Adipose-derived mesenchymal stem cell transplantation promotes adult neurogenesis in the brains of Alzheimer's disease mice

In the present study, we transplanted adipose-derived mesenchymal stem cells into the hippo-campi of APP/PS1 transgenic Alzheimer＇s disease model mice. Immunofluorescence staining revealed that the number of newly generated （BrdU＋） cells in the subgranular zone of the dentate gyrus in the hippocampus was signiifcantly higher in Alzheimer＇s disease mice after adipose-de-rived mesenchymal stem cell transplantation, and there was also a significant increase in the number of BrdU＋/DCX＋neuroblasts in these animals. Adipose-derived mesenchymal stem cell transplantation enhanced neurogenic activity in the subventricular zone as well. Furthermore, adipose-derived mesenchymal stem cell transplantation reduced oxidative stress and alleviated cognitive impairment in the mice. Based on these ifndings, we propose that adipose-derived mes-enchymal stem cell transplantation enhances endogenous neurogenesis in both the subgranular and subventricular zones in APP/PS1 transgenic Alzheimer＇s disease mice, thereby facilitating functional recovery.

Erlotinib is an orally administered, highly effective, specific epidermal growth factor receptor tyrosine kinase inhibitor, used to treat non-small cell lung cancer and pancreatic cancer. The traditional synthetic methods for Erlotinib exhibit long reaction time and safety concern. Herein, we describe a novel five-step route for the synthesis of Erlotinib in flow. These five steps comprise etherification, nitration, reduction,addition and cyclization reactions. All steps were optimized and converted to continuous flow process,which drastically reduces the reaction time and considerably improves the process safety as well as the total yield. Enabled by five continuous flow units, Erlotinib is efficiently afforded with an E-factor of 38,an overall yield of 83%, and a total residence time of 25.1 min. Majority steps in this process have been optimized for quantitative conversion, which offers the possibility of telescoping the entire process.

Fluoride is an important pollutant in wastewater,and adsorption is an effective way to remove fluoride.Because nitrogen plays an important role in adsorbent materials,computational models were developed to understand the changes in work function resulting from nitrogen doping.La-N-C-800℃,was prepared by pyrolyzing La-MOF-NH_(2)to verify the influence on the performance of removing fluoride by electrosorption.Material and electrochemical performance tests were performed to characterize La-N-C-800℃.Adsorption kinetics,adsorption thermodynamics,initial concentrations,pH,and ions competition were investigated using La-N-C-800℃for fluoride removal.In addition,density functional theory was applied to evaluate the function of nitrogen.When nitrogen atoms were added,the density of states,partial density of states,populations,and different orbits of charge were calculated to discover deep changes.Nitrogen strengthened the carbon structure and La_(2)O_(3)structure to remove fluoride.In addition,nitrogen can also act as an adsorption site in the carbon structure.These results provide design ideas for improving the performance of adsorbent materials by doping elements.

以19世纪伦敦的空气污染与儿童健康问题作为切入点,或许是一种将环境史和儿童史相结合的有意义的尝试,即儿童环境史研究路径的拓展。19世纪伦敦空气污染与因呼吸系统疾病导致的儿童死亡率上升之间存在密切关系,空气污染对从事特殊职业的儿童和普通儿童的身心健康和生活均造成了深刻影响。为避免空气污染对儿童的伤害,“转换空气”被时人认为是一种有效的应对举措。伦敦中上层社会采取了自主度假的方式,而新鲜空气慈善机构的儿童乡村假日活动在下层社会发挥了重要作用。19世纪后期,社会改革者建构了一套“身体、心理和道德为一体”的乡村体验话语体系,营造了让儿童“到乡村去”的社会语境。不过,逃避污染只是一种权宜之计,对污染源头的全面治理才是把儿童从环境污染中拯救出来的根本路径。

