Elevated and atmospheric-level methane consumption by soil methanotrophs of three grasslands in China

Background:Methane(CH4)oxidation driven by soil aerobic methanotrophs demonstrates the capacity of grassland as a CH4 sink.Methods:In this study,we compared the oxidation characteristics of atmospheric-level and elevated concentration(10%)CH4 in a typical grassland(steppe)on the Loess Plateau,an alpine meadow(meadow)on the Qinghai-Tibet Plateau,and an inland arid-area artificial grassland(pasture)in northwest China and investigated the communities of active methanotrophs and their contribution to CH4 oxidation using DNA-based stable-isotope probing and Illumina Miseq sequencing.Results:The results showed that the oxidation of atmospheric CH4 only occurred in steppe and meadow soils where the USCγgroup of methanotrophs was numerically dominant in the methanotroph community.Pasture soils,with their very low relative abundance of USCγ,did not show atmospheric CH4 oxidation.However,a DNA-stable isotope probing experiment with 10%CH4 indicated that conventional CH4 oxidizers(Methylocaldum and Methylocystis)rather than USCγcommunities assimilated significant amounts of 13CH4 for growth.Conclusions:The CH4 oxidation mechanisms in the three experimental grassland soils varied significantly.The USCγgroup may be obligate oligotrophic microorganisms or their growth requires specific unknown conditions.

Novel method for identifying the stages of discharge underwater based on impedance change characteristic

It is difficult to determine the discharge stages in a fixed time of repetitive discharge underwater due to the arc formation process being susceptible to external environmental influences. This paper proposes a novel underwater discharge stage identification method based on the Strong Tracking Filter(STF) and impedance change characteristics. The time-varying equivalent circuit model of the discharge underwater is established based on the plasma theory analysis of the impedance change characteristics and mechanism of the discharge process. The STF is used to reduce the randomness of the impedance of repeated discharges underwater, and then the universal identification resistance data is obtained. Based on the resistance variation characteristics of the discriminating resistance of the pre-breakdown, main, and oscillatory discharge stages, the threshold values for determining the discharge stage are obtained. These include the threshold values for the resistance variation rate(K) and the moment(t).Experimental and error analysis results demonstrate the efficacy of this innovative method in discharge stage determination, with a maximum mean square deviation of Scrless than 1.761.

One representative water supply system in China with nitrosamine concern: Challenges and treatment strategies

Four sampling campaigns were conducted in two years to understand the fluctuation of N-Nitrosamines(NAs) and their precursors in one drinking water treatment plant(DWTP) in East China in different seasons.This water supply system has been facing several nitrosamine challenges related with source water, including the switch of water source,high concentration of ammonium, formed NAs and NA formation potential(FP) in source water.Besides, the use of ozonation in the DWTP and chloramination in networks will increase the NDMA concentration in tap water.To address these challenges, the biopretreatment was applied in this DWTP to decrease the concentration of ammonium and NAs.The following biological activated carbon(BAC) will neutralize the nitrosamine increase brought by ozonation.The use of free chlorine in disinfection process will also decrease the NDMA formation compared with chloramination.The results will benefit other cities in China and other countries with similar impacted water sources.

炎症性肠病(IBD)是一种慢性、复发性炎症性疾病, 已成为我国重大公共卫生难题之一。抗整合素α4β7单克隆抗体主要包括维得利珠单抗(VDZ)和正在研发的阿利鲁单抗(abrilumab)。VDZ是目前唯一上市的肠道选择性抗淋巴细胞迁移生物制剂, 多项研究证明了其在IBD治疗中的疗效和安全性。如何预测、监测和评估VDZ的疗效, 如何优化VDZ用药或与其他药物联合使用的方案等, 在临床实践中受到广泛关注。现通过回顾VDZ和阿利鲁单抗的相关研究进展, 特别是已上市的VDZ在治疗IBD方面的最新研究内容, 旨在为IBD治疗的临床实践提供建议。

Mitochondrial ROS promote macrophage pyroptosis by inducing GSDMD oxidation

Disrupted mitochondrial membrane potential (MMP) and reactive oxygen species (ROS) generation are often associated with macrophage pyroptosis. It remains unclear how these forms of mitochondrial dysfunction relate to inflammasome activation and gasdermin-D (Gsdmd) cleavage, two central steps of the pyroptotic process. Here, we also found MMP collapse and ROS generation induced by Nlrp3 inflammasome activation as previous studies reported. The elimination of ROS alleviated the cleavage of Gsdmd, suggesting that Gsdmd cleavage occurs downstream of ROS release. Consistent with this result, hydrogen peroxide treatment augmented the cleavage of Gsdmd by caspase-1. Indeed, four amino acid residues of Gsdmd were oxidized under oxidative stress in macrophages. The efficiency of Gsdmd cleavage by inflammatory caspase-1 was dramatically reduced when oxidative modification was blocked by mutation of these amino acid residues. These results demonstrate that Gsdmd oxidation serves as a de novo mechanism by which mitochondrial ROS promote Nlrp3 inflammasome-dependent pyroptotic cell death.

In the Middle and Upper Yangtze region of South China,there are well developed sets of marine shale strata.Currently only Wufeng Longmaxi shale gas has been developed in scale,while the Sinian and Cambrian shale gas are still under exploration.The various shale strata show different characteristics in lithological features,such as lithofacies types and reservoir physical properties,which are due to the influence of tectonic pattern,sedimentary environment,and diagenesis caused by tectonic subsidence.This will affect the later fracturing technology and fracturing effect.The shale reservoirs of Sinian doushantuo,Cambrian Niutitang,Upper Ordovician Wufeng,and the Lower Silurian Longmaxi Formation shale were evaluated and compared with each other with respect to their sedimentary environments,lithofacies,minerals compositions,and micro pore characteristics.The reservoir characteristics of the shale and the main control factors of shale gas enrichment were summarized in this study.

GSDMB promotes non-canonical pyroptosis by enhancing caspase-4 activity

Gasdermin B (GSDMB) has been reported to be associated with immune diseases in humans, but the detailed molecular mechanisms remain unsolved. The N-terminus of GSDMB by itself, unlike other gasdermin family proteins, does not induce cell death. Here, we show that GSDMB is highly expressed in the leukocytes of septic shock patients, which is associated with increased release of the gasdermin D (GSDMD) N-terminus. GSDMB expression and the accumulation of the N-terminal fragment of GSDMD are induced by the activation of the non-canonical pyroptosis pathway in a human monocyte cell line. The downregulation of GSDMB alleviates the cleavage of GSDMD and cell death. Consistently, the overexpression of GSDMB promotes GSDMD cleavage, accompanied by increased LDH release. We further found that GSDMB promotes caspase-4 activity, which is required for the cleavage of GSDMD in non-canonical pyroptosis, by directly binding to the CARD domain of caspase-4. Our study reveals a GSDMB-mediated novel regulatory mechanism for non-canonical pyroptosis and suggests a potential new strategy for the treatment of inflammatory diseases.
