Comparative Response of SC CAKI-1 Renal Tumor to Treatment with Doxorubicin HCl and Doxorubicin Orotate

Background: Doxorubicin (DOX) is an effective treatment for many cancers across the age spectrum, but its therapeutic potential is limited because of dose-dependent relation to both progressive and irreversible cardiomyopathy leading to congestive heart failure. While decreases in cardiotoxicity have been reported with liposomal doxorubicin, the long-term cardiac effects are not known. Orotate salts of cytotoxic drugs have been shown to confer antitumor effects with a better safety profile than unconverted drug, and therefore may offer an improved approach to cancer treatment. Materials and Methods: Male, athymic NCr-nu/nu mice with subcutaneously implanted CAKI-1 human renal tumor xenografts were treated with DOX and its orotate salt (DOX-O) to evaluate antitumor activity, measured by median tumor mass doubling time and tumor weight. Nontumored male, athymic NCr-nu/nu mice were treated with DOX, DOX-O and liposomal doxorubicin formulations to determine DOX concentration in liver and heart;and to evaluate their effect on body weight. Non-tumored female, athymic NCr-nu/nu mice were treated with daunorubicin and daunorubicin orotate to evaluate tolerance. Results: DOX and DOX-O exhibited significant, similar levels of antitumor activity. Mice treated with DOX-O had a lower percentage body weight loss. In the animals treated with DOX, DOX-O, or liposomal doxorubicin, liposomal doxorubicin was associated with the lowest percentage of body weight loss, but the highest concentration of DOX in heart. In daunorubicin tolerance experiments, animals showed a better tolerance for daunorubicin orotate as measured by a smaller percentage change in body weight. Conclusions: DOX-O is effective as an antitumor therapy and may offer a less toxic alternative to DOX for maintaining therapy. The lower percentage of body weight loss in animals treated with DOX-O and daunorubicin orotate is a measure of improved tolerance and may translate into better patient outcomes.

Thermally Dependent Behaviour of Cold-Blooded Animals: Overcoming Two Temperature Barriers on the Way to Warm-Blooded

Background: The temperature preferences of cold-blooded animals are different for different groups of animals and are closely related to their evolutionary status and level of adaptive capabilities. The range of preferred temperatures for reptiles (28°C - 32°C) coincides with the zone of temperature-dependent increase in the rate of the M-cholinergic reaction in the brain, which was previously found in warm-blooded neurons. The growth of the M-cholinergic process contributes to the transition of adaptive behavior to a higher level. Of the cold-blooded, only reptiles actively use thermoregulatory behavior as a tool to achieve a temperature optimum. This paper is devoted to the study of the behavior of red-eared turtles aimed at reaching the range of preferred temperatures. Temperature conditions necessary for the survival of different groups of cold-blooded animals are compared and the reason for temperature preferences is discussed. Methods: The behavior of turtles was studied under conditions of variable solar radiation and in a 3-section terrarium with gradual temperature ranges: 23°C - 24°C;28°C - 31°C and 45°C - 50°C. Results: It was found that prolonged (up to 1.5 hours) heating at a temperature of 45°C - 50°C was the best way to achieve the preferred temperatures. This method of heating results in increasing the temperature of the shell to an average of 37°C, followed by a very slow decrease during 4 - 6 hours within the limits that closely coincided with the preferred level. Conclusion: The results obtained demonstrate that the main function of the turtle shell is to accumulate heat and keep it for a long time in a variable climate. The preferred temperature achieved in this case is necessary for the temperature-dependent transition of the rate of the M-cholinergic reaction in the brain to a higher level, which creates more significant adaptive capabilities. 城市固体废物(MSW)的收集、回收和处置过程是世界范围内的一个热点问题。问题主要在于对环境的负面影响和对自然资源的不合理利用。俄罗斯城市生活垃圾的收集、回收和处置制度不完善,家庭固体废物的初级分类也完全缺失。本文旨在设计一个莫斯科市的回收物流网络模型,根据垃圾收集点和处理点的位置,确定分拣中心在运输成本方面的最优位置分配。为解决这一问题,对城市现有的二次材料加工和回收中心的数量、类型、容量和位置进行了调查。为了确定废物收集和回收中心的最佳位置,有必要确定适合处理的固体废物数量,建立分类方法,以及储存和运输方法。对废物的性质、分类和储存有一个清晰的了解,将使计算后续处理和处置的最佳解决方案成为可能。城市生活垃分拣废物统的实施,不仅减少了对环境的负面影响,而且利用二次材料带来了经济效益。

