SWOZ1 and SWOZ2-BCNU. The cytotoxicity of BCNU to these cells was measured using the cell counting kit-8 assay. Statistical analysis was carried out by one-way analysis of variance in statistical package SPSS 13.0. Results The resistance of SWOZl and SWOZ2-BCNU against BCNU was 4.9-fold and 5.3-fold higher than that of SWOZ2. The results of quantitative RT-PCR and Western blotting confirmed that MGMT was both significantly increased in SWOZ1 and SWOZ2-BCNU compared to SOWZ2. After transfection with small interferencing RNA targeting MGMT, a decreased level of MGMT mRNA expression in SWOZ1 and SWOZ2-BCNU for more than 75% compared to negative control was found and confirmed by Western blotting. As a result, the resistance against BCNU was reversed for about 50% both in the BCNU-resistant cell lines SWOZ1 and SWOZ2-BCNU. Conclusions Silencing MGMT with specific small interferencing RNA can reverse the BCNU resistant phenotype in these glioma cell lines. MGMT may play an important role both in intrinsic and acquired BCNU-resistance in glioma.

In this paper,a digital identification method for the extraction of altitudinal belt spectra of montane natural belts is *** the sequential spectra of digital altitudinal belts in mountains at an acceptable temporal frequency and over a large area requires extensive time and work if traditional methods of field investigation are to be *** being the case,often the altitudinal belts of a whole mountain or the belts at a regional scale are represented by single ***,single points obviously cannot accurately reflect the spatial variety of altitudinal *** this context,a digital method was developed to extract the spectra of altitudinal belts from remote sensing data and SRTM DEM in the West Kunlun *** means of the 1km resolution SPOT-4 vegetation 10-day composite NDVI,the horizontal distribution of altitudinal belts were extracted through supervised classification,with a total classification accuracy of 72.23%.Then,a way of twice-scan was used to realize the automatic transition of horizontal maps to vertical *** classification results of remote-sensing data could thus be transformed automatically to sequential spectra of digital altitudinal *** upper and lower lines of the altitudinal belts were then extracted by vertical scanning of the *** between the altitudinal belts based on the montane natural zones concerning vegetation types and the geomorphological altitudinal belts were also *** a tentative method,the digital extraction method presented here is effective at digitally identifying altitudinal belts,and could be helpful in rapid information extraction over large-scale areas.

Tyrosine kinase inhibitors (TKIs) have emerged as the first-line small molecule drugs in many cancer therapies, exerting their effects by impeding aberrant cell growth and proliferation through the modulation of tyrosine kinase-mediated signaling pathways. However, there exists a substantial interindividual variability in the concentrations of certain TKIs and their metabolites, which may render patients with compromised immune function susceptible to diverse infections despite receiving theoretically efficacious anticancer treatments, alongside other potential side effects or adverse reactions. Therefore, an urgent need exists for an up-to-date review concerning the biological matrices relevant to bioanalysis and the sampling methods, clinical pharmacokinetics, and therapeutic drug monitoring of different TKIs. This paper provides a comprehensive overview of the advancements in pretreatment methods, such as protein precipitation (PPT), liquid-liquid extraction (LLE), solid-phase extraction (SPE), micro-SPE (m-SPE), magnetic SPE (MSPE), and vortex-assisted dispersive SPE (VA-DSPE) achieved since 2017. It also highlights the latest analysis techniques such as newly developed high performance liquid chromatography (HPLC) and high-resolution mass spectrometry (HRMS) methods, capillary electrophoresis (CE), gas chromatography (GC), supercritical fluid chromatography (SFC) procedures, surface plasmon resonance (SPR) assays as well as novel nanoprobes-based biosensing techniques. In addition, a comparison is made between the advantages and disadvantages of different approaches while presenting critical challenges and prospects in pharmacokinetic studies and therapeutic drug monitoring. Soil Insect Diversity and Abundance Following Different Fertilizer Treatments on the Loess Plateau of China

The presence of abundant and diverse communities of macro-arthropods is considered an indicator of sustainability in agroecosystems. This study was designed to investigate the effects of different fertilizer treatments on abundance and diversity of insects of arable loess soil on the Loess Plateau of China. These regimes included a control with no fertilizer addition or manure, treatments with application of mineral fertilizers （N, NK, NP, PK, NPK）, treatments with NPK in combination with organic materials such as wheat straw or maize stalk, treatments with two rates of organic manure application; and different crop rotations （Rot. 1： winter wheat summer maize; Rot.2： winter wheat summer maize soybean intercropping; and Rot.3： winter wheat or rapeseed summer maize soybean intercropping）. Soil macro-arthropods were collected from the plough layer （0-20 cm） and sorted by hand after each harvest in June and October 2001 and 2002. A total of 3 132 individuals were collected, from 7 orders and 55 families, dominated by Formicidae （61.72%） and Staphylinidae （14.24%）. The results showed that individuals and groups were significantly influenced by sampling dates, while groups were significantly influenced by the fertilization treatments. Soil insect biodiversity, as determined by the Shannon index, was significantly influenced by fertilization and sampling dates. The abundance of soil insects was positively and significantly correlated with soil moisture content in October 2002. Nitrogen, phosphorus and potassium fertilizers and incorporation of organic materials were favorable factors for abundance and diversity in arable loess soil.

This paper proposes a new primary lazy update protocol, PTCS （Primary Transaction Commit Schedule）. In the PTCS protocol, a serializable primary transaction schedule is generated firstly and then the secondary transactions are committed according to the serializable primary transaction schedule. PTCS protocol can guarantee serializability if the data copy graph contains no directed circles. It can also be ex tended to eliminate all requirements on the data copy graph. Compared to earlier works, PTCS protocol not only imposes a much weaker requirement on the data placement, but also avoids the deadlock caused by transaction waits and extra message overhead. The performance experiments show that the degradation of the performance caused by the replica man- agement of the PTCS protocol is tolerable.
