Effect of Acid Species on <i>Cryptococcus</i>sp. T1 and Construction of Its Bioreactor System for Continuous Neutralization of Acidic Media

Generally, the tolerance of yeasts to acid is stronger than that of bacteria. In addition, some of the yeasts are able to neutralize acidic media, which is called acid-neutralizing yeast. An acid-neutralizing yeast, Cryptococcus sp. strain T1, was previously isolated from Lake Tazawa in Japan. Here we investigated the effect of several types of acid on the neutralizing ability of strain T1, and we improved an existing bioreactor model to effectively neutralize acidic water. First we carried out a neutralizing test with strain T1 using casamino acid solution adjusted to pH 3.0 - 5.0 with each inorganic or organic acid. The solutions adjusted to pH 3.0 with nitric acid, phosphoric acid, sulfuric acid, and citric acid were neutralized by strain T1. The pH 3.0 solutions with lactic acid, formic acid, and acetic acid were not neutralized. These results demonstrated that some types of organic acid avoid neutralization by strain T1, and we hypothesized that this might be due to the sterilizing function of non-dissociative organic acid. We then constructed a bioreactor system with a column filled with T1-immobilized alginate beads. The previous research identified a problem in this bioreactor system: the high level of ammonium ions (NH+4) in the neutralized water might pollute water environments, and aluminum ions (Al3+) included in acidic water prevent the neutralization. Here, we used zeolite to get rid of the NH+4 and Al3+, and the addition of a zeolite reactor enabled the bioreactor system to neutralize the acidic water with a decrease of NH+4 in the water.

AIM To investigate the possibility of diagnosing gastric cancer from an unstained pathological tissue using Raman spectroscopy, and to compare the findings to those obtained with conventional histopathology. METHODS We produced two consecutive tissue specimens from areas with and without cancer lesions in the surgically resected stomach of a patient with gastric cancer. One of the two tissue specimens was stained with hematoxylin and eosin and used as a reference for laser irradiation positioning by the spectroscopic method. The other specimen was left unstained and used for Raman spectroscopy analysis. RESULTS A significant Raman scattering spectrum could be obtained at all measurement points. Raman scattering spectrum intensities of 725 cm^(-1) and 782 cm^(-1), are associated with the nucleotides adenine and cytosine, respectively. The Raman scattering spectrum intensity ratios of 782 cm^(-1)/620 cm^(-1), 782 cm^(-1)/756 cm^(-1), 782 cm^(-1)/1250 cm^(-1), and 782 cm^(-1)/1263 cm^(-1) in the gastric adenocarcinoma tissue were significantly higher than those in the normal stomach *** The results of this preliminary experiment suggest the feasibility of our spectroscopic method as a diagnostic tool for gastric cancer using unstained pathological specimens.

The ventral quadrant of the tracheal epithelium of the hamster,about one fourth of tracheal mucosa, was denuded by a venous needle *** stains were made with HE, PAS, and PAS-anti Brdu immunohis-tochemic technique. At the 0 h circumference cell number (CCN) was 927. *** 6 h post injured the viable cells changed in shapes and migrated from themargin to wound site. By 24 h the wound area was completely covered by a sin-gle layer of non-ciliated flattened cells, then an expotential increassing in cellregeneration occured in wound site by 48 h. The CCN had been restored to thelevel of control (1373), and the proliferative cell, composed of polygonal epi-dermoid metaplasia, 3～4 layers and reached a peak (338.8). The secretoryand ciliated cells appeared gradually from 72 h post injury, the epithelium re-stored to the normal epithelial architecture by one week post injury. In our pre-sent study either in control and non-injured epithelium or in all stages of woundsite about 70% of the Brdu positive cells contained small or confluent PAS posi-tive granules were observed. This fact indicated that secretory cells play a im-portant role in proliferation after mechanical injury and maintaining the normaltracheal pseudostratified epithelium.

Much data such as geometric image data and drawings have graph structures. Such data are called graph structured data. In order to manage efficiently such graph structured data, we need to analyze and abstract graph structures of such data. The purpose of this paper is to find knowledge representations which indicate plural abstractions of graph structured data. Firstly, we introduce a term graph as a graph pattern having structural variables, and a substitution over term graphs which is graph rewriting system. Next, for a graph G, we define a multiple layer ( g,(θ 1,…,θ k )) of G as a pair of a term graph g and a list of k substitutions θ 1,…,θ k such that G can be obtained from g by applying substitutions θ 1,…,θ k to g. In the same way, for a set S of graphs, we also define a multiple layer for S as a pair ( D,Θ ) of a set D of term graphs and a list Θ of substitutions. Secondly, for a graph G and a set S of graphs, we present effective algorithms for extracting minimal multiple layers of G and S which give us stratifying abstractions of G and S, respectively. Finally, we report experimental results obtained by applying our algorithms to both artificial data and drawings of power plants which are real world data.

