Effects of different doses of glucose and fructose on central metabolic pathways and intercellular wireless communication networks in humans

Fructose and glucose are often widely used in food processing and may contribute to many metabolic diseases.To observe the effects of different doses of glucose and fructose on human metabolism and cellular communication,volunteers were given low,medium,and high doses of glucose and fructose.Serum cytokines,glucose,lactate,nicotinamide adenine dinucleotide(NADH)and metabolic enzymes were assayed,and central carbon metabolic pathway networks and cytokine communication networks were constructed.The results showed that the glucose and fructose groups basically maintained the trend of decreasing catabolism and increasing anabolism with increasing dose.Compared with glucose,low-dose fructose decreased catabolism and increased anabolism,significantly enhanced the expression of the inflammatory cytokine interferon-γ(IFN-γ),macrophage-derived chemokine(MDC),induced protein-10(IP-10),and eotaxin,and significantly reduced the activity of isocitrate dehydrogenase(ICDH)and pyruvate dehydrogenase complexes(PDHC).Both medium and high doses of fructose increase catabolism and anabolism,and there are more cytokines and enzymes with significant changes.Furthermore,multiple cytokines and enzymes show strong relevance to metabolic regulation by altering the transcription and expression of enzymes in central carbon metabolic pathways.Therefore,excessive intake of fructose should be reduced to avoid excessive inflammatory responses,allergic reactions and autoimmune diseases.

Neural stem cells promote neuroplasticity: a promising therapeutic strategy for the treatment of Alzheimer’s disease

Recent studies have demonstrated that neuroplasticity,such as synaptic plasticity and neurogenesis,exists throughout the normal lifespan but declines with age and is significantly impaired in individuals with Alzheimer’s disease.Hence,promoting neuroplasticity may represent an effective strategy with which Alzheimer’s disease can be alleviated.Due to their significant ability to self-renew,differentiate,and migrate,neural stem cells play an essential role in reversing synaptic and neuronal damage,reducing the pathology of Alzheimer’s disease,including amyloid-β,tau protein,and neuroinflammation,and secreting neurotrophic factors and growth factors that are related to plasticity.These events can promote synaptic plasticity and neurogenesis to repair the microenvironment of the mammalian brain.Consequently,neural stem cells are considered to represent a potential regenerative therapy with which to improve Alzheimer’s disease and other neurodegenerative diseases.In this review,we discuss how neural stem cells regulate neuroplasticity and optimize their effects to enhance their potential for treating Alzheimer’s disease in the clinic.

Treatment with β-sitosterol ameliorates the effects of cerebral ischemia/reperfusion injury by suppressing cholesterol overload, endoplasmic reticulum stress, and apoptosis

β-Sitosterol is a type of phytosterol that occurs naturally in plants.Previous studies have shown that it has anti-oxidant,anti-hyperlipidemic,anti-inflammatory,immunomodulatory,and anti-tumor effects,but it is unknown whetherβ-sitosterol treatment reduces the effects of ischemic stroke.Here we found that,in a mouse model of ischemic stroke induced by middle cerebral artery occlusion,β-sitosterol reduced the volume of cerebral infarction and brain edema,reduced neuronal apoptosis in brain tissue,and alleviated neurological dysfunction;moreover,β-sitosterol increased the activity of oxygen-and glucose-deprived cerebral cortex neurons and reduced apoptosis.Further investigation showed that the neuroprotective effects ofβ-sitosterol may be related to inhibition of endoplasmic reticulum stress caused by intracellular cholesterol accumulation after ischemic stroke.In addition,β-sitosterol showed high affinity for NPC1L1,a key transporter of cholesterol,and antagonized its activity.In conclusion,β-sitosterol may help treat ischemic stroke by inhibiting neuronal intracellular cholesterol overload/endoplasmic reticulum stress/apoptosis signaling pathways.

Aluminosilicate small pore zeolites belonging to ABC-6 family play crucially important roles in the high methanol conversion with the high selectivity of light olefins,gas separation and storage,and selective catalytic reduction of NO_(x).In this work,we report a general method,called the epitaxial growth approach,for designing ABC-6 family small pore zeolites.It is mainly realized through the epitaxial growth on the nonporous SOD-type zeolite in the presence of inorganic cations(Na^(+)and K^(+))combined with a variety of organic structure directing agents(OSDAs).In this case,a series of ABC-6 family small pore zeolites such as ERI-,SWY-,LEV-,AFX-,and PTT-type zeolites have been successfully synthesized within a few hours.More importantly,the advanced focused ion beam(FIB)and the low-dose high-resolution transmission electron microscopy(HRTEM)imaging technique have been utilized for unraveling the zeolite heterojunction at the atomic level during the epitaxial growth process.It turns out(222)crystallographic planes of the SOD-type zeolite substrate provide unique pre-building units,which facilitate the growth of targeted ABC-6 family small pore zeolites along its c-axis.Moreover,the morphologies of ERI-type zeolite can also be tuned through the epitaxial growth approach,achieving a longer lifetime in the methanol conversion.

