Herb pair of Huangqi-Danggui exerts anti-tumor immunity to breast cancer by upregulating PIK3R1

Background:According to traditional Chinese medicine(TCM),drugs supplementing the vital energy,Qi,can eliminate tumors by restoring host immunity.The objective of this study is to investigate the underlying immune mechanisms of anti-tumor activity associated with Qi-supplementing herbs,specifically the paired use of Huangqi and Danggui.Methods:Analysis of compatibility regularity was conducted to screen the combination of Qi-supplementing TCMs.Using the MTT assay and a transplanted tumor mice model,the anti-tumor effects of combination TCMs were investigated in vitro and in vivo.High content analysis and flow cytometry were then used to evaluate cellular immunity,followed by network pharmacology and molecular docking to dissect the significant active compounds and potential mechanisms.Finally,the anti-tumor activity and the mechanism of the active ingredients were verified by molecular experiments.Results:There is an optimal combination of Huangqi and Danggui that,administered as an aqueous extract,can activate immunity to suppress tumor and is more effective than each drug on its own in vitro and in vivo.Based on network pharmacology analysis,PIK3R1 is the core target for the anti-tumor immunity activity of combined Huangqi and Danggui.Molecular docking analysis shows 6 components of the combined Danggui and Huangqi extract(quercetin,jaranol,isorhamnetin,kaempferol,calycosin,and suchilactone)that bind to PIK3R1.Jaranol is the most important component against breast cancer.The suchilactone/jaranol combination and,especially,the suchilactone/kaempferol combination are key for immunity enhancement and the anti-tumor effects of the extract.Conclusions:The combination of Huangqi and Danggui can activate immunity to suppress breast cancer and is more effective than the individual drugs alone.

Bioinspired Multifunctional Self-Sensing Actuated Gradient Hydrogel for Soft-Hard Robot Remote Interaction

The development of bioinspired gradient hydrogels with self-sensing actuated capabilities for remote interaction with soft-hard robots remains a challenging endeavor. Here, we propose a novel multifunctional self-sensing actuated gradient hydrogel that combines ultrafast actuation and high sensitivity for remote interaction with robotic hand. The gradient network structure, achieved through a wettability difference method involving the rapid precipitation of MoO_(2) nanosheets, introduces hydrophilic disparities between two sides within hydrogel. This distinctive approach bestows the hydrogel with ultrafast thermo-responsive actuation(21° s^(-1)) and enhanced photothermal efficiency(increase by 3.7 ℃ s^(-1) under 808 nm near-infrared). Moreover, the local cross-linking of sodium alginate with Ca^(2+) endows the hydrogel with programmable deformability and information display capabilities. Additionally, the hydrogel exhibits high sensitivity(gauge factor 3.94 within a wide strain range of 600%), fast response times(140 ms) and good cycling stability. Leveraging these exceptional properties, we incorporate the hydrogel into various soft actuators, including soft gripper, artificial iris, and bioinspired jellyfish, as well as wearable electronics capable of precise human motion and physiological signal detection. Furthermore, through the synergistic combination of remarkable actuation and sensitivity, we realize a self-sensing touch bioinspired tongue. Notably, by employing quantitative analysis of actuation-sensing, we realize remote interaction between soft-hard robot via the Internet of Things. The multifunctional self-sensing actuated gradient hydrogel presented in this study provides a new insight for advanced somatosensory materials, self-feedback intelligent soft robots and human–machine interactions.

Influence of meteorological factors on open biomass burning at a background site in Northeast China

Biomass burning(BB)is a very important emission source that significantly adversely impacts regional air quality.BB produces a large number of primary organic aerosol(POA)and black carbon(BC).Besides,BB also provides many precursors for secondary organic aerosol(SOA)generation.In this work,the ratio of levoglucosan(LG)to organic carbon(OC)and the fire hotspots map was used to identify the open biomass burning(OBB)events,which occurred in two representative episodes,October 13 to November 30,2020,and April1 to April 30,2021.The ratio of organic aerosol(OA)to reconstructed PM_(2.5)concentration(PM_(2.5)^(*))increased with the increase of LG/OC.When LG/OC ratio is higher than 0.03,the highest OA/PM_(2.5)^(*)ratio can reach 80%,which means the contribution of OBB to OA is crucial.According to the ratio of LG to K^(+),LG to mannosan(MN)and the regional characteristics of Longfengshan,it can be determined that the crop residuals are the main fuel.The occurrence of OBB coincides with farmers’preferred choices,i.e.,burning biomass in“bright weather”.The“bright weather”refers to the meteorological conditions with high temperature,low humidity,and without rain.Meteorological factors indirectly affect regional biomass combustion pollution by influencing farmers’active choices.

