Autophagy and inflammation in ischemic stroke

Appropriate autophagy has protective effects on ischemic nerve tissue,while excessive autophagy may cause cell death.The inflammatory response plays an important role in the survival of nerve cells and the recovery of neural tissue after ischemia.Many studies have found an interaction between autophagy and inflammation in the pathogenesis of ischemic stroke.This study outlines recent advances regarding the role of autophagy in the post-stroke inflammatory response as follows.(1)Autophagy inhibits inflammatory responses caused by ischemic stimulation through mTOR,the AMPK pathway,and inhibition of inflammasome activation.(2)Activation of inflammation triggers the formation of autophagosomes,and the upregulation of autophagy levels is marked by a significant increase in the autophagy-forming markers LC3-II and Beclin-1.Lipopolysaccharide stimulates microglia and inhibits ULK1 activity by direct phosphorylation of p38 MAPK,reducing the flux and autophagy level,thereby inducing inflammatory activity.(3)By blocking the activation of autophagy,the activation of inflammasomes can alleviate cerebral ischemic injury.Autophagy can also regulate the phenotypic alternation of microglia through the nuclear factor-κB pathway,which is beneficial to the recovery of neural tissue after ischemia.Studies have shown that some drugs such as resveratrol can exert neuroprotective effects by regulating the autophagy-inflammatory pathway.These studies suggest that the autophagy-inflammatory pathway may provide a new direction for the treatment of ischemic stroke.

Integrated transcriptome analysis for the hepatic and jejunal mucosa tissues of broiler chickens raised under heat stress conditions

Background:Heat stress(HS)is one of the most important threats for the current poultry industry.Therefore,many efforts have been made to ameliorate the adverse effect of HS on poultry production;however,physiological and molecular mechanisms pertaining to HS are still limited in poultry.Therefore,the objective of the current study was to investigate functional alterations based on individual and integrated transcriptomes in the liver and jejunal mucosa tissues of broiler chickens exposed to HS conditions.Results:Broiler chickens exposed to HS showed decreased growth performance and increased corticosterone concentrations in the feather.In the transcriptome analysis,the number of differentially expressed genes(DEGs)were identified in the liver and jejunal mucosa by HS conditions.In the liver,genes related to amino acid oxidation,tryptophan metabolism,lipid metabolism,oxidative phosphorylation,and immune responses were altered by HS,which support the reason why heat-stressed poultry had decreased productive performance.In the jejunal mucosa,genes related to defense systems,glutathione metabolism,detoxification of xenobiotics,and immune responses were differently expressed by HS conditions.The integrated transcriptome analysis with DEGs found in the liver and jejunal mucosa showed a considerable connectivity between the core nodes in the constructed networks,which includes glutathione metabolism,xenobiotic metabolism,carbon metabolism,and several amino acid metabolisms.Conclusions:The core network analysis may indicate that increased requirement of energy and amino acids in the jejunal mucosa of broiler chickens exposed to HS conditions is likely compromised by increased oxidation and synthesis of amino acids in the liver.Therefore,our results may provide comprehensive insights for molecular and metabolic alterations of broiler chickens raised under HS conditions,which can aid in the development of the novel strategies to ameliorate the negative effect of HS on poultry productivity and health.

Ungulate bocaparvovirus 4 and rodent bocavirus are different genotypes of the same species of virus

Bocaviruses are associated with many human infectious diseases, such as respiratory tract infections, gastroenteritis, and hepatitis. Rats are known to be reservoirs of bocaviruses, including rodent bocavirus and rat bocavirus.Recently, ungulate bocaparvovirus 4, a known porcine bocavirus, has also been found in rats. Thus, investigating bocaviruses in rats is important for determining the origin of the viruses and preventing and controlling their transmission. To the best of our knowledge, no study to date has investigated bocaviruses in the livers of rats. In this report, a total of 624 rats were trapped in southern China between 2014 and 2017. Liver and serum samples from rats were tested for the prevalence of bocaviruses using PCR. Sequences related to ungulate bocaparvovirus 4and rodent bocavirus were detected in both liver and serum samples. Interestingly, the prevalence of ungulate bocaparvovirus 4(reference strain: KJ622366.1) was higher than that of rodent bocavirus(reference strain:KY927868.1) in both liver(2.24% and 0.64%, respectively) and serum samples(2.19% and 0.44%, respectively).The NS1 regions of ungulate bocaparvovirus 4 and rodent bocavirus related sequences displayed over 84% and88% identity at the nucleic acid and amino acid levels, respectively. Furthermore, these sequences had similar genomic structure, genomic features, and codon usage bias, and shared a common ancestor. These viruses also displayed greater adaptability to rats than pigs. Our results suggested that ungulate bocaparvovirus 4 and rodent bocavirus may originate from rats and may be different genotypes of the same bocavirus species.