Nitro-aromatic compounds(NACs)are among the major components of brown carbon(BrC)in the atmosphere,causing negative impacts on regional climate,air quality,and ecological health.Due to the extensive origins,it is still a challenge to figure out the contributions and originating regions for different sources of atmospheric NACs.Here,field observations on fine particulate NACs were conducted at a coastal rural area in Qingdao,China in the winter of 2018 and 2019.The mean total concentrations of fine particulate nitro-aromatic compounds were 125.0±89.5 and 27.7±21.1 ng/m^(3)in the winter of 2018 and 2019,respectively.Among the measured eleven NACs,nitrophenols and nitrocatechols were the most abundant species.Variation characteristics and correlation analysis showed that humidity and anthropogenic primary emissions had significant influences on the NAC abundances.In this study,two tracing methods of the improved spatial concentration weighted trajectory(SCWT)model and the receptor model of positive matrix factorization(PMF)were combined to comprehensively understand the origins of NACs in fine particles at coastal Qingdao.Four major sources were identified,including coal combustion,biomass burning,vehicle exhaust,and secondary formation.Surprisingly,coal combustion was responsible for about half of the observed nitro-aromatic compounds,followed by biomass burning(~30%).The results by SCWT demonstrated that the coal combustion dominated NACs mainly originated from the Shandong peninsula and the areas to the north and southwest,while those dominated by biomass burning primarily came from local Qingdao and the areas to the west.

Understanding the risks of mercury sulfide nanoparticles in the environment: Formation, presence, and environmental behaviors

Mercury(Hg) could be microbially methylated to the bioaccumulative neurotoxin methylmercury(Me Hg), raising health concerns. Understanding the methylation of various Hg species is thus critical in predicting the Me Hg risk. Among the known Hg species, mercury sulfide(HgS) is the largest Hg reservoir in the lithosphere and has long been considered to be highly inert. However, with advances in the analytical methods of nanoparticles, HgS nanoparticles(HgS NPs) have recently been detected in various environmental matrices or organisms. Furthermore, pioneering laboratory studies have reported the high bioavailability of HgS NPs. The formation, presence, and transformation(e.g., methylation) of HgS NPs are intricately related to several environmental factors, especially dissolved organic matter(DOM). The complexity of the behavior of HgS NPs and the heterogeneity of DOM prevent us from comprehensively understanding and predicting the risk of HgS NPs. To reveal the role of HgS NPs in Hg biogeochemical cycling, research needs should focus on the following aspects: the formation pathways, the presence, and the environmental behaviors of HgS NPs impacted by the dominant influential factor of DOM. We thus summarized the latest progress in these aspects and proposed future research priorities, e.g., developing the detection techniques of HgS NPs and probing HgS NPs in various matrices, further exploring the interactions between DOM and HgS NPs. Besides, as most of the previous studies were conducted in laboratories, our current knowledge should be further refreshed through field observations, which would help to gain better insights into predicting the Hg risks in natural environment.

Benefit allocation of electricity-gas-heat-hydrogen integrated energy system based on Shapley value

The integrated energy system is an important development direction for achieving energy transformation in the context of the low-carbon development era,and an integrated energy system that uses renewable energy can reduce carbon emissions and improve energy utilization efficiency.The electric power network and the natural gas network are important transmission carriers in the en-ergy field,so the coupling relationship between them has been of wide concern.This paper establishes an integrated energy system considering electricity,gas,heat and hydrogen loads;takes each subject in the integrated energy system as the research object;anal-yses the economic returns of each subject under different operation modes;applies the Shapley value method for benefit allocation;and quantifies the contribution value of the subject to the alliance through different influencing factors to revise the benefit allocation value.Compared with the independent mode,the overall benefits of the integrated energy system increase in the cooperative mode and the benefits of all subjects increase.Due to the different characteristics of different subjects in terms of environmental benefits,collaborative innovation and risk sharing,the benefit allocation is reduced for new-energy subjects and increased for power-to-gas sub-jects and combined heat and power generation units after revising the benefit allocation,to improve the matching degree between the contribution level and the benefit allocation under the premise of increased profit for each subject.The cooperative mode effectively enhances the economic benefits of the system as a whole and individually,and provides a useful reference for the allocation of benefits of integrated energy systems.The analysis shows that the revised benefit distribution under the cooperative model increases by 3.86%,4.08%and 3.13%for power-to-gas subjects,combined heat and power generation units,and new-energy units,respectively,compared with the independent function model.

