Rhizosphere microbes influence wogonoside accumulation in perennial Scutellaria baicalensis

Dried roots of Scutellaria baicalensis Georgi are an acclaimed traditional Chinese medicine,and wogonoside content is a key indicator used to evaluate S.baicalensis quality.Rather than rising linearly with cultivation years,S.baicalensis quality initially increases and then declines.However,little is known regarding the dynamic variations in S.baicalensis rhizosphere microorganisms under long-term cultivation and underlying mechanisms of their effects on wogonoside formation and accumulation.The aim of this study was to investigate the roles of soil nutrients and rhizosphere microbes on S.baicalensis quality across different cultivation years(1-4 years).The wogonoside content(25.14 mg g^(-1))was highest in the biennial S.baicalensis and then decreased following long-term cultivation.Most soil nutrients(available nitrogen,available phosphorus,available iron,available manganese,and available zinc)were reduced significantly as cultivation years increased.Time also affected rhizosphere bacterial community structure significantly,driving it toward deterministic process(i.e.,β-nearest taxon index<-2).Available manganese and exchangeable calcium indirectly affected wogonoside formation and accumulation.Wogonoside inhibition was driven by the reduction in rhizosphere bacterial diversity,which significantly increased the relative abundance of beta-glucosidase and decreased the relative abundance of phenylalanine ammonialyase.The rhizosphere microenvironment was altered under long-term cultivation,thereby shaping rhizosphere bacterial community,reducing the community diversity,and ultimately inhibiting wogonoside formation and accumulation.Our findings may aid in understanding of the mechanisms and reasons for the reduction in S.baicalensis quality under long-term cultivation from the perspective of soil nutrients and microorganisms,which may theoretically support the future artificial cultivation and management of Chinese medicinal plants.

Task planning and collaboration of multiple robots have broad application prospects and value in the field of robotics.To improve the performance and working efficiency of our Spherical Underwater Robot(SUR),we propose a multi-robot control strategy that can realize the task planning and collaboration of multiple robots.To complete real-time information sharing of multiple robots,we first build an acoustic communication system with excellent communication performance under low noise ratio conditions.Then,the task planning and collaboration control strategy adjust the SURs so that they maintain their positions in the desired formation when the formation moves.Multiple SURs can move along desired trajectories in the expected formation.The control strategy of each SUR uses only its information and limited information of its neighboring SURs.Finally,based on theoretical analysis and experiments,we evaluate the validity and reliability of the proposed strategy.In comparison to the traditional leader–follower method,it is not necessary to designate a leader and its followers explicitly in our system;thus,important advantages,such as fault tolerance,are achieved.

Background:Aldo-keto oxidoreductase(AKR)inhibitors could reverse the resistance of several cancer cells to cis-platin,but their role in resistance remains unclear.Methods:We verified the difference of AKR1Cs expression by Western blot,RNA sequencing and qRT-PCR.The differences of AKR1Cs expression were analyzed and inferred.Use Assay of NADH and NAD^(+)content to verify the inference.The Docking experience was used to verify the affinity between MPA,MCFLA,MLS and AKR1C3.Results:Our RNA-seq results showed de novo NAD biosynthesis-related genes and NAD(P)H-dependent oxidoreductases were significantly upregulated in cis-platin-resistant HepG2 hepatic cancer cells(HepG2-RC cells)compared with HepG2 cells.At least 63 NAD(P)H-dependent reductase/oxidases were upregulated in HepG2-RC cells at least twofold.Knockdown of AKR1Cs could increase cis-platin sensitivity in HepG2-RC cells about two-fold.Interestingly,the AKR1C inhibitor meclofenamic acid could increase the cis-platin sensitivity of HepG2-RC cells about eight-fold,indicating that the knockdown of AKR1Cs only partially reversed the resistance.Meanwhile,the amount of total NAD and the ratio of NADH/NAD^(+)were increased in HepG2-RC cells compared with HepG2 cells.The ratio of NADH/NAD^(+)in HepG2-RC cells was almost seven-fold higher than in HepG2 or HL-7702 cells.Increased NADH expression could be explained as a directly operating antioxidant to scavenge cis-platin-induced radicals.Conclusion:We report here that NADH,which is produced by NAD(P)Hdependent oxidoreductases,plays a key role in the AKR-associated cis-platin resistance of HepG2 hepatic cancer cells.