目的探讨并分析中药敷贴辅助治疗小儿轮状病毒肠炎的临床效果。方法选取76例轮状病毒肠炎患儿作为本次研究的对象,依照抽签法分为试验组和对照组,各38例。对照组采取常规西药治疗,试验组在对照组基础上实施中药敷贴治疗。对2组的症状消失时间、治疗后7 d大便次数及临床疗效进行比较。结果试验组的症状消失时间短于对照组,治疗后7 d大便次数少于对照组,治疗总有效率高于对照组,差异均有统计学意义(P<0.05)。结论对轮状病毒肠炎患儿采用中药敷贴辅助治疗,能够缩短患儿的临床症状消失时间,减少大便次数,临床效果显著。

Criterion of Semi-Markov Dependent Risk model

A rigorous definition of semi-Markov dependent risk model is given. This model is a generalization of the Markov dependent risk model. A criterion and necessary conditions of semi- Markov dependent risk model are obtained. The results clarify relations between elements among semi-Markov dependent risk model more clear and are applicable for Markov dependent risk model.

Oxidized Black Phosphorus Nanosheets as an Inorganic Antiresorptive Agent

Black phosphorus(BP)is a newly discovered two-dimensional material that has promising applications from bioelectronics to biomedicine.However,facile oxidation of BP leads to changes in surface chemical composition and physical properties,often being referred to as the degradation process of BP.Degradation products of BP nanosheets,namely,oxidized BP nanosheets(oBPNSs),are routinely considered as by-products without many uses.Herein,we found that oBPNSs displayed excellent osteoclastogenesis inhibition effects without impairing cell viability.In contrast to the classic antiresorptive bisphosphonate drugs,oBPNSs showed a different mode of action by suppressing the maturation of osteoclasts.Bone resorption assays,osteoclast actin ring analysis,and tartrate-resistant acid phosphate activity assay results indicated that oBPNSs suppressed receptor activator of nuclear factor-κB(NF-κB)ligand(RANKL)-induced osteoclastogenesis in a dose-and oxidation-dependent manner.Transcriptomic and proteomic analyses indicated that oBPNSs inhibited the activation of the NF-κB signaling pathway and phosphorylation of mitogen-activated protein kinase(MAPK)in differentiated osteoclasts,as confirmed by Western blot analysis.Our results suggest that oBPNSs might be potential antiresorptive nanomaterials to treat osteoporosis.

Adjoining Batch Markov Arrival Processes of a Markov Chain

A batch Markov arrival process（BMAP） X^＊=（N, J） is a 2-dimensional Markov process with two components, one is the counting process N and the other one is the phase process J. It is proved that the phase process is a time-homogeneous Markov chain with a finite state-space, or for short, Markov chain. In this paper,a new and inverse problem is proposed firstly： given a Markov chain J, can we deploy a process N such that the 2-dimensional process X^＊=（N, J） is a BMAP？ The process X^＊=（N, J） is said to be an adjoining BMAP for the Markov chain J. For a given Markov chain the adjoining processes exist and they are not unique. Two kinds of adjoining BMAPs have been constructed. One is the BMAPs with fixed constant batches, the other one is the BMAPs with independent and identically distributed（i.i.d） random batches. The method we used in this paper is not the usual matrix-analytic method of studying BMAP, it is a path-analytic method. We constructed directly sample paths of adjoining BMAPs. The expressions of characteristic（D_k, k = 0, 1, 2· · ·）and transition probabilities of the adjoining BMAP are obtained by the density matrix Q of the given Markov chain J. moreover, we obtained two frontal Theorems. We present these expressions in the first time.

