Molecular Phylogeny and Phenotypic Characterization of Yeasts with a Broad Range of pH Tolerance Isolated from Natural Aquatic Environments

In this study, yeasts with broad range of pH tolerance were isolated and characterized from natural aquatic environments in Japan. Only a few basic and application studies of alkali-tolerant yeasts have been reported, despite the unmet industrial needs. First, we surveyed alkali-tolerant yeasts from natural aquatic environments at pH 7.6 - 9.4. We isolated 35 yeast strains that grew in pH 9.0 medium, from seven genera and nine species: 25 strains (N1, N2, through N6, N9, K1, K3 through K19) were Rhodotorula mucilaginosa;one (N7) was Rhodosporidium fluvial;one (N8) was Scheffersomyces spartinae;two (N10 and N13) were Wicherhamonyces anomalus;one (N11) was Cyberlindnera saturnus;one (S1) was Candida sp.;two (S2 and S4) were Candida intermedia;one (S3) was Candida quercuum;and one (K2) was Cryptococcus liquefacience. We examined the effects of pH on the growth of representative yeast strains. Strains K12 and S4 showed high growth at pH 3 - 10. Strains N7, N8, N10, N11, and S3 showed high growth at pH 3 - 9. Strains K2 and S1 showed high growth at pH 4 - 8. All nine of these strains had neutralizing activities from acidic media at pH 3 - 5 to pH 6 - 8. We previously isolated acid-tolerant yeasts (Cryptococcus sp. T1 [1] and Candida intermedia CeA16 [2]) from extremely acidified environments;they showed high growth at pH 3 - 9 and neutralizing activities of acidic media by releasing ammonium ions. Thus, alkali-tolerant yeasts and acid-tolerant yeasts were found to be similar species and have both high growth at a broad range of pH and neutralizing activities of acid media. Previously, we also isolated acid-tolerant, acid-neutralizing yeasts from neutral natural environments (26 strains, 12 species) [3]. Seatbelt paths of the pregnant women sitting in the rear seat of a motor vehicle

Purpose： Seatbelt use during pregnancy is important to improve maternal and fetal survival after motor vehicle collisions. However, because the rear seatbelt of a motor vehicle tends to make contact with the neck, even if it is adequately used, some pregnant women sitting in the rear seat opt not to fasten the belt. The purpose of this study is to explore seatbelt-neck contact for pregnant women sitting in the rear seat of a motor vehicle. Methods： We carried out an anthropometric study. Japanese women who were _〉30 weeks pregnant （n = 12） sat in the left side of the rear seat of a typical mid-size passenger sedan and fastened the seatbelt. Seating posture was investigated by measuring the coordinates of the anthropometric data points of the pregnant women （head, shoulder, hip joint, and knee joint）. The belt path was analyzed by measuring the clearance between the belt and the sternum or navel. Results： Among the 12 pregnant women at 33.9 week _＋ 3.3 week gestation, the shoulder belt deviated to the right side and subsequently contacted to the neck in four pregnant women （Contact group）. The height of the Contact group was significantly shorter than that of Non-contact group （152.3 cm ~ 3.0 cm vs. 159.0 cm ＋ 3.3 cm, p = 0.008）. Regarding the relative position of the seatbelt to the subject＇s body, the distances from the top of the sternum to the center of the shoulder belt were significantly shorter in Contact group （3.9 cm ~ 3.5 cm） than that in the Non-contact group （8.0 cm ~ 1.6 cm, p = 0.03）. However, no significant difference was found for the distance from the umbilicus to the center of the lap belt. Conclusion： Our findings show that because of short height and late term of pregnancy with protrusion of the abdomen, the shoulder belt deviates to the right or left, avoiding the protruded uterus, and subsequently makes contact with the neck. Seatbelt systems for rear seats need to be developed to improve passenger safety, especially for pregnant women.

Effective cofactor complex purification using nanobeads

Drug target factor complex identification is necessary for evidence based drug discovery. Previous study showed that using small chemical immobilized magnetic nanobeads the chemical target factors were effectively purified and identified. Here we succeeded to purify the chemical target factor complex, so called cofactor(s). Arginine exhibits a variety of biological activities through a complex and highly regulated set of pathways that remain incompletely understood at both the whole-body and the cellular levels. The aim of this study is to develop and validate effective purification system for arginine target complex. New arginine target protein (arginine interacting factor 4, AIF4) was purified and identified. Using recombinant AIF4 protein and arginine-immobilized magnetic nanobeads, AIF4 cofactor, AIF4-BP1, were purified. Interaction of AIF4 and AIF4-BP1 was detected in arginine-dependent manner, suggesting arginine receptor complex formation. This nanobeads technology is more than 30-fold efficient purification efficient than general purification technology.