In well stimulation treatments using hydrochloric acid,undesirable water-in-oil emulsion and acid sludge may produce and then cause operational problems in oil field *** processes intensify in the presence of Fe(Ⅲ),which are from the corroded surfaces of field equipment and/or iron-bearing minerals of the oil *** order to understand the reasons of the stability of acid emulsions,acid emulsions were prepared by mixing crude oil emulsion with 15% hydrochloric acid solutions with and without Fe(Ⅲ) and then separated into free and upper(water free) and intermediate(with water) *** is assumed that the oil phase of the free and upper layers contains the compounds which do not participate in the formation of acid emulsions,and the oil phase of the intermediate layers contains components involved in the formation of oil/acid *** composition of the oil phase of each layer of the emulsions was *** is found that the asphaltenes with a high content of sulfur,oxygen and metals as well the flocculated material of protonated non-polar oil components are concentrated at the oil/acid *** addition to the above,in the presence of Fe(Ⅲ) the Fe(Ⅲ)-based complexes with polar groups of asphaltenes are formed at the acid/oil interface,contributing to the formation of armor films which enhance the emulsion stability.

The CELLULOSE SYNTHASE-LIKE C （CSLC） family is an ancient lineage within the CELLULOSE SYNTHASE/CELLULOSE SYNTHASE-LIKE （CESA/CSL） polysaccharide synthase superfamily that is thought to have arisen before the divergence of mosses and vascular plants. As studies in the flowering plant Arabidopsis have suggested synthesis of the （1,4）-β-glucan backbone of xyloglucan （XyG）, a wall polysaccharide that tethers adjacent cellulose microfibrils to each other, as a probable function for the CSLCs, CSLC function was investigated in barley （Hordeum vulgate L.）, a species with low amounts of XyG in its walls. Four barley CSLCgenes were identified （designated HvCSLC1-4）. Phylogenetic analysis reveals three well supported clades of CSLCs in flowering plants, with barley having representatives in two of these clades. The four barley CSLCs were expressed in various tissues, with in situ PCR detecting transcripts in all cell types of the coleoptile and root, including cells with primary and secondary cell walls. Co-expression analysis showed that HvCSLC3 was coordinately expressed with putative XyG xylosyltransferase genes. Both immuno-EM and membrane fractionation showed that HvCSLC2 was located in the plasma membrane of barley suspension-cultured cells and was not in internal membranes such as endoplasmic reticulum or Golgi apparatus. Based on our current knowledge of the sub-cellular locations of polysaccharide synthesis, we conclude that the CSLC family probably contains more than one type of polysaccharide synthase.

Genetic structure and diversity of Ukrainian red clover cultivars revealed by microsatellite markers

Polymorphism of microsatellite loci of Ukrainian red clover cultivars has been studied. 87 microsatellite alleles, which occurred in different combinations, were identified. The number of alleles ranged from 7 to 10. Microsatellite allele distribution showed that 15 alleles were common for all the red clover cultivars (17.2%). The red clover cultivars were represented by homozygous and heterozygous genotypes. The observed and expected heterozygosity ranged from 0.067 to 0.269 and from 0.225 to 0.807, respectively. An analysis of molecular variance revealed that the largest proportion of variation (68.5%) resided at the intrapopulation level. Differentiation of the Ukrainian cultivars was moderately expressed (FST = 0.07).

