Protease-catalyzed Synthesis of Bz-Arg-Gly-Asp-OMe in Full Aqueous Medium

Synthesis of N-benzoyl-argininylglycylasparagine methyl ester（ Bz-Arg-Gly-Asp-OMe）, a precursor tripeptide of Arg-Gly-Asp） was catalyzed by papain under kinetic control, at alkaline pH, in a full aqueous medium. The substrates were N-benzoyl-argininylglycine ethyl ester and asparagine dimethyl ester. An aqueous solution of 0. 1 mol/L KCl/NaOH containing 8 mmol/L EDTA and 2 mmol/L DTT was selected as the reaction medium. The synthesized hydrophilic tripeptide was soluble in the reaction medium during the reaction process, however, the secondary hydro- lysis of the tripeptide product was not considerable. The effects of different factors, including water content, temperature, reaction time, and molar ratio of the substrates, on the yield of Bz-Arg-Gly-Asp-OMe were examined. The optimal reaction conditions were 0. 05 mol/L Bz-Arg-Gly-OEt and 0. 15 mol/L Asp（ -OMe）2·HCl in 0. 1 mol/L KCl/ NaOH solution（pH 8.5）, at 40 ℃, and a reaction time of 60 min, with a maximum conversion yield of 62.4%.

Toll-like receptors(TLRs) are the critical superfamily homologues that initiate sensing of the invasion of pathogens by the Toll pathway. As one of several intracellular nucleic acid-sensing TLRs, TLR13 is activated by an unmethylated motif present in the large ribosomal subunit of bacterial RNA. However, little attention has been paid to the function of TLR13 gene homologue from Laodelphax striatellus(designated as LsToll-13) in the immune response to rice stripe virus(RSV). Herein, LsToll-13 was cloned and characterized using RACE-PCR. Phylogenetic analysis showed that LsToll-13 was clustered with the TLR13 from six insects. Real-time PCR analysis demonstrated that the expression level of LsToll-13 was significantly reduced in L. striatellus with RSV infection compared with that in the naive strain. When the expression of LsToll-13 was significantly up-regulated at 6 h after bacterial infection, the expression of ribonucleoprotein(RNP) indicated that the RSV titer in the host insect was significantly suppressed. Upon knockdown of LsToll-13, using RNA interference(RNAi) in L. striatellus, the expression level of RNP was significantly increased with enhanced RSV accumulation, suggesting that LsToll-13 potentially protects L. striatellus from RSV infection. Taken together, our results indicated that LsToll-13 might be involved in the immune response of L. striatellus to RSV infection, and provided a new insight into further elucidating the molecular mechanisms of complex pathogen-host interactions and integrative pest management.

Greedy Optimization for K-Means-Based Consensus Clustering

Consensus clustering aims to fuse several existing basic partitions into an integrated one; this has been widely recognized as a promising tool for multi-source and heterogeneous data clustering. Owing to robust and high-quality performance over traditional clustering methods, consensus clustering attracts much attention, and much efforts have been devoted to develop this field. In the literature, the K-means-based Consensus Clustering(KCC) transforms the consensus clustering problem into a classical K-means clustering with theoretical supports and shows the advantages over the state-of-the-art methods. Although KCC inherits the merits from K-means,it suffers from the initialization sensitivity. Moreover, the current consensus clustering framework separates the basic partition generation and fusion into two disconnected parts. To solve the above two challenges, a novel clustering algorithm, named Greedy optimization of K-means-based Consensus Clustering(GKCC) is *** by the well-known greedy K-means that aims to solve the sensitivity of K-means initialization, GKCC seamlessly combines greedy K-means and KCC together, achieves the merits inherited by GKCC and overcomes the drawbacks of the precursors. Moreover, a 59-sampling strategy is conducted to provide high-quality basic partitions and accelerate the algorithmic speed. Extensive experiments on 36 benchmark datasets demonstrate the significant advantages of GKCC over KCC and KCC++ in terms of the objective function values and standard deviations and external cluster validity.

Membrane fouling has been investigated by using a polysulfone ultrafiltration membrane with the molecular weight cutoff of 20 kDa to treat crushed coal pressurized gasification wastewater. Under the conditions of different feed pressures, the permeate flux declines and rejection coefficients of pollutants referring to three parameters （total organic carbon （TOC）, chroma and turbidity） were studied. The membrane fouling mechanism was simulated with three classical membrane fouling models. The membrane image and pollutants were analyzed by scanning electron microscopy and gas chromatography-mass spectrography （GC-MS）. The results indicate that the permeate flux decreases with volume reduction factor before reaching a constant value. The rejection coefficients were also measured： fTOC = 70.5%,fc = 84.9% andfa- = 91%. Further analysis shows that the higher the feed pressure is, the sooner the permeate flux reaches constant value and the more sharply the permeate flux declines. Constant flux indicates a nonlinear growth with feed pressure （PF）： when Pr equals 1.2 bar, the mark for the critical flux, slight membrane fouling occurs; when PF exceeds 1.2 bar, cake layer pollution aggravates. Also the rejection coefficients of global pollutant increases slightly with PF, suggesting the possibility of cake compression when PF exceeds 1.2 bar. Through regression analysis, the fouling of polysulfone ultrafiltration membrane could be fitted very well by cake filtration model. The membrane pollutants were identified as phthalate esters and long-chain alkenes by GC-MS, and a certain amount of inorganic pollutants by X-ray photoelectron spectroscopy.

Nephrotic syndrome(NS)is the most common glomerular disease in children,affecting about 7-16 in 100,000 children under 16 years old worldwide[1].Children with NS often experience significant emotional and behavioral problems,especially with frequent relapses,steroid dependence,steroid resistance,or maternal distress[2].Psychological stress may trigger proteinuria in children with steroid-sensitive nephrotic syndrome[3].The presence of NS in a child also causes burden on family members,especially the mother[4],in association with psychological distress.

Cl-containing cerium dioxide(Ce O2) catalysts have been found to exhibit unique catalytic activities. In the present work, using density functional theory calculations with the inclusion of on-site Coulomb correction, we systematically studied the effect of Cl on the physicochemical properties of Ce O2 surfaces by substituting one subsurface O with Cl. The calculated results show that substituting an O atom with a Cl atom results in structural distortion and the reduction of one surface Ce4+ cation to Ce3+. The protruding Ce3+ cation greatly improves the adsorption energy of O2 to produce an active O2- species, and maintains the catalytic oxidation cycle of CO on Ce O2(110). These results may help us obtain a better understanding of Cl-ceria interacting systems and provide some guidance for the design of effective Ce O2-based catalysts.

A new heterometallic complex {[Cu（pic）2（H2O）2][Cd2（Hdipic）2（H2O）2Cl2]}n （Mr = 1007.73） has been synthesized under hydrothermal condition and characterized by single-crystal X-ray diffraction. It crystallizes in triclinic, space group PI^- with a = 5.8397（4）, b = 9.8664（7）, c = 14.1739（10） °A, α= 82.0150（10）, β = 80.4540（10）, γ= 82.3570（10）°, Z= 1, V= 792.47（10）°A^3, Dc = 2.112 g/cm^3,μ（MoKα） = 2.247 mm^-1, F（000） = 495, the final R = 0.0232 and wR = 0.0644 for 2748 observed reflections （I〉 2σ（I））. In the complex, seven-coordinated·Cd ions form a zigzag chain based on the alternated dinuclear Cd units. The neutral [Cu（pic）2（H2O）2] units are located at the centers of the inter space and fasten to the 1-D chain by hydrogen bonds.
