Efficacy of anti-CD20 chimeric Fab' fragment on proliferation of B lymphoma cells

The variable domain of heavy chain (VH) and light chain (VL) genes of anti-CD20 monoclonal antibody HL47 were cloned from anti-CD20 ScFv expression vector pCANBTEcd20 by PCR and ligated into vector pYZF to construct chimeric anti-CD20 Fab’ fragment expression vector pTZFcd20. Chimeric anti-CD20 Fab’ fragment was expressed in E. coli 16C9 and purified by protein G affinity chromatography. Competitive inhibition assay showed that anti-CD20 Fab’ fragment inhibited binding of HI47 to CD20 on the surface of Daudi cells. Results from MTT assay indicated that chimeric anti-CD20 Fab’fragment inhibited the proliferation of Daudi cells, IC50=69 ug/mL. Affinity of chimeric anti-CD20 Fab’ fragment was determined, Ka was about 8.9×108 (mol/L)-1.

Objective:Epithelial cancers often originate from progenitor cells,while the origin of hepatocellular carcinoma(HCC)is still ***,one of the deadliest cancers,is closely linked with liver injuries and chronic inflammation,which trigger massive infiltration of bone marrow-derived cells(BMDCs)during liver ***:To address the possible roles of BMDCs in HCC origination,we established a diethylnitrosamine(DEN)-induced HCC model in bone marrow transplanted *** and frozen tissue immunofluorescence were used to verify DENinduced HCC in the pathology of the *** cellular origin of DEN-induced HCC was further studied by single cell sequencing,single-cell nested PCR,and immunofluorescence-fluorescence in situ ***:Studies by using single cell sequencing and biochemical analysis revealed that HCC cells in these mice were coming from donor mice BMDCs,and not from recipient ***,the copy numbers of mouse orthologs of several HCC-related genes previously reported in human HCC were also altered in our mouse ***-induced HCCs exhibited a similar histological phenotype and genomic profile as human ***:These results suggested that BMDCs are an important origin of HCC,which provide important clues to HCC prevention,detection,and treatments.

High-resolution sea-level data and high-precision dating of corals in the northern South China Sea(SCS)during the Holocene provide a reference and historical background for current and future sea-level changes and a basis for scientific assessment of the evolutionary trend of coral reefs in the *** sporadic studies have been performed around Hainan Island in the northern SCS,the reconstructed sea level presents different values or is controversial because the indicative meaning of the sea-level indicators were neither quantified nor uniform ***,we determined the quantitative relationship between modern living coral and sea level by measuring the top surfaces of 27 live Porites corals from the inner reef flat along the east coast of Hainan Island and assessed the accuracy of results obtained using coral as sea-level ***,three in situ fossil Porites corals were analyzed based on elevation measurements,digital X-ray radiography,and U-Th *** survey results showed that the indicative meanings for the modern live Porites corals is(146.09±8.35)cm below the mean tide level(MTL).It suggested that their upward growth limit is constrained by the sea level,and the lowest low water is the highest level of survival for the modern live Porites *** on the newly defined indicative meanings,6 new sea-level index points(SLIPs)were obtained and 19 published SLIPs were *** SLIPs indicated a relative sea level fluctuation between(227.7±9.8)cm to(154.88±9.8)cm MTL between(5393±25)cal a BP and(3390±12)cal a BP,providing evidences of the Mid-Holocene sea-level highstand in the northern *** that,our analysis demonstrated that different sea-level histories may be produced based on different indicative meanings or *** dataset of 276 coral U-Th ages indicates that coral reef development in the northern SCS comprised the initial development,boom growth,decline,and flourishing development again.A comparison wit

中性粒细胞输注是一种临床辅助抗感染治疗方案,而用于输注的中性粒细胞的体外活性维持是影响该方案实施的有效性的关键点,目前也是相关研究的难点。因此,该文探索间充质干细胞(mesenchymal stem cell,MSC)培养上清与一些药物分子联合使用来延缓中性粒细胞的体外衰老和维持机体免疫功能,以期克服上述难题。通过分别采用不同培养条件(包括多种MSC培养上清、酸碱度调节培养基、影响中性粒细胞存活的多种药物分子及其组方等条件培养基)进行中性粒细胞的体外培养,并分别检测中性粒细胞的体外寿命及生物学功能。实验结果显示,相较于对照组(RPMI-1640培养基),MSC培养3天的培养上清可以更好地延长中性粒细胞的体外寿命,同时也可以维持中性粒细胞的ROS释放和吞噬等杀菌功能,两组之间具有显著性差异(P<0.01);通过进一步实验筛选和验证,与单纯使用MSC培养3天的培养上清相比,使用优化后的中性粒细胞条件培养基(MSC培养上清,pH为6.0,1μmol/L nicotine、10 ng/mL G-CSF、0.6 mmol/L NAC、10μmol/L DFP)显著延长了中性粒细胞的体外寿命(P<0.01)。总之,该研究发现间充质干细胞培养上清和多种药物分子组合可以更好地维持中性粒细胞的体外活性,这将为中性粒细胞的体外保存和输注研究提供较好的实验基础和理论依据。

Human embryonic stem cells （hESCs） have been successfully differentiated into hematopoietic progenitor cells with colony formation capacity and further into various kinds of blood cells including erythrocytes, megakaryocytes, neutrophils, nature killer cells and T lymphocytes . Nevertheless, the differentiation efficiency is extremely low. The differentiation usually involves complex steps such as embryonic body formation or co-culture with stromal cells, causing inconsistencies and difficulties in dissection of its molecular mechanisms [8,9]. Therefore, it is essential to establish a well-defined hematopoietic differentiation model that is independent of serum and stromal cells and much easier to manipulate genes for functional screening, thus the underlying molecular mechanism of hematopoietic differen- tiation of hESCs could be dissected more easily.

激活棕色脂肪组织(BAT)可以抑制实体肿瘤生长,然而其对正常或异常造血细胞的影响仍然不清楚。因此,该研究采用β3-肾上腺素受体激动剂米拉贝隆分别激活野生型和白血病小鼠BAT,观察和分析其对正常造血细胞以及白血病细胞的影响。通过流式分析了不同时间点米拉贝隆治疗组和对照组小鼠骨髓、脾脏、肝脏和外周血中白血病细胞的比例,以及骨髓中造血干/祖及成熟细胞的比例、数目、细胞周期、凋亡和集落形成能力;验证了脂肪组织的激活程度;通过手术移除BAT来检测米拉贝隆激活BAT在白血病中的作用。研究结果显示,米拉贝隆治疗组显著激活了BAT,减少了急性白血病小鼠的骨髓、脾脏、肝脏和外周血中白血病细胞的比例,延长了小鼠生存期。米拉贝隆对正常C57小鼠的成熟免疫细胞和骨髓造血干/祖细胞的比例和数量没有明显影响,但在白血病小鼠中治疗组骨髓中各类成熟免疫细胞的比例和绝对数均显著高于对照组,造血干/祖细胞的比例和绝对数均显著增加,细胞凋亡减少,集落形成能力增强。手术切除BAT阻断了米拉贝隆介导的抗白血病作用。研究结果表明,通过米拉贝隆预给药可以激活棕色脂肪组织,抑制白血病细胞的增殖,促进白血病小鼠正常造血干/祖细胞的增殖分化和存活,延长小鼠生存期,为今后白血病的临床治疗提供了一种新的策略。
