The optical properties of three linear conjugated oligomers （F-P, F-P-F, and P-F-P-F-P）, where phenothiazine （P） and fluorene （F） groups arrange alternately, are investigated. With the enhancement of the π-conjugated system, their absorption and emission bands both gradually red shift, and their two-photon properties are also improved. Meanwhile, their fluorescence dynamic traces are analyzed with continuous rate distribution model, exhibiting that their decay rates gradually accelerate and the rate distribution width become narrower. The quantum chemical calculation offers their molecular structures and transition mechanism, showing that the enhancement of π-conjugated system should be responsible for tile improvement of two-photon properties.

Orthotopic liver transplantation(OLT)is the only proven effective treatment for both end-stage and metabolic liver *** transplantation is a promising alternative for OLT,but the lack of available donor livers has hampered its clinical ***-like cells(HLCs)differentiated from many multi-potential stem cells can help repair damaged liver *** almost suitable cells currently identified for human use are difficult to harvest and involve invasive ***,a novel mesenchymal stem cell derived from human menstrual blood(MenSC)has been discovered and obtained easily and *** this study,we examined whether the MenSCs are able to differentiate into functional HLCs in *** three weeks of incubation in hepatogenic differentiation medium containing hepatocyte growth factor(HGF),fibroblast growth factor-4(FGF-4),and oncostain M(OSM),cuboidal HLCs were observed,and cells also expressed hepatocyte-specific marker genes including albumin(ALB),α-fetoprotein(AFP),cytokeratin 18/19(CK18/19),and cytochrome P450 1A1/3A4(CYP1A1/3A4).Differentiated cells further demonstrated in vitro mature hepatocyte functions such as urea synthesis,glycogen storage,and indocyanine green(ICG)*** intrasplenic transplantation into mice with 2/3 partial hepatectomy,the MenSC-derived HLCs were detected in recipient livers and expressed human ALB *** also showed that MenSC-derived HLC transplantation could restore the serum ALB level and significantly suppressed transaminase activity of liver injury *** conclusion,MenSCs may serve as an ideal,easily accessible source of material for tissue engineering and cell therapy of liver tissues.

为研究2型猪链球菌(SS2) pnp基因的生物学功能,本研究通过基因无痕操作技术构建SS2 05ZYH33pnp基因缺失株Δpnp以及回补株CΔpnp,经PCR和测序鉴定结果表明,正确构建了pnp基因无痕缺失株和回补株。将05ZYH33、Δpnp以及CΔpnp分别在不同温度(28℃、37℃、42℃)培养后测定OD_(600nm)值,绘制各菌株的生长曲线;利用革兰氏染色后经镜检及透射电镜观察各菌株的形态结构;通过滴定微孔平板结晶紫法检测各菌株的生物被膜形成能力;采用接触法分析各菌株的溶血特性;于含不同抗生素的THB平板上培养各菌株,分析各菌株的药物敏感性;于氧化条件与酸性条件下培养各菌株,通过统计存活率评价各菌株的氧化耐受与酸耐受能力。结果显示,缺失株的生长速率明显低于野生株和回补株,且在冷应激条件下的生存能力下降;与野生株和回补株相比,缺失株细胞壁肽聚糖层的结构更加松散、溶血能力下降、生物被膜形成能力下降、对不同类型抗生素的敏感性增强、抗氧化与抗酸能力均下降。上述结果表明pnp基因是SS2的一个重要调节因子,参与调节细菌对外界环境变化的适应能力。本研究为科学防控猪链球菌病以及研发新型抗菌药物奠定基础。

Objective Tick-borne encephalitis virus(TBEV) is an emerging pathogen in Europe and North Asia that causes tick-borne encephalitis(TBE). A simple, rapid method for detecting TBEV RNA is needed to control this disease. Methods A reverse-transcription recombinase-aided amplification(RT-RAA) assay was developed. This assay can be completed in one closed tube at 39℃ within 30 minutes. The sensitivity and specificity of RT-RAA were validated using non-infectious synthetic RNA representing a fragment of the NS5 region of the wild-type(WT) TBEV genome and the Senzhang strain. Additionally, 10 batches of tick samples were used to evaluate the performance of the RT-RAA assay. Results The analytical limit of detection of the assay was 20 copies per reaction of the TBEV synthetic transcript and 3 plaque-forming units(pfu) per reaction of TBEV titers. With the specific assay, no signal due to other arboviruses was observed. Of the 10 batches of tick samples obtained from the Changbai Mountains of China, three were TBEV-positive, which was consistent with the results of the quantitative real-time PCR assay. Conclusion A rapid, highly sensitive, specific, and easy-to-use method was developed for the detection of the TBEV Far-Eastern subtype.

