Global demand for farm animals and their meat products i.e.,pork,chicken and other livestock meat,is steadily incresing.With the ongoing life science research and the rapid development of biotechnology,it is a great opportunity to develop advanced molecular breeding markers to efficiently improve animal meat production traits.Hippo is an important study subject because of its crucial role in the regulation of organ size.In recent years,with the increase of research on Hippo signaling pathway,the integrative application of multi-omics technologies such as genomics,transcriptomics,proteomics,and metabolomics can help promote the in-depth involvement of Hippo signaling pathway in skeletal muscle development research.The Hippo signaling pathway plays a key role in many biological events,including cell division,cell migration,cell proliferation,cell differentiation,cell apoptosis,as well as cell adhesion,cell polarity,homeostasis,maintenance of the face of mechanical overload,etc.Its influence on the development of skeletal muscle has important research value for enhancing the efficiency of animal husbandry production.In this study,we traced the origin of the Hippo pathway,comprehensively sorted out all the functional factors found in the pathway,deeply analyzed the molecular mechanism of its function,and classified it from a novel perspective based on its main functional domain and mode of action.Our aim is to systematically explore its regulatory role throughout skeletal muscle development.We specifically focus on the Hippo signaling pathway in embryonic stem cell development,muscle satellite cell fate determination,myogenesis,skeletal muscle meat production and organ size regulation,muscle hypertrophy and atrophy,muscle fiber formation and its transformation between different types,and cardiomyocytes.The roles in proliferation and regeneration are methodically summarized and analyzed comprehensively.The summary and prospect of the Hippo signaling pathway within this article will provide ideas for further improving

Ag-doped CNT/HAP nanohybrids in a PLLA bone scaffold show significant antibacterial activity

Bacterial infection is a major problem following bone implant surgery.Moreover,poly-l-lactic acid/carbon nanotube/hydroxyapatite(PLLA/CNT/HAP)bone scaffolds possess enhanced mechanical properties and show good bioactiv-ityregardingbonedefectregeneration.Inthisstudy,wesynthesizedsilver(Ag)-dopedCNT/HAP(CNT/Ag-HAP)nanohybrids via the partial replacing of calcium ions(Ca2+)in the HAP lattice with silver ions(Ag+)using an ion doping technique under hydrothermal conditions.Speciﬁcally,the doping process was induced using the special lattice structure of HAP and the abundant surface oxygenic functional groups of CNT,and involved the partial replacement of Ca2+in the HAP lattice by doped Ag+as well as the in situ synthesis of Ag-HAP nanoparticles on CNT in a hydrothermal environment.The result-ing CNT/Ag-HAP nanohybrids were then introduced into a PLLA matrix via laser-based powder bed fusion(PBF-LB)to fabricate PLLA/CNT/Ag-HAP scaffolds that showed sustained antibacterial activity.We then found that Ag+,which pos-sesses broad-spectrum antibacterial activity,endowed PLLA/CNT/Ag-HAP scaffolds with this activity,with an antibacterial effectiveness of 92.65%.This antibacterial effect is due to the powerful effect of Ag+against bacterial structure and genetic material,as well as the physical destruction of bacterial structures due to the sharp edge structure of CNT.In addition,the scaffold possessed enhanced mechanical properties,showing tensile and compressive strengths of 8.49 MPa and 19.72 MPa,respectively.Finally,the scaffold also exhibited good bioactivity and cytocompatibility,including the ability to form apatite layers and to promote the adhesion and proliferation of human osteoblast-like cells(MG63 cells).

Seismic performance of HWBBF considering different design methods and structural heights

Previous research has shown that using buckling-restrained braces(BRBs)at hinged wall(HW)base(HWBB)can effectively mitigate lateral deformation of steel moment-resisting frames(MRFs)in earthquakes.Forcebased and displacement-based design methods have been proposed to design HWBB to strengthen steel MRF and this paper comprehensively compares these two design methods,in terms of design steps,advantages/disadvantages,and structure responses.In addition,this paper investigates the building height below which the HW seismic moment demand can be properly controlled.First,3-story,9-story,and 20-story steel MRFs in the SAC project are used as benchmark steel MRFs.Secondly,HWs and HWBBs are designed to strengthen the benchmark steel MRFs using force-based and displacement-based methods,called HWFs and HWBBFs,respectively.Thirdly,nonlinear time history analyses are conducted to compare the structural responses of the MRFs,HWBBFs and HWFs in earthquakes.The results show the following.1)HW seismic force demands increase as structural height increases,which may lead to uneconomical HW design.The HW seismic moment demand can be properly controlled when the building is lower than nine stories.2)The displacement-based design method is recommended due to the benefit of identifying unfeasible component dimensions during the design process,as well as better achieving the design target displacement.