A novel decellularized matrix of Wnt signaling-activated osteocytes accelerates the repair of critical-sized parietal bone defects with osteoclastogenesis, angiogenesis, and neurogenesis

Cell source is the key to decellularized matrix(DM)strategy.This study compared 3 cell types,osteocytes with/without dominant active Wnt/β-catenin signaling(daCO and WTO)and bone marrow stromal cells(BMSCs)for their DMs in bone repair.Decellularization removes all organelles and>95%DNA,and retained>74%collagen and>71%GAG,maintains the integrity of cell basement membrane with dense boundaries showing oval and honeycomb structure in osteocytic DM and smooth but irregular shape in the BMSC-DM.DM produced higher cell survival rate(90%)and higher proliferative activity.In vitro,daCO-DM induces more and longer stress fibers in BMSCs,conducive to cell adhesion,spreading,and osteogenic differentiation.8-wk after implantation of the critical-sized parietal bone defect model,daCO-DM formed tight structures,composed of a large number of densely-arranged type-I collagen under polarized light microscope,which is similar to and integrated with host bone.BV/TV(>54%)was 1.5,2.9,and 3.5 times of WTO-DM,BMSC-DM,and none-DM groups,and N.Ob/T.Ar(3.2×10^(2)/mm^(2))was 1.7,2.9,and 3.3 times.At 4-wk,daCO-DM induced osteoclastogenesis,2.3 times higher than WTO-DM;but BMSC-DM or none-DM didn't.daCO-DM increased the expression of RANKL and MCSF,Vegfa and Angpt1,and Ngf in BMSCs,which contributes to osteoclastogenesis,angiogenesis,and neurogenesis,respectively.daCO-DM promoted H-type vessel formation and nerve markersβ3-tubulin and NeuN expression.Conclusion:daCO-DM produces metabolic and neurovascularized organoid bone to accelerate the repair of bone defects.These features are expected to achieve the effect of autologous bone transplantation,suitable for transformation application.

Research on the Application of Evidence-Based Quality Control Circle to Improve the Implementation Rate of Airway Management Measures in Adult Critically Ill Patients

Objective: To explore the effect of evidence-based quality control circle (QCC) in improving the implementation rate of airway management measures in adult critically ill patients. Methods: Based on the Joanna Briggs Institute (JBI) evidence-based health care model, the best evidence of airway management in adult critically ill patients was obtained and applied to the clinic. Results: The total implementation rate of airway management measures in adult critically ill patients increased from 23.62% before the implementation of quality control circle to 88.82%, and the pulmonary infection rate in critically ill patients decreased from 42.31% to 21.74%, with statistical significance between the two groups (P 0.05). Conclusion: Evidence-based quality control circle activities can standardize the practice standards of airway management in critically ill patients, reduce the occurrence of patients’ airway related complications, and improve clinical outcomes.

Study on the mechanism of Euphorbia fischeriana Steud.- Jujubae Fructus in the treatment of hepatocirrhosis based on network pharmacology

The active components,targets,and pathways of Euphorbia fischeriana Steud.-Jujubae Fructus in treating hepatocirrhosis and the mechanism of action were explored by means of network pharmacology.Firstly,the active components and related targets of Jujubae Fructus were screened by TCMSP database and standardized by Uniprot database.The compounds of Euphorbia fischeriana Steud.were obtained by searching the literature and finally screened by PubChem database,Swiss ADME,and SwissTargetPrediction.Hepatocirrhosis targets were obtained through Genecards database,PPI network analysis was conducted on common targets of Euphorbia fischeriana Steud.-Jujubae Fructus and hepatocirrhosis by using String database,GO enrichment analysis and KEGG pathway enrichment analysis was conducted through Metascape database by using intersection targets of Euphorbia fischeriana Steud.-Jujubae Fructus and hepatocirrhosis,and the results were drawn by using Weishengxin online drawing platform.Then,the network of drug-compound-target-pathway was constructed by the software of Cytoscape3.8.0.Finally,the above results were verified by molecular docking.47 active compounds from Euphorbia fischeriana Steud.-Jujubae Fructus were screened out,which had 38 common targets,162 intersection targets,and 340 signal pathways with hepatocirrhosis,mainly involving hepatitis C,JAK-STAT signal pathway and AGE-RAGE signal pathway.Targets,such as MAPK1,AKT1,TNF,JUN,IL6 and PTGS2,play important roles in the treatment.The findings suggested that the main active ingredients of Euphorbia fischeriana Steud.-Jujubae Fructus in treating hepatocirrhosis are quercetin,scopolamine,physcion,7-deoxyrangduin,17-Hydroxyjolkinolide A,etc.Molecular docking results showed that the main active components and core targets might have a good binding capacity.This study preliminarily explored the potential mechanism of Euphorbia fischeriana Steud.-Jujubae Fructus in treating hepatocirrhosis and provided a theoretical basis for the clinical application of Euphorbia fischeriana Steud.
