Cotton is an important natural fiber crop worldwide which plays a vital role in our daily *** yield is a constant goal of cotton breeding,and lint percentage(LP)is one of the important components of cotton fiber yield.A stable QTL controlling LP,qLP_(A01.1),was identified on chromosome A01 from Gossypium hirsutum introgressed lines with *** chromosome segments in a previous *** fine-map qLP_(A01.1),an F2 population with 986individuals was established by crossing *** cultivar CCRI35 with the chromosome segment substitution line HT_390.A high-resolution genetic map including 47 loci and spanning 56.98 cM was constructed in the QTL region,and qLP_(A01.1)was ultimately mapped into an interval corresponding to an~80 kb genome region of chromosome A01in the reference genome,which contained six annotated *** data and sequence analysis revealed that S-acyltransferase protein 24(GoPAT24)might be the target gene of qLP_(A01.1).This result provides the basis for cotton fiber yield improvement via marker-assisted selection(MAS)and further studies on the mechanism of cotton fiber development.

为了使生物学类研究生具备系统完整的科研思维和实验技能,以及创新意识和创新精神,以西南大学原生物技术中心为平台,进行研究生实验室轮转训练模式的改革探索与实践。改革教学实践1年后,对38名研究生进行问卷式调查,从学生的基本情况、学生的需求、学生的认可、轮转训练效果和学生的意见反馈等方面,分析了实施研究生实验室轮转训练模式的必要性、优势和弊端。结果显示,在生物学类研究生培养模式中引入新型的实验室轮转训练初见成效,学生比较认可在开展研究生设计前进行的不同实验室之间及不同导师指导下的专项学习模式,其中94.73%的被调查学生认为有必要开设实验室轮转训练,有68.42%、55.26%学生的综合知识和专业知识分别得到了提高,与此同时,学生的实验技能及专业技能也得到了不同程度提升,为研究生课题的选择和设计,科研创新能力及综合素质的培养打下了坚实基础。

SCFP, a novel fiber-specific promoter in cotton

To investigate the expression pattern of GhSCFP which was isolated from cotton fiber cDNA library, a 1006 bp upstream fragment of the gene was cloned by chromosome walking and fused to GUS and GFP respectively. Histochemical GUS and GFP fluorescence analysis revealed that the expression of the report genes driven by the promoter sequence was detectable only in outer layer cells during the seed development in the transgentic tobaccos. In transgenic cotton, strong GUS activity was observed in spherical protrusions on 0 dpa (days post anthesis) ovule surface, and in the 2―36 dpa fiber cells, while no GUS signals were detected in the root, leaves, stem, corolla, anther and stigma. Our data demonstrated that GhSCFP upstream sequence is a cotton fiber-specific promoter and this promoter will be useful in the molecular research on fiber cell development and in cotton fiber improvements by genetic modification.

Phytosterol content and the campesterol:sitosterol ratio influence cotton fiber development: role of phytosterols in cell elongation

Phytosterols play an important role in plant growth and development, including cell division, cell elongation, embryogenesis, cellulose biosynthesis, and cell wall formation. Cotton fiber, which undergoes synchronous cell elongation and a large amount of cellulose synthesis, is an ideal model for the study of plant cell elongation and cell wall biogenesis. The role of phytosterols in fiber growth was investigated by treating the fibers with tfidemorph, a sterol biosynthetic inhibitor. The inhibition of phy- tosterol biosynthesis resulted in an apparent suppression of fiber elongation in vitro or in planta. The determination of phy- tosterol quantity indicated that sitosterol and campesterol were the major phytosterols in cotton fibers; moreover, higher con- centrations of these phytosterols were observed during the period of rapid elongation of fibers. Furthermore, the decrease and increase in campesterol：sitosterol ratio was associated with the increase and decease in speed of elongation, respectively, dur- ing the elongation stage. The increase in the ratio was associated with the transition from cell elongation to secondary cell wall synthesis. In addition, a number of phytosterol biosynthetic genes were down-regulated in the short fibers of ligon lintless-1 mutant, compared to its near-isogenic wild-type TM-1. These results demonstrated that phytosterols play a crucial role in cot- ton fiber development, and particularly in fiber elongation.

Effects of cooling rates on the mechanical properties of a Ti-based bulk metallic glass

Mechanical properties of the glassy specimens fabricated at different cooling rates with a composition of Ti40Zr25Cu12Ni3Be20 were systematically investigated. It was confirmed that faster cooling rates caused not only a larger amount of frozen-in free volume but also a higher glass transition temperature in the bulk glassy alloy. Increase in the free volume was found to favor plastic deformation and then to give rise to larger compressive plasticity, whilst the rise in the glass transition temperature seemed to be closely related to the higher yield strength. Moreover, the increase of yield strength and plasticity induced by fast cooling rates may also be associated with the residual stress generated during the fabrication process. Our results suggest that the deformation behavior of bulk metallic glasses is sensitive to various factors and influences from the other factors should be excluded as far as cooling-rate effects on bulk metallic glasses are considered.

Synergetic Control of Li+ Transport Ability and Solid Electrolyte Interphase by Boron-Rich Hexagonal Skeleton Structured All-Solid-State Polymer Electrolyte

High Li+transference number electrolytes have long been understood to provide attractive candidates for realizing uniform deposition of Li+.However,such electrolytes with immobilized anions would result in incomplete solid electrolyte interphase （SEI） formation on the Li anode because it suffers from the absence of appropriate inorganic components entirely derived from anions ***,a boron-rich hexagonal polymer structured all-solid-state polymer electrolyte （BSPE+ 10%LiBOB）with regulated intermolecular interaction is proposed to trade off a high Li+transference number against stable SEI *** Li+transference number of the as-prepared electrolyte is increased from 0.23 to 0.83 owing to the boron-rich cross-linker （BC） *** intriguingly,for the first time,the experiments combined with theoretical calculation results reveal that BOB-anions have stronger interaction with B atoms in polymer chain than TFSI-,which significantly induce the TFSI-decomposition and consequently increase the amount of LiF and Li3N in the SEI ***,a LiFePO4|BSPE+ 10%LiBOBlLi cell retains 96.7%after 400 cycles while the cell without BC-resisted electrolyte only retains 40.8%.BSPE+ 10%LiBOB also facilitates stable electrochemical cycling of solid-state Li-S *** study blazes a new trail in controlling the Li+transport ability and SEI properties,synergistically.

Cotton fiber is a raw material for the global textile industry and fiber quality is essential to its industrial *** are plant secondary metabolites that may serve as dietary components,regulate light harvesting,and scavenge reactive oxygen *** carotenoids accumulate predominantly in rapidly elongating cotton fibers,their roles in cotton fiber development remain poorly *** this study,a fiber-specific promoter proSCFP was applied to drive the expression of GhOR1Del,a positive regulator of carotenoid accumulation,to upregulate the carotenoid level in cotton fiber in *** length,strength,and fineness were increased in proSCFP:GhOR1Del transgenic cotton and abscisic acid(ABA)and ethylene contents were increased in elongating *** ABA downstream regulator GhbZIP27a stimulated the expression of the ethylene synthase gene GhACO3 by binding to its promoter,suggesting that ABA promoted fiber elongation by increasing ethylene *** findings suggest the involvement of carotenoids and ABA signaling in promoting cotton fiber elongation and provide a strategy for improving cotton fiber quality.