Introduction:Cholic acid(CA)is a natural steroid useful in treating chronic bronchitis and cholecystitis.On the other hand,its potential impact on osteoarthritis(OA)is unknown.Objective:Using an in vitro and in vivo osteoarthritis model,we sought to assess the chondroprotective properties of CA.Methods:We employed the Cell Counting Kit-8 to measure the impact of CA on chondrocyte activity to assess the toxicity of the cells.Multiple molecular biology experimental techniques were used to investigate potential signaling pathways that CA may use to prevent inﬂammation and give chondrocytes protection.Furthermore,how CA affects the OA model in Sprague-Dawley(SD)rats was evaluated.Results:CA signiﬁcantly suppressed the up-regulation of the interleukin-1β(IL-1β),cyclooxygenase-2(COX-2),and matrix metalloproteinase 13(MMP-13)and the downregulation of aggrecan and type II collagen A1(COL2A)in chondrocytes treated tumor necrosis factor-alpha(TNF-α).Differentially expressed genes(DEG)enrichment revealed IL-17,TNF,chemokine,cytokine-cytokine receptor,toll-like receptor,and nucleotide oligomerization domain-like receptor were the primary signaling pathways.The enriched DEGs included CXCL6,CCL20,MMP3,CXCL3,CXCL11,CCL5,CXCL10,MMP9,MMP13,and CXCL2;these DEGs are involved in inﬂammatory responses and their expression induced by TNF-αwas reversed by CA treatment.CA inhibits p65 nuclear translocation and inhibitory subunit kappa B alpha(IκBα)phosphorylation induced by TNF-α.Furthermore,CA attenuated protein expression of protein kinase RNA-like endoplasmic reticulum kinase(PERK),inositol-requiring transmembrane kinase/endoribonuclease 1α(IRE1α),glucose regulatory protein 78(GRP78),and sirtuin 1(SIRT1),and down-regulation of phosphorylation of AMP-activated protein kinase-α(p-AMPKα)in TNF-α-treated chondrocytes.Conclusions:CA signiﬁcantly ameliorated cartilage degradation in the OA rat model.CA alleviated the inﬂammatory response through the nuclear factor kappa B/PERK/SIRT1 axis and ameliorated cartilage degradation.

Low molecular weight supramolecular gels consist of small molecules(gelators)that self-assemble into nano-or micro-scale network structures in an appropriate solvent.Although various of gelators have been reported,the rational design of an efficient gelator is facing great challenge due to the high cost of synthesis,poor results in the practical application,and secondary pollution to the environment etc.During the last decades,worldwide water pollution is a serious challenge to human civilization.Huge amounts of organic dye wastes and water contains phenol are discharged,posing profound threat to human health and environment.Hence,it is urgent and important to design novel and environment-friendly materials to remove the phenol and organic dyes efficiently from wastewater.Herein,taking advantage of hydrogen bonding,two kinds of gelators based onα-hydroxy acid were designed and applied to effectively purify the phenol aqueous solution.First,the comparison of the gelation potential for these two geltors reveals that 1b possesses better gel ability with larger amount of solvents,lower minimal gelator concentration(MGC)and higher gelatinization stability.These results indicate the hydrogen bonding plays a key role during the gelatinization.With this in hand,gelator 1b was further utilized to sewage treatment,especially for the removal of phenol and organic dyes.The corresponding investigations are in progress.

Analysis of the Relationship between Soil pH and Clubroo of Mustard

[Objective]The paper was to study the severity of mustard clubtoot under different pH conditions and to clear the relationship between pH and mustard clubroot.[Method] Totally 288 soil samples were collected from the infected patches of clubroot in Fuling District,Chongqing City,and their pH were measured.Meanwhile,the germination rate of resting spores,the incidence rate of clubroot and vegetative growth of mustard was measured under different pH conditions,to figure out the relationship between pH and mustard clubroot.[Result] The pH of 288 pathogenic soil samples distributed from 4.0 to 7.0.The germination rates of resting spores were higher than 50% when the pH was 5.0-6.5,and the germination rates were low when the pH was 3.0,4.0,8.0,8.5,9.0 and 10.0.Greenhouse pot test showed that the incidence rate of mustard clubroot in acidic environment was higher than that in alkaline environment.The incidence of mustard clubroot was severe when the pH was 4.5-6.5; the incidence was the severest at the pH of 6.0,and the disease index reached 84.22; mustard was not attacked by clubroot at the pH of 8.0.Measurement results of various growth indicators of mustard under different pH conditions demonstrated that mustard had better growth at the pH of 5.0-7.5,and the growth was the best at the pH of 6.5,while the growth was suppressed at the pH of 4.0 and 8.0.[Conclusion]Clubroot of mustard is likely to happen under weak acidic condition.The suitable pH for the vegetative growth of mustard is 5.0-7.5.