进行了臂形草内生菌特异DNA片段的克隆及分子鉴定方法的研究。从5个臂形草品种分离的5个内生菌纯培养物中,提取基因组DNA,通过140条随机引物的RAPD分析,引物OPAK10扩增出1条约500bp的共有PCR谱带。此DNA片段命名为BE1。对BE1进行分离、纯化,经点杂交证实BE1为臂形草内生菌特异DNA片段。进一步将BE1片段进行回收、克隆和DNA序列分析。BE1在基因库中进行序列分析,未发现相关同源片段。臂形草内生菌特异DNA片段的获得,为建立一种准确、快速检测臂形草内生菌及特异内生菌方法奠定了分子生物学基础。

Attitude survey of nursing students in Madagascar and Japan

Background: Few investigations have focused on nursing students from a developing country in relation to a developed country like Japan. Objective: This study explored differences between the nursing students in Madagascar and Japan. Subjects and Methods: This is a prospective analytical study of 63 nursing students—32 Japanese and 31 Malagasy. Questionnaires were distributed to students in Madagascar and Japan in 2010 and collected for statistical analysis. Results: Malagasy students tended to think that nursing plays a psychological role, and they have positive view for this function. In contrast, Japanese students tended to think that nursing is physical work and they make a point of its outcome and the need for social and economic guarantees. Conclusion: The nursing students in Madagascar and Japan expressed different views and goals. While several factors may be involved in these differences, their distinct medical situations are most likely the major contributor.

Change of Subcutaneous Tissue Mass at the Deltoid and Thigh Areas in Japanese Infants Followed from 2 to 15 Months

We previously investigated the thickness of the epidermis and lengths from the skin surface to the muscle fascia and bone based on measurements taken from the ultrasonic echograms of Japanese infants aged 2, 3, 4, 5, 6, and 12 - 15 months. The appropriate needle length for intramuscular injection was 16 mm (5/8 inch) in Japanese infants at any age and site. In the present study, we examined these lengths in 21 infants with physical growth from 2 to 15 months. Average height increased from 58.5 to 73.6 cm and average body weight from 5835 to 9226 g until 15 months. The circumference of the thigh increased from 22.3 cm at 2 months to a maximum of 25.9 cm at 6 months, while that of the deltoid area was 15.2 cm at 2 months and 15.9 cm at 6 months. Subcutaneous mass (length from the skin surface to the muscle fascia) at the thigh increased between 3 and 4 months and decreased at 12 - 15 months and showed good relationship to the circumference of the thigh. Subcutaneous mass at the center of the deltoid area showed the same size until 6 months. Muscle mass (lengths from the muscle fascia to the bone) at thigh and deltoid areas remained the same until 6 months. Ultrasonic echogram is an effective tool for evaluating subcutaneous and muscle mass in young infants.

Purpose: To examine podoplanin expression in human pleomorphic adenomas (PA)。Methods: Immunohistochemical analysis was performed in 28 cases of PA。Results: Podoplanin expression was detected in limited modified myoepithelial elements in all cases。Strong reaction for podoplanin was observed in epitheloid and spindle cells。Conclusion: Expression of podoplanin by the limited modified myoepithelial elements may reflect the progression of PA。

Primary Fallopian Tube Adenocarcinoma Discovered with the Vaginal Cytology: A Case Report with Pathological and Immunohistochemical Investigation

Background: Primary fallopian tube cancer is an extremely rare gynecological malignancy. Aim: To discuss, through a case report, the diagnostic process by means of cytology immunohistochemistry. Case Presentation: A 47-year-old Japanese woman, who also had lung cancer, presented with enlarged para-aortic lymph node without any symptoms. Based on the vaginal cytology report suggestive of gynecologic malignant tumor (possibly fallopian tube adenocarcinoma), primary surgery comprised of total abdominal hysterectomy and bilateral salpingo-oophorectomy was performed. Histopathology and immunohistochemistry examinations revealed primary fallopian tube carcinoma with metastasis of para-aortic lymph node. She is free from recurrence and metastases 9 months after the surgery and chemotherapy. Conclusion: Although primary fallopian tube cancer is a rare gynecologic malignancy, vaginal cytology may be useful for detecting fallopian tube carcinoma.

Dissemination of <i>Staphylococcus warneri</i>in the Hair of ICU Doctors

This study reports a case of Staphylococcus warneri dissemination in the hair of ICU/ER doctors. Eleven of 23 doctors in the first survey and 11 of 24 doctors in the second survey, which were conducted after 6 months, carried S. warneri, while hair contamination of other coagulase-negative staphylococci was only sporadic. Chromosomal DNA analysis with PFGE revealed that 7 S. warneri strains isolated in the first survey and 7 strains isolated in the second survey showed similar PFGE patterns, suggesting that a cross-infection with a S. warneri strain continued over 6 months among the ICU doctors.
