Virus-induced gene silencing （VIGS） offers a powerful approach for functional analysis of individual genes by knocking down their expression. We have adopted this approach to dissect gene functions in cotton resistant to Verticfllium wilt, one of the most devastating diseases worldwide. We showed here that highly efficient VIGS was obtained in a cotton breeding line （CA4002） with partial resistance to Verticillium wilt, and GhMkk2 and Gh Ve I are required for its resistance to Verticillium wilt. Arabidopsis AtBAk1/SERk3, a central regulator in plant disease resistance, belongs to a subfamily of somatic embryogenesis receptor kinases （SERks） with five members, AtSERk1 to AtSERk5. Two BAk1 orthologs and one SERk1 ortholog were identified in the cotton genome. Importantly, GhBAk1 is required for CA4002 resistance to Verticillium wilt. Surprisingly, silencing of GhBAk1 is sufficient to trigger cell death accompanied with production of reactive oxygen species in cotton. This result is distinct from Arabidopsis in which AtBAk1 and AtSERk4 play redundant functions in cell death control. Apparently, cotton has only evolved SERk1 and BAk1 whereas AtSERk4/5 are newly evolved genes in Arabidopsis. Our studies indicate the functional importance of BAk1 in Verticillium wilt resistance and suggest the dynamic evolution of SERk family members in different plant species.

Various biological constraints including erratic and asynchronous flowering between male and female plants hinder successful hybrid development and genetic gains in greater yam breeding ***,pollen storage has gained much attention to facilitate artificial pollinations and increase the genetic *** 4-year study aimed at developing a practical long-term pollen storage technique for the successful development of yam *** pollens were collected from two Dioscorea alata males,then lyophilized(two lyophilization treatments were applied),followed by storage at room temperature(24℃–25℃)for 12 ***,the lyophilized and stored pollens were tested for viability by crossing with four female *** results showed that lyophilization is effective for achieving viable pollens after 12 months of *** 1(48 h drying)showed higher pollen germination and fertility rates than Treatment 2(72 h drying).Although we observed a reduction in viability of lyophilized pollens after 12 months of storage,we generated hybrid seedlings with success rates from 12%to 21%compared to 21%–31%when using fresh *** testing based on molecular genotyping confirmed the hybrid status of the obtained seedlings,which grew well in a *** is a practical approach for a long-term storage of greater yam pollen *** protocol will positively impact yam breeding programs particularly in developing countries.

AIM： To evaluate the efficacy and safety of laser-assisted subepithelial keratectomy （lASIk） for myopic correction done under thin flaps（120μm） and compare with results obtained under thick flaps（150μm）.METhODS： The study included 150 myopic eyes of 75 patients without previous refractive surgery who underwent lASIk prospectively. Two microkeratome heads （90 and 130） were used to created a flap with thickness of 120μm and 150μm,respectively. Thin flap group（120μm） included 75 eyes while thick flap group included 75 eyes. Follow-up period was 12 mo. Efficacy,l safety, and stability were evaluated and compared between the two ***： In 150 eyes,the mean preoperative spherical equivalent refraction was -8.65±2.6D,mean sphere was -4.4±3.5D, and mean cylinder was -1.0±1.3D. The amount of ablation was significantly larger in the thin flap （88.5±32.21μm） group than in the thick flap group （64±28.13μm）.Percentage of safety was higher in the thin flap group（94.8%） than in the thick flap group（91.7%）. There were no intraoperative complications, especially flap-related problems. Subjective symptoms of dry eye occurred in 20.7% and 33.3% of eyes in the thin and thick flap groups, respectively. CONClUSION： Thin-flap lASIk is effective and safe in correcting myopic defects. It achivevs better visual results, rapid visual recovery, and stable postoperative refraction than lASIk with thick flaps.
