Influenza causes seasonal outbreaks yearly and unpredictable pandemics with high morbidity and mortality rates.Despite significant efforts to address influenza,it remains a major threat to human public health.This issue is partially due to the lack of antiviral drugs with potent antiviral activity and broad reactivity against all influenza virus strains and the rapid emergence of drug-resistant variants.Moreover,designing a universal influenza vaccine that is sufficiently immunogenic to induce universal antibodies is difficult.Some novel epitopes hidden in the hemagglutinin(HA)trimeric interface have been discovered recently,and a number of antibodies targeting these epitopes have been found to be capable of neutralizing a broad range of influenza isolates.These findings may have important implications for the development of universal influenza vaccines and antiviral drugs.In this review,we focused on the antibodies targeting these newly discovered epitopes in the HA domain of the influenza virus to promote the development of universal anti-influenza antibodies or vaccines and extend the discovery to other viruses with similar conformational changes in envelope proteins.

Active control of flow past an elliptic cylinder using an artificial neural network trained by deep reinforcement learning

The active control of flow past an elliptical cylinder using the deep reinforcement learning(DRL)method is conducted.The axis ratio of the elliptical cylinderΓvaries from 1.2 to 2.0,and four angles of attackα=0°,15°,30°,and 45°are taken into consideration for a fixed Reynolds number Re=100.The mass flow rates of two synthetic jets imposed on different positions of the cylinderθ1andθ2are trained to control the flow.The optimal jet placement that achieves the highest drag reduction is determined for each case.For a low axis ratio ellipse,i.e.,Γ=1.2,the controlled results atα=0°are similar to those for a circular cylinder with control jets applied atθ1=90°andθ2=270°.It is found that either applying the jets asymmetrically or increasing the angle of attack can achieve a higher drag reduction rate,which,however,is accompanied by increased fluctuation.The control jets elongate the vortex shedding,and reduce the pressure drop.Meanwhile,the flow topology is modified at a high angle of attack.For an ellipse with a relatively higher axis ratio,i.e.,Γ1.6,the drag reduction is achieved for all the angles of attack studied.The larger the angle of attack is,the higher the drag reduction ratio is.The increased fluctuation in the drag coefficient under control is encountered,regardless of the position of the control jets.The control jets modify the flow topology by inducing an external vortex near the wall,causing the drag reduction.The results suggest that the DRL can learn an active control strategy for the present configuration.

Studying the spatiotemporal variations in ecosystem services and their interrelationships on the Loess Plateau against the background of the gully control and land consolidation(GCLC)project has significant implications for ecological protection and quality development of the Yellow River Basin.Therefore,in this study,we took Yan'an City,Shaanxi Province of China,as the study area,selected four typical ecosystem services,including soil conservation service,water yield service,carbon storage service,and habitat quality service,and quantitatively evaluated the spatiotemporal variation characteristics and trade-offs and synergies of ecosystem services from 2010 to 2018 using the Integrated Valuation of Ecosystem Services and Trade-offs(InVEST)model.We also analysed the relationship between the GCLC project and regional ecosystem service changes in various regions(including 1 city,2 districts,and 10 counties)of Yan'an City and proposed a coordinated development strategy between the GCLC project and the ecological environment.The results showed that,from 2010 to 2018,soil conservation service decreased by 7.76%,while the other three ecosystem services changed relatively little,with water yield service increasing by 0.56% and carbon storage service and habitat quality service decreasing by 0.16% and 0.14%,respectively.The ecological environment of Yan'an City developed in a balanced way between 2010 and 2018,and the four ecosystem services showed synergistic relationships,among which the synergistic relationships between soil conservation service and water yield service and between carbon storage service and habitat quality service were significant.The GCLC project had a negative impact on the ecosystem services of Yan'an City,and the impact on carbon storage service was more significant.This study provides a theoretical basis for the scientific evaluation of the ecological benefits of the GCLC project and the realization of a win-win situation between food security and ecological security.

The etiology of chronic kidney disease(CKD)is complex and diverse,which could be briefly categorized to glomerular-or tubularoriginated.However,the final outcomes of CKD are mainly glomerular sclerosis,endothelial dysfunction and injury,and chronic inflammation.Thus,targeted delivery of drugs to the glomeruli in order to ameliorate glomerular endothelial damage may help alleviate CKD and help enrich our knowledge.The herb tripterygium wilfordii shows therapeutic effect on kidney disease,and celastrol(CLT)is one of its active ingredients but with strong toxicity.Therefore,based on the unique structure and pathological characteristics of the glomerulus,we designed a targeted delivery system named peptides coupled CLT-phospholipid lipid nanoparticles(PC-PLNs)to efficiently deliver CLT to damaged endothelial cells and podocytes in the glomerulus for CKD treatment and research.PC-PLNs could effectively inhibit inflammation,reduce endothelial damage,alleviate CKD severity,and reduce the toxicity of CLT.We also studied the mechanism of CLT in the treatment of nephropathy and found that CLT can increase the level of NO by increasing eNOS while inhibiting the expression of VCAM-1,thus provides an anti-inflammatory effect.Therefore,our study not only offered an efficient CKD drug formulation for further development,but also provided new medical knowledge about CKD.