Transcriptome profiling highlights regulated biological processes and type III interferon antiviral responses upon Crimean-Congo hemorrhagic fever virus infection

Crimean-Congo hemorrhagic fever virus(CCHFV)is a biosafety level-4(BSL-4)pathogen that causes Crimean-Congo hemorrhagic fever(CCHF)characterized by hemorrhagic manifestation,multiple organ failure and high mortality rate,posing great threat to public health.Despite the recently increasing research efforts on CCHFV,host cell responses associated with CCHFV infection remain to be further characterized.Here,to better understand the cellular response to CCHFV infection,we performed a transcriptomic analysis in human kidney HEK293 cells by high-throughput RNA sequencing(RNA-seq)technology.In total,496 differentially expressed genes(DEGs),including 361 up-regulated and 135 down-regulated genes,were identified in CCHFV-infected cells.These regulated genes were mainly involved in host processes including defense response to virus,response to stress,regulation of viral process,immune response,metabolism,stimulus,apoptosis and protein catabolic process.Therein,a significant up-regulation of type III interferon(IFN)signaling pathway as well as endoplasmic reticulum(ER)stress response was especially remarkable.Subsequently,representative DEGs from these processes were well validated by RT-qPCR,confirming the RNA-seq results and the typical regulation of IFN responses and ER stress by CCHFV.Furthermore,we demonstrate that not only type I but also type III IFNs(even at low dosages)have substantial anti-CCHFV activities.Collectively,the data may provide new and comprehensive insights into the virus-host interactions and particularly highlights the potential role of type III IFNs in restricting CCHFV,which may help inform further mechanistic delineation of the viral infection and development of anti-CCHFV strategies.

Effect of Jianpi Bushen Sequential Formula on Adjuvant Chemotherapy of Colon Cancer:Study Protocol for a Randomized Controlled Trial

Background The side effects of chemotherapy-induced nausea and vomiting(CINV)and myelosuppression reduce the cancer patients’adherence to chemotherapy.Many Chinese patients choose Chinese medicine(CM)during chemotherapy to reduce side effects;however,the evidence is lacking.The efficacy of a CM herbal treatment protocol,Jianpi Bushen Sequential Formula(健脾补肾续贯方,JBSF)will be evaluated on chemotherapy completion rate among patients with colon cancer.Methods A multi-center double-blind randomized controlled trial(RCT)will be conducted on 400 patients with colon cancer who will receive 8 cycles of adjuvant chemotherapy with oxaliplatin and capecitabine(CAPEOX).Patients will be randomized 1:1 to receive the JBSF or placebo formula.The primary outcome is the overall chemotherapy completion rate.The secondary outcomes include individual chemotherapy completion rate,4-cycle completion rate of chemotherapy,time to treatment failure,relative dose intensity and treatment toxicity.Follow-up visits will be scheduled before every and after last chemotherapy.Discussion This study will provide evidence on whether JBSF can improve the chemotherapy completion rate and reduce side effects among patients with colon cancer.(Trial registration:Clinical Trials.gov,No.NCT03716518)。

A new species of Occidozyga Kuhl and van Hasselt,1822(Anura:Dicroglossidae)from Southern Guangxi,China

A new species of the genus Occidozyga is reported from Southern Guangxi,China,based on morphological characters and molecular analyses.morphological data revealed that the new species,Occidozyga shiwandashanensis sp.nov.,differs from its congeners by a combination of the following characters:(1)medium body size(SVL 25.2−33.8 mm in males,34.9−38.9 mm in females);(2)pale brown dorsum with irregular pale dark spots,especially on head;(3)creamy white ventral surface with brown spots on lateral margin and throat;(4)tongue fleshy,rounded,without notch,(5)tympanum hidden;(6)finger webbing absent and toe webbing full,but fourth toe not webbed to discs;(7)tips of fingers pointed,tips of toes rounded,slightly expanded into disc.The new species occurs on Mt.Shiwandashan and inhabits montane evergreen forests at 500−600 m a.s.l.
