CLP gene family,a new gene family of Cotesia vestalis bracovirus inhibits melanization of Plutella xylostella hemolymph

Polydnaviruses(PDVs)are obligatory symbionts of parasitoid wasps and play an important role in suppressing host immune defenses.Although PDV genes that inhibit host melanization are known in Microplitis bracovirus,the functional homologs in Cotesia bracoviruses remain unknown.Here,we find that Cotesia vestalis bracovirus(CvBV)can inhibit hemolymph melanization of its host,Plutella xylostella larvae,during the early stages of parasitization,and that overexpression of highly expressed CvBV genes reduced host phenoloxidase activity.Furthermore,CvBV-7-I in particular reduced host phenolox-idase activity within 12 h,and the injection of anti-CvBV-7-1 antibody increased the melanization of parasitized host larvae.Further analyses showed that CvBV-7-1 and three homologs from other Cotesia bracoviruses possessed a C-terminal leucine/isoleucine-rich region and had a similar flinction in inhibiting melanization.Therefore,a new family of bracovirus genes was proposed and named as C-terminal Leucine/isoleucine-rich Protein(CLP).Ectopic expression of CvBV-7-1 in Drosophila hemocytes increased susceptibility to bacterial repression of melanization and reduced the melanotic encapsulation of par-asitized D.melanogaster by the parasitoid Leptopilina boulardi.The formation rate of wasp pupae and the eclosion rate of C.vestalis were affected when the function of CvBV-7-1 was blocked.Our findings suggest that CLP genes from Cotesia bracoviruses encoded proteins that contain a C-terminal leucine/isoleucine-rich region and function as melanization inhibitors during the early stage of parasitization,which is important for successful parasitization.

Odorant-binding proteins(OBPs)are crucial in insect's olfactory perception,which participate in the initial step of odorant molecules transporting from the external environment to olfactory receptor neurons.To better understand the roles for OBPs in olfactory perception in Cotesia vestalis,a solitary larval endoparasitoid of diamondback moth,Plutella xylostella,we have comprehensively screened the genome of C.vestalis,and obtained 20 CvesOBPs,including 18 classic OBPs and two minus-C OBPs.Motif-pattern analysis indicates that the motifs of C.vestalis OBPs are highly conserved in Hymenoptera.The results of tissue expression analysis show that five OBPs(CvesOBP1/11/12/14/16)are highly expressed in male antennae,whereas six other OBP genes(CvesOBP7/8/13/17/18/19)are significantly transcriptionally enriched in female antennae.The results of RNA interference experiments for three most highly expressed OBP genes(CvesOBP17/18/19)in female antennae demonstrate that they are likely involved in parasitic processes of female wasps,as the wasps take a longer time to target the hosts when they are knoc ked down.

Single thiolate replacement of metal nanoclusters

Surface thiolates play important roles in evincing the structures and properties of thiolated metal nanoclusters—one type of recently emerging inorganic-organic hybrids,and thus the thiolate substitution,especially single thiolate substitution,is highly desirable for subtly tailoring the structures and properties of metal nanoclusters.However,such a single-thiolate substituting is challenging,and its influence on the metal-metal and metal-sulfur bonds remains mysterious due to the absence of the singlethiolate-substituted structure.Here,we developed a combined method,concurrently synthesized the single-thiolate-substituted nanocluster and its parent nanocluster,and successfully resolved their structures by single crystal X-ray crystallography,which reveals that the single thiolate substitute has an obvious influence on the metal-metal and metal-sulfur bond lengths although it has no effect on the absorption profile.Interestingly,the metal-metal and metal-sulfur bonds show various thermal extensibility and even the negative thermal expansion phenomena of the Au–S bond were observed in the single-thiolate-substituted nanocluster.The bond length-related stability was also observed.Overall,this study highlights a novel synthesis method and offers novel structural insights and an in-depth structure-property correlation of thiolated metal nanoclusters.