Efficacy of omarigliptin,once-weekly dipeptidyl peptidase-4 inhibitor,in patients with type 2 diabetes

Taste stimuli can elicit facial responses, and the facial responses may be useful indexes of taste sensations. In this study, we propose that eye blinking is also elicited by taste stimuli and we examined eye blink responses in six healthy young adults. Low and high concentration solutions of the four basic taste qualities (sweetness, saltiness, sourness, and bitterness) and distilled water were delivered via a silicone tube. Facial responses were recorded by a video camera and eye blink responses were identified. The number of eye blinks in the 5 s following stimulation, and the latency and duration of the first eye blink, were quantified. BACKGROUND It is widely recognized that endoscopic resection(ER) of superficial nonampullary duodenal epithelial tumors(SNADETs) is technically challenging and may carry high risks of intraoperative and delayed bleeding and *** adverse events could be more critical than those occurring in other levels of the gastrointestinal tract. Because of the low prevalence of the disease and the high risks of severe adverse events, the curability including short-and long-term outcomes have not been standardized *** To investigate the curability including short-and long-term outcomes of ER for SNADETs in a large case *** This retrospective study included cases that underwent ER for SNADETs at our university hospital between March 2004 and July 2017. Short-term outcomes of ER were measured based on en bloc and R0 resection rates as well as adverse events. Long-term outcomes included local recurrence detected on endoscopic surveillance and disease-specific mortality in patients followed up for ≥ 12 mo after *** In the study, 131 patients with 147 SNADETs were analyzed. The 147 ERs consisted of 136 endoscopic mucosal resections(EMRs)(93%) and 11 endoscopic submucosal dissections(ESDs)(7%). The median tumor diameter was 10 *** pathology diagnosis was adenocarcinoma(56/147, 38%), high-grade intraepithelial neoplasia(44/147, 30%), or low-grade intraepithelial neoplasia(47/147, 32%). The R0 resection rate was 68%(93/136) in the EMR group and73%(8/11) in the ESD group, respectively. Cap-assisted EMR(known as EMR-C)showed a higher rate of R0 resection compared to the conventional method of EMR using a snare(78% vs 62%, P = 0.06). No adverse event was observed in the EMR group, whereas delayed bleeding, intraoperative perforation, and delayed perforation in 3, 3, and 5 patients occurred in the ESD group, respectively. The purpose of this study was to propose indicators for grasping autistic characteristics of people with intellectual disabilities who were going to engage in agricultural vocational training. We analyzed the data of the Childhood Autism Rating Scale (CARS) of 44 students at the high school course of a special needs education school who participated in agricultural vocational training. The data were divided into two groups according to the scores of “General Impressions”, one of 15 question items of the CARS: one was the group whose score was 1 (usual), and the other was the group whose scores were 1.5 to 4 (unusual). The results of the Mann-Whitney U test showed significant differences between two groups in eight items including “Relating to People” (p Relating to People” with six other items. As a result, “Relating to People” was considered to be the most effective item to know the participant’s autistic characteristics at a very early stage of training. Eventually, adding two items, “Body Use” and “Imitation” which are necessary to master agricultural tasks, to “Relating to People”, we proposed three indicators.

Characteristics and Factors Associated with Obesity and Thinness among Children with Developmental Disorders

Obesity in childhood or adolescence has been recognized to be a risk factor for the onset of lifestyle-related diseases, not only in healthy children, but also in children with developmental disorders. Therefore, this study was conducted to examine the characteristics of obesity and thinness as assessed by the body fat percentage among children with developmental disorders during certain growth periods. It was also designed to investigate those factors associated with obesity and thinness based on a lifestyle and behavioral questionnaire. The subjects included 260 children from 5 to 18 years old with developmental disorders. The results of the study showed that a decrease in thinness and increase in obesity with ageing exhibited more noticeable trends among those children with mental retardation. The factors associated with obesity in children with developmental disorders were characterized by the dietary content, eating behaviors, and food preferences particular to such children, as well as low physical activity and a family history of obesity. The results of this study suggested the importance of continuous guidance along with family participation in order to improve obesity among children with developmental disorders, while focusing on the characteristics of certain growth periods.
