Element Segregation and Solidification Behavior of a Nb, Ti, Al Co-Strengthened Superalloy ЭК151

The as-cast microstructure, element segregation and solidification behavior of a multi-alloyed superalloy ЭК151 have been investigated. The results show that the severe element segregation leads to the complicated precipitations at the inter-dendritic region, including η-Ni3(Ti, Nb), eutectic(γ + γ’) and Laves, which shows the characteristics of both Ti, Al-strengthened and Nb-strengthened alloys. Differential thermal analysis, heating and quenching tests reveal the solidification sequence as follows: Liquids →γ matrix →(Nb, Ti)C →η-Ni 3(Ti, Nb) →eutectic( γ+γ’) → Laves. The melting points are between 1250 and 1260 °C for(Nb, Ti)C, between 1200 and 1210 °C for η phase, between 1180 and 1190 °C for eutectic(γ+γ’) and Laves. γ’ initially precipitates from matrix at 1150 °C and achieves the maximum precipitation at 1130 °C. According to the microstructure evolution captured during solidification and composition analysis by an energy dispersive spectrometer and electron probe microanalyzer,(Nb, Ti)/Al ratio is put forward to explain the formation of η-Ni3(Ti, Nb) and eutectic( γ+γ’). The solidification of γ matrix increased the Nb, Ti concentration in the residual liquids, so the high(Nb, Ti)/Al ratio would result in the formation of η-Ni3(Ti, Nb); the precipitation of the phase consumed Nb and Ti and decreased the(Nb, Ti)/Al ratio in the liquid, which led to the precipitation of eutectic(γ + γ’). Laves formed by the sides of η-Ni3(Ti, Nb) and in front of the eutectic( γ + γ’) after Al, Ti were further depleted by the two phases and Cr, Co, Mo were rejected to liquids.

The effect of phosphorus on the precipitations of γ",γ’ and δ phases and associated tensile properties in IN718C alloy are investigated in this *** is revealed that P atoms are dissolved in the grain interior to a relatively high degree and hence influence the precipitation behaviors in the grain interior and improve the tensile strength of IN718C alloy.γ" and γ’ phases did not precipitate in the alloy without P addition during air cooling,while γ" and γ’ phases precipitated in the grain interior during air cooling in the alloys with P addition,and the amounts of γ" and γ’ phases increased with increasing P ***,the Vickers micro-hardness in the as-cast state increased gradually with increasing P *** double-aging state,the sizes of γ" and γ’ phases in the alloys with P addition were larger than that in the alloy without P addition,while the sizes were invariable when the P content(wt%)was higher than ***,the micro-hardness and tensile strength of IN718C alloy treated by double aging increased first and then kept invariable with increasing P *** precipitations of δ phases both in the grain interior and on grain boundaries were inhibited by P *** inhibitory effect of P on δ phase enhanced gradually with increasing content of P,but the plasticity increased first and then *** is more,the crack tended to propagate into the matrix around the particles(Laves phases and NbC carbides)in the alloys without P addition at the beginning of the tensile fracture,while it tended to propagate along the interfaces between the matrix and those particles in the alloys with P addition,which resulted from the synthetical effect of P on γ" γ’ and δ phases.

Objective:This paper is to explore a method of transferring human SDF-1 and its mutant SDF-154 intrakine gene into COS-7 cells for determining their expression and subcelluar localization of the fusion *** could offer feasibility for inhibiting the metastasis of malignant tumors by phonotypic knockout for blocking functional expression of receptor on the ***:Amplify the target gene with PCR from the constructed plasimid SDF-WT-Gly×4-DecPET-30a(+) with a C-terminal retention signal fragment KDEL. After the pcDNA3.1 SDF-1KDEL,pcDNA3.1 SDF-154KDEL,pEGFPSDF-1KDEL and pEGFPSDF-154KDEL eukaryotic expression vectors were constructed and the DNA sequence was accurate,they were transferred into COS-7 cells with liposome . The exogenous expressions were observed, fusion protein SDF-1His and SDF-154His were confirmed by Western blot,and the SDF-1EGFP and SDF-154EGFP were determined by Laser Scanning Confocal Microscopy. Results:Four expression vectors were constructed successfully, the fusion protein SDF-1 KDELHis and SDF-1 54 KDELHis expressed in COS-7 *** localization analysis showed that SDF-1KDELEGFP and SDF-154KDELEGFP were located mainly in endoplasmic ***:Four expression vectors pcDNA3.1 SDF-1KDEL, pcDNA3.1SDF-154KDEL, pEGFPSDF-1KDEL and pEGFPSDF-154KDEL were constructed successfully, which could express in eukaryotic cell and locate mainly in the endoplasmic reticulum .