Solvent-free nanofluids hold promise for many technologically significant applications.The liquid-like behavior,a typical rheological property of solvent-free nanofluids,has aroused considerable interests.However,there has been still lack of efficient methods to predict and control the liquid-like behavior of solvent-free nanofluids.In this paper,we propose a semi-discrete dynamic system with stochastic excitation describing the temperature change effects on the rheological property of multiwall carbon nanotubes(MWCNTs)modified by grafting sulfonic acid terminated organosilanes as corona and tertiary amine as canopy,which is a typical covalent-type solvent-free nanofluid system.The vibration of the grafting branches is simulated by employing a structure-preserving approach,and the shear force of grafting branches at the fixed end is computed subsequently.By taking the shear forces as an excitation acting on the MWCNTs,the axial motion of the MWCNTs is solved with the 7-point Gauss-Kronrod quadrature rule.The critical temperature associated with the appearance of the liquid-like behavior as well as the upper bound of the moving speed of the modified MWCNTs is determined,which can be used to predict and control the liquid-like behavior of the modified MWCNTs in engineering applications.

Microspheres can break the diffraction limit and magnify nano-structure imaging,and with its advantages of low cost and label-free operation,microsphere-assisted imaging has become an irreplaceable tool in the life sciences and for precision measurements.However,the tiny size and limited imaging field of traditional solid microspheres cause difficulties when imaging large sample areas.Alternatively,droplets have similar properties to those of microspheres,with large surface curvature and refractive-index difference from the surrounding environment,and they can also serve as lenses to focus light for observation and imaging.Previous work has shown that droplets with controllable size can be generated using an optical tweezer system and can be driven by optical traps to move precisely like solid microspheres.Here,a novel microdroplet-assisted imaging technology based on optical tweezers is proposed that better integrates the generation,manipulation,and utilization of droplets.

Community-Based Management and Treatment Services for Psychosis-China, 2019

Introduction:Community-based services for psychosis are an important part of mental health services in China.We analyzed community-based follow-up and treatment services for psychosis in China in 2019 to provide evidence for policymaking and services delivering.Methods:Data from the National Information System for Psychosis were used to analyze usage of the information system and the registration,management,and treatment situation of patients with psychosis in China in 2019 and compared the results of 2019 with that of 2018.Results:In 2019,100%of cities and counties used the information system and 6,230,157 patients were registered.In 2019,there were 5,944,724 registered patients(94.05%)in China under community-based follow-up services and 5,375,252 patients(85.04%)had regular follow-ups;5,295,657 patients(83.78%)were treated by taking antipsychotics and 3,354,251(53.07%)patients took medication regularly;4,974,314 patients at home(87.81%)were in stable condition;and there was a significant difference in patients’stable condition and medication-taking rates among eastern,central,and western regions(p<0.05).Compared with 2018,the nationwide regular management rate,medication-taking rate,regular medication-taking rate,and patients’stable rate increased by 2.35%,2.48%,11.29%,and 7.21%respectively.Conclusions and Implications for Public Health Practice:Compared with 2018,the level of management and treatment improved significantly but still needs to be further strengthened,especially in western China.

High density and uniform distribution of the gold nanoparticles functionalized single-stranded DNA modified reduced graphene oxide nanocomposites were obtained by non-covalent interaction.The positive gold nanoparticles prepared by phase inversion method exhibited good dimensional homogeneity and dispersibility,which could readily combine with single-stranded DNA modified reduced graphene oxide nanocomposites by electrostatic interactions.The modification of single-stranded DNA endowed the reduced graphene oxide with favorable biocompatibility and provided the preferable surface with negative charge for further assembling of gold nanoparticles to obtain gold nanoparticles/single-stranded DNA modified reduced graphene oxide nanocomposites with better conductivity,larger specific surface area,biocompatibility and electrocatalytic characteristics.The as-prepared nanocomposites were applied as substrates for the construction of cholesterol oxidase modified electrode and well realized the direct electron transfer between the enzyme and electrode.The modified gold nanoparticles could further catalyze the products of cholesterol oxidation catalyzed by cholesterol oxidase,which was beneficial to the enzyme-catalyzed reaction.The as-fabricated bioelectrode exhibited excellent electrocatalytic performance for the cholesterol with a linear range of 7.5–280.5μmol·L^(−1),a low detection limit of 2.1μmol·L^(−1),good stability and reproducibility.Moreover,the electrochemical biosensor showed good selectivity and acceptable accuracy for the detection of cholesterol in human serum samples.

BACKGROUND:Chlorfenapyr is used to kill insects that are resistant to organophosphorus insecticides.Chlorfenapyr poisoning has a high mortality rate and is difficult to treat.This article aims to review the mechanisms,clinical presentations,and treatment strategies for chlorfenapyr poisoning.DATA RESOURCES:We conducted a review of the literature using PubMed,Web of Science,and SpringerLink from their beginnings to the end of October 2023.The inclusion criteria were systematic reviews,clinical guidelines,retrospective studies,and case reports on chlorfenapyr poisoning that focused on its mechanisms,clinical presentations,and treatment strategies.The references in the included studies were also examined to identify additional sources.RESULTS:We included 57 studies in this review.Chlorfenapyr can be degraded into tralopyril,which is more toxic and reduces energy production by inhibiting the conversion of adenosine diphosphate to adenosine triphosphate.High fever and altered mental status are characteristic clinical presentations of chlorfenapyr poisoning.Once it occurs,respiratory failure occurs immediately,ultimately leading to cardiac arrest and death.Chlorfenapyr poisoning is diflcult to treat,and there is no specific antidote.CONCLUSION:Chlorfenapyr is a new pyrrole pesticide.Although it has been identified as a moderately toxic pesticide by the World Health Organization(WHO),the mortality rate of poisoned patients is extremely high.There is no specific antidote for chlorfenapyr poisoning.Therefore,based on the literature review,future efforts to explore rapid and effective detoxification methods,reconstitute intracellular oxidative phosphorylation couplings,identify early biomarkers of chlorfenapyr poisoning,and block the conversion of chlorfenapyr to tralopyril may be helpful for emergency physicians in the diagnosis and treatment of this disease.
