Raman spectroscopy for the diagnosis of unlabeled and unstained histopathological tissue specimens

AIM To investigate the possibility of diagnosing gastric cancer from an unstained pathological tissue using Raman spectroscopy, and to compare the findings to those obtained with conventional histopathology. METHODS We produced two consecutive tissue specimens from areas with and without cancer lesions in the surgically resected stomach of a patient with gastric cancer. One of the two tissue specimens was stained with hematoxylin and eosin and used as a reference for laser irradiation positioning by the spectroscopic method. The other specimen was left unstained and used for Raman spectroscopy analysis. RESULTS A significant Raman scattering spectrum could be obtained at all measurement points. Raman scattering spectrum intensities of 725 cm^(-1) and 782 cm^(-1), are associated with the nucleotides adenine and cytosine, respectively. The Raman scattering spectrum intensity ratios of 782 cm^(-1)/620 cm^(-1), 782 cm^(-1)/756 cm^(-1), 782 cm^(-1)/1250 cm^(-1), and 782 cm^(-1)/1263 cm^(-1) in the gastric adenocarcinoma tissue were significantly higher than those in the normal stomach *** The results of this preliminary experiment suggest the feasibility of our spectroscopic method as a diagnostic tool for gastric cancer using unstained pathological specimens.

Robust Surface Reconstruction of Plant Leaves from 3D Point Clouds

The automation of plant phenotyping using 3D imaging techniques is ***,conventional methods for reconstructing the leaf surface from 3D point clouds have a trade-off between the accuracy of leaf surface reconstruction and the method’s robustness against noise and missing *** mitigate this trade-off,we developed a leaf surface reconstruction method that reduces the effects of noise and missing points while maintaining surface reconstruction accuracy by capturing two components of the leaf(the shape and distortion of that shape)separately using leaf-specific *** separation simplifies leaf surface reconstruction compared with conventional methods while increasing the robustness against noise and missing *** evaluate the proposed method,we reconstructed the leaf surfaces from 3D point clouds of leaves acquired from two crop species(soybean and sugar beet)and compared the results with those of conventional *** result showed that the proposed method robustly reconstructed the leaf surfaces,despite the noise and missing points for two different leaf *** evaluate the stability of the leaf surface reconstructions,we also calculated the leaf surface areas for 14 consecutive days of the target *** result derived from the proposed method showed less variation of values and fewer outliers compared with the conventional methods.

Could we remain simply applying discount rate to evaluate the life quality of our future generations?

When we try to estimate future environmental costs resulting from our current economic activities,we usually calculate the present value of the consequences for our future generations by utilizing a discount rate as a standard economic *** popularity of this procedure is solely supported by its simplicity which assumes:(1)a perfect financial market over even 100 years,and(2)existence of a stable time preference between two consecutive periods for an individual *** apparent deficiency of this approach is that the present value of life quality of future generations varies to a large extent along an arbitrarily chosen discount *** a matter of fact,the discount rate,which could reflect the time preference as2%or 5%,matters when we predict current strategies of environmental protection for future *** applying a discount rate to evaluate the quality of our future generation,without clarifying the actual production mechanism behind this,is almost to the same as ignoring the fact that all of the goods are produced through an actual production process and that environmental degradation reduces the efficiency of that *** greatest concern for our future generations should not be given by an assumed discount rate,since the discount rate itself is determined by financial market conditions at certain points in time.

Adsorption of urea,creatinine,and uric acid from three solution types using spherical activated carbon and its recyclability

In this paper,we propose that the urinary toxins from the wastewater be adsorbed on an adsorbent such as spherical activated carbon and the latter be regenerated by subjecting it to high temperatures to recycle activated carbon and also to recycle the water used in *** studied the adsorption of artificial waste dialysate,which is a mixed solution of urea,creatinine,and uric acid,and the separate solutions for each of these and found that their extents of adsorption onto the spherical activated carbon material were nearly *** amount of adsorption was approximately 1.4 mg·g^−1 for urea,18 mg·g^−1 for creatinine,and 20 mg·g^−1 for uric *** urea,creatinine,and uric acid adsorbed onto the spherical activated carbon decomposed on heat treatment at 500℃,and the adsorption capacity of the spherical activated carbon was *** study successfully demonstrated that the spherical activated carbon can be recycled in the waste dialysate treatment process.

