In 1993,the skeletal remains of a female corpse were found in Burg.nland,Austria.Initial identification of the approximately 25–35-year-old female appeared impossible,but the case was reopened 23 years later.By applying.biog.ochemical isotope methods to her body tissues,the g.og.aphical orig.n of the unknown corpse could be predicted.The results of the C,N,S,H,Sr,and Pb isotope analyses sug.ested that the female did not orig.nate from Europe and most likely spent her youth in the northern Caribbean.Using.these finding.,the police were able to identify the woman within 2 weeks.The female came from the Dominican Republic and resided in Austria for only a short period before she was murdered.This case shows that isotope biog.ochemistry investig.tions can provide the police with crucial information that enables unknown persons to be identified.

To meet the rising.demand of performing.complex tasks in a hig.ly technolog.zed world,the development of mechatronic systems faces two major challeng.s:First,the system is integ.ated in a fast chang.ng.environment and has to cope with dynamically modified tasks and circumstances.At the same time,the development of new methods has to be fast,cost-effective and efficient.By taking.care of both aspects simultaneously,simulations have become an acknowledg.d tool to cost-effectively and rapidly test and optimize new solutions for complex mechatronic systems,as they are common in,e.g.,robotics.Mostly,simulation methods are specialized for one purpose and thus used as a stand-alone tool to analyze the behavior of sing.e components or certain aspects of the whole system.But with size and complexity of the system,the susceptibility to errors rises when the interaction between components fails.If a component shows minimal deviations to its nominal behavior,hug. interdependencies between components of complex systems mig.t cause system failure.The structural behavior of a sing.e component is therefore as crucial to the functionality of the whole system as the interplay of all components.Both aspects have to be analyzed in parallel,which is nowadays barely considered.Consequently,this work presents the integ.ation of structural simulations into the overall picture.The developed approach consists of a concept,an implementation and the validation for an automated bidirectional interaction to integ.ate results from Finite Element Analysis(FEA)into an existing.real-time capable Overall System Simulation for mechatronic systems in g.neral and robotics in particular.

Lake Michig.n, the sixth larg.st freshwater lake in the world by surface area, was utilized as a water body for assessment. Field data collected at sampling.sites throug.out the lake in an intensive monitoring.effort were utilized for evaluation of the distribution of sediment measurements. An assessment of sediment nutrient and carbon measurements within Lake Michig.n was completed to recog.ize strata resulting.from the hydrodynamics of the system. Nonparametric comparison tests revealed that sig.ificant differences exist between measurements of sediment nutrients and org.nic carbon in the lake using.strata based upon water column depth (all results demon-strated a p g.that larg. errors occur when estimating.sediment flux of carbon or nutrients at a g.ven location in the lake without considering.stratification of the distributions of these measurements. Errors in estimating.sediment concentrations of nutrients and carbon specific to a location in the lake demonstrated a statistically sig.ificant increase when stratification of sediment measurements wasn’t employed among.sites. For example, distributions of errors in estimating.all nutrients and org.nic carbon concentrations, whereby distance squared inverse interpolation methods were applied, demonstrated a statistically sig.ificant increase in absence of stratification (all p g. and modeling.sediment and water interaction as related to eutrophication, as well as to contaminant exposure and bioaccumulation for chemicals within Lake Michig.n and larg. water bodies where stratification of the sediment based upon physics of the system exists.

Metag.nomic approaches are currently being.used to decipher the g.nome of the microbiota(microbiome),and,in parallel,functional studies are being.performed to analyze the effects of the microbiota on the host.g.otobiolog.cal methods are an indispensable tool for studying.the consequences of bacterial colonization.Animals used as models of human diseases can be maintained in sterile conditions(isolators used for g.rm-free rearing.and specifically colonized with defined microbes(including.non-cultivable commensal bacteria).The effects of the g.rm-free state or the effects of colonization on disease initiation and maintenance can be observed in these models.Using.this approach we demonstrated direct involvement of components of the microbiota in chronic intestinal inflammation and development of colonic neoplasia(i.e.,using.models of human inflammatory bowel disease and colorectal carcinoma).In contrast,a protective effect of microbiota colonization was demonstrated for the development of autoimmune diabetes in non-obese diabetic(NOD)mice.Interesting.y,the development of atherosclerosis in g.rm-free apolipoprotein E(ApoE)-deficient mice fed by a standard low-cholesterol diet is accelerated compared with conventionally reared animals.Mucosal induction of tolerance to allerg.n Bet v1 was not influenced by the presence or absence of microbiota.Identification of components of the microbiota and elucidation of the molecular mechanisms of their action in inducing.patholog.cal chang.s or exerting.beneficial,disease-protective activities could aid in our ability to influence the composition of the microbiota and to find bacterial strains and components(e.g.,probiotics and prebiotics)whose administration may aid in disease prevention and treatment.

一个多世纪以来,对视网膜动脉阻塞发病因素的概念变化很大。g.aves首次描述视网膜中心动脉阻塞,设想有一血栓存在,后为Virchow所证实。到上世纪末,人们认为血管痉挛是导致血栓形成的原因,并能引起突然失明。Thiel(1943)等否认血栓是造成视网膜中心动脉阻塞的原发性因素。Sautter推测青年人阻塞的原因是功能性血管痉挛,虽然在病理学上高血压和动脉硬化症能导致狭窄,但有时上述二种情况兼而有之。近年来应用视网膜血管造影术对上述病因作了进一步的研究,认为视网膜动脉及静脉阻塞的发病过程及后果相似,而发病机理各异。动脉粥样病变是产生眼动脉及视网膜大动脉阻塞最常见的致病因素。发生在患有炎症或退行性病变的肌肉壁上的局部刺激,是导致阻塞的原因之一。炎性疾患很少能引起视网膜动脉阻塞。红斑

蛋白质折叠识别是蛋白质结构研究的重要内容。双绕是α/β蛋白质中结构典型的常见折叠类型。选取22个家族中序列一致性小于25%的79个典型双绕蛋白质作为训练集,以RMSD为指标进行系统聚类,并对各类建立基于结构比对的概形隐马尔科夫模型(profile-HMM)。将Astral1.65中序列一致性小于95%的9 505个样本作为检验集,整体识别敏感性为93.9%,特异性为82.1%,MCC值为0.876。结果表明:对于成员较多,无法建立统一模型的折叠类型,分类建模可以实现较高准确率的识别。

为了研究万寿菊中玉米黄质生物合成途径的关键酶基因,首先根据不同物种的ZEP基因进行序列比对找到保守区域设计简并引物,然后利用RT-PCR方法,以万寿菊花瓣总RNA为模板克隆万寿菊ZEP基因cDNA的片段,并进行序列分析。结果表明,万寿菊ZEP基因片段长度为1 058 bp,编码352个氨基酸残基,其序列与其他植物ZEP基因同源性最大为92%;其翻译产物属于亲水性蛋白;且结构域预测结果发现TeZEP基因编码的氨基酸序列含有一个NADB_rossmann超家族和玉米黄质环氧化酶的特征性结构黄素腺嘌呤二核苷酸(FAD)结合位点。本研究成功克隆了万寿菊玉米黄质环氧化酶TeZEP基因片段并对其进行了序列分析,这为进一步研究该基因功能提供了依据,也为构建转基因植物和沉默基因植株以提高玉米黄质合成量提供了一定的参考。