The role of proteasomes in tumorigenesis

Protein homeostasis is the basis of normal life activities,and the proteasomefamily plays an extremely important function in this process.The proteasome 2os is a concentric circle structure with twoαrings and twoβrings overlapped.The proteasome 2os can perform both ATP-dependent and non-ATP-dependent ubiquitination proteasome degradation by binding to various subunits(such as 19S,11S,and 200 PA),which is performed by its active subunitβ1,β2,andβ5.The proteasome can degrade misfolded,excess proteins tomaintain homeostasis.At the same time,it can be utilized by tumors to degrade over-proliferate and unwanted proteins to support their growth.Proteasomes can affect the development of tumors from several aspects including tumor signaling pathways such as NF-kB and p53,cell cycle,immune regulation,and drug resistance.Proteasome-encoding genes have been found to be overexpressed in a variety of tumors,providing a potential novel target for cancer therapy.In addition,proteasome inhibitors such as bortezomib,carfilzomib,and ixazomib have been put into clinical application as the first-line treatment of multiple myeloma.More and more studies have shown that it also has different therapeutic effects in other tumors such as hepatocellular carcinoma,non-small cell lung cancer,glioblastoma,and neuroblastoma.However,proteasome inhibitors are not much effective due to their tolerance and singleness in other tumors.Therefore,further studies on their mechanisms of action and drug interactions are needed to investigate their therapeutic potential.

Psychological interventions for depression in children and adolescents:A bibliometric analysis

BACKGROUND Depression has gradually become a common psychological disorder among children and adolescents.Depression in children and adolescents affects their physical and mental development.Psychotherapy is considered to be one of the main treatment options for depressed children and adolescents.However,our understanding of the global performance and progress of psychological interventions for depression in children and adolescents(PIDCA)research is limited.AIM To identify collaborative research networks in this field and explore the current research status and hotspots through bibliometrics.METHODS Articles and reviews related to PIDCA from January 2010 to April 2023 were identified from the Web of Science Core Collection database.The Charticulator website,CiteSpace and VOSviewer software were used to visualize the trends in publications and citations,the collaborative research networks(countries,institutions,and authors),and the current research status and hotspots.RESULTS Until April 16,2023,1482 publications were identified.The number of documents published each year and citations had increased rapidly in this field.The United States had the highest productivity in this field.The most prolific institution was the University of London.Pim Cuijpers was the most prolific author.In the context of research related to PIDCA,both reference co-citation analysis and keywords co-occurrence analysis identified 10 research hotspots,including thirdwave cognitive behavior therapy,short-term psychoanalytic psychotherapy,cognitive behavioral analysis system of psychotherapy,family element in psychotherapy,modular treatment,mobile-health,emotion-regulation-based transdiagnostic intervention program,dementia risk in later life,predictors of the efficacy of psychological intervention,and risks of psychological intervention.CONCLUSION This bibliometric study provides a comprehensive overview of PIDCA from 2010 to present.Psychological intervention characterized as psychological-process-focused,short,family-involved,modular,inte

Utilizing a metal-forging inspired chain combing strategy to enhance properties and expand applications of Nylon 66 plastic via heat inducing

The mechanical properties of engineering plastics can be enhanced through effective surface mechanical treatment(SMT),which can be applied to various types of engineering plastics,eliminating the limitations of conventional polymer processing and could potentially extend their applications and improve their performance and reliability as structural-functional materials.Inspired by metal forging,this work proposes a simple and effective SMT strategy to enhance the mechanical properties of polyamide 66(PA66).Tensile tests have shown that SMTed PA66 samples exhibit significant improvements in both Young’s modulus and ultimate tensile strength(UTS),with a 2104 MPa Young’s modulus,almost double that of the pristine samples,and a 35.56%increase in UTS,reaching 183 MPa.Additionally,the modulus within the localized SMTed surface layer could reach up to 14 GPa,which is approximately 14 times higher than that of the pristine sample.The Vickers hardness within the localized SMTed surface layer can be doubled,reaching 10.72 Hv,and the crystallinity can increase by approximately 20%compared to the untreated region.Furthermore,force field molecular dynamics(FFMD)simulations were conducted to investigate the ternary relationship between the SMT method,PA66’s molecular structure,and its properties.The combination of MD simulations and versatile structural characterizations provides evidence that the SMT method’s mechanism,under heat induction,results in a chain-combing procedure that changes the polymer’s molecular morphology microscopically and enhances its mechanical properties macroscopically.