Molecular evidence for biochemical diversification of phenolamide biosynthesis in rice plants

Two phenolamides （PAs）, p-coumaroylputrescine and feruloylputrescine strongly accumulate in rice （Oryza sativa cv. Nipponbare） leaves subjected to attack of chewing and sucking herbivores. Here we identified and characterized in vitro three novel rice genes that mediated coumaroyI-CoA/ feruloyI-CoA conjugation to polyamines, putrescine and agmatine. Interestingly, two genes were highly specific for their polyamine substrates, encoding putresdne N-hydrox- ycinnamoyltransferase and agmatine N-hydroxycinnamoyl- transferase, while the third enzyme could use both polyamines and it was therefore annotated as putrescine/ agmatine N-hydroxycinnamoyltransferase. All genes were preferentially expressed in rice roots and developing flowers, and in addition, the putrescine/agmatine N-hydroxycinnamoyl- transferase transcripts were strongly induced by wounding in the young rice leaves. Because the wound response of this gene was only partially suppressed in the jasmonoyI-L-isoleucine deficient plants （Osjarl）, it suggests that its upregulation （as well as inducible PAs in rice） may be largely independent of jasmonoyI-L-isoleucine signaling pathway. The finding of three closely related genes with a similar and/or overlapping activity in PA biosynthesis provides another striking example of rapid diversification of plant metabolism in response to environmental stresses in nature.

Optimal Total Sleep Duration per Day to Maintain Health-Related Quality of Life in Male Collegiate Athletes:A Cross-Sectional Study

Purpose This study investigated the optimal total sleep duration per day required by collegiate athletes to maintain the physical and mental health-related quality of life(HRQOL),compared with non-athlete *** In this cross-sectional study,a questionnaire survey was conducted to assess demographic variables,lifestyle and sleep habits,and HRQOL in 392 collegiate students(non-athletes,n=174;athletes,n=218).Physical component summary(PCS)and mental component summary(MCS)were assessed using the short-form-8 health *** with both good PCS and MCS were defined as having a good *** confirm an association between the total sleep duration per day and good HRQOL,logistic regression analyses were conducted in non-athlete students and collegiate athletes ***,receiver-operating curve(ROC)analyses were performed for the detection of the cut-off point of total sleep duration per day sufficient to maintain a good *** The average total sleep duration per day was 7 h 19 min for collegiate athletes,and 78.9%of them had a worse *** cut-off point of total sleep duration per day to maintain good HRQOL for collegiate athletes was 7.92 h(area under ROC,0.64;P=0.038;sensitivity,75.4%;specificity,57.9%),which was longer than 6.79 h for non-athlete *** Collegiate athletes required longer nocturnal sleep than non-athlete ***,their habitual nocturnal sleep duration was shorter compared to their optimal duration;around 70%of them faced chronic insufficient *** sleep habits and sleep education is important in maintaining their good health-related quality of life.

Comparison of Effect of Brassinosteroid and Gibberellin Biosynthesis Inhibitors on Growth of Rice Seedlings

Brassinosteroid（BR） and gibberellin（GA） are two predominant plant hormones that regulate plant cell elongation. Mutants disrupt the biosynthesis of these hormones and display different degrees of dwarf phenotypes in rice. Although the role of each plant hormone in promoting the longitudinal growth of plants has been extensively studied using genetic methods, their relationship is still poorly understood. In this study, we used two specific inhibitors targeting BR and GA biosynthesis to investigate the roles of BR and GA in growth of rice seedlings. Yucaizol, a specific inhibitor of BR biosynthesis, and Trinexapac-ethyl, a commercially available inhibitor of GA biosynthesis, were used. The effect of Yucaizol on rice seedlings indicated that Yucaizol significantly retarded stem elongation. The IC_（50） value was found to be approximately 0.8 μmol/L. Yucaizol also induced small leaf angle phenocopy in rice seedlings, similarly to BR-deficient rice, while Trinexapac-ethyl did not. When Yucaizol combined with Trinexapac-ethyl was applied to the rice plants, the mixture of these two inhibitors retarded stem elongation of rice at lower doses. Our results suggest that the use of a BR biosynthesis inhibitor combined with a GA biosynthesis inhibitor may be useful in the development of new technologies for controlling rice plant height.