Rational design of unrestricted pRN1 derivatives and their application in the construction of a dual plasmid vector system for Saccharolobus islandicus

Saccharolobus islandicus REY15A represents one of the very few archaeal models with versatile genetic tools,which include efficient genome editing,gene silencing,and robust protein expression systems.However,plasmid vectors constructed for this crenarchaeon thus far are based solely on the pRN2 cryptic plasmid.Although this plasmid coexists with pRN1 in its original host,early attempts to test pRN1-based vectors consistently failed to yield any stable host-vector system for Sa.islandicus.We hypothesized that this failure could be due to the occurrence of CRISPR immunity against pRN1 in this archaeon.We identified a putative target sequence in orf904 encoding a putative replicase on pRN1(target N1).Mutated targets(N1a,N1b,and N1c)were then designed and tested for their capability to escape the host CRISPR immunity by using a plasmid inter-ference assay.The results revealed that the original target triggered CRISPR immunity in this archaeon,whereas all three mutated targets did not,indicating that all the designed target mutations evaded host immunity.These mutated targets were then incorporated into orf904 individually,yielding corresponding mutated pRN1 backbones with which shuttle plasmids were constructed(pN1aSD,pN1bSD,and pN1cSD).Sa.islandicus transformation revealed that pN1aSD and pN1bSD were functional shuttle vectors,but pN1cSD lost the capability for replication.These results indicate that the missense mutations in the conserved helicase domain in pN1c inactivated the replicase.We further showed that pRN1-based and pRN2-based vectors were stably maintained in the archaeal cells either alone or in combination,and this yielded a dual plasmid system for genetic study with this important archaeal model.

In this paper,a novel ternary eutectic salt Na_(2)CO_(3)-Li_(2)CO_(3)-LiF was designed and investigated for concentrated solar power(CSP).The FactSage software was used to predict the composition and eutectic point of Na_(2)CO_(3)-Li_(2)CO_(3)-LiF.The microstructure,thermophysical properties,and thermal stability of eutectic salts were experimentally measured using various analytical methods.With a mass ratio of 57%:32%:11%,the eutectic salt exhibited excellent thermal storage properties with a fusion enthalpy of 413 J/g and a melting point of 426.8℃.The excellent thermal stability of the eutectic salt was reflected by a weight loss of only 0.8% at 600℃.

Effect of fermented Rosa roxburghii Tratt fruit juice on gut microbiota in a dextran sulfate sodium-induced colitis mouse model

This study mainly investigated the regulatory effect of Rosa roxburghii Tratt fruit juice fermented by Lacticaseibacillus paracasei SR10-1(LAB-RRTJ)on modulating gut microbiota in dextran sulfate sodium(DSS)-induced ulcerative colitis in mice.Compared to control group,DSS induction decreased body weight of mice,indexes of Shannon,Simpson,Chao1 and Faith_pd,and increased disease activity index(DAI)and levels of interleukin 1β(IL-1β),IL-6,tumor necrosis factorα(TNF-α)and interferon-γ(IFN-γ);And this induction also led to an increase in Proteobacteria,Verrucomicrobia and Actinobacteria at phylum level,harmful bacterial species richness at genus level,and relative richness of S.sciuri,Desulfovibrio C21_c20,R.gnavus and Akkermansia muciniphila at species level,and a decrease in Firmicutes at phylum level and relative richness of B.acidifaciens in mice.LAB-RRTJ increased body weight of mice with DSS induced ulcerative colitis(UC)and indexes of Shannon,Simpson,Chao1 and Faith_pd,reduced DAI and the content of four infl ammatory factors and improved gut microbiota imbalance in DSS induced UC mice.Besides,the number of operational taxonomic units(OTUs)increased,α-diversity andβ-diversity were restored and similar to those in mice in the control group after LAB-RRTJ treatment.Compared with the positive drug treatment group,LAB-RRTJ has a better effect on regulating gut microbiota diversity in colitis mice.Correlation analysis showed that infl ammatory factors were positively correlated with harmful bacteria and negatively correlated with beneficial bacteria which commonly found in some colitis mice.Taken together,our study demonstrated that LAB-RRTJ could alleviate DSS-induced colitis in mice through the modulation of infl ammatory cytokines and gut microbiota composition.