Universal dynamic scaling and Contact dynamics in quenched quantum gases

Recently universal dynamic scaling is observed in several systems,which exhibit a spatiotemporal self-similar scaling behavior,analogous to the spatial scaling near phase transition.The latter one arises from the emergent continuous scaling symmetry.Motivated by this,we investigate the possible relation between the scaling dynamics and the continuous scaling symmetry in this paper.We derive a theorem that the scaling invariance of the quenched Hamiltonian and the initial density matrix can lead to the universal dynamic scaling.It is further demonstrated both in a two-body system analytically and in a many-body system numerically.For the latter one,we calculate the dynamics of quantum gases quenched from the zero interaction to a finite interaction via the non-equilibrium high-temperature virial expansion.A dynamic scaling of the momentum distribution appears in certain momentum-time windows at unitarity as well as in the weak interacting limit.Remarkably,this universal scaling dynamics persists approximately with smaller scaling exponents even if the scaling symmetry is fairly broken.Our findings may offer a new perspective to interpret the related experiments.We also study the Contact dynamics in the BEC−BCS crossover.Surprisingly,the half-way time displays a maximum near unitarity while some damping oscillations occur on the BEC side due to the dimer state,which can be used to detect possible two-body bound states in experiments.

Historical Changes and Multi-scenario Prediction of Land Use and Terrestrial Ecosystem Carbon Storage in China

Terrestrial carbon storage(CS)plays a crucial role in achieving carbon balance and mitigating global climate change.This study employs the Shared Socioeconomic Pathways and Representative Concentration Pathways(SSPs-RCPs)published by the Intergovernmental Panel on Climate Change(IPCC)and incorporates the Policy Control Scenario(PCS)regulated by China’s land management policies.The Future Land Use Simulation(FLUS)model is employed to generate a 1 km resolution land use/cover change(LUCC)dataset for China in 2030 and 2060.Based on the carbon density dataset of China’s terrestrial ecosystems,the study analyses CS changes and their relationship with land use changes spanning from 1990 to 2060.The findings indicate that the quantitative changes in land use in China from 1990 to 2020 are characterised by a reduction in the area proportion of cropland and grassland,along with an increase in the impervious surface and forest area.This changing trend is projected to continue under the PCS from 2020 to 2060.Under the SSPs-RCPs scenario,the proportion of cropland and impervious surface predominantly increases,while the proportions of forest and grassland continuously decrease.Carbon loss in China’s carbon storage from 1990 to 2020 amounted to 0.53×10^(12)kg,primarily due to the reduced area of cropland and grassland.In the SSPs-RCPs scenario,more significant carbon loss occurs,reaching a peak of8.07×10^(12)kg in the SSP4-RCP3.4 scenario.Carbon loss is mainly concentrated in the southeastern coastal area and the Beijing-TianjinHebei(BTH)region of China,with urbanisation and deforestation identified as the primary drivers.In the future,it is advisable to enhance the protection of forests and grassland while stabilising cropland areas and improving the intensity of urban land.These research findings offer valuable data support for China’s land management policy,land space optimisation,and the achievement of dual-carbon targets.

Covalent organic frameworks(COFs),a rapidly developing category of crystalline conjugated organic polymers,possess highly ordered structures,large specific surface areas,stable chemical properties,and tunable pore microenvironments.Since the first report of boroxine/boronate ester-linked COFs in 2005,COFs have rapidly gained popularity,showing important application prospects in various fields,such as sensing,catalysis,separation,and energy storage.Among them,COFs-based electrochemical(EC)sensors with upgraded analytical performance are arousing extensive interest.In this review,therefore,we summarize the basic properties and the general synthesis methods of COFs used in the field of electroanalytical chemistry,with special emphasis on their usages in the fabrication of chemical sensors,ions sensors,immunosensors,and aptasensors.Notably,the emerged COFs in the electrochemiluminescence(ECL)realm are thoroughly covered along with their preliminary applications.Additionally,final conclusions on state-of-the-art COFs are provided in terms of EC and ECL sensors,as well as challenges and prospects for extending and improving the research and applications of COFs in electroanalytical chemistry.