Stimuli-responsive luminescent materials have attracted significant attention in the development of smart photoactive materials for both fundamental research and technological applications.In this work,a new copper iodide cluster(1)with aggregationinduced emission(AIE)characteristic,was obtained combining Cu_(4)I_(4) core with rhodamine B derivative ligand.1 has reversible and distinct multi-stimuli-responsive luminescence for external temperature,volatile organic compound,and mechanical force.Significantly,1 exhibited unusual large blue shift(84 nm)after being ground,which originated from the change of intermolecular interactions.Moreover,1 exhibits high oxygen quenching efficiency for 82.2% at 1 bar.Mechanistic studies showed that the multi-stimuli-responsive luminescence properties of 1 can be attributed to the regulation of cluster-centered luminescence process,metal-to-ligand charge transfer process,halide-to-metal charge transfer luminescence process and aggregationinduced barrier to oxygen process.This work not only reports an AIE copper iodide cluster,but also provides a new strategy to develop multi-stimuli-responsive luminescence materials.

Fe/S oxidation-coupled arsenic speciation transformation mediated by AMD enrichment culture under different pH conditions

Arsenic(As)speciation transformation in acid mine drainage(AMD)is comprehensively affected by biological and abiotic factors,such as microbially mediated Fe/S redox reactions and changes in environmental conditions(pH and oxidation-reduction potential).However,their combined impacts on arsenic speciation transformation remain poorly studied.Therefore,we explored arsenic transformation and immobilization during pyrite dissolution mediated by AMD enrichment culture under different acidic pH conditions.The results for incubation and mineralogical transformation of solid residues show that in the presence of AMD enrichment culture,pH 2.0,2.5,and 3.0 are more conducive to the formation of jarosites and ferric arsenate,which could immobilize high quantities of dissolved arsenic by adsorption and coprecipitation.The pH conditions significantly affect the initial adsorption of microbial cells to the minerals and the evolution of microbial community structure,further infuencing the biodissolution of pyrite and the release and oxidation process of Fe/S.The results of Fe/S/As speciation transformation of the solid residues show that the transformation of Fe,S,and As in solution is mainly regulated by pH and potential values,which imposed significantly different effects on the formation of secondary minerals and thus arsenic oxidation and immobilization.The above results indicated that arsenic transformation is closely related to the Fe/S oxidation associated with pyrite bio-oxidation,and this correlation is critically regulated by the pH conditions of the system.

ZNF554 Inhibits Endometrial Cancer Progression via Regulating RBM5 and Inactivating WNT/β-Catenin Signaling Pathway

Objective:Uterine corpus endometrial carcinoma(UCEC),a kind of gynecologic malignancy,poses a significant risk to women’s health.The precise mechanism underlying the development of UCEC remains elusive.Zinc finger protein 554(ZNF554),a member of the Krüppel-associated box domain zinc finger protein superfamily,was reported to be dysregulated in various illnesses,including malignant tumors.This study aimed to examine the involvement of ZNF554 in the development of UCEC.Methods:The expression of ZNF554 in UCEC tissues and cell lines were examined by qRT-PCR and Western blot assay.Cells with stably overexpressed or knocked-down ZNF554 were established through lentivirus infection.CCK-8,wound healing,and Transwell invasion assays were employed to assess cell proliferation,migration,and invasion.Propidium iodide(PI)staining combined with fluorescence-activated cell sorting(FACS)flow cytometer was utilized to detect cell cycle distribution.qRT-PCR and Western blotting were conducted to examine relative mRNA and protein levels.Chromatin immunoprecipitation assay and luciferase reporter assay were used to explore the regulatory role of ZNF554 in RNA binding motif 5(RBM5).Results:The expression of ZNF554 was found to be reduced in both UCEC samples and cell lines.Decreased expression of ZNF554 was associated with higher tumor stage,decreased overall survival,and reduced disease-free survival in UCEC.ZNF554 overexpression suppressed cell proliferation,migration,and invasion,while also inducing cell cycle arrest.In contrast,a decrease in ZNF554 expression resulted in the opposite effect.Mechanistically,ZNF554 transcriptionally regulated RBM5,leading to the deactivation of the Wingless(WNT)/β-catenin signaling pathway.Moreover,the findings from rescue studies demonstrated that the inhibition of RBM5 negated the impact of ZNF554 overexpression onβ-catenin and p-glycogen synthase kinase-3β(p-GSK-3β).Similarly,the deliberate activation of RBM5 reduced the increase inβ-catenin and p-GSK-3βcaused by the suppression of ZNF554.In v

To the Editor:Air pollution can lead to respiratory and cardiovascular diseases(CVDs),and its impact on human health is very significant.Air particles less than 0.1µm can pass through the blood brain barrier,reach the brain,and enter the central nervous system,causing physiological and pathological changes.A study shows that long-term exposure to fine particulate matter(PM_(2.5))will increase the risk of CVDs among Chinese residents.^([1])Another study found that a short time exposure to low concentrations of air pollutants may also lead to the onset of acute coronary syndrome within one hour.^([2])A meta-analysis based on seven databases showed that environmental air pollution was positively correlated with elevated blood pressure and hypertension.^([3])Mounting evidence indicates that air pollution can cause cardiovascular and cerebrovascular diseases and affect human health furtherly.

