Metabolic profiles and morphological characteristics of leaf tips among different sweet potato(Ipomoea batatas Lam.)varieties

Sweet potato leaf tips have high nutritional value,and exploring the differences in the metabolic profiles of leaf tips among different sweet potato varieties can provide information to improve their qualities.In this study,a UPLC-Q-Exactive Orbitrap/MS-based untargeted metabolomics method was used to evaluate the metabolites in leaf tips of 32 sweet potato varieties.Three varieties with distinct overall metabolic profiles(A01,A02,and A03),two varieties with distinct profiles of phenolic acids(A20 and A18),and three varieties with distinct profiles of flavonoids(A05,A12,and A16)were identified.In addition,a total of 163 and 29 differentially expressed metabolites correlated with the color and leaf shape of sweet potato leaf tips,respectively,were identified through morphological characterization.Group comparison analysis of the phenotypic traits and a metabolite-phenotypic trait correlation analysis indicated that the color differences of sweet potato leaf tips were markedly associated with flavonoids.Also,the level of polyphenols was correlated with the leaf shape of sweet potato leaf tips,with lobed leaf types having higher levels of polyphenols than the entire leaf types.The findings on the metabolic profiles and differentially expressed metabolites associated with the morphology of sweet potato leaf tips can provide useful information for breeding sweet potato varieties with higher nutritional value.

Tumor-derived exosomes induce initial activation by exosomal CD19 antigen but impair the function of CD19-specific CAR T-cells via TGF-βsignaling

Tumor-derived exosomes (TEXs) enriched in immune suppressive molecules predominantly drive T-cell dysfunction and impair antitumor immunity. Chimeric antigen receptor (CAR) T-cell therapy has emerged as a promising treatment for refractory and relapsed hematological malignancies, but whether lymphoma TEXs have the same impact on CAR T-cell remains unclear. Here, we demonstrated that B-cell lymphoma-derived exosomes induce the initial activation of CD19-CAR T-cells upon stimulation with exosomal CD19. However, lymphoma TEXs might subsequently induce CAR T-cell apoptosis and impair the tumor cytotoxicity of the cells because of the upregulated expression of the inhibitory receptors PD-1, TIM3, and LAG3 upon prolonged exposure. Similar results were observed in the CAR T-cells exposed to plasma exosomes from patients with lymphoma. More importantly, single-cell RNA sequencing revealed that CAR T-cells typically showed differentiated phenotypes and regulatory T-cell (Treg) phenotype conversion. By blocking transforming growth factor β (TGF-β)-Smad3 signaling with TGF-β inhibitor LY2109761, the negative effects of TEXs on Treg conversion, terminal differentiation, and immune checkpoint expression were rescued. Collectively, although TEXs lead to the initial activation of CAR T-cells, the effect of TEXs suppressed CAR T-cells, which can be rescued by LY2109761. A treatment regimen combining CAR T-cell therapy and TGF-β inhibitors might be a novel therapeutic strategy for refractory and relapsed B-cell lymphoma.

Impact of oxygen incorporation on interface optimization and defect suppression for efficiency enhancement in Cu2ZnSn（S,Se）4 solar cells

Kesterite Cu2ZnSn（S,Se）4（CZTSSe） solar cells suffer from severe carrier recombination, limiting the photovoltaic performance. Unfavorable energy band alignment at the p-n junction and defective front interface are two main causes. Herein, oxygen incorporation in CZTSSe via absorber air-annealing was developed as a strategy to optimize its surface photoelectric property and reduce the defects. With optimized oxygen incorporation conditions, the carrier separation and collection behavior at the front interface of the device is improved. In particular, it is found that oxygen incorporated absorber exhibits increased band bending, larger depletion region width, and suppressed absorber defects. These indicate the dynamic factors for carrier separation become stronger. Meanwhile, the increased potential difference between grain boundaries and intra grains combined with the decreased concentration of interface deep level defect in the absorber provide a better path for carrier transport. As a consequence, the champion efficiency of CZTSSe solar cells has been improved from 9.74% to 12.04% with significantly improved open-circuit voltage after optimized air-annealing condition. This work provides a new insight for interface engineering to improve the photoelectric conversion efficiency of CZTSSe devices.

临界滑动面的合理确定是边坡稳定性分析的关键问题之一。传统的边坡滑动面假设方法构造生成的潜在滑动面形式单一,且无法考虑复杂边坡中土层强度参数变化对临界滑动面形状的影响,从而降低了边坡稳定性分析结果的可靠性。基于此,提出一种新的广义对数螺旋型滑面模型,该模型可生成包含传统圆弧滑动面和对数螺旋滑动面在内的多种形式的潜在滑动面;并针对非均质边坡中各土层抗剪强度不同的特点,采用多中心分段构造策略,将该滑面模型应用于非均质边坡稳定性分析中。基于极限平衡理论计算滑动面安全系数并搜索临界滑动面,对边坡进行稳定性评估。选取多个均质边坡及多种型式的非均质边坡(水平分层、倾斜分层和斜折线分层)经典算例与现有研究成果以及数值模拟软件获得的结果进行对比分析,验证该滑面模型的合理性与优势性。研究结果表明:文中方法计算所得安全系数与其他方法相比更小,相对误差在5%以内。能够得到传统滑动面假设方法下难以搜索到的临界滑动面,并与数值模拟结果吻合度较好,稳定性分析结果更接近实际情况。由此说明,此方法用于均质边坡及非均质边坡稳定性分析是合理可行的,有望为工程实践提供更优的临界滑动面解决方案。

