Combo Packet:An Encryption Traffic Classification Method Based on Contextual Information

With the increasing proportion of encrypted traffic in cyberspace, the classification of encrypted traffic has becomea core key technology in network supervision. In recent years, many different solutions have emerged in this field.Most methods identify and classify traffic by extracting spatiotemporal characteristics of data flows or byte-levelfeatures of packets. However, due to changes in data transmission mediums, such as fiber optics and satellites,temporal features can exhibit significant variations due to changes in communication links and transmissionquality. Additionally, partial spatial features can change due to reasons like data reordering and retransmission.Faced with these challenges, identifying encrypted traffic solely based on packet byte-level features is significantlydifficult. To address this, we propose a universal packet-level encrypted traffic identification method, ComboPacket. This method utilizes convolutional neural networks to extract deep features of the current packet andits contextual information and employs spatial and channel attention mechanisms to select and locate effectivefeatures. Experimental data shows that Combo Packet can effectively distinguish between encrypted traffic servicecategories (e.g., File Transfer Protocol, FTP, and Peer-to-Peer, P2P) and encrypted traffic application categories (e.g.,BitTorrent and Skype). Validated on the ISCX VPN-non VPN dataset, it achieves classification accuracies of 97.0%and 97.1% for service and application categories, respectively. It also provides shorter training times and higherrecognition speeds. The performance and recognition capabilities of Combo Packet are significantly superior tothe existing classification methods mentioned.

Anti virulence strategy has been considered as one of the most promising approaches to combat drug-rcesistant bacterial infections.Porc-forming toxins(PFTs)are the largest class of bacterial toxins,inficting their virulence ffect through creating pores on the cell membrane.However,curent solutions for eliminating PFTs are mostly designed based on their molecular structure,requiring customized design for different interactions.In the present study,we employed erythroliposome(denoted as RM-PL),a biomimetic platform constructed by artificial lipid membranes and natural erythrocyle membranes,to necutralize different hemolytic PFTs regardless of their molecular structure.When tested with model PFTs,including a-hemolysin,listeriolysin O,and streptolysin O,RM-PL could completely inhibit toxin-induced hemolysis in a concentration-dependent manner.In vivo studies further confirmed that RM-PL.could eficiently neuralize various toxins and save animals'lives without causing damage to 0rgans or tissues.In addition,we explored the underlying mechanisms of this efficient detoxification ability and found that it was mainly macrophages in the spleen and the liver that took up RM-PL-absorbed toxins through a variety of endocytosis pathways and digested them in lysosomes.In summary,the biomimetic RM-PL presented a promising system for broad-spectrum and powerful toxin neutralization with a mech-anism of lysosome-mediaed loxin degradation.

Platelets buoy up cancer metastasis via arresting cancer cells, enhancing their adhesion, and facilitating their extravasation through the vasculature. When deprived of intracellular and granular contents, platelet decoys could prevent metastatic tumor formation. Inspired by these, we developed nanoplatesomes by fusing platelet membranes with lipid membranes(P-Lipo) to restrain metastatic tumor formation more efficiently. It was shown nanoplateletsomes bound with circulating tumor cells(CTC)efficiently, interfered with CTC arrest by vessel endothelial cells, CTC extravasation through endothelial layers, and epithelial-mesenchymal transition of tumor cells as nanodecoys. More importantly, in the mouse breast tumor metastasis model, nanoplateletsomes could decrease CTC survival in the blood and counteract metastatic tumor growth efficiently by inhibiting the inflammation and suppressing CTC escape. Therefore, nanoplatelesomes might usher in a new avenue to suppress lung metastasis.

Tumor-targeted immunotherapy is a remarkable breakthrough,offering the inimitable advantage of specific tumoricidal effects with reduced immune-associated cytotoxicity.However,existing platforms suffer from low efficacy,inability to induce strong immunogenic cell death(ICD),and restrained capacity of transforming immune-deserted tumors into immune-cultivated ones.Here,an innovative platform,perfluorooctyl bromide(PFOB)nanoemulsions holding MnO_(2) nanoparticles(MBP),was developed to orchestrate cancer immunotherapy,serving as a theranostic nanoagent for MRI/CT dual-modality imaging and advanced ICD.By simultaneously depleting the GSH and eliciting the ICD effect via highintensity focused ultrasound(HIFU)therapy,the MBP nanomedicine can regulate the tumor immune microenvironment by inducing maturation of dendritic cells(DCs)and facilitating the activation of CD8^(+)and CD4^(+)T cells.The synergistic GSH depletion and HIFU ablation also amplify the inhibition of tumor growth and lung metastasis.Together,these findings inaugurate a new strategy of tumor-targeted immunotherapy,realizing a novel therapeutics paradigm with great clinical significance.

