Constructing 3D railway scenes helps improve the railway information construction ability and the comprehensive railway management ***,the existing modeling methods have disadvantages such as low flexibility in scene configuration,weak extensibility of scene objects and poor reusability of modeling *** enable multitype 3D railway scenes to be automatically constructed,a template-based knowledge reuse method was proposed in this ***,based on parsing of modeling operation characteristics and modeling knowledge expression,a modeling knowledge template was designed to store modeling knowledge and modeling operations in a parameterized *** contents of this template were described in terms of semantic information,geometric information,topologic information and reference system information,and the method for instantiating the modeling knowledge template was ***,a scene configuration template was designed to organize the instantiated modeling knowledge files and scene *** the use of a flexible tree structure,the scene configuration template enables extensibility of scene ***,the two types of templates were parsed in terms of scene organization,linear referencing information,geometric and topologic relationships,and semantic *** this way,different types of 3D railway scenes were generated *** a prototype system was developed,experiments on automatic modeling of two 3D railway scenes were carried out to verify the template-based knowledge reuse *** experimental results show that the proposed method can be used to generate multitype 3D railway scenes *** this method,model combination can be configured,model types can be extended and modeling knowledge can be reused.

Understanding the dynamics of gas-liquid two-phase flows is a challenge in the fields of nonlinear *** first construct and analyze a recurrence network from Chen's chaotic system and find that the network local statistic is feasible to characterize chaotic dynamics associated with unstable periodic *** we construct recurrence networks from gas-liquid two-phase flow experimental signals and associate the network topological statistic with the flow pattern *** results indicate that the recurrence network could be a powerful tool for the dynamic characterization of experimental gas-liquid two-phase flows.

Immunoassay is a powerful technique that uses highly specific antigenantibody interactions to detect biochemical targets such as proteins and *** a diagnostic tool,immunoassay is employed in the screening,diagnosis,and prognosis of diseases,which are crucial for the grasp and control of patient conditions in clinical *** the rapid development of nanotechnology,immunoassays based on nanoprobes have attracted more and more attention due to the advantages of high sensitivity,specificity,stability,and *** nanoprobes are nanoscale particles that can act as signal carriers or targeting agents for *** this paper,we review the recent advances in various types of nanoprobes for immunoassays,such as colloidal gold,quantum dots,magnetic nanoparticles,nanozymes,aggregation-induced emission,and up-conversion *** effect of the nanoprobe construction and synthesis methods on their detection performance deserves to be studied in *** also compare their detection ranges and limits in different immunoassay methods,such as lateral flow immunoassays,fluorescent immunoassays,and surface-enhanced Raman scattering ***,we discuss the benefits and challenges of nanoprobes in immunoassays and provide insights into their future *** study aims to offer a comprehensive and critical perspective on the role of nanoprobes in the field of immunoassays.

Cancer stem cells(CSCs)play a pivotal role in tumor initiation,proliferation,metastasis,drug resistance,and ***,targeting CSCs has emerged as a promising avenue for cancer ***,3-phosphoglycerate dehydrogenase(PHGDH)has been identified as being intricately associated with the regulation of numerous cancer stem ***,reports detailing the functional regulators of PHGDH that can mitigate the stemness across cancer types are *** this study,the novel“molecular glue”LXH-3-71 was identified,and it robustly induced degradation of PHGDH,thereby modulating the stemness of colorectal cancer cells(CRCs)both in vitro and in ***,LXH-3-71 was observed to form a dynamic chimera,between PHGDH and the DDB1-CRL E3 *** insights not only elucidate the anti-CSCs mechanism of the lead compound but also suggest that degradation of PHGDH may be a more viable therapeutic strategy than the development of PHGDH ***,compound LXH-3-71 was leveraged as a novel ligand for the DDB1-CRL E3 ligase,facilitating the development of new PROTAC molecules targeting EGFR and CDK4 degradation.

