Lung cancer ranks the top of malignancies that cause cancer-related deaths *** leaves of Morus alba L are traditional Chinese medicine widely applied in respiratory *** previous work has demonstrated the anti-lung cancer effect of secondary metabolites of mulberry leaf,but their mechanism of action has still not fully *** synthesized Moracin N(MAN)-Probe conjugated with alkyne to label lung cancer cells and identified protein targets by chemical proteomic *** and its probe exerted similar growth-inhibitory effect on human lung cancer *** proteomic results showed that MAN targeted the programmed death ligand 1(PD-L1)checkpoint pathway and T cell receptor(TCR)signaling pathway,indicating its immune-regulatory ***-free surface plasmon resonance(SPR)results showed the direct interaction of MAN with PD-L1 *** docking analysis demonstrated that MAN bound to E158 residue of PD-L1 *** downregulated the expression levels of PD-L1 in a time-and dose-dependent manner and disrupted the PD-L1/programmed death 1(PD-1)binding,including other secondary metabolites of mulberry leaves Guangsangon E(GSE)and Chalcomoracin(CMR).Human peripheral blood mononuclear cells(PBMCs)co-cultured with MAN-treated A549 cells,resulting in the increase of CD8^(+)GZMB^(+)T cells and the decrease of CD8^(+)PD-1^(+)T *** suggested that MAN exerts anti-cancer effect through blocking the PD-L1/PD-1 *** vivo,MAN combined with anti-PD-1 antibody significantly inhibited lung cancer development and metastasis,indicating their synergistic *** together,secondary metabolites of mulberry leaves target the PD-L1/PD-1 signaling,enhance T cell-mediated immunity and inhibit the tumorigenesis of lung *** modulatory effect on tumor microenvironment makes them able to enhance the therapeutic efficacy of immune checkpoint inhibitors in lung cancer.

差异化收费作为调控高速公路流量时空分布的手段,研究该条件下货车出行行为可为制定差异化收费方案提供量化分析依据。利用甘肃省张掖至酒泉段货车出行意向调查数据,建立MNL模型、路径选择分别位于上下层的2种NL模型以及CNL模型,标定结果表明CNL模型拟合精度最高。对CNL模型进行选择枝效用弹性分析,利用标定后的选择模型结合现状数据进行模拟,对优惠组合条件-流量曲面进行梯度分析。经弹性系数分析,夜间-非收费公路选择枝的直接弹性系数为-0.044与-0.13,说明夜间行驶于非收费公路的货车驾驶员更易因效用函数变化改选其他方案;经曲面梯度分析,可得当折扣组合坐标为(0.318,0.354)时梯度的模取得最大值,即优惠额在白天31.8%、夜间35.4%时对货车吸引效率最高,优惠比例超过此优惠组合时吸引效率将逐渐递减。

人口状况是一个国家最基本、最重要的国情,也是开展交通规划需要考虑的基本要素。在我国人口出现新情况、新特征和新趋势的背景下,需要加快推动开展适应人口变化的交通规划工作。本文结合美国、日本与英国在人口总量、年龄结构、空间分布与城乡人口流动的发展变动特点,分析归纳了国外在规划机制方法、设施建设养护、城市群交通、城乡交通一体化等方面的规划经验,重点从拓展人口参数应用、扩大交通运输有效投资、优化城市群快速运输通道、加强城乡交通一体化、完善无障碍出行环境等方面,研究提出我国交通规划发展的启示与建议。研究结果对于促进我国交通规划与人口变化的良性互动发展具有一定意义。