Characteristics of volatile organic compounds,NO_2,and effects on ozone formation at a site with high ozone level in Chengdu

Chengdu is a megacity in the southwest of China with high ozone(O_3) mixing ratio.Observation of volatile organic compounds(VOCs), NO_2 and O_3 with high temporal resolution was conducted in Chengdu to investigate the chemical processes and causes of high O_3 levels. The hourly mixing ratios of VOCs, NO2, and O_3 were monitored by an online system from 28 August to 7 October, 2016. According to meteorological conditions,Chengdu, with relative warm weather and low wind speed, is favorable to O_3 formation.Part of the O_3 in Chengdu may be transported from the downtown area. In O_3 episodes,the average mixing ratios of NO_2 and O_3 were 20.20 ppbv and 47.95 ppbv, respectively. In non-O_3 episodes, the average mixing ratios of NO_2 and O_3 were 16.38 ppbv and 35.15 ppbv,respectively. The average mixing ratio of total VOCs(TVOCs) was 40.29 ppbv in non-O_3 episodes, which was lower than that in O_3 episodes(53.19 ppbv). Alkenes comprised51.7% of the total O_3 formation potential(OFP) in Chengdu, followed by aromatics which accounted for 24.2%. Ethylene, trans-pentene, propene, and BTEX(benzene, ethylbenzene,toluene, m/p-xylene, o-xylene) were also major contributors to the OFP in Chengdu. In O_3 episodes, intensive secondary formations were observed during the campaign. Oxygenated VOCs(OVOCs), such as acetone, Methylethylketone(MEK), and Methylvinylketone(MVK)were abundant. Isoprene rapidly converted to MVK and Methacrolein(MACR) during O_3 episodes. Acetone was mainly the oxidant of C3-C5 hydrocarbons.

Tumor-resident microbiota in breast cancer promotes cancer initiation and malignant progression.However,targeting microbiota to improve the effects of breast cancer therapy has not been investigated in detail.Here,we evaluated the microbiota composition of breast tumors and found that enterotoxigenic Bacteroides fragilis(ETBF)was highly enriched in the tumors of patients who did not respond to taxane-based neoadjuvant chemotherapy.ETBF,albeit at low biomass,secreted the toxic protein BFT-1 to promote breast cancer cell stemness and chemoresistance.Mechanistic studies showed that BFT-1 directly bound to NOD1 and stabilized NOD1 protein.NOD1 was highly expressed on ALDH+breast cancer stem cells(BCSCs)and cooperated with GAK to phosphorylate NUMB and promote its lysosomal degradation,thereby activating the NOTCH1-HEY1 signaling pathway to increase BCSCs.NOD1 inhibition and ETBF clearance increase the chemosensitivity of breast cancer by impairing BCSCs.

文旅融合视域下,古树名木资源保护与利用有其独特价值。龙胜各族自治县现存古树4464株,隶属于43科86属125种,主要树种有枫香树、木荷、杉木、华南五针松、马尾松、樟、水青冈等12种,累计占古树总量的80.94%;关键树种为银杉。根据树龄划分,该县有特级古树3株,一级古树114株,二级古树381株,三级古树3465株,分别占比0.07%、2.55%、8.53%、77.62%,古树名木树龄及相应的数量构成总体呈金字塔形。从生长场所看,古树主要分布在远郊野外,计2519株;其次是乡村街道,计1900株;最少是城区,仅45株。从权属情况看,古树权属以集体和国有为主,个人所有为辅,绝大部分古树名木权属清晰。从生长质量看,古树正常株有4272株,衰弱株152株,濒危株9株,死亡株31株,绝大多数古树名木生长环境良好。在对古树名木资源保护的基础之上,充分发挥古树名木的康养旅游、科普旅游、民俗旅游价值,是促进文旅融合、赋能旅游高质量发展的可行性路径。

在以人为中心的发展理念下,社区参与从“幕后文本”走向“台前行动”,破解知其义而收效甚微的迷思需要更具操作性的研究工具予以应对,源于西方理论、始于中国实践的影像发声法提供了可参考的增强路径。文章从影像发声法的概念、起源、行动过程等介绍了基于该方法的社区参与式设计之可行性,强调问题关切、批判对话和赋权变革的三阶目标,指出其具有丰富研究数据、参与门槛低和赋权于民等众多优势,但同时也需要注意伦理道德和强化影响深度;之后以加拿大社区健康环境项目为例,阐述了影像发声法的介入过程并展开阶段性反思;最后,从因地制宜、视听互构、程序正义、实施保障和空间治理等层面讨论了影像发声法对我国参与式社区规划的实践启示,旨在促进人人能够享有社区权利。