Coronavirus disease 2019(COVID-19)is a pandemic with no specific drugs and high *** most urgent need is to find effective *** sought to determine whether hydroxychloroquine(HCQ)application may reduce the death risk of critically ill COVID-19 *** this retrospective study,we included 550 critically ill COVID-19 patients who need mechanical ventilation in Tongji Hospital,Wuhan,from February 1,2020 to April 4,*** 550 patients received comparable basic treatments including antiviral drugs and antibiotics,and 48 of them were treated with oral HCQ treatment(200 mg twice a day for 7–10 days)in addition to the basic *** endpoint is fatality of patients,and inflammatory cytokine levels were compared between HCQ and non-hydroxychloroquine(NHCQ)*** found that fatalities are 18.8%(9/48)in HCQ group,which is significantly lower than 47.4%(238/502)in the NHCQ group(Pning of the treatment to 5.2(3.0–23.4)pg mL–1(P<0.05)at the end of the treatment in the HCQ group but there is no change in the NHCQ *** data demonstrate that addition of HCQ on top of the basic treatments is highly effective in reducing the fatality of critically ill patients of COVID-19 through attenuation of inflammatory cytokine ***,HCQ should be prescribed as a part of treatment for critically ill COVID-19 patients,with possible outcome of saving lives.

We herein describe AncPhore,a versatile tool for drug discovery,which is characterized by pharmacophore feature analysis and anchor pharmacophore(i.e.,most important pharmacophore features)steered molecular fitting and virtual *** analyses of numerous protein-ligand complexes using AncPhore revealed that anchor pharmacophore features are biologically important,commonly associated with protein conservative characteristics,and have significant contributions to the binding *** evaluation of AncPhore showed that it had substantially improved prediction ability on different types of target proteins including metalloenzymes by considering the specific contributions and diversity of anchor pharmacophore *** demonstrate the practicability of AncPhore,we screened commercially available chemical compounds and discovered a set of structurally diverse inhibitors for clinically relevant metallo-β-lactamases(MBLs);of them,4 and 6 manifested potent inhibitory activity to VIM-2,NDM-1 and IMP-1 *** analyses of VIM-2:4 complex revealed the precise inhibition mode of 4 with VIM-2,highly consistent with the defined anchor pharmacophore ***,we also identified new hit compounds by using AncPhore for indoleamine/tryptophan 2,3-dioxygenases(IDO/TDO),another class of clinically relevant *** work reveals anchor pharmacophore as a valuable concept for target-centered drug discovery and illustrates the potential of AncPhore to efficiently identify new inhibitors for different types of protein targets.

Objective This study aimed to investigate the effect and underlying mechanism of Fructus lycii in improving exercise *** A network pharmacological approach was used to explore potential mechanisms of action of Fructus *** muscle C2C12 cells and immunofluorescence were employed to verify the effect and mechanism of the representative components in Fructus lycii predicted by network pharmacological *** Six potential active components,namely quercetin,β-sitosterol,stigmasterol,7-Omethylluteolin-6-C-beta-glucoside_qt,atropine,and glycitein,were identified to have potency in improving exercise fatigue via multiple pathways,such as the PI3K-Akt,neuroactive ligand-receptor interaction,IL-17,TNF,and MAPK signaling *** immunofluorescence results indicated that quercetin,a significant active component in Fructus lycii,increased the mean staining area of 2-NBDG,TMRM,and MitoTracker,and decreased the area of CellRox compared to the ***,the protein expression levels of p-38 MAPK,p-MAPK,p-JNK,p-PI3K,and p-AKT markedly increased after quercetin *** Fructus lycii might alleviate exercise fatigue through multiple components and *** these,quercetin appears to improve exercise fatigue by enhancing energy metabolism and reducing oxidative *** PI3K-AKT and MAPK signaling pathways also appear to play a role in this process.

A common problem for Zn alloys is the trade-off between antibacterial ability and *** paper proposes a strategy to solve this problem by increasing release ratio of Ca^(2+)ions,which is realized by significant refinement of CaZn13 particles through bottom circulating water-cooled casting(BCWC)and *** with conventionally fabricated Zn-0.3Ca alloy,the BCWC-rolled alloy shows higher antibacterial abilities against *** and ***,meanwhile much less toxicity to MC3T3-E1 ***,plasticity,degradation uniformity,and ability to induce osteogenic differentiation in vitro of the alloy are *** elongation up to 49%,which is the highest among Zn alloys with Ca,and is achieved since the sizes of CaZn_(13) particles and Zn grains are small and *** a result,the long-standing problem of low formability of Zn alloys containing Ca has also been solved due to the elimination of large CaZn_(13) *** BCWC-rolled alloy is a promising candidate of making GBR membrane.