以中国知网收录的80篇装配式建筑风险研究领域的文献为研究对象,基于文献计量和文本分析研究装配式建筑项目风险识别方法、风险分类及常见风险。研究结果表明,文献综述是最常用的风险识别方法;环境、资源、施工、管理、供应链为装配式建筑项目常见风险类别;自然灾害、恶劣气候及不可抗力,作业人员风险意识淡薄,装配式构件质量问题,设计方案可行性与集成性差,施工现场吊装作业环境及周边环境不利为装配式建筑项目常见风险因素。针对常见风险因素,提出了对策建议,以期为装配式建筑项目风险管理提供借鉴。

如何通过资源型集群的转型升级和区域创新网络的创新驱动，避免路径锁定实现可持续发展，是我国众多资源枯竭型城市正在面对的难题。本文首先使用数据包络分析法测度了资源型集群的经济发展效率和区域创新网络的创新效率，然后以空间地理因素为切入点，构建了资源型集群和区域创新网络的空间计量模型，探索由技术、人员、信息等因素产生的空间溢出效应对资源型集群和区域创新网络的作用机制。研究发现：资源型城市中的产业集群和区域创新网络存在显著的“虹吸效应”;区域创新网络对资源型集群的支撑作用更为明显，资源型集群对区域创新网络的贡献相对较少；区域创新网络中的技术、基础设施和人力资源具有显著空间溢出效应，资源型集群的空间溢出效应则不显著。

利用具有大带宽、低时延、高可用、可调度特性的5G网络,连接站区运输生产业务系统、现场信息采集设备和人员携带的智能终端等,构建铁路车站作业自动通知系统,替代传统的人工电话通知模式,实现作业通知的智能生成、自动触发、及时可信传达和交互过程全程自动管控,可以达到平稳生产运行、提高作业效率、保障作业质量和安全、减轻人员劳动强度等应用效果。从需求分析入手,梳理铁路车站作业人员沟通交互的4个方面内容,提炼出作业通知的3种分类和通用的业务流程,针对当前存在的问题提出建设基于5G通信的车站作业自动通知系统的技术目标,指出需要研究的关键技术,即泛在与融合的高可用网络通信技术、车站作业实时数据在线自动集成处理技术、车站生产业务事件智能检测技术和车站作业通知交互过程自动管控技术,并对系统架构与功能实现等方面进行必要阐述。

Electrochemical mass spectrometry(EC-MS)is a powerful tool to capture and analyze the intermediates and products during elec-trochemical reactions.This hyphenated technique combines electrochemistry with mass spectrometry using specific apparatuses,which helps researchers study mechanisms of redox reactions by in situ detecting chemical composition changes.Recently,various EC-MS methods have been applied in a series of electrochemical reactions to reveal the mechanisms,mainly in the areas of electro-chemical sensors,organic electrochemistry,and electrocatalysis.In this review,we intend to summarize the recent advances in re-al-time analysis of different types of electrochemical reactions by EC-MS and offer an outlook on the perspectives in these areas.

The latest view suggests the inactive core,surface pulverization,and poly sulfide shuttling effect of metal sulfides are responsible for their low capacity and poor cycling performance in sodium-ion batteries(SIBs).Whereas overcoming the above problems based on conventional nanoengineering is not efficient enough.In this work,erythrocyte-like CuS microspheres with an elastic buffering layer of ultrathin poly aniline(PANI) were synthesized through one-step selfassembly growth,followed by in situ polymerization of aniline.When CuS@PANI is used as anode electrode in SIBs,it delivers high capacity,ultrahigh rate capability(500 mAh gat 0.1 A g,and 214.5 mAh gat 40 A g),and superior cycling life of over 7500 cycles at 20 A g.A series of in/ex situ characterization techniques were applied to investigate the structural evolution and sodium-ion storage mechanism.The PANI swollen with electrolyte can stabilize solid electrolyte interface layer,benefit the ion transport/charge transfer at the PANI/electrolyte interface,and restrain the size growth of Cu particles in confined space.Moreover,finite element analyses and density functional simulations confirm that the PANI film effectively buffers the volume expansion,suppresses the surface pulverization,and traps the poly sulfide.

为了提高核酸提取装置的移液精度和稳定性,以基于空气置换原理、活塞驱动方式的移液模块作为研究对象,提出了一种基于正弦函数的S型加减速曲线算法,并将其与匀加减速、抛物线型加减速曲线算法进行了仿真对比和实验验证。结果表明:采用基于正弦函数的S型加减速曲线算法的核酸提取装置,在移液量为10μL、20μL、50μL、100μL和200μL的不同工作条件下,移液精度均取得了显著的提升,同时实验数据的CV值满足JJG 646-2006移液器检定规程。