Design and experiment of a Chinese chive harvester

Recognizing the labor intensity of current domestic Chinese chive harvesting and the need for its mechanized harvesting,a self-propelled electric Chinese chive harvester is designed.The machine is composed of a control system,supporting mechanism,cutting parts,conveying and clamping mechanisms,frame,and collection system.It can complete Chinese chive feeding,cutting,transmission,and binning operations at one time.It has a cutting width of 0.25 m and wheelbases of 260 mm and 460 mm.Two 12 V,42 Ah lead-acid batteries were used in series as the power supply.The mechanical properties of Chinese chive were analyzed,and shear and compression tests were carried out to determine that the average value of the maximum shear force on the chives is 129 N.Up to a compressive force of 225 N,Chinese chives are not damaged.According to the physical characteristics of Chinese chives and the requirements of the harvesting operations,design and calculation of the key components of the harvester’s transmission mechanism and the strength of the whole machine frame were completed.An embedded control system and Cortex-M3 microprocessor were used to control the harvester and realize its functions of rolling and harvesting while regulating its speed and protecting its motor.Using the Box-Behnken test design method,the machine’s operating speed,cutter speed,and cutter height were selected as influencing factors,and the lodging rate and loss rate were evaluated as a test.A mathematical regression model of influencing factors and evaluation indexes was established to analyze the influence of these factors and indexes and optimize test parameters.The optimal parameter combination is determined to be:operating speed,1.1 km/h;cutter speed,220 r/min;cutter height 2.0 cm.The parameters were verified from field tests and the results showed that under the optimized parameter combination,the average lodging rate was 3.96% and the average loss rate was 3.23%,indicating that the Chinese chive harvester meets the agronomic requirements of Chinese chiv

A novel live attenuated vaccine candidate protects chickens against subtype B avian metapneumovirus

Avian metapneumovirus(aMPV) is a highly contagious pathogen that causes acute upper respiratory tract diseases in chickens and turkeys, resulting in serious economic losses. Subtype B aMPV has recently become the dominant epidemic strain in China. We developed an attenuated aMPV subtype B strain by serial passaging in Vero cells and evaluated its safety and efficacy as a vaccine candidate. The safety test showed that after the 30th passage, the LN16-A strain was fully attenuated, as clinical signs of infection and histological lesions were absent after inoculation.The LN16-A strain did not revert to a virulent strain after five serial passages in chickens. The genomic sequence of LN16-A differed from that of the parent wild-type LN16(wtLN16) strain and had nine amino acid mutations. In chickens, a single immunization with LN16-A induced robust humoral and cellular immune responses, including the abundant production of neutralizing antibodies, CD4^(+) T lymphocytes, and the Th1(IFN-γ) and Th2(IL-4 and IL-6)cytokines. We also confirmed that LN16-A provided 100% protection against subtype B aMPV and significantly reduced viral shedding and turbinate inflammation. Our findings suggest that the LN16-A strain is a promising live attenuated vaccine candidate that can prevent infection with subtype B aMPV.

就地固化联合复合地基对堆载邻近桩基的影响

路桥并线路基修筑引起既有桥桩产生挠曲、侧向位移,严重时可能会造成重大坍塌事故的发生。结合工程现场试验,针对道路施工对邻近桥桩影响问题提出就地固化联合复合地基加固新方法,联合数值模型分析就地固化深度、固化土弹性模量和复合地基相关因素对邻近桩基的保护效果。研究结果表明:搅拌桩加固影响深度为1.5倍桩长左右,素混凝土桩施工会对被动桩在0.75倍桩长左右处产生最大水平位移。就地固化(2.0 m)+水泥搅拌桩法(1.8 m)可极大减小被动桩浅层水平位移,保护桥桩的安全。当固化土水泥掺量为5%~7%、水泥搅拌桩水泥掺量为12%~18%时,提高水泥掺量对减小被动桩水平位移有着明显的作用。同时,将普通褥垫层水泥搅拌桩替换为就地固化+水泥搅拌桩复合地基,桩身最大负弯矩减小近16.6%。

Flexural behavior of high-strength,steel-reinforced,and prestressed concrete beams

To study the flexural behavior of prestressed concrete beams with high-strength steel reinforcement and high-strength concrete and improve the crack width calculation method for flexural components with such reinforcement and concrete,12 specimens were tested under static loading.The failure modes,flexural strength,ductility,and crack width of the specimens were analyzed.The results show that the failure mode of the test beams was similar to that of the beams with normal reinforced concrete.A brittle failure did not occur in the specimens.To further understand the working mechanism,the results of other experimental studies were collected and discussed.The results show that the normalized reinforcement ratio has a greater effect on the ductility than the concrete strength.The cracking-and peak-moment formulas in the code for the design of concrete(GB 50010-2010)applied to the beams were both found to be acceptable.However,the calculation results of the maximum crack width following GB 50010-2010 and EN 1992-1-1:2004 were considerably conservative.In the context of GB 50010-2010,a revised formula for the crack width is proposed with modifications to two major factors:the average crack spacing and an amplification coefficient of the maximum crack width to the average spacing.The mean value of the ratio of the maximum crack width among the 12 test results and the relative calculation results from the revised formula is 1.017,which is better than the calculation result from GB 50010-2010.Therefore,the new formula calculates the crack width more accurately in high-strength concrete and high-strength steel reinforcement members.Finally,finite element models were established using ADINA software and validated based on the test results.This study provides an important reference for the development of high-strength concrete and highstrength steel reinforcement structures.