Uncontrollable hemorrhage remains staple trouble in surgical procedures and a leading cause after major trauma.The bleeding issue may trigger various pathologic scenarios that can lead to tissue morbidities and mortalities,and currently available on-site hemostatic agents are confined to a narrow therapeutic index and may carry the risk of immunogenicity.Inspired by the crucial role of platelets in the process of thrombus,a platelet-mimetic plateletsome with wound targeting and blood coagulation properties is developed for hemorrhage control.Plateletsome is formulated by integrating platelet membranes with functionalized synthetic liposomes and exhibits superior wound targeting and effective hemostasis properties.It presents less blood loss and shorter hemostasis time than the platelet membrane vehicles or the conventional liposomes in the mouse tail transection model.The strong homing of the biomimetic plateletsome to the thrombus was also confirmed,demonstrating the potential of this engineered cell membrane vesicle as a biomimetic hemostat for bleeding treatment.

SARS-CoV-2 has been marked as a highly pathogenic coronavirus of COVID-19 disease into the human population,causing over 5.5 million confirmed cases worldwide.As COVID-19 has posed a global threat with significant human casualties and severe economic losses,there is a pressing demand to further understand the current situation and develop rational strategies to contain the drastic spread of the virus.Although there are no specific antiviral therapies that have proven effective in randomized clinical trials,currently,the rapid detection technology along with several promising therapeutics for COVID-19 have mitigated its drastic transmission.Besides,global institutions and corporations have commenced to parse out effective vaccines for the prevention of COVID-19.Herein,the present review will give exhaustive details of extensive researches concerning the drug discovery and therapeutic options for COVID-19 as well as some insightful discussions of the status of COVID-19.

Cancer immunotherapy has veered the paradigm of cancer treatment.Despite recent advances in immunotherapy for improved antitumor efficacy,the complicated tumor microenvironment(TME)is highly immunosuppressive,yielding both astounding and unsatisfactory clinical successes.In this regard,clinical outcomes of currently available immunotherapy are confined to the varied immune systems owing in large part to the lack of understanding of the complexity and diversity of the immune context of the TME.Various advanced designs of nanomedicines could still not fully surmount the delivery barriers of the TME.The immunosuppressive TME may even dampen the efficacy of antitumor immunity.Recently,some nanotechnology-related strategies have been inaugurated to modulate the immunosuppressive cells within the tumor immune microenvironment(TIME)for robust immunotherapeutic responses.In this review,we will highlight the current understanding of the immunosuppressive TIME and identify disparate subclasses of TIME that possess an impact on immunotherapy,especially those unique classes associated with the immunosuppressive effect.The immunoregulatory cell types inside the immunosuppressive TIME will be delineated along with the existing and potential approaches for immunosuppressive cell modulation.After introducing the various strategies,we will ultimately outline both the novel therapeutic targets and the potential issues that affect the efficacy of TIME-based nanomedicines.

针对区块链系统加密货币交易记录中存在的盗币异常行为,文章基于Ada Boost模型提出一种具有隐私保护功能的异常交易检测方案。该方案采用加法同态加密和矩阵混淆技术,在有效识别并预测异常交易的同时,保证交易数据的隐私性。此外,在云外包环境中设计实现方案的底层协议,并证明了方案的正确性和隐私保护性质。与同类协议相比,该方案在保证隐私性的同时,具有较高的检测准确率和召回率,平均每条记录的检测时间为毫秒级,适用于真实加密货币交易的检测场景。

Material Engineering in Gut Microbiome and Human Health

Tremendous progress has been made in the past decade regarding our understanding of the gut microbiome’s role in human health.Currently,however,a comprehensive and focused review marrying the two distinct fields of gut microbiome and material research is lacking.To bridge the gap,the current paper discusses critical aspects of the rapidly emerging research topic of“material engineering in the gut microbiome and human health.”By engaging scientists with diverse backgrounds in biomaterials,gut-microbiome axis,neuroscience,synthetic biology,tissue engineering,and biosensing in a dialogue,our goal is to accelerate the development of research tools for gut microbiome research and the development of therapeutics that target the gut microbiome.For this purpose,state-of-the-art knowledge is presented here on biomaterial technologies that facilitate the study,analysis,and manipulation of the gut microbiome,including intestinal organoids,gut-on-chip models,hydrogels for spatial mapping of gut microbiome compositions,microbiome biosensors,and oral bacteria delivery systems.In addition,a discussion is provided regarding the microbiome-gut-brain axis and the critical roles that biomaterials can play to investigate and regulate the axis.Lastly,perspectives are provided regarding future directions on how to develop and use novel biomaterials in gut microbiome research,as well as essential regulatory rules in clinical translation.In this way,we hope to inspire research into future biomaterial technologies to advance gut microbiome research and gut microbiome-based theragnostics.