The Bidirectional Effects of Arsenic on miRNA-21: A Systematic Review and Meta-analysis

Objective Arsenic is a metalloid environmental carcinogen involved in the occurrence and development of many cancers. miRNA-21 plays a crucial role in arsenic-induced carcinogenesis. We aimed to elucidate the mechanism by which miRNA-21 influences arsenic-induced cancer. Methods We used meta-analysis of published studies to determine how arsenic induces cancerous cells through miRNA-21. Results Low-dose arsenic exposure（≤ 5 μmol/L） can increase miRNA-21 and phosphorylated signal transducter and activator of transcription 3（pSTAT3） expression, and decrease programmed cell death protein 4（PDCD4） and protein sprouty homolog 1（Spry1） expression. High-dose arsenic exposure（〉 5 μmol/L）, can increase miRNA-21 expression, and decrease Spry1 and E-cadherin expression. Short-term arsenic exposure（≤ 24 h） can increase miRNA-21 and pS TAT3 expression, and decrease PDCD4 expression. Moreover, long-term arsenic exposure（〉 24 h） can increase the miRNA-21, STAT3, and pSTAT3 expression, and decrease PDCD4 expression. We found that activation of miRNA-21 and pSTAT3 were most pronounced following long-term arsenic exposure at low doses, and the effects on PDCD4 expression were most pronounced following short-term arsenic exposure at low doses. miRNA-21 inhibitors increased the expression of tumor suppressor genes PDCD4, PTEN, and Spry1 and miRNA-21-mimics suppressed the expression of these tumor suppressor genes. Conclusion Arsenic can cause cancer by activating miRNA-21 and inhibiting the expression of PDCD4, PTEN, and Spry1.

As a crucial transcription factor for spermatogenesis,GATA-binding protein 4(GATA4)plays important roles in the functioning of Sertoli and Leydig *** knockout of GATA4 in mice results in age-dependent testicular atrophy and loss of ***,whether GATA4 is associated with human azoospermia has not been ***,we analyzed the GATA4 gene by direct sequencing of samples obtained from 184 Chinese men with idiopathic nonobstructive azoospermia(NOA).We identified a missense mutation(c.191G>A,p.G64E),nine single-nucleotide polymorphisms(SNPs),and one rare variant(c.^(*)84C>T)in the 3′untranslated region(UTR).Functional studies demonstrated that the p.G64E mutation did not affect transactivation ability of GATA4 for spermatogenesis-related genes(claudin-11 and steroidogenic acute regulatory protein,Star),and the 3′UTR rare variant c.^(*)84C>T did not generate microRNA-binding sites to repress GATA4 *** our knowledge,this is the first report to investigate the association between GATA4 and azoospermia;our results indicate that mutations in GATA4 may not be pathogenic for NOA in Chinese men.

Dear Editor,Fan et *** shown that whey protein,such as that found in human,bovine,and goat milk,could effectively inhibit SARS-CoV-2 infection,but they did not identify the components in whey protein having this activity.1 Since p-lactoglobulin(P-LG)is the main component of whey protein,we herein tested the anti-SARS-CoV-2 activity of p-LG and 3-hydroxyphthalic anhydride(3HP)-modified P-LG(3HP-P-LG),an active component in anti-human papillomavirus(HPV)biological dressing(JB01-BD)that has been used in clinics since 2013 to block cervical infection of HPV.

环境管控单元是“三线一单”中落实生态环境准入清单的基本载体,其划定结果直接影响清单的编制和管控要求的落实。本研究分析了现有环境管控单元划定方法,总结了行政边界拟合法和要素分区聚类法2种主要划定方法的优缺点,发现拟合法划定结果应用优势明显,但易出现优先保护单元划定偏小、优先保护区所在单元主导功能不一致的问题。本研究结合市、镇2级单元实际案例,提出结合地形地貌和交通网络优化环境管控单元边界的方法,为“三线一单”更新调整提供环境管控单元优化思路。