Improvement of Forage Production Sustainability in the Leningrad Region under Climate Change

The increase of probability and depth of weather anomalies, which is important characteristic of climate change process, leads to higher risk of loss of perennial grasses forage yield, rise of price volatility on forage grain market. Increase of sustainability of dairy production supposes development of adapted to new weather conditions technologies of forage production, which are characterized by different investment and current costs. It generates a need of economic evaluation and comparative analysis of technologies taking into account climate change forecast and need of sustainable forage production guarantee. Purpose of the research project is the development of approaches to assessing adaptive technologies in forage production, increasing of dairy farming sustainability to changes in the external environment, including climate change. We assessed changes in forage production technologies in the Leningrad region of Russia, analyzed the implications of these changes for production and financial performance of dairy farming, proposed and tested methods for determination of the optimum harvesting time of perennial grasses. Special attention is paid to the long-term effects of changing the structure of cows＇ diet--shortening the productive use of animals, which reduces the efficiency of dairy herd use and limits the possibility of expanded reproduction. The primary method of the study is the analysis of milk production statistics in the Leningrad region and the Russian Federation. As a result of the study, the necessity to improve the production and investment planning quality, as well as the use of forecasts during planning of dairy production, is grounded.

Heterogeneity of the Neural Composition of Cortical Regions as a Condition for a Wide Range Regulating Spontaneous Activity

On the slices of sensorimotor and frontal cortex in layer V of guinea pigs the diversity of neuronal spontaneous activity, the mechanism of its origin and functional specificity were studied. In both regions, neurons that did not have spontaneous activity predominated (39% in the sensorimotor cortex and 32% in the frontal cortex) over neurons with any other firing levels. The iontophoretic application of the excitatory transmitter, glutamate, caused activation spike reactions in all registered neurons;moreover, short-term activation reactions to glutamate had a significantly longer after-discharge in neurons of the frontal cortex (up to 2500 ms and more) compared to reactions of neurons of the sensorimotor region. This means that postexcitatory hyperpolarization in nerve cells of the frontal cortex is less expressed and, therefore, they have a lower density of K+ channels on their membranes. With an increase in the level of spontaneous activity, K+ membrane permeability decreases, which is confirmed by the appearance of a long activation reaction to acetylcholine (which blocks K+ channels), exactly when spontaneous firing appears in “silent” neurons. Despite the fact that spontaneous activity is formed by glutamatergic excitation, its considerable diversity is associated with the structural and membrane characteristics of neurons, which determine the different degrees of EPSPs attenuation on the way of moving along dendrites. Acetylcholine regulates this process in different ways, in accordance with different states of K+ membrane permeability. Therefore, the lower content of K+ channels on the membranes of neurons of the frontal cortex does not allow regulating spontaneous activity in the same range as occurs in neurons of the sensorimotor region. Factors Affecting Clinical Depression in Diabetic Geriatric Population

Introduction: Diabetes mellitus is a common metabolic disorder affecting about 10% - 25% of the elderly population. The mechanisms, linking diabetes and major depressive disorders are unknown. Obesity is the risk factor for both diabetes mellitus and depression. The goal of this study was to explore if BMI of elderly diabetic patients with depression differed from BMI of the diabetic individuals without depression. Further, we aimed to explore the association of age, gender, health insurance status, ethnicity influence and systolic blood pressure (SBP) and diastolic blood pressure (DBP) with clinical depression in geriatric diabetics. Methods: Nursing home residents were recruited from an outpatient internal medicine clinic in a teaching university setting. After given written consent, patients received Public Health Questionnaire-9 (PHQ-9) in either English or Spanish language. Results: While the clinically depressed diabetic participants did not differ from that of the non-depressed diabetic participants with respect to age (70.6 ± 6.1 vs. 72.1 ± 4.8;p = 0.111), gender (47.2% of males vs. 57.1% of females;p = 0.754), or ethnicity (69% explain vs. 71%, p = 1.000), race did appear to differ in depression status: Caucasian, African-American, and others were respectively 37%, 48%, and 15% vs. 32%, 16%, 52%, p = 0.0003). Neither the BMI (p > 0.499) nor Hemoglobin A1c (p > 0.839) differed between the clinically depressed and non-depressed participants when controlled for these race differences. Conclusion: In our sample, diabetic African Americans are three times more likely to be depressed. Diabetic Caucasians also experience higher rates of clinical depression. However, diabetics with depression did not differ from diabetics without depression with regard to their age, gender, BMI, HbA1C, health insurance status, systolic blood pressure or diastolic blood pressure.