Assemblies of covalent organic framework microcrystals:multiple-dimensional manipulation for enhanced applications

Covalent organic frameworks (COFs) are well known as the next generation of shape-persistent zeolite analogues, which have brought new impetus to the development of porous organic materials as well as two-dimensional polymers. Since the advent of COFs in 2005, many striking findings have definitely proven their great potentials expanding applications across energy,environment and healthcare fields. With thorough exploration over a decade, research interest has been drawn on the scientific challenges on chemistry, while making full play of COF values has remained far from satisfactory yet. Thus opening an avenue to modulating COF assemblies on the multi-scale is no longer just an option, but a necessity for matching the application requirements with enhanced performances. In this mini-review, we summarize the recent progress on design of nanoscale COFs with varying forms. Detailed description is concentrated on the synthetic strategies of COF assemblies such as spheres, fibers,tubes, coatings and films, thereby shedding light on the flexible manipulation over dimensions, compositions and ***, the advanced applications of nanoscale COFs have been discussed here with comparison of their bulky counterparts.

Prediction and stratification for the surgical adverse events after minimally invasive esophagectomy:A two-center retrospective study

BACKGROUND Minimally invasive esophagectomy(MIE)is a widely accepted treatment for esophageal cancer,yet it is associated with a significant risk of surgical adverse events(SAEs),which can compromise patient recovery and long-term *** preoperative identification of high-risk patients is critical for improving *** To establish and validate a risk prediction and stratification model for the risk of SAEs in patients with *** This retrospective study included 747 patients who underwent MIE at two centers from January 2019 to February *** were separated into a train set(n=549)and a validation set(n=198).After screening by least absolute shrinkage and selection operator regression,multivariate logistic regression analyzed clinical and intraoperative variables to identify independent risk factors for SAEs.A risk stratification model was constructed and validated to predict the probability of *** SAEs occurred in 10.2%of patients in train set and 13.6%in the validation *** with SAE had significantly higher complication rate and a longer hospital stay after *** key independent risk factors identified included chronic obstructive pulmonary disease,a history of alcohol consumption,low forced expiratory volume in the first second,and low albumin *** stratification model has excellent prediction accuracy,with an area under the curve of 0.889 for the training set and an area under the curve of 0.793 for the validation *** The developed risk stratification model effectively predicts the risk of SAEs in patients undergoing MIE,facilitating targeted preoperative interventions and improving perioperative management.

基底膜相关基因在类风湿关节炎不同中医证型中的转录组学分析及药物预测

背景:基底膜基因与类风湿关节炎的发生发展密切相关,但不同中医证型下基底膜相关基因在其发病机制中的作用尚不明确。目的:基底膜基因联合转录组学分析探讨类风湿关节炎5种不同中医证型的发病机制差异,并进行潜在治疗药物预测。方法:基于GEO数据库整理类风湿关节炎相关中医证型芯片数据以及基底膜相关基因,利用R-limma包筛选差异表达基因,Mfuzz包进行表达趋势分析。借助STRING数据库构建PPI网络,UPset筛选关键基因,通过R-clusterProfiler包对差异表达基因进行GSEA及富集分析,同时绘制ROC曲线评估基底膜相关核心靶点对5种证型分别的诊断价值。基于CIBERSORT核心算法计算每种证型的免疫浸润情况。最后,利用SymMap和COREMINE数据库预测可靶向基底膜相关核心基因治疗类风湿关节炎不同证型的潜在中药及小分子药物。结果与结论:①在5种类风湿关节炎中医证型中(风湿痹阻证、寒湿痹阻证、肝肾亏虚证、气血两虚证及血瘀阻络证)分别筛选出67,47,59,57,55个基底膜相关差异表达基因,其中关键靶点分别为5,7,5,3,5个。③各证型中富集最多的为细胞基质黏附、免疫细胞迁移及胶原代谢等生物过程以及细胞外基质受体相互作用、PI3K-Akt、局灶性粘连和Rap1信号通路等。④药物预测结果显示,土茯苓、川乌、绵马贯众及黄精是通过影响基底膜基因治疗5种类风湿关节炎中医证型最具潜力的中药。⑤结果证实,基底膜相关基因的异常表达可能是通过调控细胞附着、免疫细胞迁移及炎症反应等多个途径影响类风湿关节炎的发生和发展,在不同证型中有着差异表现,其中ITGA6可作为5种类风湿关节炎证型中共同的诊断标志物。清热解毒类中药可能是类风湿关节炎不同证型治疗中可以贯穿始终的潜在有效药物。