大麻素受体1通过G_(αi/o)/RhoA信号通路促进急性肺损伤小鼠巨噬细胞M1极化

目的·探讨阻断大麻素受体1(cannabinoid receptor 1,CB1)对小鼠急性肺损伤(acute lung injury,ALI)的影响及其潜在的分子机制。方法·将40只小鼠随机分为空白对照组、AM281(CB1拮抗剂)对照组、脂多糖(lipopolysaccharide,LPS)组、LPS+AM281组,每组10只。利用LPS诱导建立小鼠ALI模型。经苏木精-伊红(hematoxylin and eosin,H-E)染色观察各组小鼠肺组织病理表现并计算炎症评分。通过反转录和实时荧光定量聚合酶链反应(real-time fluorescent quantitative polymerase chain reaction,qPCR)检测各组小鼠肺巨噬细胞M1型标志物[肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)、IL-12]和M2型标志物[精氨酸酶(arginase,Arg)、甘露糖受体C型2(mannose receptor,C type 2,Mrc2)、巨噬细胞半乳糖型凝集素1(macrophage galactose-type lectin 1,Mgl1)]的mRNA水平。体外培养人髓系白血病单核细胞THP-1,通过免疫荧光技术检测THP-1细胞中CB1和CB2的表达,并在进一步阻断CB1及抑制G_(αi/o)/RhoA信号通路后,检测M1型标志物的mRNA水平。结果·LPS组小鼠肺组织损伤明显,炎症评分明显升高;阻断CB1后,与LPS组相比,LPS+AM281组小鼠肺损伤得到改善,表现为肺泡壁毛细血管充血改善、肺间质及肺泡腔内炎症细胞浸润减少,炎症评分下降(P=0.007)。与对照组相比,LPS组小鼠肺组织中M1型标志物水平上调,而在阻断CB1后巨噬细胞极性发生改变,M1/M2比例发生反转(均P<0.05)。体外研究发现,巨噬细胞表达CB1和CB2,利用花生四烯基-2-氯乙酰胺(arachidonyl-2-chloroethylamide,ACEA)激活CB1可上调M1型标志物的表达,阻断CB1及选择性抑制G_(αi/o)/RhoA信号通路后,M1型标志物的表达均明显下调(均P<0.05)。结论·CB1在ALI中通过G_(αi/o)/RhoA信号通路促进巨噬细胞向M1极化,阻断CB1可改善肺损伤。

Effect of the withdrawal rate on the microstructure and thermal durability of a third-generation single crystal superalloy

Single crystal Ni-based superalloys are the typical structural materials for high-pressure turbine blades,and their microstructure is critical in determining their mechanical *** withdrawal rate is a key parameter affecting the microstructure during the single crystal growth *** the present work the effect of the withdrawal rate on the microstructure of a third-generation single crystal superalloy containing 6.8 wt% Re has been investigated,and the creep resistance of the alloy *** results showed that increased withdrawal rate refined the dendritic structure,reduced dendritic arm spacing,promoted the growth of secondary tertiary dendrites and decreased solidification segregation with a reduced size of γ’ *** porosity density of the ascast alloy first decreased and then increased with the withdrawal rate,while the minimum porosity densityoccurred when the alloy was under the solidification condition of withdrawal rate of 4.5 mm/*** maximum creep rupture life of 326.4 h of the heat-treated alloys under the test condition of 1100℃/140 MPa also appeared at the alloys under the withdrawal rates of 4.5 mm/*** is believed that the minimum porosity density and reduced size of the γ’ phase may be the main reasons for the enhanced creep rupture life of the alloys with withdrawal rates of 4.5 mm/*** investigation provides theoretical support and a practical basis for the development of third-generation single crystal superalloys.

Objective To study the relationship between PK, AE and SNPs in Chinese renal carcinoma patients. Methods Establish a high performance liquid chromatography-tandam mass spectrometry(HPLC-MS/MS) method for determining the concentration of sunitinib and N-desethyl sunitinib(SU12662) in human plasma. Research the frequencies of adverse events after orally administrated sunitinib occurred in Chinese renal carcinoma patients and the relationship between AEs and concentration of sunitinib and SU12662. Multiple SNaPShot detection method to genotyped CYP3 A4(rs35599367),CYP3 A5(rs776746), ABCB1(rs2032582, rs1128503, rs1045642), ABCG2(rs2231142), PDGFRα(rs1800812), and FLT3(rs1933437). Using univariate and multivariate logistic regression, to analyze the relationship between the genotype and AE. Population PK analysis was performed by Pheonix(Version 1.4) with NLME(nonlinear mixed effect modeling). First order conditional estimates(FOCE) was applied for all estimations. Diagnostic plots, Bootstrap and Visual Predictive Check(VPC) were used to to evaluate the stability and validity of the final model. Study the population PK of sunitinib in Chinese renal carcinoma patients. Explore the impact factors in the population PK of sunitinib. Results The HPLC-MS/MS method for determining the concentration of sunitinib and N-desethyl sunitinib(SU12662) in human plasma was successfully established. The calibration curve for plasma sunitinib was linear in the range of 0.5-200 ng·mL-1(r2=0.9975). The calibration curve for plasma SU12662 was linear in the range of 0.25-100.00 ng·mL-1(r2=0.9998), while the intra-dayand inter-day precisions were all less than15% and the accuracies were with in the range of 15%. Hand-foot syndrome(HFS), hypertension, thrombocytopenia, diarrhea and leucopenia are the most common AEs that happened after administrating sunitinib.