Track-Pattern-Based Characteristics of Extratropical Transitioning Tropical Cyclones in the Western North Pacific

Based on the Regional Specialized Meteorological Center(RSMC)Tokyo-Typhoon Center best-track data and the NCEP-NCAR reanalysis dataset,extratropical transitioning(ET)tropical cyclones(ETCs)over the western North Pacific(WNP)during 1951–2021 are classified into six clusters using the fuzzy c-means clustering method(FCM)according to their track patterns.The characteristics of the six hard-clustered ETCs with the highest membership coefficient are shown.Most tropical cyclones(TCs)that were assigned to clusters C2,C5,and C6 made landfall over eastern Asian countries,which severely threatened these regions.Among landfalling TCs,93.2%completed their ET after landfall,whereas 39.8%of ETCs completed their transition within one day.The frequency of ETCs over the WNP has decreased in the past four decades,wherein cluster C5 demonstrated a significant decrease on both interannual and interdecadal timescales with the expansion and intensification of the western Pacific subtropical high(WPSH).This large-scale circulation pattern is favorable for C2 and causes it to become the dominant track pattern,owning to it containing the largest number of intensifying ETCs among the six clusters,a number that has increased insignificantly over the past four decades.The surface roughness variation and three-dimensional background circulation led to C5 containing the maximum number of landfalling TCs and a minimum number of intensifying ETCs.Our results will facilitate a better understanding of the spatiotemporal distributions of ET events and associated environment background fields,which will benefit the effective monitoring of these events over the WNP.

Comprehensive analyses of the proteome and ubiquitome revealed mechanism of high temperature accelerating petal abscission in tree peony

Tree peony(Paeonia suffruticosa Andrews)is a well-known ornamental plant with high economic value,but the short fluorescence is a key obstacle to its ornamental value and industry development.High temperature accelerates flower senescence and abscission,but the associated mechanisms are poorly understood.In this study,the tandem mass tag(TMT)proteome and label-free quantitative ubiquitome from tree peony cut flowers treated with 20℃for 0 h(RT0),20℃or 28℃for 60 h(RT60 or HT60)were examined based on morphological observation,respectively.Totally,6970 proteins and 1545 lysine ubiquitinated(Kub)sites in 844 proteins were identified.Hydrophilic residues(such as glutamate and aspartate)neighboring the Kub sites were in preference,and 36.01%of the Kub sites were located on the protein surface.The differentially expressed proteins(DEPs)and Kub-DEPs in HT60 vs RT60 were mainly enriched in ribosomal protein,protein biosynthesis,secondary metabolites biosynthesis,flavonoid metabolism,carbohydrate catabolism,and auxin biosynthesis and signaling revealed by GO and KEGG analysis,accompanying the increase of endogenous abscisic acid(ABA)accumulation and decrease of endogenous indoleacetic acid(IAA)level.Additionally,the expression patterns of six enzymes(SAMS,ACO,YUC,CHS,ANS and PFK)putatively with Kub modifications were analyzed by proteome and real-time quantitative RT-PCR.The cell-free degradation assays showed PsSAMS and PsACO proteins could be degraded via the 26 S proteasome system in tree peony flowers.Finally,a working model was proposed for the acceleration of flower senescence and abscission by high temperature.In summary,all results contributed to understanding the mechanism of flower senescence induced by high temperature and prolonging fluorescence in tree peony.

Cobalt-nitrogen co-doped porous carbon sphere as highly efficient catalyst for liquid-phase cyclohexane oxidation with molecular oxygen and the active sites investigation

The selective oxidation of cyclohexane to cyclohexanone and cyclohexanol(KA oil)is a challenging issue in the chemical industry.At present the industrial conversion of cyclohexane to cyclohexanone and cyclohexanol is normally controlled at less than 5%selectivity.Thus,the development of highly active and stable catalysts for the aerobic oxidation of cyclohexane is necessary to overcome this low-efficiency process.Therefore,we have developed a cobalt-nitrogen co-doped porous sphere catalyst,Co-NC-x(x is the Zn/Co molar ratio,where x=0,0.5,1,2,and 4)by pyrolyzing resorcinol-formaldehyde resin microspheres.It achieved 88.28%cyclohexanone and cyclohexanol selectivity and a cyclohexane conversion of 8.88%under Co-NC-2.The results showed that the introduction of zinc effectively alleviated the aggregation of Co nanoparticles and optimized the structural properties of the material.In addition,Co0 and pyridinic-N are proposed to be the possible active species,and their proportion efficiently increased in the presence of Zn^(2+)species.In this study,we developed a novel strategy to design highly active catalysts for cyclohexane oxidation.
